| 2 | Formation history and protolith characteristics of granulite facies metamorphic rock in Central Cathaysia deduced from U-Pb and Lu-Hf isotopic studies of single zircon grains显示文摘The petrochemical as well as zircon U-Pb and Lu-Hf isotopic studies of granulite facies metamorphic rock from the Taoxi Group in eastern Nanling Range, Central Cathaysia indicate that its protolith is the sedimentary rock with low maturation index. The clastic materials are mostly from middle Neoproterozoic (~736 Ma) granitoid rocks with minor Neoarchaean and Paleoproterozoic rocks. The timing of this Neoproterozoic magmatism is in agreement with the second period of magmatism widespread surrounding the Yangtze Block. Hf isotopic data indicate that the Neoproterozoic granitoids resulted from the recycled Paleoproterozoic mantle-derived crustal materials. The sedimentary rock was deposited in Late Neoproterozoic Era, and carried into low crust in Early Paleozoic. The partial melting of the meta-sedimentary rock took place at about 480 Ma and subsequently granulite facies metamorphism occurred at ca. 443 Ma. The zircons forming during this time interval (Early Paleozoic) show large Hf isotope variations, and their -Hf(t) values increase from -13.2 to +2.36 with decreasing age, suggesting the injection of mantle-derived materials during partial melting and metamorphism processes in the Early Paleozoic. Calculation results show that this metamorphic rock, if evolved to Mesozoic, has similar isotopic composition to the nearby Mesozoic high Si peraluminous granites, implying that this kind of granulite facies metamorphic rock is probably the source material of some Mesozoic peraluminous granitoids in eastern Nanling Range. | YU Jinhai ZHOU Xinmin Y. | 5 | Association of Dietary Pattern during Pregnancy and Gestational Diabetes Mellitus: A Prospective Cohort Study in Northern China显示文摘Objective To examine the association of maternal dietary patterns during pregnancy with gestational diabetes mellitus(GDM) in northern China.Methods The dietary intakes of pregnant women were recorded twice by 24‐hour dietary recalls for three days prior to having been diagnosed with GDM, at 5‐15 and 24‐28 gestational weeks, respectively.GDM was diagnosed, and serum glycosylated hemoglobin(HbA1c) was measured at 24‐28 weeks.Dietary patterns were assessed by factor analysis. The association of the dietary pattern with GDM and HbA1c was examined by multiple logistic models.Results Of 753 participants, 64(8.5%) were diagnosed with GDM. Four dietary patterns were identified: Western pattern(dairy, baked/fried food and white meat), traditional pattern(light‐colored vegetables, fine grain, red meat and tubers), mixed pattern(edible fungi, shrimp/shellfish and red meat)and prudent pattern(dark‐colored vegetables and deep‐sea fish). Compared with the prudent pattern,both the Western pattern and the traditional pattern were associated with an increased risk of GDM(a OR = 4.40, 95% CI: 1.58‐12.22; a OR = 4.88, 95% CI: 1.79‐13.32) and a high level of HbA1c(a OR = 12.37,95% CI: 1.47‐103.91; a OR = 26.23, 95% CI: 2.54‐270.74). | 9 | Probiotic bacteria change Escherichia coli-induced gene expression in cultured colonocytes:Implications in intestinal pathophysiology显示文摘AIM: To investigate the change in eukaryotic gene expression profile in Caco-2 cells after infection with strains of Escherichia coli and commensal probiotic bacteria. METHODS: A 19 200 gene/expressed sequence tag gene chip was used to examine expression of genes after infection of Caco-2 cells with strains of normal flora E. coli,Lactobacillus plantarum,and a combination of the two. RESULTS: The cDNA microarray revealed up-regulation of 155 and down-regulation of 177 genes by E. coli . L. plantarum up-regulated 45 and down-regulated 36 genes. During mixed infection,27 genes were up-regulated and 59 were down-regulated,with nullification of stimulatory/inhibitory effects on most of the genes. Expression of several new genes was noted in this group. CONCLUSION: The commensal bacterial strains used in this study induced the expression of a large number of genes in colonocyte-like cultured cells and changed the expression of several genes involved in important cellular processes such as regulation of transcription,protein biosynthesis,metabolism,cell adhesion,ubiquitination,and apoptosis. | 12 | Effects of serum containing natural cerebrolysin on glucose-regulated protein 78 and CCAAT enhancer-binding protein homologous protein expression in neuronal PC12 cells following tunicamycin-induced endoplasmic reticulum stress显示文摘BACKGROUND:Glucose-regulated protein 78(GRP78),a marker of endoplasmic reticulum stress,can prolong cell survival.Alternatively,CCAAT enhancer-binding protein homologous protein(CHOP),a transcription factor specific for endoplasmic reticulum stress,can cause cell cycle arrest and cell apoptosis. OBJECTIVE:To study the protective effects of serum containing natural cerebrolysin on endoplasmic reticulum stress in tunicamycin-induced neuronal PC12 cells,and analyze the influence on GRP78 and CHOP expressions. DESIGN,TIME AND SETTING:A parallel controlled study was performed at the Institute of Integrated Western and Traditional Chinese Medicine,Shenzhen Hospital,Southern Medical University,between March 2006 and August 2008. MATERIALS:Adult Sprague-Dawley rats were perfused with natural Cerebrolysin aqueous extract(0.185 g/kg/d) to produce serum containing natural Cerebrolysin.Physiological saline was used to produce blank serum.PC12 cell line was provided by Shanghai Institute of Cell Biology, Chinese Academy of Science.Tunicamycin was provided by Sigma(St.Louis,USA),and natural Cerebrolysin,containing ginseng,rhizoma gastrodiae,and gingko leaf(1:2:2),by Shengzhen Institute of Integrated Western and Traditional Chinese Medicine. METHODS:PC12 cells were treated with DMEM culture media containing 10%blank serum (normal control group),tunicamycin(1μg/mL;model group),and 5%,10%,and 15%serum containing natural cerebrolysin and tunicamycin(1μg/mL;low-,moderate-,and high-dose serum containing natural cerebrolysin groups),for 2 hours. MAIN OUTCOME MEASURES:PC12 cells were treated with tunicamycin for 48 hours after which apoptosis was measured using the TUNEL method to calculate apoptotic index.GRP78 expression was detected using immunocytochemistry.After 24 hours of treatment with tunicamycin,GRP78 and CHOP mRNA expressions were measured using RT-PCR. RESULTS:The apoptotic index and CHOP mRNA expression were in the model group and three cerebrolysin groups were significantly increased when compared to the normal control group(P<0.05).In contrast,GRP78 mRNA and protein expressions were significantly decreased(P<0.05).
