1Human CMTM2/CKLFSF2 enhances the ligand-induced transactivation of the androgen receptor显示文摘CKLF (chemokine-like factor)-like MARVEL (MAL and related proteins for vesicle trafficking and mem-brane link domain) transmembrane domain containing (CMTM) is a novel gene family. One member of this family, CMTM2, also named chemokine-like factor superfamily 2 (CKLFSF2), is expressed highly in the testis and moderately in the prostate, marrow and peripheral blood cells. However, the function of human CMTM2 remains unknown. Here, we found that CMTM2 was upregulated in 5α-dihydrotestos- terone (DHT)-treated LNCaP cells. We investigated the relationship between CMTM2 and the androgen receptor. Our results showed that CMTM2 enhanced DHT-mediated androgen receptor (AR) transacti-vation and the expression of prostate specific antigen (PSA). We also observed that CMTM2 enhanced the AR protein level, which was reversed by silencing endogenous CMTM2 expression, which sug-gested that CMTM2 might play an important role in maintaining the AR protein level. We also found that CMTM2 suppressed Akt activation. A previous study showed that Akt could phosphorylate AR at Ser210 and Ser790 and lead to AR ubiquitylation and degradation as well as suppression of AR activity. 2Interaction of hTCF4 by yeast two-hybrid system显示文摘To investigate the interaction of hTCF4, the yeast two-hybrid system has been used for testing the interaction of mutants of hTCF4 with themselves. Mutants of hTCF4 (hTCF4 Ⅰ ) and hTCF4 Ⅱ) have been obtained by polymerase chain reaction (PCR). Bait (hTCF4 Ⅰ -pDBLeu and hTCF4Ⅱ-pDBLeu) and prey (hTCF4 Ⅰ -pPC86 and hTCF4Ⅱ-pPC86) have been constructed by DNA recombination for yeast two-hybrid. Interaction of hTCF4Ⅱ with itself is found in the reverse yeast two-hybrid system (GIBCOBRL Co.). However, no interactions are found in hTCF4Ⅱ with hTCF4 Ⅰ and in hTCF4 Ⅰ with hTCF4 Ⅰ . 3Aberrant activity of Wnt/Frizzled signaling pathway in renal cancer cell lines显示文摘The expression of Wnt, Wnt receptor-Frizzled, and several other key components in Wnt pathway in renal cancer cell lines was studied. The result of semi-quantitative RT-PCR has shown that the expression level of Wnt5A and hFz5 mRNA were higher in renal cancer cell line (GRC-1) than in normal renal cell line (HK-2). This result has been confirmed by in situ hybridization. The expression of p-catenin protein was obviously higher in GRC-1 than in HK-2 (P<0.01), but there were no different expressions of its mRNA between 3 lines. The reasons of the overexpression of β-catenin has been investigated by means of immunocytochemistry, SSCP and so on, no mutation of β-catenin gene and APC were found. That means that the overexpression of Wnt5A/hFz5 might be the reason of overexpression of β-catenin. 7Thoracic interstitial injection of drug-liposomes in mice for treating atherosclerosis显示文摘Intervaginal space injection(ISI)is a novel mode of administration investigated over the last decade.After injecting nanoparticles into the intervaginal space,they can be transported along low flow resistance channels into the interstitial space.This transport has a certain delivery direction,and site-specific injection can work on specific organs or tissues.In this study,the thorax,a new ISI site in the interstitial surrounding the internal thoracic artery named the thoracic interstitial injection(tISI)was investigated.To prove the targeting ability of the tISI,two sizes of gold nanoparticles(AuNPs)(47 and 87 nm)were administered to mice.After 1 h,the biodistribution of AuNPs in the tissues was measured via single particle inductively coupled plasma mass spectrometry(spICP-MS).The results showed that the concentration of AuNPs in the aorta after tISI injection was significantly higher than that after intravenous injection.Moreover,fewer nanoparticles with larger particle sizes were observed to have entered the blood and were better targeted to the aorta.Thereafter,tanshinone IIa sodium sulfonate liposomes were administered for the treatment of aortic atherosclerosis.The proportion of aortic plaques in atherosclerotic Apoe-/-mice administered via tISI was significantly lower than that in other model animals(P<0.001).Furthermore,the proteoglycan content and CD68-positive cell count in the plaques were significantly reduced.The vascular elastic fibers at the plaque site were thickened,and fractures were reduced.tISI was,therefore,determined to be an effective strategy for the treatment of atherosclerotic aortic plaques.Tun Yan Huizhen Sun Yahong Shi Ya Gao Xi Lu Kai Li Yuting Zhu Qiang Zhang Tingting Li Zhongxian Li Xiaohan Zhou Yuting Guo Yinglu Ji Xiaoli Shi Dong Han 2023Nano Research2023,16,4:0

9Census report on Chinese urological surgeons显示文摘Objective:To grasp the general situation of Chinese urological surgeons and the status quo of their scientific research,work and training,thus providing valuable recommendations for urological talent team construction in future.Methods:The survey respondentswere the urological surgeons,who held the Certificate ofMedical Practitioner in the People’s Republic of China,whose scope of practice was confined to urological surgery.The urological surgeons involved in the project completed an online questionnaire survey.All the data were collected through the internet.Results:There were a total of 18981 urological surgeons in China in 2015,of whom 15875 from 2602 hospitals participated in this project,with a mean age of 39.64 years old.In 2015,1949631 cases of surgery were performed,including 493723 cases of open surgery,1146444 cases of endoscopic/laparoscopic surgery(robot-assisted laparoscopic surgery were excluded),6259 robot-assisted surgery and other types of urological surgery.Besides,Chinese urological surgeons published 1358 monographs as well as 14558 academic papers,and also obtained 2064 scientific funds in 2015.A total of 92122 person-time participated in academic conferences.Urological surgeons with higher educational degrees as well as higher academic titles and from Eastern China or higher-level hospitals hadmore opportunities to participate in further education and training.Yinghao Sun Liqun Zhou Junhua Zhang Nianzeng Xing Qian Zhang Xiuzhong Hu Linhui Wang Yinglu Guo 2020Asian Journal of Urology2020,7,2:0
10A specific assay for JmjC domain-containing lysine demethylase and its application to inhibitor screening显示文摘Dear Editor, Chromatin is the macromolecular complex of DNA and histone proteins, which provides the scaffold for the packaging of our entire genome. In the basic unit of chromatin, the nucleosomes contain 147 base pairs of DNA, which is wrapped around a histone octamer, with two each of histones H2A, H2B, H3, and H4. Several types of post-translational modifications, including acetylation, phosphorylation, ubiquitination, and methylation, are possible on the tails of histones (Zhang et al., 2015). Combinations of these modifications determine the chromatin structure and transcriptional regulation of genes. Histone methylation is highlighted because of the specific dynamics related to gene regulation. Aberrant histone methylation has been observed in many types of tumors (Albert and Helin, 2010) and may contribute to the neoplastic transformation of cells (Chi et al., 2010). Histone lysine methylation is regulated by two classes of enzymes that have opposing activities: histone lysine methyltransferases (KMTs) and histone lysine demethylases (KDMs).
