| 1 | Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. | 2 | Genotypic Characterization of Methicillin-resistant Staphylococcus aureus Isolated from Pigs and Retail Foods in China显示文摘Objective To investigate the genotypic diversity of Methicillin-resistant Staphylococcus aureus(MRSA) isolated from pigs and retail foods from different geographical areas in China and further to study the routes and rates of transmission of this pathogen from animals to food. Methods Seventy-one MRSA isolates were obtained from pigs and retail foods and then characterized by multi-locus sequencing typing(MLST), spa typing, multiple-locus variable number of tandem repeat analysis(MLVA), pulsed-field gel electrophoresis(PFGE), and antimicrobial susceptibility testing. Results All isolated MRSA exhibited multi-drug resistance(MDR). Greater diversity was found in food-associated MRSA(7 STs, 8 spa types, and 10 MLVA patterns) compared to pig-associated MRSA(3 STs, 1 spa type, and 6 MLVA patterns). PFGE patterns were more diverse for pig-associated MRSA than those of food-associated isolates(40 vs. 11 pulse types). Among the pig-associated isolates, CC9-ST9-t899-MC2236 was the most prevalent clone(96.4%), and CC9-ST9-t437-MC621(20.0%) was the predominant clone among the food-associated isolates. The CC9-ST9 isolates showed significantly higher antimicrobial resistance than other clones. Interestingly, CC398-ST398-t034 clone was identified from both pig-and food-associated isolates. Of note, some community-and hospital-associated MRSA strains(t030, t172, t1244, and t4549) were also identified as food-associated isolates. Conclusion CC9-ST9-t899-MC2236-MDR was the most predominant clone in pigs, but significant genetic diversity was observed in food-associated MRSA. | 3 | Molecular organic geochemical peculiarities of lacustrine core sediments in Fildes Peninsula, King George Island, Antarctica显示文摘The peculiarities of lipids in lacustrine core sediments from Fildes Peninsula, King George Island, Antarctica, are reported. The origin of high-molecular-weight n-alkanes, alkenes, fatty acids, alcohols is supposed to be related to the abundant moss occurring in the sediments. Abundant iso- and ante/so-fatty acids that occur in the sediments indicate a relatively strong bacterial activity in non-ice-cover zone than in other regions in Antarctica. The C18:2 unsaturated fatty acids mainly originate from autochthonous algae in the sediment pool, and the C18:2/C18:0 ratios in the profile are controlled more by paleotemperatures than source assemblies. Long-chain alkenones were identified in freshwater lacustrine sediment in Antarctica for the first time, of which Prymnesiophyceae is assumed to be the source organism. | 5 | High-voltage Ride Through Strategy for DFIG Considering Converter Blocking of HVDC System显示文摘This paper presents a P-Q coordination based highvoltage ride through(HVRT) control strategy for doubly fed induction generators(DFIGs) based on a combined Q-V control and P-V de-loading control. The active/reactive power injection effect of DFIG on transient overvoltage is firstly analyzed and the reactive power capacity evaluation of DFIG considering its de-loading operation is then conducted. In the proposed strategy, the reactive power limit of DFIG can be flexibly extended during the transient process in coordination with its active power adjustment. As a result, the transient overvoltage caused by DC bipolar block can be effectively suppressed. Moreover, key outer loop parameters such as Q-V control coefficient and deloading coefficient can be determined based on the voltage level of point of common coupling(PCC) and the available power capacity of DFIG. | 6 | Blood Pressure Associated with Arsenic Methylation and Arsenic Metabolism Caused by Chronic Exposure to Arsenic in Tube Well Water显示文摘Objective The effects of arsenic exposure from drinking water, arsenic metabolism, and arsenic methylation on blood pressure(BP) were observed in this study. Methods The BP and arsenic species of 560 participants were determined. Logistic regression analysis was applied to estimate the odds ratios of BP associated with arsenic metabolites and arsenic methylation capability. Results BP was positively associated with cumulative arsenic exposure(CAE). Subjects with abnormal diastolic blood pressure(DBP), systolic blood pressure(SBP), and pulse pressure(PP) usually had higher urinary i As(inorganic arsenic), MMA(monomethylated arsenic), DMA(dimethylated arsenic), and TAs(total arsenic) than subjects with normal DBP, SBP, and PP. The i As%, MMA%, and DMA% differed slightly between subjects with abnormal BP and those with normal BP. The PMI and SMI were slightly higher in subjects with abnormal PP than in those with normal PP. Conclusion Our findings suggest that higher CAE may elevate BP. Males may have a higher risk of abnormal DBP, whereas females have a higher risk of abnormal SBP and PP. Higher urinary i As may increase the risk of abnormal BP. Lower PMI may elevate the BP. | 8 | Involvement of the Mitochondrion-dependent and the Endoplasmic Reticulum Stress-signaling Pathways in Isoliquiritigenin-induced Apoptosis of HeLa Cell显示文摘Objective Isoliquiritigenin(ISL),a licorice chalconoid,is considered to be a bioactive agent with chemopreventive potential.This study investigates the mechanisms involved in ISL-induced apoptosis in human cervical carcinoma HeLa cells.Methods Cell viability was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)assay.Apoptosis was determined by flow cytometry using an Annexin V-FITC Apoptosis Detection Kit.The intracellular ROS levels were assessed using a 2,7-dichlorofluorescein probe assay.The mitochondrial membrane potential was measured with the dual-emission potential-sensitive probe 5,5',6,6'-tetra-chloro-1,1',3,3'-tetraethyl-imidacarbocyanine iodide(JC-1).The degradation of poly-ADP-ribose polymerase(PARP)protein,the phosphorylation of PKR-like ER kinase(PERK),the phosphorylation of the α-subunit of eukaryotic initiation factor 2(eIF2α),the expression of the 78 kD glucose-regulated protein(GRP 78),and the activation of caspase-12 were analyzed via western blot analysis.Results ISL significantly inhibited the proliferation,the increase in ROS levels and apoptotic rates of HeLa cells in a concentration-dependent manner.Moreover,ISL induced mitochondrial dysfunction,caspase activation,and PARP cleavage,which displayed features of mitochondria dependent on apoptotic signals.Besides,exposure of HeLa cells to ISL triggered endoplasmic reticulum(ER)stress,as indicated by the increase in p-eIF2α and GRP78 expression,ER stress-dependent apoptosis is caused by the activation of ER-specific caspase-12.Conclusion The findings from our study suggest that ISL-induced oxidative stress causes HeLa cell apoptosis via the mitochondrion-dependent and the ER stress-triggered signaling pathways. | YUAN Xuan ZHANG Bo GAN Lu WANG Zhen Hua YU Ba Cui LIU Liang Liang ZHENG Qiu Sheng WANGZhi Ping | 2013 | Biomedical and Environmental Sciences2013,26,4: | 8 |

| 10 | Toxicity of Graphene Quantum Dots in Zebrafish Embryo显示文摘Objective To evaluate the bio-safety of graphene quantum dots(GQDs), we studied its effects on the embryonic development of zebrafish. Methods In vivo, biodistribution and the developmental toxicity of GQDs were investigated in embryonic zebrafish at exposure concentrations ranging from 12.5-200 μg/m L for 4-96 h post-fertilization(hpf). The mortality, hatch rate, malformation, heart rate, GQDs uptake, spontaneous movement, and larval behavior were examined. Results The fluorescence of GQDs was mainly localized in the intestines and heart. As the exposure concentration increased, the hatch and heart rate decreased, accompanied by an increase in mortality. Exposure to a high level of GQDs(200 μg/mL) resulted in various embryonic malformations including pericardial edema, vitelline cyst, bent spine, and bent tail. The spontaneous movement significantly decreased after exposure to GQDs at concentrations of 50, 100, and 200 μg/mL. The larval behavior testing(visible light test) showed that the total swimming distance and speed decreased dose-dependently. Embryos exposed to 12.5 μg/mL showed hyperactivity while exposure to higher concentrations(25, 50, 100, and 200 μg/mL) caused remarkable hypoactivity in the light-dark test. Conclusion Low concentrations of GQDs were relatively non-toxic.
