| 2 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas. | Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. | 3 | Optimization of parameters on photocatalytic degradation of chloramphenicol using TiO_2 as photocatalyist by response surface methodology显示文摘The experimental design methodology was applied for modeling and optimizing the operation parameters on photocatalytic degradation of chloramphenicol (CAP) using TiO 2 as photocatalyst in a photoreactor. Three experimental parameters (including pH, TiO 2 concentration and CAP initial concentration) were adopted to obtain the preliminary information. The multivariate experimental design was employed to establish a quadratic model as a functional relationship between the degradation rate of CAP and three experimental parameters. The interaction effects and optimal parameters were obtained by using Design Expert software. The optimal values of the operation parameters under the related constraint conditions were found at pH 6.4, TiO 2 concentration of 0.94 g/L and CAP initial concentration of 19.97 mg/L, respectively. The degradation rate of CAP approached 85.97% under optimal conditions. The regression analysis with R 2 value of 0.9519 had a good agreement between the experimental results and the predictive values. | 4 | Effects of ambient air pollution on allergic rhinitis among preschool children in Changsha, China显示文摘Prevalence of allergic rhinitis has rapidly increased among Chinese children,but the reasons are unclear.Recent findings have suggested that exposure to outdoor air pollutants may increase the risk of allergic rhinitis,but the results were inconsistent.This study further investigated the effect of outdoor air pollutants on allergic rhinitis among preschool children.A standardized questionnaire on health,home and environmental factors was conducted for 4988 children aged 1–8 in the city of Changsha,and the concentrations of PM10(particle diameter 10 m),sulfur dioxide(SO2)and nitrogen dioxide(NO2)during 2006–2011 were acquired from the official web of Changsha Environmental Protection Agency.Results showed that the prevalence of children’s doctor diagnosed rhinitis was 8.4%(95%confidence interval[CI]7.0%–10.0%).It was found that the prevalence of rhinitis was not associated with site-specific background concentrations of air pollutants,but significantly positively correlated with age-related accumulative personal exposure of PM10,SO2and NO2.We conclude that age-related accumulative personal exposure to ambient air pollution may play an important role in the development of rhinitis. | LU Chan DENG QiHong OU CuiYun LIU WeiWei SUNDELL Jan | 2013 | Chinese Science Bulletin2013,58,34: | 7 |
| 5 | Genome evolution trend of common carp (Cyprinus carpio L.) as revealed by the analysis of microsatellite loci in a gynogentic family显示文摘Genome evolution arises from two main ways of duplication and reduction. Fish specific genome duplication (FSGD) may have occurred before the radiation of the teleosts. Common carp (Cyprinus carpio L.) has been considered to be a tetraploid species, because of its chromosome numbers (2n=100) and its high DNA content. Using 69 microsatellite primer pairs, the variations were studied to better understand the genome evolution (genome duplication and diploidization) of common carp from a gynogenetic family. About 48% of primer pairs were estimated to amplify duplicates based on the number of PCR amplification per individual. Segregation patterns in the family suggested a partially duplicated genome structure and disomic inheritance. This indicates that the common carp is tetraploid and polyploidy occurred by allotetraploidy. Two primer pairs (HLJ021 and HLJ332) were estimated to amplify reduction based on the number of PCR amplification per individual. One allele in HLJ002 locus and HLJ332 locus was clearly lost in the gynogenetic family and the same as in six wild populations. Segregation patterns in the family suggested a partially diplodization genome structure. | 6 | A consensus linkage map of common carp (Cyprinus carpio L.) to compare the distribution and variation of QTLs associated with growth traits显示文摘The ability to detect and identify quantitative trait loci (QTLs) in a single population is often limited. Analyzing multiple populations in QTL analysis improves the power of detecting QTLs and provides a better understanding of their functional allelic variation and distribution. In this study, a consensus map of the common carp was constructed, based on four populations, to compare the distribution and variation of QTLs. The consensus map spans 2371.6 cM across the 42 linkage groups and comprises 257 microsatellites and 421 SNPs, with a mean marker interval of 3.7 cM/marker. Sixty-seven QTLs affecting four growth traits from the four populations were mapped to the consensus map. Only one QTL was common to three populations, and nine QTLs were detected in two populations. However, no QTL was common to all four populations. The results of the QTL comparison suggest that the QTLs are responsible for the phenotypic variability observed for these traits in a broad array of common carp germplasms. | 7 | Shrinkage of Mt. Bogda Glaciers of Eastern Tian Shan in Central Asia during 1962–2006显示文摘Many small mountain glaciers have been reported undergoing strong shrinkage, and it is therefore important to understand how they respond to climate change. The availability of topographic maps from 1962, Landsat TM imagery from 1990 and ASTER(Advanced Spaceborne Thermal Emission and Radiometer) imagery from 2006 and field investigation of some glaciers allow a comprehensive analysis of glacier change based on glacier size and topography on Mt. Bogda. Results include:(1) an overall loss of a glacierized area by 31.18±0.31 km^2 or 21.6% from 1962 to 2006,(2) a marked dependence of glacier area shrinkage on initial size, with smaller glaciers experiencing higher shrinkage levels,(3) the disappearance of 12 small glaciers,(4) a striking difference in area loss between the southern and northern slopes of 25% and 17%, respectively. A subset of the investigated glaciers shows that the area 57.45±0.73 km2 in 1962 reduced to 54.79±0.561 km^2 in 1990 and 48.88±0.49 km^2 in 2006, with a relative area reduction of 4.6% during 1962–1990, and 10.8% during 1990–2006. The corresponding volume waste increased from 6.9% to 10.2%. Three reference glaciers were investigated in 1981 and revisited in 2009. Their terminus experienced a marked recession. Meteorological data from stations around Mt. Bogda reveals that glacier shrinkage is correlated with winter warming and an extension of the ablation period. | 8 | Genesis of Geothermal Fluid in Typical Geothermal Fields in Western Sichuan,China显示文摘The hydrogeochemical characteristics of geothermal fluids can reveal the genesis of geothermal systems and act as important references for developing and using geothermal resources.This study presents hydrogeochemical processes and thermal cycle mechanisms of typical geothermal fields in Western Sichuan.Based on the geological conditions in Western Sichuan,29 hot springs in three geothermal fields in the Batang and Litang areas were selected for hydrochemical and isotopic(δD andδ18O)analyses.Furthermore,the temperature of the thermal reservoir was calculated and the upflow cooling process of the hot springs was analyzed.Most of the subterranean hot waters in Batang and Litang are of the HCO3-Na hydrochemical type.The ion variation in Batang is primarily affected by water-rock interactions.There is a strong positive correlation between Na+,B?,and Cl?in Litang,suggesting that they have the same material source.The Na+and metaboric acid content is relatively high,which indicates that the groundwater runoff in both areas is relatively long-lasting,with reduced flow velocity;moreover,the metasilicic acid content is relatively high,which supports this conclusion.Both hydrogen and oxygen isotopes plot near the atmospheric precipitation line,indicating that groundwater recharge is functionally obtained from precipitation.The calculated thermal storage temperatures in Batang and Litang were 88–199℃ and 96–154℃,respectively.The proportion of cold water mixing in Batang was 64%–67%,while that in Litang was 60%–68%.According to the calculated results,the initial thermal cycle depth of the Batang area(4540–4780 m)was greater than that of the Litang area(3150–3960 m).The enthalpy of the deep parental geothermal fluid in Batang was 1550 J/g with a Cl^(-)concentration of 37 mg/L,while that in Litang was 2100 J/g with a Cl^(-)concentration of 48 mg/L. | ZHAO Jiayi WANG Guiling ZHANG Cuiyun XING Linxiao LI Man ZHANG Wei | 2021 | Acta Geologica Sinica(English Edition)2021,95,3: | 2 |
| 9 | Effect of Starch Sources on the Release Rates of Herbicides Encapsulated显示文摘The effects of starch sources on the behaviors of starch matrices and on the rates of herbicides released from the matrices were studied for slow release. The starches considered include native corn starch, wheat starch, potato starch and cassava starch. The matrices were prepared through encapsulating 2,4-dichlorophenoxyacetic or 2,4,5-trichlorophenoxyacetic acids as model herbicides with hot-gelatinized starch pastes. The encapsulation was evaluated in terms of herbicide content, swellability, encapsulation efficiency, and release rate. The results show that starch sources play an important role on the matrix behaviors and on release rates. The rate of 2,4-D released follows the order: wheat starch < potato starch < corn starch < cassava starch. And for the rate of 2,4,5-T, this order is nearly the same only with an exception that the late two kinds of starch are similar.
