1MAP kinase gene STK1 is required for hyphal, conidial, and appressorial development, toxin biosynthesis, pathogenicity, and hypertonic stress response in the plant pathogenic fungus Setosphaeria turcica显示文摘The mitogen-activated protein kinase(MAPK), a key signal transduction component in the MAPK cascade pathway, regulates a variety of physiological activities in eukaryotes. However, little is known of the role MAPK plays in phytopathogenic fungi. In this research, we cloned the MAPK gene STK1 from the northern corn leaf blight pathogen Setosphaeria turcica and found that the gene shared high homology with the high osmolality glycerol(HOG) MAPK gene HOG1 of Saccharomyces cerevisiae. In addition, gene knockout technology was employed to investigate the function of STK1. Gene knockout mutants(KOs) were found to have altered hyphae morphology and no conidiogenesis, though they did show similar radial growth rate compared to the wild-type strain(WT). Furthermore, microscope observations indicated that STK1 KOs did not form normal appressoria at 48 h post-inoculation on a hydrophobic surface. STK1 KOs had reduced virulence, a significantly altered Helminthosporium turcicum(HT)-toxin composition, and diminished pathogenicity on the leaves of susceptible inbred corn OH43. Mycelium morphology appeared to be significantly swollen and the radial growth rates of STK1 KOs declined in comparison with WT under high osmotic stress. These results suggested that STK1 affects the hyphae development, conidiogenesis, and pathogenicity of S. turcica by regulating appressorium development and HT-toxin biosynthesis. 2Growth inhibitory effect of wild-type Kras2 gene on a colonic adenocarcinoma cell line显示文摘AIM: To observe the growth inhibitory effect of wild-type Kras2 gene on a colonic adenocarcinoma cell line Caco-2. METHODS: Recombinant plasmid pCI-neo-Kras2 with wild type Kras2 open reading frame was constructed. The Caco-2 cells were transfected with either pCI-neo or pCI-neo-Kras2 using Lipofectamine 2000. The expression of wild type Kras 2 was examined by Northern blot analysis. And the expression of wild type Kras2 protein was examined by Western blot analysis. The effects of wild-type Kras2 on cell proliferation were analyzed by monotetrazolium (MTT) assay, meanwhile analyses of cell cycle and spontaneous apoptosis rate were carried out by flow cytometry (FCM). RESULTS: The plasmid of pCI-neo-Kras 2 was successfully established. The growth rate of cells transfected with pCI-neo-Kras2 was significantly lower than the control cells transfected with the empty pCI-neo vector (P < 0.05). Cell cycle analysis revealed arrest of the pCI-neo-Kras2 transfected cells in G0/G1 phases, decreased DNA synthesis and decreased fractions of cells in S phase. The proliferative index of cells transfected with pCI-neo-Kras2 was decreased compared with the control cells (49.78% vs 64.21%),while the apoptotic rate of Caco-2 cells with stable Kras 2 expression increased (0.30% vs 0.02%). 3Establishment of a diagnostic model of coronary heart disease in elderly patients with diabetes mellitus based on machine learning algorithms显示文摘OBJECTIVE To establish a prediction model of coronary heart disease(CHD)in elderly patients with diabetes mellitus(DM)based on machine learning(ML)algorithms.METHODS Based on the Medical Big Data Research Centre of Chinese PLA General Hospital in Beijing,China,we identified a cohort of elderly inpatients(≥60 years),including 10,533 patients with DM complicated with CHD and 12,634 patients with DM without CHD,from January 2008 to December 2017.We collected demographic characteristics and clinical data.After selecting the important features,we established five ML models,including extreme gradient boosting(XGBoost),random forest(RF),decision tree(DT),adaptive boosting(Adaboost)and logistic regression(LR).We compared the receiver operating characteristic curves,area under the curve(AUC)and other relevant parameters of different models and determined the optimal classification model.The model was then applied to 7447 elderly patients with DM admitted from January 2018 to December 2019 to further validate the performance of the model.RESULTS Fifteen features were selected and included in the ML model.The classification precision in the test set of the XGBoost,RF,DT,Adaboost and LR models was 0.778,0.789,0.753,0.750 and 0.689,respectively;and the AUCs of the subjects were 0.851,0.845,0.823,0.833 and 0.731,respectively.Applying the XGBoost model with optimal performance to a newly recruited dataset for validation,the diagnostic sensitivity,specificity,precision,and AUC were 0.792,0.808,0.748 and 0.880,respectively.CONCLUSIONS The XGBoost model established in the present study had certain predictive value for elderly patients with DM complicated with CHD.Hu XU Wen-Zhe CAO Yong-Yi BAI Jing DONG He-Bin CHE Po BAI Jian-Dong WANG Feng CAO Li FAN 2022Journal of Geriatric Cardiology2022,19,6:3
4Stk2,a Mitogen-Activated Protein Kinase from Setosphaeria turcica,Specifically Complements the Functions of the Fus3 and Kss1 of Saccharomyces cerevisiae in Filamentation,Invasive Growth,and Mating Behavior显示文摘Setosphaeria turcica,an essential phytopathogenic fungus,is the primary cause of serious yield losses in corn;however,its pathogenic mechanism is poorly understood.We cloned STK2,a newly discovered mitogen-activated protein kinase gene with a deduced amino acid sequence that is 96%identical to MAK2 from Phaeosphaeria nodorum,56%identical to KSS1 and 57%identical to FUS3 from Saccharomyces cerevisiae.To deduce Stk2 function in S.turcica and to identify the genetic relationship between STK2 and KSS1/FUS3 from S.cerevisiae,a restructured vector containing the open reading frame of STK2 was transformed into a fus3/kss1 double deletion mutant of S.cerevisiae.The results show that the STK2complementary strain clearly formed pseudohyphae and ascospores,and the strain grew on the surface of the medium after rinsing with sterile water and the characteristics of the complementary strain was the same as the wild-type strain.Moreover,STK2 complemented the function of KSS1 in filamentation and invasive growth,as well as the mating behavior of FUS3 in S.cerevisiae,however,its exact functions in S.turcica will be studied in the future research.GU Shou-qin YANG Yang LI Po ZHANG Chang-zhi FAN Yu ZHANG Xiao-yu TIAN Lan HAO Zhi-min CAO Zhi-yan GONG Xiao-dong FAN Yong-shan HAN Jian-min DONG Jin-gao 2013Journal of Integrative Agriculture2013,12,12:3
5Efficacy of Shenfu decoction on sepsis in rats with condition induced by cecal ligation and puncture显示文摘OBJECTIVE:To evaluate the efficacy of Shenfu decoction(SFD)prepared with a traditional Chinese formula,on sepsis in rats with the condition induced by cecal ligation and puncture(CLP),and to study the possible mechanism underlying its action.METHODS:Forty clean-grade male Sprague-Dawley rats were divided randomly into three groups:normal control group(NCG,n=10),model control group(MCG,n=15)and Shenfu decoction group(SFDG,n=15).Sham-operated rats in NCG were served as operation control,while rats in both MCG and SFDG were exposed to CLP,a procedure to develop experimental sepsis.Rats in SFDG were administered with SFD by gavage(3 mg/g of body weight,twice a day)2 h prior to CLP and directly after successful CLP,while rats in NCG and MCG were gavaged with equivalent volume of sterilized water.Rats in all groups were starved with free access to drink.After 24 h of administration,the mortality of rats in each group was assessed.The indicators of inflammatory response[the peritoneal inflammation by Simon's method Classification as well as serum concentrations of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)by enzyme linked immunosorbent assay(ELISA)]in survival rats were evaluated.The indicators of gut barrier[The intestinal mucosal injury index,serum concentrations of D-lactic acid and secretory IgA(sIgA)in intestinal mucosa by ELISA,as well as gut microbiota by16S rRNA gene sequencing]in survival rats were evaluated.RESULTS:The mortality(20%)of rats in SFDG was lower than that(33.3%)of the MCG(P<0.01).The mortality(20%)of rats in SFDG was lower than that(33.3%)of the MCG(χ2=6.533,P=0.011).Compared with the MCG,the peritoneal inflammation as well as serum concentrations of TNF-αand IL-6 decreased significantly in SFDG(all P<0.01).Compared with the MCG,the IMII,serum concentrations of D-lactic acid,sIgA in intestinal mucosa were alleviated by SFD treatment(all P<0.01).Increase in levels of Proteobacteria and reduction levels of Bacteroidetes induced by sepsis were observed,and these two disturbed gut microbiota phyla could be regulated after SFD treatment.Increase in levels of Proteobacteria and reduction levels of Bacteroidetes induced by sepsis were observed,and these two disturbed gut microbiota phyla could be regulated after SFD treatment.CONCLUSION:SFD may play a protective role in sepsis by alleviating sepsis-induced inflammatory response and gut barrier damage in rats.Liu Jin Liu Fusheng Liang Tengxiao Cao Po Li Jing Zhang Yin Liu Yang 2020Journal of Traditional Chinese Medicine2020,40,4:2
6Silk-based hydrogel incorporated with metal-organic framework nanozymes for enhanced osteochondral regeneration显示文摘Osteochondral defects(OCD)cannot be efficiently repaired due to the unique physical architecture and the pathological microenvironment including enhanced oxidative stress and inflammation.Conventional strategies,such as the control of implant microstructure or the introduction of growth factors,have limited functions failing to manage these complex environments.Here we developed a multifunctional silk-based hydrogel incorporated with metal-organic framework nanozymes(CuTA@SF)to provide a suitable microenvironment for enhanced OCD regeneration.The incorporation of CuTA nanozymes endowed the SF hydrogel with a uniform microstructure and elevated hydrophilicity.In vitro cultivation of mesenchymal stem cells(MSCs)and chondrocytes showed that CuTA@SF hydrogel accelerated cell proliferation and enhanced cell viability,as well as had antioxidant and antibacterial properties.Under the inflammatory environment with the stimulation of IL-1β,CuTA@SF hydrogel still possessed the potential to promote MSC osteogenesis and deposition of cartilage-specific extracellular matrix(ECM).The proteomics analysis further confirmed that CuTA@SF hydrogel promoted cell proliferation and ECM synthesis.In the full-thickness OCD model of rabbit,CuTA@SF hydrogel displayed successfully in situ OCD regeneration,as evidenced by micro-CT,histology(HE,S/O,and toluidine blue staining)and immunohistochemistry(Col I and aggrecan immunostaining).Therefore,CuTA@SF hydrogel is a promising biomaterial targeted at the regeneration of OCD.Zhicheng Cao Hongmei Wang Jialin Chen Yanan Zhang Qingyun Mo Po Zhang Mingyue Wang Haoyang Liu Xueyang Bao Yuzhi Sun Wei Zhang Qingqiang Yao 2023Bioactive Materials2023,,2:2

10A novel order reduction method for nonlinear dynamical system under external periodic excitations显示文摘The concept of approximate inertial manifold (AIM) is extended to develop a kind of nonlinear order reduction technique for non-autonomous nonlinear systems in second-order form in this paper.Using the modal transformation,a large nonlinear dynamical system is split into a 'master' subsystem,a 'slave' subsystem,and a 'negligible' subsystem.Accordingly,a novel order reduction method (Method I) is developed to construct a low order subsystem by neglecting the 'negligible' subsystem and slaving the 'slave' subsystem into the 'master' subsystem using the extended AIM.As a comparison,Method II accounting for the effects of both 'slave' subsystem and the 'negligible' subsystem is also applied to obtain the reduced order subsystem.Then,a typical 5-degree-of-freedom nonlinear dynamical system is given to compare the accuracy and efficiency of the traditional Galerkin truncation (ignoring the contributions of the slave and negligible subsystems),Method I and Method II.It is shown that Method I gives a considerable increase in accuracy for little computational cost in comparison with the standard Galerkin method,and produces almost the same accuracy as Method II.Finally,a 3-degree-of-freedom nonlinear dynamical system is analyzed by using the analytic method for showing predominance and convenience of Method I to obtain the analytically reduced order system.CAO DengQing,WANG JinLin & HUANG WenHu School of Astronautics,Harbin Institute of Technology,PO Box 137,Harbin 150001,China 2010Science China(Technological Sciences)2010,53,3:1
