1Effect of hepatocyte apoptosis induced by TNF-α on acute severe hepatitis in mouse models显示文摘AIM To study the effect of hepatocyteapoptosis and necrosis induced by TNF-α on thepathogenesis of acute severe hepatitis(ASH).METHODS The model of ASH was prepared inD-galactosamine(GAIN)sensitized BALB/c miceby injection of either endotoxin(ET)or tumornecrosis factor-α(TNF-α).Morphologicalchanges of apoptotic hepatocytes were studiedby both light and electron microscope and in siteend labeling method(ISEL).Molecular biologicalchanges of DNA ladder were observed byelectrophoresis of extract from liver tissues.Biochemical changes were measured by alanineaminotransferase(ALT),asparticaminotransferase(AST)and TNF-α.The relationbetween apoptosis and necrosis was evaluatedsimultaneously.RESULTS The sequence of hepatocyteapoptosis,necrosis,and final death from ASHwas observed both in GAIN/ET and GAIN/TNF-agroup.Apoptosis was prominent at 3.5 h and 5 hafter injection of inducer,while necrosis becamedominant at 9 h after challenge.The appearanceof apoptosis was earlier in GAIN/TNF-α groupthan that in GAIN/ ET group.Pretreatment ofmice with antiTNF IgG1 may completely preventthe liver injury induced by GalN/ET.CONCLUSION TNF-α can cause liver damageby inducing hepatic apoptosis and necrosis inmice with endotoxemia.Guo Qing Zang Xia Qiu Zhou Hong Yu Qing Xie Guo Ming Zhao Bin Wang Qing Guo Yue Qin Xiang Dan Liao Department of Infectious Diseases,Rujin Hospital,Shanghai Second Medical University,Shanghai 200025,China 2000World Journal of Gastroenterology2000,6,5:30

3MicroRNA-143 suppresses gastric cancer cell growth and induces apoptosis by targeting COX-2显示文摘AIM:To investigate the function of microRNA-143(miR-143)in gastric cancer and explore the target genes of miR-143.METHODS:A quantitative real-time reverse transcription-polymerase chain reaction(RT-PCR)analysis was performed to evaluate miR-143 expression in gastric cancer cell lines.After transfecting gastric cancer cells with miR-143-5p and miR-143-3p precursors,Alamar blue and apoptosis assays were used to measure the respective proliferation and apoptosis rates.Cyclooxygenase-2(COX-2)expression was determined by realtime RT-PCR and Western blot assays after miR-143transfection.Reporter plasmids were constructed,and a luciferase reporter assay was used to identify the miR-143 binding site on COX-2.RESULTS:Both miR-143-5p and miR-143-3p were significantly downregulated in multiple gastric cancer cell lines.Forced miR-143-5p and miR-143-3p expression in gastric cancer cells produced a profound cytotoxic effect.MiR-145-5p transfection into gastric cancer cells resulted in a greater growth inhibitory effect(61.23%±3.16%vs 46.58%±4.28%,P<0.05 in the MKN-1cell line)and a higher apoptosis rate(28.74%±1.93%vs 22.13%±3.31%,P<0.05 in the MKN-1 cell line)than miR-143-3p transfection.Further analysis indicated that COX-2 expression was potently suppressed by miR-143-5p but not by miR-143-3p.The activity of a luciferase reporter construct that contained the 3’-untranslated region(UTR)of COX-2 was downregulated by miR-143-5p(43.6%±4.86%,P<0.01)but not by miR-143-3p.A mutation in the miR-145-5p binding site completely ablated the regulatory effect on luciferase activity,which suggests that there is a direct miR-145-5p binding site in the 3’-UTR of COX-2.CONCLUSION:Both miR-143-5p and miR-143-3p function as anti-oncomirs in gastric cancer.However,miR-143-5p alone directly targets COX-2,and it exhibits a stronger tumor suppressive effect than miR-143-3p.Xiao-Li Wu Bin Cheng Pei-Yuan Li Huan-Jun Huang Qiu Zhao Zi-Li Dan Dean Tian Peng Zhang 2013World Journal of Gastroenterology2013,19,43:15
4Autophagy and tight junction proteins in the intestine and intestinal diseases显示文摘The intestinal epithelium(IE) forms an indispensible barrier and interface between the intestinal interstitium and the luminal environment. The IE regulates water, ion and nutrient transport while providing a barrier against toxins, pathogens(bacteria, fungi and virus) and antigens. The apical intercellular tight junctions(TJ) are responsible for the paracellular barrier function and regulate transepithelial flux of ions and solutes between adjacent cells. Increased intestinal permeability caused by defects in the IE TJ barrier is considered an important pathogenic factor for the development of intestinal inflammation, diarrhea and malnutrition in humans and animals. In fact, defects in the IE TJ barrier allow increased antigenic penetration, resulting in an amplified inflammatory response in inflammatory bowel disease(IBD), necrotizing enterocolitis and ischemia-reperfusion injury. Conversely, the beneficial enhancement of the intestinal TJ barrier has been shown to resolve intestinal inflammation and apoptosis in both animal models of IBD and human IBD. Autophagy(self-eating mechanism) is an intracellular lysosome-dependent degradation and recycling pathway essential for cell survival and homeostasis.Dysregulated autophagy has been shown to be directly associated with many pathological processes,including IBD. Importantly, the crosstalk between IE TJ and autophagy has been revealed recently. We showed that autophagy enhanced IE TJ barrier function by increasing transepithelial resistance and reducing the paracellular permeability of small solutes and ions, which is, in part, by targeting claudin-2,a cation-selective, pore-forming, transmembrane TJ protein, for lysosome(autophagy)-mediated degradation. Interestingly, previous studies have shown that the inflamed intestinal mucosa in patients with active IBD has increased claudin-2 expression. In addition, inflammatory cytokines(for example,tumor necrosis factor-α, interleukin-6, interleukin-13, and interleukin-17) whose levels are increased in IBD patients cause an increase in claudin-2 expression and a claudin-2-dependent increase in TJ permeability. Thus, the role of claudin-2 in intestinal pathological processes has been attributed, in part, to the increase of intestinal TJ permeability. 7Basic Characteristics,Spatial Disparity and Its Major Influencing Factors of Service Industry in China显示文摘Based on the analysis of its basic characteristics,this article investigated the disparities of Chinese service industry among the three regions(the eastern China,the western China and the middle China) and inter-provincial disparities of that in the three regions by Theil coefficient and cluster analysis.Then,major factors influencing its spatial disparity were explored by correlation analysis and regression analysis.The conclusions could be drawn as follows.1) The development of Chinese service industry experienced three phases since the 1980s:rapid growth period,slow growth period,and recovery period.From the proportion of value-added and employment,its development was obviously on the low level.From the composition of industrial structure,traditional service sectors were dominant,but modern service sectors were lagged.Moreover,its spatial disparity was distinct.2) The level of Chinese service industry was divided into five basic regional ranks:well-developed,developed,relatively-developed,underdeveloped and undevel-oped regions.As a whole,the overall structure of spatial disparity was steady in 1990-2005.But there was notable gradient disparity in the interior structure of service industry among different provinces.Furthermore,the overall disparity expanded rapidly in 1990-2005.The inter-provincial disparity of service industry in the three regions,especially in the eastern China,was bigger than the disparity among the three regions.And 3) the level of economic development,the level of urban development,the scale of market capacity,the level of transportation and telecommunication,and the abundance of human resources were major factors influencing the development of Chinese service industry.SHEN Yuming QIU Ling REN Wangbing CAO Yi HU Dan SONG Yujing 2009Chinese Geographical Science2009,19,4:6

10IL-17-triggered downregulation of miR-497 results in high HIF-1α expression and consequent IL-1β and IL-6 production by astrocytes in EAE mice显示文摘Interleukin 17(IL-17)is increasingly recognized as a key factor that contributes to the pathogenesis of multiple sclerosis(MS)and its experimental mouse autoimmune encephalomyelitis(EAE)model.However,the roles and regulatory mechanisms of IL-17-induced pro-inflammatory cytokine production in EAE mice remain largely unclear.In this study,the expression of IL-17,hypoxia inducible factor-1α(HIF-1α),IL-1β,IL-6 and microRNA-497(miR-497),as well as their intrinsic associations,was investigated using EAE model mice and cultured astrocytes exposed to IL-17 in vitro.We observed markedly increased production of IL-17,HIF-1α,IL-1βand IL-6 in the brain tissues of EAE mice,while the expression and secretion of HIF-1α,IL-1βand IL-6 were also significantly increased when cultured primary astrocytes from mice were stimulated with IL-17.Meanwhile,the expression of miR-497 was downregulated both in vivo and in vitro.Subsequent in vitro experiments revealed that IL-17 induced the production of IL-1βand IL-6 in astrocytes through the upregulation of HIF-1αas a transcriptional factor,indicating that IL-17-mediated downregulation of miR-497 enhanced HIF-1αexpression.Furthermore,astrocytespecific knockdown of IL-17RA and HIF-1αor astrocyte-specific overexpression of miR-497 by infection with different lentiviral vectors containing an astrocyte-specific promotor markedly decreased IL-1βand IL-6 production in brain tissues and alleviated the pathological changes and score of EAE mice.Collectively,these findings indicate that decreased miR-497 expression is responsible for IL-17-triggered high HIF-1αexpression and consequent IL-1βand IL-6 production by astrocytes in EAE mice.Kai Shan Rongrong Pang Chenhui Zhao Xiaomei Liu Wenxing Gao Jing Zhang Dan Zhao Yingwei Wang Wen Qiu 2017Cellular & Molecular Immunology2017,14,11:5
11Berberine-Promoted CXCR4 Expression Accelerates Endothelial Repair Capacity of Early Endothelial Progenitor Cells in Persons with Prehypertension显示文摘Objective: To evaluate whether the berberine treatment can improve endothelial repair capacity of early endothelial progenitor cells(EPCs) from prehypertensive subjects through increasing CXC chemokine receptor 4(CXCR4) signaling. Methods: EPCs were isolated from prehypertensive and healthy subjects and cultured. In vivo reendothelialization capacity of EPCs from prehypertensive patients with or without in vitro berberine treatment was examined in a nude mouse model of carotid artery injury. The protein expressions of CXCR4/Janus kinase-2(JAK-2) signaling of in vitro EPCs were detected by Western blot analysis. Results: CXCR4 signaling and alteration in migration and adhesion functions of EPCs were evaluated. Basal CXCR4 expression was significantly reduced in EPCs from prehypertensive patients compared with normal subjects(P<0.01). Also, the phosphorylation of JAK-2 of EPCs, a CXCR4 downstream signaling, was significantly decreased(P<0.01). Berberine promoted CXCR4/JAK-2 signaling expression of in vitro EPCs(P<0.01). Transplantation of EPCs pretreated with berberine markedly accelerated in vivo reendothelialization(P<0.01). The increased in vitro function and in vivo reendothelialization capacity of EPCs were inhibited by CXCR4 neutralizing antibody or pretreatment with JAK-2 inhibitor AG490, respectively(P<0.01). 12Anterior Cingulate Cortex Mediates Hyperalgesia and Anxiety Induced by Chronic Pancreatitis in Rats显示文摘Central sensitization is essential in maintaining chronic pain induced by chronic pancreatitis(CP),but cortical modulation of painful CP remains elusive.Here,we examined the role of the anterior cingulate cortex(ACC)in the pathogenesis of abdominal hyperalgesia in a rat model of CP induced by intraductal administration of trinitrobenzene sulfonic acid(TNBS).TNBS treatment resulted in long-term abdominal hyperalgesia and anxiety in rats.Morphological data indicated that painful CP induced a significant increase in FOS-expressing neurons in the nucleus tractus solitarii(NTS)and ACC,and some FOS-expressing neurons in the NTS projected to the ACC.In addition,a larger portion of ascending fibers from the NTS innervated pyramidal neurons,the neural subpopulation primarily expressing FOS under the condition of painful CP,rather than GABAergic neurons within the ACC.CP rats showed increased expression of vesicular glutamate transporter 1,and increased membrane trafficking and phosphorylation of the N-methyl-D-aspartate receptor(NMDAR)subunit NR2B and theα-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid receptor(AMPAR)subunit GluR1 within the ACC.Microinjection of NMDAR and AMPAR antagonists into the ACC to block excitatory synaptic transmission significantly attenuated abdominal hyperalgesia in CP rats,which was similar to the analgesic effect of endomorphins injected into the ACC.Specifically inhibiting the excitability of ACC pyramidal cells via chemogenetics reduced both hyperalgesia and comorbid anxiety,whereas activating these neurons via optogenetics failed to aggravate hyperalgesia and anxiety in CP rats.Taken together,these findings provide neurocircuit,biochemical,and behavioral evidence for involvement of the ACC in hyperalgesia and anxiety in CP rats,as well as novel insights into the cortical modulation of painful CP,and highlights the ACC as a potential target for neuromodulatory interventions in the treatment of painful CP.Dan Ren Jia-Ni Li Xin-Tong Qiu Fa-Ping Wan Zhen-Yu Wu Bo-Yuan Fan Ming-Ming Zhang Tao Chen Hui Li Yang Bai Yun-Qing Li 2022Neuroscience Bulletin2022,38,4:4
13Cardioprotective mechanisms of salvianic acid A sodium in rats with myocardial infarction based on proteome and transcriptome analysis显示文摘Ischemic heart diseases(IHDs)cause great morbidity and mortality worldwide,necessitating effective treatment.Salvianic acid A sodium(SAAS)is an active compound derived from the well-known herbal medicine Danshen,which has been widely used for clinical treatment of cardiovascular diseases in China.This study aimed to confirm the cardioprotective effects of SAAS in rats with myocardial infarction and to investigate the underlying molecular mechanisms based on proteome and transcriptome profiling of myocardial tissue.The results showed that SAAS effectively protected against myocardial injury and improved cardiac function.The differentially expressed proteins and genes included important structural molecules,receptors,transcription factors,and cofactors.Functional enrichment analysis indicated that SAAS participated in the regulation of actin cytoskeleton,phagosome,focal adhesion,tight junction,apoptosis,MAPK signaling,and Wnt signaling pathways,which are closely related to cardiovascular diseases.SAAS may exert its cardioprotective effect by targeting multiple pathways at both the proteome and transcriptome levels.This study has provided not only new insights into the pathogenesis of myocardial infarction but also a road map of the cardioprotective molecular mechanisms of SAAS,which may provide pharmacological evidence to aid in its clinical application.Dan Jia Cheng-zhong Zhang Yan Qiu Xiao-fei Chen Lin Jia Alex F.Chen Yi-feng Chai Zhen-yu Zhu Jin Huang Chuan Zhang 2019Acta Pharmacologica Sinica2019,40,12:4
14Simultaneous Determination of Six Quaternary Ammonium Alkaloids in Coptidis Rhizoma by UPLC显示文摘Objective To establish a new,rapid,and reliable reversed-phase ultra performance liquid chromatography (RP-UPLC)method for the simultaneous determination of six quaternary ammonium alkaloids(QAAs)in Coptidis Rhizoma.Methods The effect of different experimental parameters on the analysis of QAAs by RP-UPLC was evaluated.Results Optimal resolution was achieved with an Acquity UPLC BEH C18 column using a gradient elution profile and a mobile phase consisting of water spiked with 10 mmol/L ammonium bicarbonate(A,pH adjusted to 10.0 by ammonia water)and acetonitrile(B),at a flow rate of 0.30 mL/min and wavelength of 345 nm. 15Stern-Geary Constant for X80 Pipeline Steel in the Presence of Different Corrosive Microorganisms显示文摘The Stern-Geary constant(B value)is indispensable to measure the corrosion rate in the microbiologically influenced corrosion(MIC)systems.Linear polarization resistance(LPR)and weight loss methods were used to study the variation of B values for X80 pipeline steel in the presence of Pseudomonas aeruginosa,Acetobacter aceti and Desulfovibrio vulgaris.The results showed that B values in the presence of three different bacteria were 35.60±0.55 mV,33.00±1.00 mV and 58.60±0.55 mV,respectively,suggesting that the change of corrosion system significantly affected the B values of X80 pipeline steel.This work further indicated that the determination of B values is necessary to accurately measure the MIC rate by LPR method.Yu-Peng Sun Chun-Tian Yang Chun-Guang Yang Da-Ke Xu Qi Li Lu Yin Cheng-Shuo Qiu Dan liu Ke Yang 2019Acta Metallurgica Sinica(English Letters)2019,32,12:3
16Application of microcalorimetry of Escherichia coli growth and discrimin antanalysis to the quality assessment of a Chinese herbal injection(Yinzhihuang)显示文摘This paper describes a novel approach to the quality control of a Chinese herbal injection based on microca lorimetric determination of its effect on the biothermal active fingerprint(BTAF)of Escherichia coli(E.coli)growth.Yinzhihuang Injection(YZHI)was selected for this proof of concept study.Reference samples of Y ZHI were collected and compared with test(stressed)samples prepared under different stress conditions.The BTAF of E.coli growth was found to be affected by YZHI and the changes were analyzed on the basis of eleven biothermokinetic parameters.Similarity and multivariate statistical analysis were used to investi-gate the differences between reference and test samples and discriminant analysis was used to delineate the altered samples.Reference samples were found to have coincident BTAFs with similarity index>0.99.Stressed samples showed differences in the BTAF which increased in line with decreased quality.Discriminant formulae were developed based on a sensitivity parameter which could identify all altered samples.In conclusion,BTAF can be used to assess the quality of YZHI both qualitatively and quantitatively and has the potential to provide a sensitive method for quality control of Chinese herbal injections.Yongshen Ren Ping Zhang Dan Yan Yan Yan Longhu Chen Lingling Qiu Zhinan Mei Xiaohe Xiao 2012Acta Pharmaceutica Sinica B2012,2,3:3
17Effect of acupuncture plus conventional treatment on brain activity in ischemic stroke patients: a regional homogeneity analysis显示文摘OBJECTIVE: To determine differences in cerebral activity evoked by acupuncture and conventional stroke treatment, and identify the treatment targets.METHODS: In total, 21 patients were randomly divided into two groups. Group A(11 patients) received both acupuncture and conventional treatment, while group B(10 patients) received conventional treatment only. Resting-state functional magnetic resonance imaging(f MRI) was performed on each participant before and after treatment. Regional homogeneity analysis was performed to investigate the potential mechanism of acupuncture treatment by comparing differences in cerebral activity between treatments.RESULTS: Group A showed higher Re Ho in the frontal lobe(BA6, BA46), supra-marginal gyrus(BA40),middle temporal gyrus(BA21), cerebellum, and insula. Group B showed higher Re Ho in the frontal lobe(BA6) and parietal lobe(BA3, BA7).CONCLUSION: Acupuncture and conventional treatment triggered relatively different clinical efficacy and brain responses. Acupuncture treatment more significantly improved the symptoms of stroke patients. 20Poly(amide-6-b-ethylene oxide)/[Bmim][Tf2N] blend membranes for carbon dioxide separation显示文摘Poly(amide-6-b-ethylene oxide)(Pebax1657)/1-butyl-3-methylimidazo-lium bis[trifluoromethyl)sulfonyl]-imide([Bmim][Tf_2N]) blend membranes with different [Bmim][Tf_2N] contents were prepared via solution casting and solvent evaporation method. The permeation properties of the blend membranes for CO_2, N_2,CH_4 and H_2 were studied, and the physical properties were characterized by differential scanning calorimeter(DSC) and X-ray diffraction(XRD). Results showed that [Bmim][Tf_2N] was dispersed as amorphous phase in the blend membranes, which caused the decrease of Tg(PE) and crystallinity(PA). With the addition of [Bmim][Tf_2N], the CO_2 permeability increased and reached up to approximately 286 Barrer at 40 wt%[Bmim][Tf_2N], which was nearly double that of pristine Pebax1657 membrane. The increase of CO_2 permeability may be attributed to high intrinsic permeability of [Bmim][Tf_2N], the increase of fractional free of volume(FFV) and plasticization effect. However, the CO_2 permeability reduced firstly when the [Bmim][Tf_2N]content was below 10 wt%, which may be due to that the small ions of [Bmim][Tf_2N] in the gap of polymer chain inhibited the flexibility of polymer chain; the interaction between Pebax1657 and [Bmim][Tf_2N]decreased the content of EO units available for CO_2 transport and led to a more compact structure. For Pebax1657/[Bmim][Tf_2N] blend membranes, the permeabilities of N_2, H2 and CH4decreased with the increase of feed pressure due to the hydrostatic pressure effect, while CO_2 permeability increased with the increase of feed pressure for that the CO_2-induced plasticization effect was stronger than hydrostatic pressure effect.Yongtao Qiu Jizhong Ren Dan Zhao Hui Li Maicun Deng 2016Journal of Energy Chemistry2016,25,1:3
