| 1 | Whole-genome sequencing to detect mutations associated with resistance to insecticides and Bt proteins in Spodoptera frugiperda显示文摘The fall armyworm(FAW),Spodoptera frugiperda,is a major pest native to the Americas that has recently invaded the Old World.Point mutations in the target-site proteins acetylcholinesterase-1(ace-1),voltage-gated sodium channel(VGSC)and ryanodine receptor(RyR)have been identified in S.frugiperda as major resistance mechanisms to organophosphate,pyrethroid and diamide insecticides respectively.Mutations in the adenosine triphosphate-binding cassette transporter C2 gene(ABCC2)have also been identified to confer resistance to Cry IF protein.In this study,we applied a whole-genome sequencing(WGS)approach to identify point mutations in the target-site genes in 150 FAW individuals collected from China,Malawi,Uganda and Brazil.This approach revealed three amino acid substitutions(A201S,G227A and F290V)of S.frugiperda ace-1,which are known to be associated with organophosphate resistance.The Brazilian population had all three ace-1 point mutations and the 227A allele(mean frequency=0.54)was the most common.Populations from China,Malawi and Uganda harbored two of the three ace-1 point mutations(A201S and F290V)with the 290V allele(0.47-0.58)as the dominant allele.Point mutations in VGSC(T929I,L932F and L1014F)and RyR(I4790M and G4946E)were not detected in any of the 150 individuals.A novel 12-bp insertion mutation in exon 15 of the ABCC2 gene was identified in some of the Brazilian individuals but absent in the invasive populations.Our results not only demonstrate robustness of the WGS-based genomic approach for detection of resistance mutations,but also provide insights for improvement of resistance management tactics in S.frugiperda. | Fang Guan Jianpeng Zhang Huiwen Shen Xingliang Wang Amanda Padovan Tom K.Walsh Wee Tek Tay Karl H.J.Gordon William James Cecilia Czepak Michael Hilary Otim Donald Kachigamba Yidong Wu | 2021 | Insect Science2021，28，3： | 6 |

| 12 | NDFIP1 limits cellular TAZ accumulation via exosomal sorting to inhibit NSCLC proliferation显示文摘NDFIP1 has been previously reported as a tumor suppressor in multiple solid tumors,but the function of NDFIP1 in NSCLC and the underlying mechanism are still unknown.Besides,the WW domain containing proteins can be recognized by NDFIP1,resulted in the loading of the target proteins into exosomes.However,whether WW domain-containing transcription regulator 1(WWTR1,also known as TAZ)can be packaged into exosomes by NDFIP1 and if so,whether the release of this oncogenic protein via exosomes has an effect on tumor development has not been investigated to any extent.Here,we first found that NDFIP1 was low expressed in NSCLC samples and cell lines,which is associated with shorter OS.Then,we confirmed the interaction between TAZ and NDFIP1,and the existence of TAZ in exosomes,which requires NDFIP1.Critically,knockout of NDFIP1 led to TAZ accumulation with no change in its mRNA level and degradation rate.And the cellular TAZ level could be altered by exosome secretion.Furthermore,NDFIP1 inhibited proliferation in vitro and in vivo,and silencing TAZ eliminated the increase of proliferation caused by NDFIP1 knockout.Moreover,TAZ was negatively correlated with NDFIP1 in subcutaneous xenograft model and clinical samples,and the serum exosomal TAZ level was lower in NSCLC patients.In summary,our data uncover a new tumor suppressor,NDFIP1 in NSCLC,and a new exosome-related regulatory mechanism of TAZ. | Yirui Cheng Xin Lu Fan Li Zhuo Chen Yanshuang Zhang Qing Han Qingyu Zeng Tingyu Wu Ziming Li Shun Lu Cecilia Williams Weiliang Xia | 2023 | Protein & Cell2023，14，2： | 0 |
| 13 | Effects of 'Candidatus Liberibacter solanacearum'(haplotype B)on Bactericera cockerelli fitness and vitellogenesis显示文摘'Candidatus Liberibacter solanacearum'(Lso)are phloem-restricted and unculturable Gram-negative bacteria.Presently five haplotypes have been identified worldwide;but only haplotypes A and B are associated with the vector Bactericera cockerelli(Sulc.)in the Americas.Previous studies showed that Lso-infection reduces B.cockerelli reproductive output and that Lso haplotype B is more pathogenic than Lso haplotype A.To understand the interaction of Lso haplotype B and B.cockerelli,the fitness of Lso-free and Lso B-infected insects,and the expression of vitellogenin(5cKg7-like),a gene involved directly in the insect reproduction were analyzed.Statistical differences in the number of eggs oviposited,and the total number of progeny nymphs and adults were found among crosses of insects with or without Lso.Significant differences in sex proportions were found between Lso B-infected and Lso-free crosses:a higher proportion of Fi adult females were obtained from Lso B-infected mothers.A significant reduction of BcVgl-like was observed in crosses performed with Lso B-infected females compared to the Lso-free insects.In female cohorts of different age,a significant reduction of BcVgl-like expression was measured in 7-d-old Lso B-infected females(virgin and mated)compared with 7-d-old Lso-free females(virgin and mated),respectively.The reduction of BcVgl-like transcript was associated with a lower number of developing oocytes observed in female5s reproductive systems.Overall,this study represents the first step to understand the interaction of Lso B with B.cockerelli,highlighting the effect of Lso B infection on egg production,BcVgl-like expression,and oocyte development. | Angelica Albuquerque Tomilhero Frias Freddy Ibanez Azucena Mendoza William Mario de Carvalho Nunes Cecilia Tamborindeguy | 2020 | Insect Science2020，27，1： | 0 |

| 15 | Screening for colorectal neoplastic lesions following acute diverticulitis: Would a sigmoidoscopy suffice?显示文摘AIM: To investigate the yield of colorectal malignant or premalignant lesions during colonoscopy performed following an episode of acute diverticulitis.METHODS: A prospectively maintained electronic database of a public teaching hospital(Modbury Hospital, South Australia) was searched for international classification of diseases codes for acute diverticulitis from July 2007 to June 2011. The electronic database and each patient's medical records were reviewed for demographic data, clinical presentation, investigation results, colonoscopy details and surgical intervention.RESULTS: Two hundred and nineteen patients were diagnosed with acute diverticulitis with a median age of 60 years(range 24-93). One hundred and thirty-nine patients(63.5%) had follow-up screening colonoscopy, with the median interval between the episode of acute diverticulitis and colonoscopy being 8 wk(range: 1-66). Colonoscopy revealed polyps in 21 patients(15%) and no cases of colorectal cancer. Of the 21 patients with polyps, there were 14 patients(10%) with tubular/villous adenomas(13 in rectosigmoid region and 1 in descending colon). CONCLUSION: Detection of colorectal cancer in patients undergoing routine colonoscopy following acute diverticulitis is rare. However, colonic polyps in the left colon are noted. A flexible sigmoidoscopy is an adequate screening tool in such patients. | 17 | Hepatitis C virus burden:Treating and educating people without prejudice显示文摘Hepatitis C virus(HCV)infection has a worldwide incidence of 1.1%.In Italy,60%of people who inject drugs(PWIDs)and are receiving assistance for substance use disorder are infected with HCV.However,this subset of patients has extremely limited access to care due to multiple factors,including alcohol abuse,psychological comorbidities,and homeless status.AIM To describe the impact of our HCV-dedicated service for substance use disorder(SSUD)service on PWIDs receiving anti-HCV therapy.METHODS A dedicated,multidisciplinary team was set up at the SSUD of Trento in October 2020 to provide antiviral treatment to HCV RiboNucleic Acid-positive patients with an active or previous history of substance abuse.The treatment was followed by a health education program.Patients were treated with Direct-Acting Antivirals(DAAs).Data were retrospectively analyzed to assess the efficacy of our dedicated program in terms of therapy completion,HCV eradication,and compliance(primary endpoint).The rate of HCV reinfection and DAA-related toxicity were also assessed(secondary endpoints).RESULTS A total of 40 patients were enrolled in the study:28(70.0%)were treated with Sofosbuvir/Velpatasvir,while 12(30.0%)received Glecaprevir/Pibrentasvir.At the time of inclusion in the study,36 patients were receiving opioid agonist maintenance therapy,whilst another 4 had just finished the treatment.37.5%had a history of alcoholism and 42.5%received concomitant psychiatric treatment.All 40 patients(100.0%)completed the therapy cycle and 92.5%of patients adhered to the program.All patients tested negative for viral load at the end of the treatment.There were no significant drug interactions with common psychiatric treatments and no side effects were observed.The sustained virological response was achieved in 92.5%of cases with good tolerability,although two patients discontinued treatment temporarily.After HCV eradication,one patient died from an overdose,another from complications of cirrhosis,and one reinfection occurred.CONCLUSION Very high adherence to therapy and good tolerability was observed in our series of HCV patients treated at the SSUD,regardless of the substance abuse condition.Further validation in a larger population is required. | Elettra Merola Elisa Menotti Giovanna Branz Andrea Michielan Sonia Seligmann Annora Ratti Flora Agugiaro Luisa Moser Giovanni Vettori Anna Franceschini William Mantovani Riccardo Pertile Giovanni de Pretis Cecilia Pravadelli | 2022 | World Journal of Hepatology2022，14，7： | 0 |
