| 1 | Research progress on FASN and MGLL in the regulation of abnormal lipid metabolism and the relationship between tumor invasion and metastasis显示文摘Tumorigenesis involves metabolic reprogramming and abnormal lipid metabolism,which is manifested by increased endogenous fat mobilization,hypertriglyceridemia,and increased fatty acid synthesis.Fatty acid synthase(FASN)is a key enzyme for the de novo synthesis of fatty acids,and monoacylglycerol esterase(MGLL)is an important metabolic enzyme that converts triglycerides into free fatty acids.Both enzymes play an important role in lipid metabolism and are associated with tumor-related signaling pathways,the most common of which is the PI3K–AKT signaling pathway.They can also regulate the immune microenvironment,participate in epithelial–mesenchymal transition,and then regulate tumor invasion and metastasis.Current literature have shown that these two genes are abnormally expressed in many types of tumors and are highly correlated with tumor migration and invasion.This article introduces the structures and functions of FASN and MGLL,their relationship with abnormal lipid metabolism,and the mechanism of the regulation of tumor invasion and metastasis and reviews the research progress of the relationship of FASN and MGLL with tumor invasion and metastasis. | Jingyue Zhang Yawen Song Qianqian Shi Li Fu | 2021 | Frontiers of Medicine2021,15,5: | 10 |
| 2 | Mutation profile and its correlation with clinicopathology in Chinese hepatocellular carcinoma patients显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most common causes of cancer worldwide.Although many studies have focused on oncogene characteristics,the genomic landscape of Chinese HCC patients has not been fully clarified.Methods:A total of 165 HCC patients,including 146 males and 19 females,were enrolled.The median age was 55 years(range,27-78 years).Corresponding clinical and pathological information was collected for further analysis.A total of 168 tumor tissues from these patients were selected for next-generation sequencing(NGS)-based 450 panel gene sequencing.Genomic alterations including single nucleotide variations(SNV),short and long insertions and deletions(InDels),copy number variations,and gene rearrangements were analyzed.Tumor mutational burden(TMB)was measured by an algorithm developed in-house.The top quartile of HCC was classified as TMB high.Results:A total of 1,004 genomic alterations were detected from 258 genes in 168 HCC tissues.TMB values were identified in 160 HCC specimens,with a median TMB of 5.4 Muts/Mb(range,0-28.4 Muts/Mb)and a 75%TMB of 7.7 Muts/Mb.The most commonly mutated genes were TP53,TERT,CTNNB1,AXIN1,RB1,TSC2,CCND1,ARID1A,and FGF19.SNV was the most common mutation type and C:G>T:A and guanine transformation were the most common SNVs.Compared to wild-type patients,the proportion of Edmondson grade III-IV and microvascular invasion was significantly higher in TP53 mutated patients(P<0.05).The proportion of tumors invading the hepatic capsule was significantly higher in TERT mutated patients(P<0.05).The proportion of Edmondson grade I-II,alpha fetoprotein(AFP)<25μmg/L,and those without a history of hepatitis B was significantly higher in CTNNB1 mutated patients(P<0.05).CTNNB1 mutations were associated with TMB high in HCC patients(P<0.05).Based on correlation analysis,the mutation of TP53 was independently correlated with microvascular invasion(P=0.002,OR=3.096)and Edmondson grade III-IV(P=0.008,OR=2.613).The mutation of TERT was independently correlated with tumor invasion of the liver capsule(P=0.001,OR=3.030),and the mutation of CTNNB1 was independently correlated with AFP(<25μmg/L)(P=0.009,OR=3.414).Conclusions:The most frequently mutated genes of HCC patients in China were TP53,TERT,and CTNNB1,which mainly lead to the occurrence and development of HCC by regulating the P53 pathway,Wnt pathway,and telomere repair pathway.There were more patients with microvascular invasion and Edmondson III-IV grade in TP53 mutated patients and more patients with hepatic capsule invasion in TERT mutated patients,while in CTNNB1 mutated patients,there were more patients with Edmondson I-II grade,AFP<25μmg/L,and a non-hepatitis B background.Also,the TMB values were significantly higher in CTNNB1 mutated patients than in wild type patients. | Shuo Wang Huasheng Shi Tao Liu Manjiang Li Sanshun Zhou Xuan Qiu Zusen Wang Weiyu Hu Weidong Guo Xiaoqian Chen Honglin Guo Xiaoliang Shi Junping Shi Yunjin Zang Jingyu Cao Liqun Wu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,2: | 8 |
| 3 | Malicious Synchrophasor Detection Based on Highly Imbalanced Historical Operational Data显示文摘By maliciously manipulating the synchrophasors produced by phasor measurement units in power systems,cyber attackers can mislead the control center into taking wrong actions.From the viewpoint of machine learning,normal and malicious synchrophasors may exhibit different spatial distribution characteristics when mapped into a latent space.Hence,a malicious synchrophasor detector can be acquired by training a classification model with instances derived from historical operational synchrophasor data.However,malicious synchrophasors occur infrequently in practice.It is likely to incur a great deal of effort and may even introduce inevitable experience errors when extracting and labeling a sufficient number of malicious synchrophasors from historical operational data for training.For most existing detectors,if they are directly trained with highly imbalanced datasets,their performances may severely deteriorate.In this paper,a novel type of malicious synchrophasor detector is developed based on a combinatorial use of data rebalancing,Bagging-based ensemble learning,and the widely recognized eXtreme Gradient Boosting(XGBoost)classifier.Experiments show that although fewer malicious instances are provided,the proposed detector is still capable of detecting malicious synchrophasors. | Jingyu Wang Zhengwei Sun Bin Bao Dongyuan Shi | 2019 | CSEE Journal of Power and Energy Systems2019,5,1: | 7 |
| 4 | Turbidite and bottom-current evolution revealed by anisotropy of magnetic susceptibility of redox sediments in the Ulleung Basin,Sea of Japan显示文摘Anisotropy of magnetic susceptibility(AMS) from above and below 7.31 m in a core from the southern Ulleung Basin shows clear differences on an equal area projection of the lower hemisphere.Rather concentrated steep inclination of K 3 and horizontal inclination of K 1 and K 2(K1 ≥K2 ≥K3) are located within the upper part,and the latter two axes lie perpendicular to each other near the bedding plane.In contrast,random distribution of the three axes and extremely high value of shape parameters(Q=(K1-K2)/[(K1+K2)/2-K3]) are evident in the lower part,indicating complete destruction of the original sedimentary structure.This result is consistent with data from X-radiographs,which show numerous conglomerates,distortions and cleavages in the lower part.According to age models by Liu et al.(2010) and Zou et al.(2010),the bottom age at 7.31 m is 48 cal ka BP,and the time domain is discussed below.The degree of AMS(P) is low,1-1.08,and linearly related to the foliation(F)(R2 =0.95,N=176).The relationship between F and linearity(L) implies oblate aligning patterns that are typically sedimentary in origin.At least five redox couplets were found with the aid of S ratios and other rock magnetic parameters,and in most cases,the original signals of climate survived early diagenesis.The paleomagnetically reoriented AMS show corresponding changes with millennial events in the last 48 cal ka.Clearly tilted K 3 directions and reduction of P and F occurred within DO1-BA warm events,when melt water pulse 1A nd the YD cold event took place.The turbulent conditions therein,synchronous with coarsening of sediments,provide evidence of strong bottom currents and possible directional changes,as evidenced by different K3 tilting directions.A NE current direction in the last 4.5 cal ka is consistent with in situ measurements of bottom currents.Responses of AMS,mainly to climatic modulation,show on the one hand,the limited influence of diagenesis on rock magnetic signals and,on the other hand,the dramatic change of hydrodynamic conditions and terrigenous inputs during rapid sea level rise during the last deglaciation. | GE ShuLan SHI XueFa LIU YanGuang WANG KunShan ZOU JianJun DIAO JingYu ZHU ZhiWei WANG ChunJuan | 2012 | Chinese Science Bulletin2012,57,6: | 7 |
| 5 | An approximate nonlinear modified Mohr-Coulomb shear strength criterion with critical state for intact rocks显示文摘In this paper, the Mohr-Coulomb shear strength criterion is modified by mobilising the cohesion and internal friction angle with normal stress, in order to capture the nonlinearity and critical state concept for intact rocks reported in the literature. The mathematical expression for the strength is the same as the classical form, but the terms of cohesion and internal friction angle depend on the normal stress now,leading to a nonlinear relationship between the strength and normal stress. It covers both the tension and compression regions with different expressions for cohesion and internal friction angle. The strengths from the two regions join continuously at the transition of zero normal stress. The part in the compression region approximately satisfies the conditions of critical state, where the maximum shear strength is reached. Due to the nonlinearity, the classical simple relationship between the parameters of cohesion, internal friction angle and uniaxial compressive strength from the linear Mohr-Coulomb criterion does not hold anymore. The equation for determining one of the three parameters in terms of the other two is supplied. This equation is nonlinear and thus a nonlinear equation solver is needed. For simplicity, the classical linear relationship is used as a local approximation. The approximate modified Mohr-Coulomb criterion has been implemented in a fracture mechanics based numerical code FRACOD,and an example case of deep tunnel failure is presented to demonstrate the difference between the original and modified Mohr-Coulomb criteria. It is shown that the nonlinear modified Mohr-Coulomb criterion predicts somewhat deeper and more intensive fracturing regions in the surrounding rock mass than the original linear Mohr-Coulomb criterion. A more comprehensive piecewise nonlinear shear strength criterion is also included in Appendix B for those readers who are interested. | 6 | Effectiveness of ozonated saline in the treatment of VX2 tumors in rabbits显示文摘Objective To investigate the efficacy, safety, and associated mechanisms of injected ozonated saline in the treatment of VX2 tumors. Methods A total of 90 rabbits bearing VX2 tumors on their left hind legs were randomly divided into three groups. The control group(A) received normal saline, while groups B and C received 20 μg/mL and 40 μg/mL O_3/O_2 ozonated saline, respectively. Rabbits were anesthetized and 2 mL of blood was drawn directly from the heart to measure serum concentrations of interleukin(IL-6) and tumor necrosis factor(TNF-α). The skin covering the VX2 tumor was cut in each rabbit and the maximum and vertical diameters of the tumors were measured under direct visualization. Several milliliters of saline, saline pre-treated with 20 μg/mL O_3/O_2, or saline pre-treated with 40 μg/mL O_3/O_2 were directly injected into the tumors of groups A, B, and C, respectively(injection volume(milliliter) =1/2 volume of the tumor, V = 1/2ab^2). On days 4, 8 and 12 following treatment, 10 rabbits were randomly selected from each group for blood sample collection, and serum IL-6 and TNF-α were measured. The tumor growth rate was calculated by measuring the maximum and vertical diameters of the VX2 tumors under direct visualization. All selected rabbits were euthanized and the tumors, livers, and lungs were removed for pathological examination. The tumor necrosis rate was calculated by cutting the tumors into half along the longitudinal axis and measuring the maximum diameters of the intratumoral necrotic regions. Results The average tumor volume in the three groups increased to different degrees at each time point; however, the average tumor growth rates in groups B and C were substantially lower than that in group A, exhibiting a statistically significant difference. The difference in the tumor growth rate between group B and group C was not statistically significant. The serum concentrations of IL-6 and TNF-α increased in the three groups at each time point, with larger increases occurring in groups B and C; however, the greater increases did not reach statistical significance. Although the diameters of the necrotic areas were larger in both groups B and C than that in group A, significant differences in necrotic area diameters were only found when comparing groups A and C on days 4 and 12 following treatment. Conclusion Direct injection of different concentrations of ozonated saline into VX2 tumors significantly increased intratumoral necrosis and reduced the tumor growth rate. | 8 | Integrative Analysis Reveals Enhanced Regulatory Effects of Human Long Intergenic Non-Coding RNAs in Lung Adenocarcinoma显示文摘Although there is an accumulating appreciation of the key roles that long intergenic non-coding RNAs(linc RNAs) play in diverse cellular processes, our knowledge of how linc RNAs function in cancer remains sparse. Here, we present a comprehensive landscape of RNA-seq transcriptome profiles of lung adenocarcinomas and their paired normal counterparts to unravel gene regulation rules of linc RNAs.Consistent with previous findings of co-expression between neighboring protein-coding genes, linc RNAs were typically co-expressed with their neighboring genes, which was found in both cancerous and normal tissues. By building a mathematical model based on correlated gene expression, we distinguished an additional subset of linc RNAs termed 'regulatory linc RNAs', representing their dominant roles in gene regulation. The number of regulatory linc RNAs was significantly higher in cancerous compared to normal tissues, and most of them positively regulated protein-coding genes in trans. Functional validation, using knockdown, determined that regulatory linc RNA,GAS5, affected its predicted protein-coding targets. Moreover, we discovered hundreds of differentially expressed regulatory linc RNAs with inclusion of some cancer-associated linc RNAs.
