| 3 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas. | Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. | 4 | Modeling experimental study on weathering-leaching of Emeishan basalt and its relation with metallogenesis显示文摘The Emeishan basalt has a high Cu background value (170×10-6 on average), and thus provides a ore-forming material source for copper ores. The collected samples were exposed in basalt lavas of the third segment of the Emeishan basalt eruptive cycle. By using a set of automatically cycling glass apparatuses, weathering-leaching experiments by three kinds of rainwater on the collected samples were carried out in the open system (at normal temperature and normal pressure). The analysis results showed that the most intensive export of Cu occurred under acidic rainwater-induced weathering-leaching conditions, almost 2-3 orders of magnitude the export of Cu by mod-ern air rainwater, and 1-2 orders of magnitude higher than the export of Cu by CO2 rainwater. In addition, the total amount of Zn, Cu, U, Ni and Sr exported by acidic rainwater are greater than 1%. | 6 | Effectiveness and safety of Shenmai injection in treatment of shock:a Meta-analysis显示文摘OBJECTIVE:To evaluate the effectiveness and safety of Shenmai injection for shock.METHODS: Randomized controlled trials(RCTs)that evaluated the therapeutic effect of Shenmai injection on shock(including septic shock, cardiogenic shock, hypovolemic shock, neurogenic shock and anaphylactic shock) were included in this analysis. The major electronic databases were searched until May 2015. The methodological quality of the trials was assessed according to the Cochrane Handbook. Review Manager 5.3 and Stata 12.0 software were applied for data analysis.RESULTS: Thirty RCTs involving 2038 participants were included. The methodological quality of the trials was generally passable. The combined use of Shenmai injection and conventional medicine was significantly more effective at managing shock compared to conventional medicine alone in the outcomes of total effective rate [risk ratio(RR 1.25,95% confidence interval(CI) 1.18 to 1.31] and mortality rate [risk difference(RD)-0.10, 95% CI-0.17 to-0.02]. Likewise, improvements were observed in other metrics. Three trials reported adverse events, but no trial reported serious adverse effects.CONCLUSION: Our results indicated the potential effectiveness of Shenmai injection combined with conventional medicine treatment for shock. | 9 | Dual-axial self-attention network for text classification显示文摘Text classification is an important task in natural language processing and numerous studies aim to improve the accuracy and efficiency of text classification models. In this study, we propose an effective and efficient text classification model which is based on self-attention solely. The recently proposed multidimensional self-attention significantly improved the performance of self-attention. However, existing models suffer from two major limitations:(1) the previous multi-dimensional self-attention models are quite timeconsuming;(2) the dependencies of elements along the feature axis are not taken into account. To overcome these problems, in this paper, a much more computational efficient multi-dimensional self-attention model is proposed, and two parallel self-attention modules, called dual-axial self-attention, are applied to capture rich dependencies along the feature axis as well as the text axis. | 16 | Simulation on solidification of an Al-Ni alloy under electromagnetic stirring显示文摘The microstructure of Al-Ni alloy has a significant influence on its performance, while electromagnetic stirring is one of the most effective methods for control of solidification structure of Al alloy. To investigate the effect of electromagnetic stirring on the solidification of the ingot, the solidification of the Al-50Ni alloy in vacuum with electromagnetic stirring was described by numerical simulation in this paper; and a three dimensional mathematical model was established. The electromagnetic field was simulated by ANSYS software and the thermal-flow field was simulated by FLUENT software. The coupling between the electromagnetic field and the thermal-flow field was implemented by user-defined subroutines. It is found that the current intensity has significant influences on the fluid flow and the microstructure of the alloy. | 17 | Effects of Exogenous Manganese (Mn) on Mineral Elements, Polyamines and Antioxidants in Apple Rootstock Malus robusta Rehd.显示文摘Manganese(Mn)is one of the essential microelements in all organisms.However,high level of Mn is deleterious to plants.In this study,the effects of exogenous manganese application on mineral element,polyamine(PA)and antioxidant accumulation,as well as polyamine metabolic and antioxidant enzyme activities,were investigated in Malus robusta Rehd.,a widely grown apple rootstock.High level of Mn treatments decreased endogenous Mg,Na,K and Ca contents,but increased Zn content,in a Mn-concentration-dependent manner.Polyamine metabolic assays revealed that,except the content of perchloric acid insoluble bound(PIS-bound)spermine,which increased significantly,the contents of putrescine(Put),spermidine(Spd)and spermine(Spm)all decreased progressively,accompanied with the decreased activities of arginine decarboxylase(ADC,EC 4.1.1.19)and ornithine decarboxylase(ODC,EC 4.1.1.17),and the increased activities of diamine oxidase(DAO,EC 1.4.3.6)and polyamine oxidase(PAO,EC 1.5.3.3).Further antioxidant capacity analyses demonstrated that contents of anthocyanin,non-protein thiols(NPT)and soluble sugar,and the activities of guaiacol peroxidase(POD,EC 1.11.1.7),catalase(CAT,EC 1.11.1.6)and superoxide dismutase(SOD,EC 1.15.1.1),also increased upon different concentrations of Mn treatments.Our results suggest that endogenous ion homeostasis is affected by high level of Mn application,and polyamine and antioxidant metabolism is involved in the responses of M.robusta Rehd.plants to high level of Mn stress. | Dazhuang Qi Meixia Liang Fudong Jiang Jianzhao Li Xuqiang Qiao Hongxia Zhang | 2020 | Phyton-International Journal of Experimental Botany2020,89,4: | 1 |
