2Mechanisms on inhibition of photosynthesis in Kandelia obovata due to extreme cold events under climate change显示文摘Introduction:Mangroves that grow at the latitudinal extremes of their distribution are susceptible to extreme cold events.Successive enhancement of low temperature stress(seLTS)is a typical characteristic of extreme cold events.Low temperature stress can inhibit mangrove photosynthesis,which often inhibits the growth and development of mangroves.However,the possible reasons for impairment to photosynthesis of mangroves due to extreme cold events remain unclear.Methods:Kandelia obovata seedlings in a growth chamber were exposed to 5℃/−2℃(day/night)for 36 h(−2℃ for 16 h)with 12 h light per day at 600μmol m^(−2) s^(−1) photosynthetic photon flux density(PPFD)(a low temperature stress,aLTS),then the plants were transferred to the control condition(15℃/10℃(day/night)and allowed to recover for 5 days(R1).The other seedlings were subjected to low temperature treatment with a day/night temperature of 5℃/−1℃ in a growth chamber for 24 h.Then these plants were transferred to 5℃/−2℃(day/night)under the same light and climate conditions for 36 h(two low temperature stresses,tLTS).Following the successive enhancement of low temperature treatment,these plants were returned to 15℃/10℃(day/night)for another 5-day recovery period(R2).Results:Results showed that aLTS treatment significantly reduced leaf net photosynthetic rate(P_(n))and stomatal conductance(G_(s)),while increased intercellular CO_(2) concentration(C_(i)).Photosynthetic activity of leaves quickly recovered after the plants were returned to control temperature for 5 days(R1).However,decreases in leaf Pn were more obvious under tLTS treatment than those under aLTS treatment.This reduced Pn might be attributed to stomatal and non-stomatal limitations.Moreover,non-stomatal limitation played a major role in reducing Pn during tLTS treatment,as proven by reduced ribulose 1,5-biphosphate carboxylase(RuBPCase)activity.This limitation also enhanced lipid peroxidation in chloroplasts,decreased sucrosemetabolizing enzymes and ratios of both chlorophyll a/chlorophyll b(Chl a/b)and chlorophyll/carotenoids(Chl/Car),and increased protease senescence.Damages of tLTS treatment to photosynthesis were insufficiently alleviated even after the plants were returned to control temperature for 5 days(R2).Conclusions:Successive enhancement of low temperature depressed successful recovery of leaf photosynthesis of K.obovata seedlings mainly by impairing pigment synthesis,antioxidant metabolism,and sucrose translocation,as well as accelerating senescence of endopeptidase.Furthermore,our results suggest that decreases in sucrose content in leaves might also account for increases in reactive oxygen species(ROS)in chloroplasts.Chunfang Zheng Jianwu Tang Jinong Chen Weicheng Liu Jianbiao Qiu Xin Peng Yong Ye 2016Ecological Processes2016,5,1:6

6Deletion of a Dna K protein gene causes seedling green-revertible albino by retarding chloroplast development in rice显示文摘Green-revertible albino mutants are important sources for studying chloroplast structure, chloroplast development, chlorophyll biosynthesis, and plant photosynthesis. In the present study, we characterized a greenrevertible albino mutant gra(k), which was obtained from the tissue-cultured rice Kitaake. The mutant gra(k) exhibited albino on its first three leaves. The leaf color started to turn green at the four-leaf stage. The chlorophyll contents were deeply reduced at the seedling stage, and the chloroplast development was delayed in gra(k). The greenrevertible albino(gra) phenotype of the mutant gra(k) was temperature dependent. The main agronomic traits,including plant height, tilling number per plant, seed setting rate, and thousand-grain weight, slightly decreased in gra(k) comparing to those in the wild-type Kitaake.Genetic analysis showed that the gra phenotype was controlled by a single recessive nucleic gene. By using 5,168 recessive F2individuals derived from the cross of gra(k) 9 Jodan, the locus of the gene Gra(k) was delimited in a DNA region of 200 kb between the makers B-31 and P11 on chromosome 5. Sequencing analysis indicated thatthe three functionally annotated genes, LOC_Os05g23700,LOC_Os05g23720, and LOC_Os05g23740, were all deleted in the 200 kb region in the mutant gra(k). Transgenic test revealed that the gra(k) plants over-expressing LOC_Os05g23740CDS were restored to normal green as the wild-type Kitaake. 9Establishment of a real-time PCR for quantifying transforming growth factor beta1 in blood of hepatocellular carcinoma patients显示文摘Background: The carcinogenesis of hepatocellular carcinoma (HCC) is a multi-factorial, multistep and complex process. Its prognosis is poor and early detection is of the utmost importance. Transforming growth factor β1 (TGF-β1) message RNA (mRNA) has been reported to be elevated in HCC patients using Northern blotting. However, little work has been done about the detection of TGF-β1 mRNA levels in peripheral blood of patients with HCC using the real-time polymerase chain reactions (PCR) method. Objective: To assess the prognostic value of quantitative levels of TGF-β1 mRNA in peripheral blood of patients with HCC, and to investigate the relationship between the expression of TGF-β1 mRNA in peripheral blood and many diagnostic and pathological factors. Methods: We developed an optimized Taqman real-time PCR to quantify TGF-β1 mRNA in peripheral blood of 53 patients with HCC and 44 healthy volunteers. In addition, blood was collected from patients with HCC for measuring levels of total bilirubin (TBil), prealbumin, albumin, alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma glutamyltranspeptidase (GGT), alpha-L-fucosidase (AFU), alpha fetoprotein (AFP), carcino-embryonic antigen (CEA), carbohydrate antigen 19-9 (CA19-9), viral load and platelet counts. Statistical analysis was performed using the SPSS software system (SPSS 10.0). Results: In real-time PCR, fluorescence was detectable in all blood specimens from patients with HCC and healthy volunteers. The levels of TGF-β1 mRNA expression in patients with HCC were significantly higher compared to that in healthy volunteers (P<0.000 1), suggesting an association of the activated TGF-β1 gene transcription with hepato- carcinogenesis. Patients with HCC were divided into 2 groups according to their TGF-β1 mRNA above (group A, n=28)or below (group B, n=25) the mean level. Statistical results demonstrated that TGF-β1 mRNA expression level was correlated with patients age, serum levels of CEA, CA19-9 and viral copy number (P<0.05). 12Detection of coalbed fractures with P-wave azimuthal AVO in 3-D seismic exploration显示文摘The detection of fractures is important for production and safety in coal fields. Subsurface fractures result in azimuthal anisotropy of the seismic wave, and the amplitude of reflection wave varies with offset and azimuth. In case of weak anisotropy, the reflection coefficients of P-wave are concisely denoted as the analytic function of fracture parameters. For the purpose of predicting the coal-bed fracture distribution through analyzing variation of the reflection amplitudes with offset and azimuth, 3-D seismic data with full-azimuth were acquired in a coal field in Huainan, Anhui Province. The careful analysis and process of seismic data showed that the reflection amplitude of the primary coalbed varied with azimuth in much consistent with the theoretical model.
