1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 9Co-seismic Coulomb stress changes on the northern Tanlu fault zone caused by the Tohoku-Oki M_(W)9.0 earthquake显示文摘Based on the spherical earth dislocation theory and a fault slip model of the Tohoku-Oki M_(W)9.0 earthquake,the co-seismic Coulomb failure stress changes(ΔCFS)on the northern Tanlu fault zone at depths of 0–40 km are calculated.By comparing two sets of results from the spherical earth dislocation theory and the semi-infinite space one,the effect of earth curvature on the calculation results is analyzed quantitatively.First,we systematically summarize previous researches related to the northern Tanlu fault zone,divide the fault zone as detailed as possible,give the geometric parameters of each segment,and establish a segmented structural model of the northern Tanlu fault zone.Second,we calculate the Coulomb stress changes on the northern Tanlu fault zone by using the spherical earth dislocation theory.The result shows the Coulomb stress changes are no more than 0.003 MPa,which proves the great earthquake did not significantly change the stress state of the fault zone.Finally,we quantitatively analyze the disparities between the results of semi-infinite space dislocation theory and the spherical earth one.The average disparity between them is about 7.7%on the northern Tanlu fault zone and is 16.8%on the Fangzheng graben,the maximum disparity on this graben reaches up to 25.5%.It indicates that the effect of earth curvature can not be ignored.So it’s necessary to use the spherical earth dislocation theory instead of the semi-infinite space one to study the Coulomb stress change in the far field.Fei Chen Tai Liu Yawen She Xing Huang Guangyu Fu 2020Earthquake Science2020,33,1:1

12S1PR1 serves as a viable drug target against pulmonary fibrosis by increasing the integrity of the endothelial barrier of the lung显示文摘Idiopathic pulmonary fibrosis(IPF)is a progressive lung disease with unclear etiology and limited treatment options.The median survival time for IPF patients is approximately 2–3 years and there is no effective intervention to treat IPF other than lung transplantation.As important components of lung tissue,endothelial cells(ECs)are associated with pulmonary diseases.However,the role of endothelial dysfunction in pulmonary fibrosis(PF)is incompletely understood.Sphingosine-1-phosphate receptor 1(S1PR1)is a G protein-coupled receptor highly expressed in lung ECs.Its expression is markedly reduced in patients with IPF.Herein,we generated an endothelial-conditional S1pr1 knockout mouse model which exhibited inflammation and fibrosis with or without bleomycin(BLM)challenge.Selective activation of S1PR1 with an S1PR1 agonist,IMMH002,exerted a potent therapeutic effect in mice with bleomycin-induced fibrosis by protecting the integrity of the endothelial barrier.These results suggest that S1PR1 might be a promising drug target for IPF therapy.Mengyao Hao Rong Fu Jun Tai Zhenhuan Tian Xia Yuan Yang Chen Mingjin Wang Huimin Jiang Ming Ji Fangfang Lai Nina Xue Liping Bai Yizhun Zhu Xiaoxi Lv Xiaoguang Chen Jing Jin 2023Acta Pharmaceutica Sinica B2023,13,3:1

14Detection of FOXO1 break-apart status by fluorescence in situ hybridization in atypical alveolar rhabdomyosarcoma显示文摘The morphologies of alveolar rhabdomyosarcoma(ARMS) are various. Some cases entirely lack an alveolar pattern and instead display a histological pattern that overlaps with embryonal rhabdomyosarcoma(ERMS). The method of pathological diagnosis of ARMS and ERMS has been updated in the 4th edition of the World Health Organization's guidelines for classification of skeletal muscle tumors. Under the new guidelines, there is still no molecular test to distinguish between these two subtypes of rhabdomyosarcoma(RMS). In the present study, we applied fluorescent in situ hybridization(FISH) and found that the Forkhead box O1(FOXO1) gene broke apart, amplified, and displayed an aneuploid signal that was related to the RMS pathological subtype.Aside from the fact that FOXO1 break-apart and its amplification were correlated with atypical ARMS, aneuploidies were usually found in atypical ERMS. 15Atomically dispersed Ni anchored on polymer-derived mesh-like Ndoped carbon nanofibers as an efficient CO_(2) electrocatalytic reduction catalyst显示文摘Efficient electroreduction of CO_(2) into CO and other chemicals turns greenhouse gases into fuels and value-added chemicals,holding great promise for a closed carbon cycle and the alleviation of climate changes.However,there are still challenges in the large-scale application of CO_(2) electroreduction due to the sluggish kinetics.Herein we develop a self-assembly strategy to synthesize a highly efficient CO_(2) reduction electrocatalyst with atomically dispersed Ni-N_(4) active centers anchored on polymerderived mesh-like N-doped carbon nanofibers(Ni-N_(4)/NC).The Ni-N_(4)/NC exhibits high selectivity for CO_(2) reduction reaction with CO Faradaic efficiency(CO FE)above 90% over a wide potential range from−0.6 to−1.0 V vs.RHE.The catalyst reaches a maximum CO FE up to 98.4% at−0.8 V with a TOF of 1.28×10^(5) h^(–1) and Tafel slope of 113 mV·dec^(–1).The catalyst also exhibits remarkable stability,with little change in current density and CO FE over a 10-hour durability test at–0.8 V vs.RHE.This method provides a new route for the synthesis of highly efficient CO_(2) reduction electrocatalyst.Tai Cao Rui Lin Shoujie Liu Weng-Chon(Max)Cheong Zhi Li Konglin Wu Youqi Zhu Xiaolu Wang Jian Zhang Qiheng Li Xiao Liang Ninghua Fu Chen Chen Dingsheng Wang Qing Peng Yadong Li 2022Nano Research2022,15,5:0
