| 1 | Oxymatrine liposome attenuates hepatic fibrosis via targeting hepatic stellate cells显示文摘AIM:To investigate the potential mechanism of ArgGly-Asp(RGD) peptide-labeled liposome loading oxymatrine(OM) therapy in CCl 4-induced hepatic fibrosis in rats.METHODS:We constructed a rat model of CCl 4 induced hepatic fibrosis and treated the rats with different formulations of OM.To evaluate the antifibrotic effect of OM,we detected levels of alkaline phosphatase,hepatic histopathology(hematoxylin and eosin stain and Masson staining) and fibrosis-related gene expression of matrix metallopeptidase(MMP)-2,tissue inhibitor of metalloproteinase(TIMP)-1 as well as type Ⅰ procollagen via quantitative real-time polymerase chain reaction.To detect cell viability and apoptosis of hepatic stellate cells(HSCs),we performed 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-diphenytetrazoliumromide assay and flow cytometry.To reinforce the combination of oxymatrine with HSCs,we constructed fluorescein-isothiocyanate-conjugated Arg-Gly-Asp peptide-labeled liposomes loading OM,and its targeting of HSCs was examined by fluorescent microscopy.RESULTS:OM attenuated CCl 4-induced hepatic fibrosis,as defined by reducing serum alkaline phosphatase(344.47 ± 27.52 U/L vs 550.69 ± 43.78 U/L,P < 0.05),attenuating liver injury and improving collagen deposits(2.36% ± 0.09% vs 7.70% ± 0.60%,P < 0.05) and downregulating fibrosis-related gene expression,that is,MMP-2,TIMP-1 and type Ⅰ procollagen(P < 0.05).OM inhibited cell viability and induced apoptosis of HSCs in vitro.RGD promoted OM targeting of HSCs and enhanced the therapeutic effect of OM in terms of serum alkaline phosphatase(272.51 ± 19.55 U/L vs 344.47 ± 27.52 U/L,P < 0.05),liver injury,collagen deposits(0.26% ± 0.09% vs 2.36% ± 0.09%,P < 0.05) and downregulating fibrosis-related gene expression,that is,MMP-2,TIMP-1 and type Ⅰ procollagen(P < 0.05).Moreover,in vitro assay demonstrated that RGD enhanced the effect of OM on HSC viability and apoptosis.CONCLUSION:OM attenuated hepatic fibrosis by inhibiting viability and inducing apoptosis of HSCs.The RGD-labeled formulation enhanced the targeting efficiency for HSCs and the therapeutic effect. | Ning-Li Chai Qiang Fu Hui Shi Chang-Hao Cai Jun Wan Shi-Ping Xu Ben-Yan Wu | 2012 | World Journal of Gastroenterology2012,18,31: | 27 |
| 2 | Simulation research on a minimum root-mean-square error rotation-fitting algorithm for gravity matching navigation显示文摘Gravity/inertial combination navigation is a leading issue in realizing passive navigation onboard a submarine. A new rotation-fitting gravity matching algorithm, based on the Terrain Contour Matching (TERCOM) algorithm, is proposed in this paper. The algorithm is based on the principle of least mean-square-error criterion, and searches for a certain matched trajectory that runs parallel to a trace indicated by an inertial navigation system on a gravity base map. A rotation is then made clockwise or counterclockwise through a certain angle around the matched trajectory to look for an optimal matched trajectory within a certain angle span range, and through weighted fitting with another eight suboptimal matched trajectories, the endpoint of the fitted trajectory is considered the optimal matched position. In analysis of the algorithm reliability and matching error, the results from simulation indicate that the optimal position can be obtained effectively in real time, and the positioning accuracy improves by 35% and up to 1.05 nautical miles using the proposed algorithm compared with using the widely employed TERCOM and SITAN methods. | 4 | Review of low timing jitter mode-locked fiber lasers and applications in dual-comb absolute distance measurement显示文摘Passively mode-locked fiber lasers emit femtosecond pulse trains with excellent short-term stability. The quantum-limited timing jitter of a free running femtosecond erbium-doped fiber laser working at room temperature is considerably below one femtosecond at high Fourier frequency. The ultrashort pulse train with ultralow timing jitter enables absolute time-of-flight measurements based on a dual-comb implementation, which is typically composed of a pair of optical frequency combs generated by femtosecond lasers. Dead-zone-free absolute distance measurement with sub-micrometer precision and kHz update rate has been routinely achieved with a dual-comb configuration, which is promising for a number of precision manufacturing applications, from large step-structure measurements prevalent in microelectronic profilometry to three coordinate measurements in large-scale aerospace manufacturing and shipbuilding. In this paper, we first review the sub-femtosecond precision timing jitter characterization methods and approaches for ultralow timing jitter mode-locked fiber laser design. Then, we provide an overview of the state-of-the-art dual-comb absolute ranging technology in terms of working principles, experimental implementations, and measurement precisions. Finally, we discuss the impact of quantum-limited timing jitter on the dual-comb ranging precision at a high update rate. | 6 | Relations Between Photosynthetic Parameters and Seed Yields of Adzuki Bean Cultivars (Vigna angularis)显示文摘The study comparatively examined the leaf photosynthetic capacities of different adzuki bean cultivars, high-yield 2000-75 and Jihong 9218, and low-yield Hongbao 1 and Wanxuan 1 from flowering to ripening. It showed that after flowering, the leaves of the cultivars gradually aged, the leaf chlorophyll (Chl.), soluble protein (SP) contents, net photosynthetic rates (P n ), transpiration rates (T r ) and stomatal conductance (G s ) of the cultivars tended to decline, but the leaf intercellular CO 2 concentration (C i ) of the cultivars tended to rise. The leaf photosynthetic capacities of the cultivars decreased gradually from the lower to the upper nodes. The dry seed yields of the cultivars were positively correlated with their leaf Chl., SP, P n, and T r and G s , and negatively associated with their leaf C i . At the late growth stages, the high-yield cultivars maintained higher leaf Chl. contents, SP contents, P n , T r , and G s than the low-yield cultivars, indicating that leaf photosynthetic capacity was one of important yield-affecting factors of adzuki bean. | 7 | H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak显示文摘AIM To investigate the protective efficacy ofH2 strain attenuated live hepatitis A vaccines(H2-strain vaccines)in hepatitis A(HA)outbreaks.METHODS With the permission of theirparents,5551 pre-school and grade 1-3 primaryschool children were inoculated with 1 dose(106.5TCID50)of H2-strain vaccines in anonrandomized,controlled trial conducted inFucheng County,Hebei Province in May 1997.Another 6485 children in the same grades andcompatible in gender and age were enrolled ascontrols.Epidemiological and serological surveywas conducted to evaluate the protectiveefficacy of the vaccines.ELISA was used todetect serum IgM anti-HAV.RESULTS HA outbreak started in early May1998,peaked in the middle of the same month,and lasted about 80 days.Overall 302 HA caseswere found,192(53.58%)were 5-9 years old.One vaccinee and 25 control cases were found tohave hepatitis A,which account for 0.28%(1/356)and 5.92%(25/422)of all vaccinees andcontrols in the 14 villages,respectively.The protective efficacy of vaccines was 95.27%(95% Cl:85.83%-104.72%).In subjects testedfor anti-HAV IgM from 13 villages,1(0.40%)overt and 11(4.06%)asymptomatic HAV caseswere found in 271 vaccinees,but 21(6.69%)ofovert and asymptomatic ones were found in 314controls.CONCLUSION H2 strain vaccines were excellent in preventing overt hepatitis A, but not so effective in preventing asymptomatic hepatitis A virus infection. | 8 | Impacts of different drying strategies on drying characteristics,the retention of bio-active ingredient and colour changes of dried Roselle显示文摘The drying kinetics of Roselle(Hibiscus sabdariffa L) of variety Terengganu(UMKL-1) and the quality attribution of Roselle were studied. The experiments were conducted using four different drying methods, including solar greenhouse drying(SD), solar greenhouse with intermittent heat pump drying(SIHP), hot air drying(HA) and heat pump drying(HP). Among the four drying methods, HP achieved the highest drying rate at a range from0.054 g H_2 O·(g DM)^(-1) ·min^(-1) to 0.212 g H_2 O·(g DM)^(-1)·min^(-1) while SD had the lowest drying rate, measured at 0.042 g H_2 O·(g DM)^(-1)·min^(-1).The analysis on colour kinetics revealed that there is no significant colour loss(p > 0.05) observed from HP's dried Roselle. | 10 | Real world effectiveness of PCSK-9 inhibitors combined with statins versus statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease in China(RWE-PCSK study)显示文摘BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for MACE.RESULTS In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). There was no significant difference between PCSK-9 inhibitor and statins-based treatment in reducing the risk of MACE(hazard ratio = 2.52, 95% CI: 0.49-12.97, P = 0.250).CONCLUSIONS In the real world, PCSK-9 inhibitors combined with statins could significantly reduce LDL-C levels among patients with very high risk of ASCVD in China. For this pur- pose, the C60 derivative C60(C(COOH)2)2, a thera- peutic agent for degeneration diseases of central nervous system, was synthesized, purified and characterized. Its interaction with the living cell and penetration of the cellular membrane were in situ studied using the real time imaging technique, and its potential cytotoxicity was also examined by flow cy- tometry. The results indicate that C60(C(COOH)2)2 can easily enter cells, and is mainly located in cytoplasm by fluorescein labeling. Furthermore, C60(C(COOH)2)2 can carry the molecule that cannot cross cellular membranes into cells, because fluo- rescein compound itself cannot enter the cell or ad- here to membrane. | 13 | Plasma αl-antitrypsin: A Neglected Predictor of Angiographic Severity in Patients with Stable Angina Pectoris显示文摘Background: As an acute phase protein, ct I-antitrypsin (AAT) has been extensively studied in acute coronary syndrome, but it is unclear whether a relationship exists between AAT and stable angina pectoris (SAP). The purpose of the present study was to investigate the association between AAT plasma levels and SAP. Methods: Overall, 103 SAP patients diagnosed by coronary angiography and clinical manifestations and 118 control subjects matched for age and gender were enrolled in this case-control study. Plasma levels of AAT, high-sensitivity C-reactive protein (hsCRP), lipid profiles and other clinical parameters were assayed for all participants. The severity of coronary lesions was evaluated based on the Gensini score (GS) assessed by coronary angiography. Results: Positively correlated with the GS (r = 0.564, P 〈 0.001), the plasma AAT level in the SAP group was significantly higher than that in the control group (142.08 ± 19.61 mg/dl vs. 125.50 ± 19.67 mg/dl, P 〈 0.001). The plasma AAT level was an independent predictor tbr both SAP (odds ratio [OR] = 1.037, 95% confidence interval [C/]: 1.020-1.054, P 〈 0.001) and a high GS (OR = 1.087, 95% CI: 1.051-1.124, P 〈 0.001) in a multivariate logistic regression model. In the receiver operating characteristic curve analysis, plasma AAT level was found to have a larger area under the curve (AUC) for predicting a high GS (AUC = 0.858, 95% CI: 0.788-0.929, P 〈 0.001 ) than that of hsCRP (AUC = 0.665, 95% CI: 0.557-0.773, P = 0.006; Z = 2.9363, P 〈 0.001), with an optimal cut-off value of 137.85 mg/dl (sensitivity: 94.3%, specificity: 68.2%). Methods Abnormal recognition of facial expressions of emotion was assayed according to Japanese and Caucasian facial expressions of emotion (JACFEE), Wisconsin card sorting test (WCST), positive and negative symptom scale, and Hamilton anxiety and depression scale, respectively, in 88 paranoid schizophrenia patients and 75 healthy volunteers. Results Patients scored higher on the Positive and Negative Symptom Scale and the Hamilton Anxiety and Depression Scales, displayed lower JACFEE recognition accuracies and poorer WCST performances. The JACFEE recognition accuracy of contempt and disgust was negatively correlated with the negative symptom scale score while the recognition accuracy of fear was positively with the positive symptom scale score and the recognition accuracy of surprise was negatively with the general psychopathology score in patients. Moreover, the WCST could predict the JACFEE recognition accuracy of contempt, disgust, and sadness in patients, and the perseverative errors negatively predicted the recognition accuracy of sadness in healthy volunteers. The JACFEE recognition accuracy of sadness could predict the WCST categories in paranoid schizophrenia patients.
