| 2 | Achievements in cancer research supported by National Natural Science Foundation of China显示文摘For the past two decades,great achievements have been made in cancer research of China and the majority of them were supported by the National Natural Science Foundation of China(NSFC).In this article,we reviewed the funding history of NSFC including the number of proposals,the amount of grants,distribution of supported program categories during the period of 1987 to 2012 and the representative achievements made by continuous support of NSFC.Also,the impacts of NSFC support on discipline development and talent fostering were analyzed.From 1987 to 2012,a total of 8738 proposals related to cancer research were funded by NSFC.The percentages of the Research Program,Talent-Training Program,and the Research Environment Program were 61.19%,36.32%,and 2.49%,respectively.Under the support of NSFC,cancer research in China has achieved a significantly rapid and balanced development in both disciplines and scientist resources. | HONG Wei LI Cui ZHANG LiPing XU YanYing ZHANG Jun HU Hai DONG ErDan | 2013 | Chinese Science Bulletin2013,58,1: | 5 |

| 5 | Heart failure research in China: current status and future direction显示文摘Heart failure is a major public health problem worldwide. Despite advances in the therapy and care of heart failure, morbidity and mortality remain persistently high. Recent years have witnessed major breakthroughs in the investigations of pathogenesis, prevention, and treatment of heart failure in China. It is noteworthy that the continuing and growing support from funding agencies in China including the National Natural Science Foundation of China has yielded substantial effects on the basic and clinical research in heart failure in the last decade. In this review, we provide an update on the current status of heart failure research, both basic and translational/clinical, in China, including funding and publications. | 6 | Pathological matrix stiffness promotes cardiac fibroblast differentiation through the POU2F1 signaling pathway显示文摘Cardiac fibroblast(CF)differentiation into myofibroblasts is a crucial cause of cardiac fibrosis,which increases in the extracellular matrix(ECM)stiffness.The increased stiffness further promotes CF differentiation and fibrosis.However,the molecular mechanism is still unclear.We used bioinformatics analysis to find new candidates that regulate the genes involved in stiffnessinduced CF differentiation,and found that there were binding sites for the POU-domain transcription factor,POU2F1(also known as Oct-1),in the promoters of 50 differentially expressed genes(DEGs)in CFs on the stiffer substrate.Immunofluorescent staining and Western blotting revealed that pathological stiffness upregulated POU2F1 expression and increased CF differentiation on polyacrylamide hydrogel substrates and in mouse myocardial infarction tissue.A chromatin immunoprecipitation assay showed that POU2F1 bound to the promoters of fibrosis repressors IL1R2,CD69,and TGIF2.The expression of these fibrosis repressors was inhibited on pathological substrate stiffness.Knockdown of POU2F1 upregulated these repressors and attenuated CF differentiation on pathological substrate stiffness(35 kPa).Whereas,overexpression of POU2F1 downregulated these repressors and enhanced CF differentiation.In conclusion,pathological stiffness upregulates the transcription factor POU2F1 to promote CF differentiation by inhibiting fibrosis repressors.Our work elucidated the crosstalk between CF differentiation and the ECM and provided a potential target for cardiac fibrosis treatment. | Mingzhe Li Jimin Wu Guomin Hu Yao Song Jing Shen Junzhou Xin Zijian Li Wei Liu Erdan Dong Ming Xu Youyi Zhang Han Xiao | 2021 | Science China(Life Sciences)2021,64,2: | 3 |
| 7 | Extracting and Measuring Uncertain Biomedical Knowledge from Scientific Statements显示文摘Purpose:Given the information overload of scientific literature,there is an increasing need for computable biomedical knowledge buried in free text.This study aimed to develop a novel approach to extracting and measuring uncertain biomedical knowledge from scientific statements.Design/methodology/approach:Taking cardiovascular research publications in China as a sample,we extracted subject-predicate-object triples(SPO triples)as knowledge units and unknown/hedging/conflicting uncertainties as the knowledge context.We introduced information entropy(IE)as potential metric to quantify the uncertainty of epistemic status of scientific knowledge represented at subject-object pairs(SO pairs)levels.Findings:The results indicated an extraordinary growth of cardiovascular publications in China while only a modest growth of the novel SPO triples.After evaluating the uncertainty of biomedical knowledge with IE,we identified the Top 10 SO pairs with highest IE,which implied the epistemic status pluralism.Visual presentation of the SO pairs overlaid with uncertainty provided a comprehensive overview of clusters of biomedical knowledge and contending topics in cardiovascular research.Research limitations:The current methods didn’t distinguish the specificity and probabilities of uncertainty cue words.The number of sentences surrounding a given triple may also influence the value of IE.Practical implications:Our approach identified major uncertain knowledge areas such as diagnostic biomarkers,genetic polymorphism and co-existing risk factors related to cardiovascular diseases in China.These areas are suggested to be prioritized;new hypotheses need to be verified,while disputes,conflicts,and contradictions need to be settled.Originality/value:We provided a novel approach by combining natural language processing and computational linguistics with informetric methods to extract and measure uncertain knowledge from scientific statements. | Xin Guo Yuming Chen Jian Du Erdan Dong | 2022 | Journal of Data and Information Science2022,7,2: | 3 |

| 9 | Renin--angiotensin system inhibitor is associated with the reduced risk of all-cause mortality in COVID-19 among patients with/without hypertension显示文摘Consecutively hospitalized patients with confirmed coronavirus disease 2019(COVID-19)in Wuhan,China were retrospectively enrolled from January 2020 to March 2020 to investigate the association between the use of renin–angiotensin system inhibitor(RAS-I)and the outcome of this disease.Associations between the use of RAS-I(angiotensin-converting enzyme inhibitor(ACEI)or angiotensin receptor blocker(ARB)),ACEI,and ARB and in-hospital mortality were analyzed using multivariate Cox proportional hazards regression models in overall and subgroup of hypertension status.A total of 2771 patients with COVID-19 were included,with moderate and severe cases accounting for 45.0%and 36.5%,respectively.A total of 195(7.0%)patients died.RAS-I(hazard ratio(HR)=0.499,95%confidence interval(CI)0.325–0.767)and ARB(HR=0.410,95%CI 0.240–0.700)use was associated with a reduced risk of all-cause mortality among patients with COVID-19.For patients with hypertension,RAS-I and ARB applications were also associated with a reduced risk of mortality with HR of 0.352(95%CI 0.162–0.764)and 0.279(95%CI 0.115–0.677),respectively.RAS-I exhibited protective effects on the survival outcome of COVID-19.ARB use was associated with a reduced risk of all-cause mortality among patients with COVID-19. | Huai-yu Wang Suyuan Peng Zhanghui Ye Pengfei Li Qing Li Xuanyu Shi Rui Zeng Ying Yao Fan He Junhua Li Liu Liu Shuwang Ge Xianjun Ke Zhibin Zhou Gang Xu Ming-hui Zhao Haibo Wang Luxia Zhang Erdan Dong | 2022 | Frontiers of Medicine2022,16,1: | 0 |

| 13 | FMO3-TMAO axis modulates the clinical outcome in chronic heart-failure patients with reduced ejection fraction:evidence from an Asian population显示文摘The association among plasma trimethylamine-N-oxide(TMAO),FMO3 polymorphisms,and chronic heart failure(CHF)remains to be elucidated.TMAO is a microbiota-dependent metabolite from dietary choline and carnitine.A prospective study was performed including 955 consecutively diagnosed CHF patients with reduced ejection fraction,with the longest follow-up of 7 years.The concentrations of plasma TMAO and its precursors,namely,choline and carnitine,were determined by liquid chromatography-mass spectrometry,and the FMO3 E158K polymorphisms(rs2266782)were genotyped.The top tertile of plasma TMAO was associated with a significant increment in hazard ratio(HR)for the composite outcome of cardiovascular death or heart transplantation(HR=1.47,95%CI=1.13-1.91,P=0.004)compared with the lowest tertile.After adjustments of the potential confounders,higher TMAO could still be used to predict the risk of the primary endpoint(adjusted HR=1.33,95%CI=1.01-1.74,P=0.039).This result was also obtained after further adjustment for carnitine(adjusted HR=1.33,95%CI=1.01-1.74,P=0.039).The FM03 rs2266782 polymorphism was associated with the plasma TMAO concentrations in our cohort,and lower TMAO levels were found in the AA-genotype.Thus,higher plasma TMAO levels indicated increased risk of the composite outcome of cardiovascular death or heart transplantation independent of potential confounders,and the FMO3 AA-genotype in rs2266782 was related to lower plasma TMAO levels. | Haoran Wei Mingming Zhao Man Huang Chenze Li Jianing Gao Ting Yu Qi Zhang Xiaoqing Shen Liang Ji Li Ni Chunxia Zhao Zeneng Wang Erdan Dong Lemin Zheng Dao Wen Wang | 2022 | Frontiers of Medicine2022,16,2: | 0 |
| 14 | A novel β_(2)-AR agonist,Higenamine,induces β-arrestin-biased signaling显示文摘The biased ligands in G protein-coupled receptors(GPCRs)have opened new avenues for developing safer and more effective drugs.However,the identification of such biased ligands as drug candidates is highly desirable.Here,we report that Higenamine,a compound isolated from a Chinese herb,functions as a novel β-arrestin-biased ligand of the β_(2)-adrenergic receptor(β_(2)-AR).The radioligand binding assays demonstrated that Higenamine was the ligand of β_(2)-AR.Higenamine induced phosphorylation of extracellular signal-regulated kinase 1/2(ERK1/2),which can be blocked by propranolol,an inhibitor of β_(2)-AR.The Gi protein inhibitor,pertussis toxin,had no effect on the phosphorylation of ERK1/2 induced by Higenamine.Furthermore,Higenamine induced ERK1/2 phosphorylation through transactivation of Epithelial growth factor receptor(EGFR).We also found that Higenamine-induced-ERK1/2 phosphorylation is dependent on β-arrestin1/2,and HG inhibits Doxorubicin-induced cardiomyocyte apoptosis.Our results identify Higenamine as a novel biased ligand via the β-arrestin-dependent pathway.These findings give us a better understanding of Higenamine’s potential role in designing diagnostic and therapeutic strategies. | Nana Zhang Haibo Zhu Zijian Li Erdan Dong | 2022 | Science China(Life Sciences)2022,65,7: | 0 |
