| 1 | Chinese Atherosclerosis Risk Evaluation(CARE II)study:a novel cross-sectional,multicentre study of the prevalence of high-risk atherosclerotic carotid plaque in Chinese patients with ischaemic cerebrovascular events-design and rationale显示文摘Background:Carotid atherosclerotic plaque is identified as one of the main sources of ischaemic stroke.However,the prevalence of carotid high-risk atherosclerotic plaque in Chinese patients with ischaemic cerebrovascular events has been inconsistently reported and needs to be investigated in a large population.Objectives:The primary objective of CARE II study was to determine the prevalence and characteristics of high-risk features of atherosclerotic plaques in the carotid arteries in Chinese patients with recent ischaemic stroke or transient ischaemia attack(TIA).The relationship between carotid plaque features and cerebral infarcts,the differences of carotid plaque patterns among different regions of China and the gender specific characteristics of carotid plaque will be also determined.Study design:The CARE II study will enrol 1000 patients with recent ischaemic stroke or TIA and carotid plaque from 13 hospitals and medical centres across China.In this cross-sectional,non-randomised,observational,multicentre study,all patients will undergo carotid artery MRI of bilateral carotid arteries and routine brain MRI with standardised protocols.The MRI will be interpreted at core reading centres to evaluate the characteristics of morphology and compositions of carotid plaque.Conclusions:This is a cross-sectional,multicentre study to investigate the prevalence and characteristics of high-risk atherosclerotic carotid plaque in Chinese patients with stroke and TIA by using high-resolution MRI of vessel wall.This trial is sufficiently powered to demonstrate the prevalence of carotid high-risk plaque and to explore regional differences in Chinese patients who suffered stroke. | Xihai Zhao Rui Li Daniel S Hippe Thomas S Hatsukami Chun Yuan CARE-II Investigators | 2017 | Stroke & Vascular Neurology2017,2,1: | 21 |

| 3 | Activation of human fibroblast-like synoviocytes by uric acid crystals in rheumatoid arthritis显示文摘Hyperuricemia-mediated uric acid crystal formation may cause joint inflammation and provoke the destruction of joints through the activation of inflammasome-mediated innate immune responses.However,the immunopathological effects and underlying intracellular regulatory mechanisms of uric acid crystal-mediated activation of fibroblast-like synoviocytes(FLS)in rheumatoid arthritis(RA)have not been elucidated.Therefore,we investigated the in vitro effects of monosodium urate crystals,alone or in combination with the inflammatory cytokines tumor-necrosis factor(TNF)-a or interleukin(IL)-1b,on the activation of human FLS from RA patients and normal control subjects and the underlying intracellular signaling mechanisms of treatment with these crystals.Monosodium urate crystals were able to significantly increase the release of the inflammatory cytokine IL-6,the chemokine CXCL8 and the matrix metalloproteinase(MMP)-1 from both normal and RA-FLS(all P,0.05).Moreover,the additive or synergistic effect on the release of IL-6,CXCL8 and MMP-1 from both normal and RA-FLS was observed following the combined treatment with monosodium urate crystals and TNF-a or IL-1b.Further experiments showed that the release of the measured inflammatory cytokine,chemokine and MMP-1 stimulated by monosodium urate crystals were differentially regulated by the intracellular activation of extracellular signal-regulated kinase and c-Jun N-terminal kinase pathways but not the p38 mitogen-activated protein kinase pathway.Our results therefore provide a new insight into the uric acid crystal-activated immunopathological mechanisms mediated by distinct intracellular signal transduction pathways leading to joint inflammation in RA. | Da P Chen Chun K Wong Lai S Tam Edmund K Li Christopher WK Lam | 2011 | Cellular & Molecular Immunology2011,8,6: | 5 |
| 4 | Molecular and Epidemiological Characterization of Infant Botulism in Beijing, China显示文摘Laboratory-based pathogen isolation, identification, and toxicity determination were performed on samples from a suspected case of infant botulism. Mice injected with cultures generated from the enema sample and ingested Powered infant formula(PIF) presented typical signs of botulism. Antitoxins to polyvalent botulinum neurotoxins(BoNTs) and monovalent BoNT type B antitoxin had protective effects. Clostridium botulinum isolated from the enema and residual PIF samples were positive for type B toxin. Pulsed-field gel electrophoresis(PFGE) revealed that the two strains of C. botulinum isolated from the two samples produced indistinguishable pulsotypes. | 5 | Down-scaled regional ocean modeling system (ROMS) for high-resolution coastal hydrodynamics in Korea显示文摘A down-scaled operational oceanographic system is developed for the coastal waters of Korea using a regional ocean modeling system(ROMS).The operational oceanographic modeling system consists of atmospheric and hydrodynamic models.The hydrodynamic model,ROMS,is coupled with wave,sediment transport,and water quality modules.The system forecasts the predicted results twice a day on a 72 h basis,including sea surface elevation,currents,temperature,salinity,storm surge height,and wave information for the coastal waters of Korea.The predicted results are exported to the web-GIS-based coastal information system for real-time dissemination to the public and validation with real-time monitoring data using visualization technologies.The ROMS is two-way coupled with a simulating waves nearshore model,SWAN,for the hydrodynamics and waves,nested with the meteorological model,WRF,for the atmospheric surface forcing,and externally nested with the eutrophication model,CE-QUAL-ICM,for the water quality.The operational model,ROMS,was calibrated with the tidal surface observed with a tide-gage and verified with current data observed by bottom-mounted ADCP or AWAC near the coastal waters of Korea.To validate the predicted results,we used real-time monitoring data derived from remote buoy system,HF-radar,and geostationary ocean color imager(GOCI).This down-scaled operational coastal forecasting system will be used as a part of the Korea operational oceanographic system(KOOS) with other operational oceanographic systems. | LIM Hak-Soo KIM Chang S PARK Kwang-Soon SHIM Jae Seol CHUN Insik | 2013 | Acta Oceanologica Sinica2013,32,9: | 4 |

| 7 | Differences in atheroma between Caucasian and Asian subjects with anterior stroke:A vessel wall MRI study显示文摘Background and purpose While extracranial carotid artery stenosis is more common among Caucasians and intracranial artery stenosis is more common among Asians,the differences in atherosclerotic plaque characteristics have not yet been extensively examined.We sought to investigate plaque location and characteristics within extracranial carotid and intracranial arteries in symptomatic Caucasians and Chinese using vessel wall MRI.Methods Subjects with recent anterior circulation ischaemic stroke were recruited and imaged at two sites in the USA and China using similar protocols.Both extracranial carotid and intracranial arteries were reviewed to determine plaque location and characteristics.Results The prevalence of extracranial carotid plaque in Caucasians and Chinese was 73.1%and 49.1%,respectively(p=0.055).Prevalence of intracranial plaque was 38.5%and 69.1%in Caucasians and Chinese,respectively(p=0.02).Furthermore,42% of Caucasians and 16%of Chinese had high-risk plaque(HRP)features(intraplaque haemorrhage,luminal surface disruption)in the extracranial carotid artery(p=0.03).The prevalence of HRP features in intracranial arteries was not significantly different between the two cohorts(4%vs 11%;p=0.42).Conclusions Differences in the location and characteristics of cerebrovascular atherosclerosis were identified by vessel wall MRI in US Caucasian and Chinese subjects with recent anterior circulation ischaemic stroke.Extracranial carotid plaques with HRP features were more common in Caucasians.Intracranial plaques were more common in Chinese subjects,but no significant difference between the two cohorts in intracranial HRP prevalence was found.Larger studies using vessel wall imaging to investigate racial differences in cerebrovascular disease may inform underlying mechanisms of HRP development and may ultimately help guide appropriate therapy. | Hiroko Watase Mi Shen Binbin Sui Peiyi Gao Dong Zhang Jie Sun Niranjan Balu Daniel S Hippe Gail P Jarvik Xihai Zhao Rui Li Shuo Chen Chun Yuan Thomas S Hatsukami | 2021 | Stroke & Vascular Neurology2021,6,1: | 4 |
| 8 | The heterodimeric structure of heterogeneous nuclear ribonucleoprotein C1/C2 dictates 1,25-dihydroxyvitamin D-directed transcriptional events in osteoblasts显示文摘Heterogeneous nuclear ribonucleoprotein(hnRNP) C plays a key role in RNA processing but also exerts a dominant negative effect on responses to 1,25-dihydroxyvitamin D(1,25(OH)2D) by functioning as a vitamin D response element-binding protein(VDRE-BP). hnRNPC acts a tetramer of hnRNPC1(huC1) and hnRNPC2(huC2), and organization of these subunits is critical to in vivo nucleic acid-binding. Overexpression of either huC1 or huC2 in human osteoblasts is sufficient to confer VDRE-BP suppression of 1,25(OH)2D-mediated transcription. However, huC1 or huC2 alone did not suppress 1,25(OH)2D-induced transcription in mouse osteoblastic cells. By contrast, overexpression of huC1 and huC2 in combination or transfection with a bone-specific polycistronic vector using a ‘‘self-cleaving'' 2A peptide to co-express huC1/C2 suppressed1,25D-mediated induction of osteoblast target gene expression. Structural diversity of hnRNPC between human/NWPs and mouse/rat/rabbit/dog was investigated by analysis of sequence variations within the hnRNP CLZ domain. The predicted loss of distal helical function in hnRNPC from lower species provides an explanation for the altered interaction between huC1/C2 and their mouse counterparts. | 10 | Ipsilateral breast tumor recurrence in early stage breast cancer patients treated with breast conserving surgery and adjuvant radiation therapy: Concordance of biomarkers and tumor location from primary tumor to in-breast tumor recurrence显示文摘BACKGROUND Patients with an in-breast tumor recurrence(IBTR)after breast-conserving therapy have a high risk of distant metastasis and disease-related mortality.Classifying clinical parameters that increase risk for recurrence after IBTR remains a challenge.AIM To describe primary and recurrent tumor characteristics in patients who experience an IBTR and understand the relationship between these characteristics and disease outcomes.METHODS Patients with stage 0-II breast cancer treated with lumpectomy and adjuvant radiation were identified from institutional databases of patients treated from 2003-2017 at our institution.Overall survival(OS),disease-free survival,and local recurrence-free survival(LRFS)were estimated using the Kaplan Meier method.We identified patients who experienced an isolated IBTR.Concordance of hormone receptor status and location of tumor from primary to recurrence was evaluated.The effect of clinical and treatment parameters on disease outcomes was also evaluated.RESULTS We identified 2164 patients who met the eligibility criteria.The median follow-up for all patients was 3.73[interquartile range(IQR)2.27-6.07]years.Five-year OS was 97.7%(95%CI:96.8%-98.6%)with 28 deaths;5-year LRFS was 98.0%(97.2-98.8)with 31 IBTRs.We identified 37 patients with isolated IBTR,19(51.4%)as ductal carcinoma in situ and 18(48.6%)as invasive disease,of whom 83.3%had an in situ component.Median time from initial diagnosis to IBTR was 1.97(IQR:1.03-3.5)years.Radiotherapy information was available for 30 of 37 patients.Median whole-breast dose was 40.5 Gy and 23 patients received a boost to the tumor bed.Twenty-five of thirty-two(78.1%)patients had concordant hormone receptor status,HER-2 receptor status,and estrogen receptor(ER)(P=0.006)and progesterone receptor(PR)(P=0.001)status from primary to IBTR were significantly associated.There were no observed changes in HER-2 status from primary to IBTR.The concordance between quadrant of primary to IBTR was 10/19[(62.2%),P=0.008].Tumor size greater than 1.5 cm(HR=0.44,95%CI:0.22-0.90,P=0.02)and use of endocrine therapy upfront(HR=0.36,95%CI:0.18-0.73,P=0.004)decreased the risk of IBTR.CONCLUSION Among patients with early stage breast cancer who had breast conserving surgery treated with adjuvant RT,ER/PR status and quadrant were highly concordant from primary to IBTR.Tumor size greater than 1.5 cm and use of adjuvant endocrine therapy were significantly associated with decreased risk of IBTR. | Juhi M Purswani Fauzia Shaikh S Peter Wu Jennifer Chun Kim Freya Schnabel Nelly Huppert Carmen A Perez Naamit K Gerber | 2020 | World Journal of Clinical Oncology2020,11,1: | 2 |
