| 4 | A bone-targeted engineered exosome platform delivering siRNA to treat osteoporosis显示文摘The complex pathogenesis of osteoporosis includes excessive bone resorption,insufficient bone formation and inadequate vascularization,a combination which is difficult to completely address with conventional therapies.Engineered exosomes carrying curative molecules show promise as alternative osteoporosis therapies,but depend on specifically-functionalized vesicles and appropriate engineering strategies.Here,we developed an exosome delivery system based on exosomes secreted by mesenchymal stem cells(MSCs)derived from human induced pluripotent stem cells(iPSCs).The engineered exosomes BT-Exo-siShn3,took advantage of the intrinsic anti-osteoporosis function of these special MSC-derived exosomes and collaborated with the loaded siRNA of the Shn3 gene to enhance the therapeutic effects.Modification of a bone-targeting peptide endowed the BT-Exo-siShn3 an ability to deliver siRNA to osteoblasts specifically.Silencing of the osteoblastic Shn3 gene enhanced osteogenic differentiation,decreased autologous RANKL expression and thereby inhibited osteoclast formation.Furthermore,Shn3 gene silencing increased production of SLIT3 and consequently facilitated vascularization,especially formation of type H vessels.Our study demonstrated that BT-Exo-siShn3 could serve as a promising therapy to kill three birds with one stone and implement comprehensive anti-osteoporosis effects. | Yongzhi Cui Yuanyuan Guo Li Kong Jingyu Shi Ping Liu Rui Li Yongtao Geng Weihang Gao Zhiping Zhang Dehao Fu | 2022 | Bioactive Materials2022,7,4: | 3 |
| 5 | Pre-chamber turbulent jet ignition of methane/air mixtures with multiple orifices in a large bore constant volume chamber: effect of air-fuel equivalence ratio and pre-mixed pressure显示文摘Liquefied natural gas (LNG), mainly composed of methane, is in progress to substitute diesel fuel in heavyduty marine engine for practical, economic, and environmental considerations. However, natural gas is relatively difficult to be ignited in a large bore combustion chamber. A combustion enhancement technique called prechamber turbulent jet ignition (TJI) can permit combustion and flame propagation in a largebore volume. To investigate the effect of air-fuel equivalence ratio and pre-mixed pressure on pre-chamber TJI of methane/air mixtures with multiple orifices in a large bore volume, experimental tests and computational simulations were implemented to study the discharge of hot turbulent jets from six orifices of the pre-chamber. Different initial pressures and air-fuel equivalence ratios were considered to analyze the characteristics of TJI. The asymmetry of the turbulent jet actuated from six different orifices were found due to the asymmetric orientation of the spark plug, resulting in the inhomogeneous distribution of combustion in the constant volume chamber, which should be considered seriously in the marine engine design. | 6 | A Smart Charging Algorithm Based on a Fast Charging Station Without Energy Storage System显示文摘With the growing popularity of electric vehicles(EV),there is an urgent demand to solve the stress placed on grids caused by the irregular and frequent access of EVs.The traditional direct current(DC)fast charging station(FCS)based on a photovoltaic(PV)system can effectively alleviate the stress of the grid and carbon emission,but the high cost of the energy storage system(ESS)and the under utilization of the grid-connected interlinking converters(GIC)are not very well addressed.In this paper,the DC FCS architecture based on a PV system and ESS-free is first proposed and employed to reduce the cost.Moreover,the proposed smart charging algorithm(SCA)can fully coordinate the source/load properties of the grid and EVs to achieve the maximum power output of the PV system and high utilization rate of GICs in the absence of ESS support for FCS.SCA contains a self-regulated algorithm(SRA)for EVs and a grid-regulated algorithm(GRA)for GICs.While the DC bus voltage change caused by power fluctuations does not exceed the set threshold,SRA readjusts the charging power of each EV through the status of the charging(SOC)feedback of the EV,which can ensure the power rebalancing of the FCS.The GRA would participate in the adjustment process once the DC bus voltage is beyond the set threshold range.Under the condition of ensuring the charging power of all EVs,a GRA based on adaptive droop control can improve the utilization rate of GICs.At last,the simulation and experimental results are provided to verify the effectiveness of the proposed SCA. | Qifu Cheng Lei Chen Qiuye Sun Rui Wang Dazhong Ma Dehao Qin | 2021 | CSEE Journal of Power and Energy Systems2021,7,4: | 2 |

| 9 | CUDA-Zero:a framework for porting shared memory GPU applications to multi-GPUs显示文摘As the prevalence of general purpose computations on GPU, shared memory programming models were proposed to ease the pain of GPU programming. However, with the demanding needs of more intensive workloads, it's desirable to port GPU programs to more scalable distributed memory environment, such as multi-GPUs. To achieve this, programs need to be re-written with mixed programming models (e.g. CUDA and message passing). Programmers not only need to work carefully on workload distribution, but also on scheduling mechanisms to ensure the efficiency of the execution. In this paper, we studied the possibilities of automating the process of parallelization to multi-GPUs. Starting from a GPU program written in shared memory model, our framework analyzes the access patterns of arrays in kernel functions to derive the data partition schemes. To acquire the access pattern, we proposed a 3-tiers approach: static analysis, profile based analysis and user annotation. Experiments show that most access patterns can be derived correctly by the first two tiers, which means that zero efforts are needed to port an existing application to distributed memory environment. | 11 | Screening of Potential Biomarkers for Gastric Cancer with Diagnostic Value Using Label-free Global Proteome Analysis显示文摘Gastric cancer(GC)is known as a top malignant type of tumors worldwide.Despite the recent decrease in mortality rates,the prognosis remains poor.Therefore,it is necessary to find novel biomarkers with early diagnostic value for GC.In this study,we present a large-scale proteomic analysis of 30 GC tissues and 30 matched healthy tissues using label-free global proteome profiling.Our results identified 537 differentially expressed proteins,including 280 upregulated and 257 downregulated proteins.The ingenuity pathway analysis(IPA)results indicated that the sirtuin signaling pathway was the most activated pathway in GC tissues whereas oxidative phosphorylation was the most inhibited.Moreover,the most activated molecular function was cellular movement,including tissue invasion by tumor cell lines.Based on IPA results,15 hub proteins were screened.Using the receiver operating characteristic curve,most of hub proteins showed a high diagnostic power in distinguishing between tumors and healthy controls.A four-protein(ATP5B-ATP5O-NDUFB4-NDUFB8)diagnostic signature was built using a random forest model.The area under the curve(AUC)values of this model were 0.996 and 0.886 for the training and testing sets,respectively,suggesting that the four-protein signature has a high diagnostic power.This signature was further tested with independent datasets using plasma enzyme-linked immune sorbent assays,resulting in an AUC value of 0.778 for distinguishing GC tissues from healthy controls,and using immunohistochemical tissue microarray analysis,resulting in an AUC value of 0.805.In conclusion,this study identifies potential biomarkers and improves our understanding of the pathogenesis,providing novel therapeutic targets for GC. | Yongxi Song Jun Wang Jingxu Sun Xiaowan Chen Jinxin Shi Zhonghua Wu Dehao Yu Fei Zhang Zhenning Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 1 |
| 12 | Exergy losses in premixed flames of dimethyl ether and hydrogen blends显示文摘A second-law thermodynamic analysis was conducted for stoichiometric premixed dimethyl ether(DME)/hydrogen(H2)/air flames at atmospheric pressure.The exergy losses from the irreversibility sources,i.e.,chemical reaction,heat conduction and species diffusion,and those from partial combustion products were analyzed in the flames with changed fuel blends.It is observed that,regardless of the fuel blends,chemical reaction contributes most to the exergy losses,followed by incomplete combustion,and heat conduction,while mass diffusion has the least contribution to exergy loss.The results also indicate that increased H2 substitution decreases the exergy losses from reactions,conduction,and diffusion,primarily because of the flame thickness reduction at elevated H2 substitution.The decreases in exergy losses by chemical reactions and heat conduction are higher,but the exergy loss reduction by diffusion is slight.However,the exergy losses from incomplete combustion increase with H2 substitution,because the fractions of the unbumed fuels and combustion intermediates,e.g.,H2 and OH radical,increase.The overall exergy losses in the DME/H2 flames decrease by about 5%with increased H2 substitution from 0%to 100%. | Tongbin ZHAO Jiabo ZHANG Dehao JU Zhen HUANG Dong HAN | 2019 | Frontiers in Energy2019,13,4: | 1 |

| 14 | Interaction of citrate with Pt(100) surface investigated by cyclic voltammetry towards understanding the structure-tuning effect in nanomaterials synthesis显示文摘This study aims to understand the effects of functional agents such as capping agents, stabilizers, surfactants and additives in shape-controlled synthesis of nanomaterials. The well-defined Pt(100) single crystal surface was used as a model to investigate its interaction with citrate, a capping agent that is often used in shape-controlled synthesis of nanomaterials. It demonstrated that, through a systematic study of electrochemical cyclic voltammetry, the presence of citrate in solution could increase the current peak density of hydrogen adsorption at high potential (j p,L ), while decrease proportionally the current peak density of hydrogen adsorption at low potential (j p,S ). Furthermore, the increase of citrate concentration shifted negatively the peak potentials (E p,L and E p,S ) of both j p,L and j p,S . The results indicated that the interaction of citrate with Pt(100) surface could induce increasing the (100) surface domains of two-dimensional long range order (2D-(100)), and decreasing the (100) surface domains of one-dimensional short range order (1D-(100)). It also revealed that the interaction of citrate with Pt(100) surface could stabilize the 2D-(100) structure. | 20 | Pan-cancer analyses demonstrate that ANKRD6 is associated with a poor prognosis and correlates with M2 macrophage infiltration in colon cancer显示文摘Objective:Ankyrin repeat domain-containing protein 6(ANKRD6)is an ankyrin repeat-containing protein which is structurally related to vertebrate inversin and Drosophila Diego.However,the correlations between ANKRD6 and tumor-infiltrating immune cells in cancers is not clear.Methods:ANKRD6 expression was analyzed by Oncomine,Tumor Immune Estimation Resource(TIMER)and Gene Expression Profiling Interactive Analysis(GEPIA).PrognoScan and GEPIA were used to evaluate the influence of ANKRD6 on clinical prognosis.TIMER and CIBERSORT were used to analyze correlations between ANKRD6 expression levels and tumor immune cell infiltrates.Immunohistochemical analysis of the relationship between ANKRD6 expression and overall survival,as well as the relationship between ANKRD6 expression and M2 macrophage infiltration,was performed.Results:High level of ANKRD6 expression was associated with poor prognosis of colon cancer.ANKRD6 expression level was positively correlated with infiltrating levels of CD8+T cells,CD4+T cells,macrophages,neutrophils and dendritic cells in colon cancer by using TIMER.Using CIBERSORT,we found that in plasma cells,CD8+T cells,CD4+memory resting T cells,follicular helper T cells and activated natural killer cells were significantly lower in the ANKRD6-high group than in the ANKRD6-low group.M0 and M2 macrophages were significantly higher in the ANKRD6-high group than in the ANKRD6-low group.Immunohistochemistry confirmed that M2 macrophage infiltration in the ANKRD6-high group significantly increased.Conclusions:The high ANKRD6 expression is associated with poor prognosis of colon cancer.ANKRD6 expression is positively correlated with M2 macrophage infiltration in colon cancer. | Rui Bai Dehao Wu Zhong Shi Wangxiong Hu Juan Li Yuanyuan Chen Weiting Ge Ying Yuan Shu Zheng | 2021 | Chinese Journal of Cancer Research2021,33,1: | 1 |
