3Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans.Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang 2020Chinese Medical Journal2020,,9:99
4A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 5Current status and future tendency of lake eutrophication in China显示文摘Current trophic status and trend of Chinese freshwater lakes were investigated in this study. The results showed that all lakes studied were commonly undergoing the eutrophication process, water quality decreased and lake's ecosystem is being declined. Most of the urban lakes are facing serious eutrophication. Many medium-sized lakes are in metrophic or eutrophic status, some local water are even approaching the hypertrophic level. The famous five freshwater lakes in China have entered into eutrophication in the condition of higher nutrient load. Lake Taihu, Hongze and Caohu are already in eutrophic state. Eutrophic lakes are mainly distributed in the middle and lower reaches of Yangtze River and Yungui plateau. Lake eutrophication developed rapidly. Among the 34 lakes studied in 1970's, most of lakes were in the mesotrophic status,mesotrophic water area accounted for 91.8%. With the nine year of 1978-1987 the area percentage of oligotrophic lakes decreased from 3.2% to 0.53%, and that of eutrophic lakes increased from 5.0% to 55.01%. Recent data showed 57.5% lakes were in eutrophic and hypertrophic status of the 40 surveyed lakes.Eutrophic trend of Lake Taihu, Chaohu and Xuanwu in the region of the middle and lower reaches of Yangtze River was predicated using the ecological stress model. The results showed that in 2008 Lake Taihu, Chaohu and Xuanwu might be of eutrophication, eutrophication and hypertrophication, respectively if no control measurement is taken. Provided the pollution water treatment rate is 60% in 2030, approximately 30 billion ton pollution water would still be discharged directly in the lakes. Therefore, in 2030 the urban lakes in China might be eutrophication or hypertrophication, and most of the medium-sized lakes at the urban-rural fringe might be in eutrophication or hypertrophication. The famous five biggest freshwater lakes in China might be eutrophication if control countermeasures are taken as now.Lake eutrophication has become a serious environmental problem in China. 6Spatiotemporal precipitation variations in the arid Central Asia in the context of global warming显示文摘This study analyzed the temporal precipitation variations in the arid Central Asia (ACA) and their regional differences during 1930-2009 using monthly gridded precipitation from the Climatic Research Unit (CRU). Our results showed that the annual precipitation in this westerly circulation dominated arid region is generally increasing during the past 80 years, with an apparent increasing trend (0.7 mm/10 a) in winter. The precipitation variations in ACA also differ regionally, which can be divided into five distinct subregions (I West Kazakhstan region, II East Kazakhstan region, III Central Asia Plains region, IV Kyrgyzstan region, and V Iran Plateau region). The annual precipitation falls fairly even on all seasons in the two northern subregions (regions I and II, approximately north of 45°N), whereas the annual precipitation is falling mainly on winter and spring (accounting for up to 80% of the annual total precipitation) in the three southern subregions. The annual precipitation is increasing on all subregions except the southwestern ACA (subregion V) during the past 80 years. A significant increase in precipitation appeared in subregions I and III. The long-term trends in annual precipitation in all subregions are determined mainly by trends in winter precipitation. Additionally, the precipitation in ACA has significant interannual variations. The 2-3-year cycle is identified in all subregions, while the 5-6-year cycle is also found in the three southern subregions. Besides the inter-annual variations, there were 3-4 episodic precipitation variations in all subregions, with the latest episodic change that started in the midto late 1970s. The precipitations in most of the study regions are fast increasing since the late 1970s. Overall, the responses of ACA precipitation to global warming are complicated. 8Effects of ground-level ozone (O_3) pollution on the yields of rice and winter wheat in the Yangtze River Delta显示文摘Effects of elevated O3 on the yields of rice and winter wheat were studied by using open-top chambers(OTCs). Results showed that compared to the control treatment, 200 ppb, 100 ppb, 50 ppb treatments caused a 80.4%, 58.6% and 10.5% decrease in grain yields per winter wheat plant and a 49.1%, 26.1% and 8.2% decrease in grain yield per rice plant, respectively. 92018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. 12Electroacupuncture improves learning and memory functions in a rat cerebral ischemia/reperfusion injury model through PI3K/Akt signaling pathway activation显示文摘Electroacupuncture has been widely used to treat cognitive impairment after cerebral ischemia,but the underlying mechanism has not yet been fully elucidated.Studies have shown that autophagy plays an important role in the formation and development of cognitive impairment,and the phosphoinositide 3-kinase(PI3K)/Akt signaling pathway plays an important role in autophagy regulation.To investigate the role played by the PI3K/Akt signaling pathway in the electroacupuncture treatment of cerebral ischemia/reperfusion rat models,we first established a rat model of cerebral ischemia/reperfusion through the occlusion of the middle cerebral artery using the suture method.Starting at 2 hours after modeling,electroacupuncture was delivered at the Shenting(GV24)and Baihui(GV20)acupoints,with a dilatational wave(1-20 Hz frequency,2 mA intensity,6 V peak voltage),for 30 minutes/day over 8 consecutive days.Our results showed that electroacupuncture reduced the infarct volume in a rat model of cerebral ischemia/reperfusion injury,increased the mRNA expression levels of the PI3K/Akt signaling pathwayrelated factors Beclin-1,mammalian target of rapamycin(mTOR),and PI3K,increased the protein expression levels of phosphorylated Akt,Beclin-1,PI3K,and mTOR in the ischemic cerebral cortex,and simultaneously reduced p53 mRNA and protein expression levels.In the Morris water maze test,the latency to find the hidden platform was significantly shortened among rats subjected to electroacupuncture stimulation compared with rats without electroacupuncture stimulation.In the spatial probe test,the number of times that a rat crossed the target quadrant was increased in rats subjected to electroacupuncture stimulation compared with rats without electroacupuncture stimulation.Electroacupuncture stimulation applied to the Shenting(GV24)and Baihui(GV20)acupoints activated the PI3K/Akt signaling pathway and improved rat learning and memory impairment.This study was approved by the Animal Ethics Committee of the First Affiliated Hospital of Henan University of Traditional Chinese Medicine,China(approval No.8150150901)on March 10,2016.Hui-Ling Wang Fei-Lai Liu Rui-Qing Li Ming-Yue Wan Jie-Ying Li Jing Shi Ming-Li Wu Jun-Hua Chen Wei-Juan Sun Hong-Xia Feng Wei Zhao Jin Huang Ren-Chao Liu Wen-Xue Hao Xiao-Dong Feng 2021Neural Regeneration Research2021,16,6:48
13Epidemiology and risk factors of colorectal cancer in China显示文摘In China, colorectal cancer(CRC) ranked fourth and fifth in the highest incidence and mortality rates of all malignancies in 2018, respectively. Although these rates are below the world average, China placed first worldwide in the number of new CRC cases and CRC-related deaths because of its comparatively large population. This disease represents a threat to the health of population and incurs a heavy economic burden on the society and individuals. CRC has various risk factors, including age, sex, lifestyle, genetic factors, obesity, diabetes, gut microbiota status, and precancerous lesions. Furthermore, incidence and mortality rates of CRC are closely related to socioeconomic development levels, varying according to regional and population characteristics. Prevention is the main strategy to reduce incidence and mortality rates of CRC. This can be achieved through strategies stimulating lifestyle changes, healthy diet habits, and early screening for high-risk individuals. To reduce the burden of CRC, public health officials should promote prevention and management of modifiable risk factors through national policies. The rising incidence and mortality rates of CRC in China may be timely curbed by clarifying specific epidemiological characteristics, optimizing early screening strategies, and strictly implementing diagnosis and treatment guidelines. 15Expression of vascular endothelial growth factor and its role in oncogenesis of human gastric carcinoma显示文摘AIM To establish the role of vascular endothelial growth factor (VEGF) in the oncogenesisof human gastric carcinoma more directly.METHODS The expression of VEGF and its receptor kinase-domain insert containing receptor (KDR) in human gastric cancer tissue were observed by immunohistochemical staining. VEGF levels were manipulated in human gastric cancer cell using eukaryotic expression constructs designed to express the complete VEGF165 complimentary DNA in either the sense or antisense orientation. The biological changes of the cells were observed in which VEGF was up-regulated or downregulated.RESULTS VEGF-positive rate was 50%, and VEGF was mainly localized in the cytoplasm and membrane of the tumor cells, while KDR was mainly located in the membrane of vascular endothelial cells in gastric cancer tissues and peri-cancerous tissue. In 2 cases of 50 specimens, the gastric cancer cells expressed KDR,localized in both the cytoplasm and membrane.Introduction of VEGF165 antisense into human gastric cancer cells ( SGC-7901, immunofluorescence intensity,31.6%)) resulted in a significant reduction in VEGFspecific messenger RNA and total and cell surface VEGF protein ( immunofluorescence intensity, 8.9%)(P<0.05). Conversely, stable integration of VEGF165 in the sense orientation resulted in an increase in cellular and cell surface VEGF (immunofluorescence intensity,75.4%) (P<0.05). 16Permian Alkaline Granites in Central Inner Mongolia and Their Geodynamic Significance显示文摘Alkaline granites (Rb-Sr ages 276-286 Ma)occurring in the Bayan Ul-East Ujimqin belt at the southern margin of the Siberian plate originated in a tensional tectonic environment about 60 Ma earlier than the Late Devonian to Early Carboniferous collision between the Siberian and Sino-Korean plates. They belong to post-orogenic A-type granites and may be used as an indicator of the end of the orogeny. At the northern margin of the Sino-Korean plate, however, only late-orogenic calc-alkaline granites occurred during the late Caboniferous-Permian, and alkaline syenites did not appear until the Late Triassic. 17Molecular cloning and functional characterization of a DREB1/CBF-like gene (GhDREB1L) from cotton显示文摘The transcription factors DREB1s/CBFs play important roles in the regulation of plant resistance to environmental stresses and are quite useful for generating transgenic plants tolerant to these stresses. In the present work, a cDNA encoding DREB1/CBF-like protein (GhDREB1L) from cotton was isolated, and its sequence features, DNA binding preference, and expression patterns of the transcripts were also characterized. GhDREB1L contained one conserved AP2/ERF domain and its amino acid sequence was similar to the DREB1/CBF group of the DREB family from other plants. The DNA-binding domain of GhDREB1L was successfully expressed as a fusion protein in Escherichia coli BL21 (DE3) and purified by Ni-NTA affinity chromatography. Electrophoretic mobility shift assay revealed that the purified GhDREB1L fusion protein had a specific binding activity with the previously characterized DRE ele-ment (core sequence, ACCGAC) and also with the DRE-like sequence (core sequence, GCCGAC) in the promoter of the dehydration-responsive late embryogenesis-abundant gene LEA D113. Semi-quantita- tive RT-PCR showed that GhDREB1L was induced in the cotton cotyledons by low temperature, as well as drought and NaCl treatments. 20The abnormal expression of retinoic acid receptor-β,p53 and Ki67 protein,n normal,premalignant and malignant esophageal tissues显示文摘AIM: Esophageal cancer remains a significant healthiproblem worldwide. It is important to investigate alterationsin expression of retinoic acid receptor-β, p53 and Ki67proteins in esophageal carcinogenesis.METHODS: To find biomarkers for early identification ofesophageal cancer, we analyzed the retinoic acid receptor-j3,p53 protein and the proliferation marker Ki67 in surgicalspecimens of normal, mildly, and severely dysplastic andmalignant esophageal tissues by in situ hybridization ofRNA and immunohistochemistry.RESULTS: RAR-β was expressed in 94.3 %(33/35) of normalmucosae, 67.8 %(19/28) of the mild, 58.1% (18/31) of thesevere ieaions and 53.2 % (116/218) of tumor samples. RAR-β mRNA was expressed in 62.7 % (42/67), 55.1% (43/78) and29.2 % (7/24) of well, moderated and poorly differentiatedSSCs. The p53 and Ki67 proteins were 5.9 % (2/34) of thenormal mucosa. P53 and Ki67 stained positively in 10.7 % (3/28) and 21.4 % (6/28) of mild dysplasia, and 51.6 %(16/31)and 58. 1% (18/31) of severely dysplasia respectively.Samples from esophageal cancer showed no higher levers ofp53 and Ki67 expression than seen in severely dysplasticlesions.
