2Puerarin reduces increased c-fos, c-jun, and type Ⅳ collagen expression caused by high glucose in glomerular mesangial cells显示文摘Aim: Increased expression of c-fos, c-jun and type Ⅳ collagen (CoⅣ) in glomerular mesangial cells (GMC) are important characteristics of diabetic nephropathy. Both c-los and c-jun regulate the gene expression of extracellular matrix components, and CoⅣ is the main component of the extracellular matrix. It has been reported that puerarin inhibits aggregation of the extracellular matrix in diabetic rats by an as yet unknown mechanism. The aim of this study is to investigate the effect of puerarin on c-fos, c-jun and CoⅣ expression in GMC cultured in medium containing 5.6 or 27.8 mmol/L glucose. Methods: The expressions of c-fos and c-jun were measured at the protein level using flow cytometry. CoⅣ content was detected using radioimmunoassay. Protein kinase C (PKC) activity was measured using liquid scintillation counting. Results: Puerarin (10^(-5) mmol/L) significantly ameliorated the high-glucose effect on c-fos, c-jun and CoⅣ expression. This effect is accompanied by a reduced PKC activity in these cells. Coclusion: Our results suggest that reduced PKC activity and expression of c-fos and c-jun in GMC might participate in the mechanisms underlying the therapeutic effect of puerarin on diabetic nephropathy.Cai-ping MAO Zhen-lun GU 2005Acta Pharmacologica Sinica2005,26,8:19

4From intrinsic dielectric loss to geometry patterns: Dual-principles strategy for ultrabroad band microwave absorption显示文摘As electromagnetic absorbers with wide absorption bandwidth are highly pursued in the cutting-edge electronic and telecommunication industries, the traditional dielectric or magnetic bulky absorbers remain concerns of extending the effective absorption bandwidth. In this work, a dual-principle strategy has been proposed to make a better understanding of the impact of utilizing conductive absorption fillers coupled with implementing artificial structures design on the absorption performance. In the comparison based on the microscopic studies, the carbon nanotubes (CNTs)-based absorbers are confined to narrow operating bandwidth and relatively fixed response frequency range, which can not fulfill the ever-growing demands in the application. With subsequent macroscopic structure design based on the CNTs-based dielectric fillers, the artificial patterns show much more broadened absorption bandwidth, covering the majority of C-band, the whole X-band, and Ku-band, due to the tailored electromagnetic parameters and more reflections and scatterings. 6Electroacupuncture regulates glucose-inhibited neurons in treatment of simple obesity显示文摘The glucose-inhibited neurons present in the lateral hypothalamic area are regarded as glucose detectors. This structure is involved in the regulation of food intake through extracellular blood glucose concentrations, and plays a crucial role in obesity onset. In the present study, obesity models established with high fat feeding were treated with electroacupuncture at Zusanli (ST36)/ Inner Court (ST44) on the left side and Tianshu (ST25) bilaterally. We found that electroacupuncture could effectively reduce body weight and the fat-weight ratio, and decrease serum leptin, resistin, tumor necrosis factor alpha, and neuropeptide Y levels, while increase serum adiponectin and cholecystokinin-8 levels. This treatment altered the electrical activity of glucose-inhibited neurons in the lateral hypothalamic area, with electroacupuncture at Zusanli/ Inner Court exerting an inhibitory effect, while electroacupuncture at bilateral Tianshu exerting an excitatory effect. 7Effects of tripchlorolide on the epididymides and testes of rats显示文摘Aim: To further evaluate the antifertility effects of tripchlorolide, a derivative of triptolide produced at the extraction procedure of Tripterygium wilfordii Hook. f., in male rats and to investigate its sites and possible mechanisms of action.Methods: In male rats, tripchlorolidte was given by oral garage at a dose of 50 ug'kg^-1, d^-1 for 5 weeks, fertility wasassessed by mating tests, and biochemical indices and light microscopic obsefvation of the epididymides and testes werealso performed. Results: Administration of tripchlorolide at 50 ,ug, kg ^-1, d^-1 for 3 weeks did not influence the fertilityof male rats, but 5-week treatment rendered the rats infertile. The density and motility of spennatozoa collected fromcanda epididymides were reduced significandy. The epididymal weights, as well as the L-camitine concentration and a-glucosidase content in the epididymal fluid were decreasd. There were no significant differences in a-glucosidase andacid phosphatase (ACP) in caput epididyma/homogenatas between the control and the experimental rats. Obvioos mor-phological changes were observed in the epididymal spematozoa, mainly including head and tail separation or acrosomecurving, Sloughed speprmatids were found in the semirfifeous and epididymal tubules. In testicular homogenates,tripchlorolide had no influence on the lactate dehydrogenase-C4(LDH-C4) and hyaluronidase activities. No apparentlesions were observed in the seminiferous and epididymal epithelium. 9Sequence analysis of the gene correlated with cytoplasmic male sterility (CMS) in rape-seed (Brassica napus) Polima and Shaan 2A显示文摘orf224 is a CMS-related mitochondrial gene discovered in Polima cytoplasm. Shaan 2A CMS line is the parent of the first rapeseed hybrid cultivar Qinyou No. 2 that has been grown in many regions of China. In this work, genomic DNA of Polima CMS line and Shaan 2A CMS line were used as templates, two primers of specific oligonucleo-tides at 5’ and 3’ ends were used, PCR was performed, the amplification fragments were cloned into pGEM-T Easy vectors and DNA sequences were determined. The CMS-associated gene, orf224-1 present in Shaan 2A CMS line, has a sequence highly homologous to the orf224 of the Polima CMS line, except for one nucleotidc at position +398. There were only one base (AAC→AGC) and one amino acid (Asn→Ser) differences between the two. The homologies of the two sequences in nucleotide and amino acid were 99.9% and 99.6%, respectively. 10The promoter analysis of the human C17orf25 gene, a novel chromosome 17pl3.3 gene显示文摘The human C17orf25 gene (Accession No. AF177342) is one of thirteen genes cloned from a region displaying a high score of loss of heterozygosity within chromosome 17p13.3 in human hepatocellular car-cinoma in China[l]. To unveil the underlying mechanisms for the transcription regulation of this gene and understand its implication to the hepatocellular carcinogenesis, we looked into the relevant aspects by both bioinformatic and experimental executions. We found: 1, The abundant expression of the C17orf25 gene was evident in all the cell lines and tissue samples tested, showing little hepatoma-selectivity; 2, Its tran-scription starts at a single site, locating at -60 from the translation initiation codon; 3, A 58 bp fragment containing the transcription start, extending from -112 to -55, represents the minimal promoter; 4, The consensus sequence within this fragment recognized by SP1 contributes predominantly to the activity of the minimal promoter; 5, The bioinformatic analysis suggests that the C17ort25 gene may encode a protein in the family of the glyoxalase. 11Effects of tallowtree seed coat on seed germination显示文摘We measured physiological parameters including water uptake, in-vitro embryo germination ratio, and seed coat structure observed by scanning electron microscopy (SEM) to explore the influence of seed coat on the germination of seeds of tallow tree (Sapium sebiferum (Linn) Roxb.). Tallow tree seeds had good water permeability. We found that germination of cabbage seeds was inhibited when cabbage seeds were soaked in extracted solutions from tallow tree seed coat. Seed coat structure at the side of the radicle appeared to be a barrier to seed germination. We tested methods to break tallow tree seed dormancy. 12Activity of HDV ribozymes to trans-cleave HCV RNA显示文摘AIM: To explore whether HDV ribozymes have the abilityto trans-cleave HCVRNA.METHODS: Three HDV genomic ribozymes weredesigned and named RzC1, RzC2 and RzC3. Thesubstrate RNA contained HCVRNA 5'-noncoding regionand 5'-fragment of C region (5'-NCR-C). All theribozymes and HCV RNA 5'-NCR-C were obtained bytranscription in vibo from their DNA templates, and HCVRNA 5'-NCR-C was radiolabelled at its 5'-end Undercertain pH, temperature, appropriate concentration ofMg2+ and deionized formamide, these ribozymes wererespectively or simultaneously mixed with HCVRNA 5'-NCR-C and reacted for a certain time. The trans-cleavage reaction was stopped at different time points,and the products were separated with polyacrylamidegel electrophoresis (PAGE), displayed byautoradiography. Percentage of trans-deaved productswas measured to indicate the activity of HDV ribozymes.RESULTS: RzC1 and RzC2 could trans-cleave 26 % and21.8 % of HCV RNA 5'-NCR-C under our reactionconditions with 2.5 mol. L-1 deionized formamiderespectively. The percentage of HCV RNA 5'-NCR-Ctrans-cleaved by RzC1, RzC2 or combined usage of thethree ribozymes increased with time, up to 24.9 %, 20.3 %and 37.3 % respectively at 90 min point. 13Genetic diversity of pepper(Capsicum spp.) germplasm resources in China reflects selection for cultivar types and spatial distribution显示文摘Pepper(Capsicum spp.) is an important vegetable crop in the world. Now the pepper in China contributes one-third of the world's peppers production. Genetic diversity of the pepper germplasm of China is expected interesting to know. To explore the structure of genetic diversity in Chinese pepper germplasm resources and possible relationship with cultivar types or geographic origin, we sampled and compared 372 Gen Bank pepper accessions(local cultivars and landraces) from 31 provinces, autonomous regions and municipalities of China and 31 additional accessions from other countries. These accessions were genotyped using 28 simple sequence repeat(SSR) markers spanning the entire pepper genome. We then investigated the genetic structure of the sampled collection using model-based analysis in STRUCTURE v2.3.4 and examined genetic relationships by the unweighted pair-group method of mathematical averages(UPGMA) in MEGA. In addition to geographic origin, we evaluated eight plant and fruit traits. In total, 363 alleles were amplified using the 28 SSR primers. Gene diversity, polymorphism information content and heterozygosity of the 28 SSR loci were estimated as 0.09–0.92, 0.08–0.92 and 0.01–0.34, respectively. The UPGMA cluster analysis clearly distinguished Capsicum annuum L. from other cultivated pepper species. Population structure analysis of the 368 C. annuum accessions uncovered three genetic groups which also corresponded to distinct cultivar types with respect to the plant and fruit descriptors. The genetic structure was also related to the geographic origin of the landraces. 14Prediction of microvascular invasion in solitary hepatocellular carcinoma≤5 cm based on computed tomography radiomics显示文摘BACKGROUND Liver cancer is one of the most common malignant tumors,and ranks as the fourth leading cause of cancer death worldwide.Microvascular invasion(MVI)is considered one of the most important factors for recurrence and poor prognosis of liver cancer.Thus,accurately identifying MVI before surgery is of great importance in making treatment strategies and predicting the prognosis of patients with hepatocellular carcinoma(HCC).Radiomics as an emerging field,aims to utilize artificial intelligence software to develop methods that may contribute to cancer diagnosis,treatment improvement and evaluation,and better prediction.AIM To investigate the predictive value of computed tomography radiomics for MVI in solitary HCC≤5 cm.METHODS A total of 185 HCC patients,including 122 MVI negative and 63 MVI positive patients,were retrospectively analyzed.All patients were randomly assigned to the training group(n=124)and validation group(n=61).A total of 1351 radiomic features were extracted based on three-dimensional images.The diagnostic performance of the radiomics model was verified in the validation group,and the Delong test was applied to compare the radiomics and MVIrelated imaging features(two-trait predictor of venous invasion and radiogenomic invasion).RESULTS A total of ten radiomics features were finally obtained after screening 1531 features.According to the weighting coefficient that corresponded to the features,the radiomics score(RS)calculation formula was obtained,and the RS score of each patient was calculated.The radiomics model exhibited a better correction and identification ability in the training and validation groups[area under the curve:0.72(95%confidence interval:0.58-0.86)and 0.74(95%confidence interval:0.66-0.83),respectively].Its prediction performance was significantly higher than that of the image features(P<0.05).CONCLUSION Computed tomography radiomics has certain predictive value for MVI in solitary HCC≤5 cm,and the predictive ability is higher than that of image features.Peng Liu Xian-Zhen Tan Ting Zhang Qian-Biao Gu Xian-Hai Mao Yan-Chun Li Ya-Qiong He 2021World Journal of Gastroenterology2021,27,17:6
15Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 16Obstructive sleep apnea aggravates neuroinflammation and pyroptosis in early brain injury following subarachnoid hemorrhage via ASC/HIF-1α pathway显示文摘Obstructive sleep apnea can worsen the prognosis of subarachnoid hemorrhage.Howeve r,the underlying mechanism remains unclear.In this study,we established a mouse model of subarachnoid hemorrhage using the endovascular perforation method and exposed the mice to intermittent hypoxia for 8 hours daily for 2 consecutive days to simulate sleep apnea.We found that sleep apnea aggravated brain edema,increased hippocampal neuron apoptosis,and worsened neurological function in this mouse model of subarachnoid hemorrhage.Then,we established an in vitro HT-22 cell model of hemin-induced subarachnoid hemorrhage/intermittent hypoxia and found that the cells died,and lactate dehydrogenase release increased,after 48 hours.We further investigated the underlying mechanism and found that sleep apnea increased the expression of hippocampal neuroinflammatory factors interleukin-1β,interleukin-18,inte rleukin-6,nuclear factorκB,pyro ptosis-related protein caspase-1,pro-caspase-1,and NLRP3,promoted the prolife ration of astrocytes,and increased the expression of hypoxia-inducible factor 1αand apoptosis-associated speck-like protein containing a CARD,which are the key proteins in the hypoxia-inducible factor 1α/apoptosis-associated speck-like protein containing a CARD signaling pathway.We also found that knockdown of hypoxia-inducible factor 1αexpression in vitro greatly reduced the damage to HY22 cells.These findings suggest that sleep apnea aggravates early brain injury after subarachnoid hemorrhage by aggravating neuroinflammation and pyroptosis,at least in part through the hypoxia-inducible factor 1α/apoptosis-associated speck-like protein containing a CARD signaling pathway.Jun Xu Qian Li Chen-Yu Xu Shan Mao Jia-Jia Jin Wei Gu Ying Shi Chun-Fang Zou Liang Ye 2022Neural Regeneration Research2022,17,11:6
17Use of Organic Solvents to Extract Organochlorine Pesticides (OCPs) from Aged Contaminated Soils显示文摘Problems associated with organochlorine pesticide (OCP)-contaminated sites in China have received wide attention. To solve such problems, innovative ex-situ methods of site remediation are urgently needed. We investigated the feasibility of the extraction method with different organic solvents, ethanol, 1-propanol, and three fractions of petroleum ether, using a soil collected from Wujiang (WJ), China, a region with long-term contamination of dichlorodiphenyltrichloroethanes (DDTs). We evaluated different influential factors, including organic solvent concentration, washing time, mixing speed, solution-to-soil ratio, and washing temperature, on the removal of DDTs from the WJ soil. A set of relatively better parameters were selected for extraction with 100 mL L 1 petroleum ether (60-90℃): washing time of 180 min, mixing speed of 100 r min 1 , solution-to-soil ratio of 10:1, and washing temperature of 50℃. These selected parameters were also applied on three other seriously OCP-polluted soils. 20Genetic and Antibiotic Resistance Characteristics ofCampylobacterjejuni Isolated from Diarrheal Patients,Poultry and Cattle in Shenzhen显示文摘Objective To investigate genetic and antibiotic resistance characteristics of Campylobacter jejuni(C. jejuni) isolated from Shenzhen. Methods Multilocs sequence typing and agar dilution methods were used to define the genotype and antibiotic resistance of C. jejuni, respectively. Results In total, 126 C. jejuni strains were isolated. The prevalence of C. jejuni was 5.3% in diarrheal patients. The prevalence in poultry meat(36.5%) was higher than that in cattle meat(1.1%). However, the prevalence in poultry cloacal swabs(27.0%) was lower than that in cattle stool(57.3%). Sixty-two sequence types were obtained, among which 27 of the STs and 10 alleles were previously unreported. The most frequently observed clonal complexes were ST-21(11.9%), ST-22(10.3%), and ST-403(7.1%). ST-21, ST-45, ST-354, ST-403, and ST-443 complexes overlapped between isolates from patients and cattle, whereas ST-45 and ST-574 complexes overlapped between isolates from patients and poultry. All C. jejuni were resistant to at least one antibiotic. The highest resistance rate was toward ciprofloxacin(89.7%), followed by tetracycline(74.6%), and nalidixic acid(69.0%). Conclusion This is the first report of the genotypes and antibiotic resistance of C. jejuni in Shenzhen.
