1WHEAT STRIPE RUST AND INTEGRATION OF SUSTAINABLE CONTROL STRATEGIES IN CHINA显示文摘Stripe (yellow) rust caused by Puccinia striiformis f. sp. tritici occurs in almost allwheat-producing regions of the world. Severe countrywide epidemics in Chinahave caused substantial yield losses. Growing resistant cultivars is the beststrategy to control this disease but the pathogen can overcome resistance inwheat cultivars. The high variation in the virulence of the pathogen combinedwith the large areas of susceptible wheat cultivars enables the pathogenpopulation to increase rapidly and disperse over long distances under favorableenvironmental conditions, resulting in severe pandemics within croppingseasons. Current stripe rust control measures are based on many years ofresearch including the underlying epidemiology regarding year-to-year survivalof the pathogen, pathways of pathogen dispersal within seasons and years, therole of P. striiformis sexual hybridization, the use of resistance sources inbreeding programs, and year-round surveillance of national wheat crops that arepresent in different parts of the country throughout the year. All these strategiesdepend on accurate prediction of epidemics, more precise use of fungicides tomeet national requirements and better deployment of resistance genes. 2Genomic analysis,trajectory tracking,and field surveys reveal sources and long-distance dispersal routes of wheat stripe rust pathogen in China显示文摘Identifying sources of phytopathogen inoculum and determining their contributions to disease outbreaks are essential for predicting disease development and establishing control strategies.Puccinia striiformis f.sp.tritici(Pst),the causal agent of wheat stripe rust,is an airborne fungal pathogen with rapid virulence variation that threatens wheat production through its long-distance migration.Because of wide variation in geographic features,climatic conditions,and wheat production systems,Pst sources and related dispersal routes in China are largely unclear.In the present study,we performed genomic analyses of 154 Pst isolates from all major wheat-growing regions in China to determine Pst population structure and diversity.Through trajectory tracking,historical migration studies,genetic introgression analyses,and field surveys,we investigated Pst sources and their contributions to wheat stripe rust epidemics.We identified Longnan,the Himalayan region,and the Guizhou Plateau,which contain the highest population genetic diversities,as the Pst sources in China.Pst from Longnan disseminates mainly to eastern Liupan Mountain,the Sichuan Basin,and eastern Qinghai;that from the Himalayan region spreads mainly to the Sichuan Basin and eastern Qinghai;and that from the Guizhou Plateau migrates mainly to the Sichuan Basin and the Central Plain.These findings improve our current understanding of wheat stripe rust epidemics in China and emphasize the need for managing stripe rust on a national scale.Yuxiang Li Jichen Dai Taixue Zhang Baotong Wang Siyue Zhang Conghao Wang Jiguang Zhang Qiang Yao Mingju Li Chengyun Li Yuelin Peng Shiqin Cao Gangming Zhan Fei Tao Haifeng Gao Weili Huang Xiaojun Feng Yingwen Bai Zhuoma Qucuo Hongsheng Shang Chong Huang Wancai Liu Jiasui Zhan Xiangming Xu Xianming Chen Zhensheng Kang Xiaoping Hu 2023Plant Communications2023,4,4:1

4A novel ambigrammatic mycovirus,PsV5,works hand in glove with wheat stripe rust fungus to facilitate infection显示文摘Here we describe a novel narnavirus,Puccinia striiformis virus 5(PsV5),from the devastating wheat stripe rust fungus P.striiformis f.sp.tritici(Pst).The genome of PsV5 contains two predicted open reading frames(ORFs)that largely overlap on reverse strands:an RNA-dependent RNA polymerase(RdRp)and a reverseframe ORF(rORF)with unknown function.Protein translations of both ORFs were demonstrated by immune technology.Transgenic wheat lines overexpressing PsV5(RdRp-rORF),RdRp ORF,or rORF were more susceptible to Pst infection,whereas PsV5-RNA interference(RNAi)lines were more resistant.Overexpression of PsV5(RdRp-rORF),RdRp ORF,or rORF in Fusarium graminearum also boosted fungal virulence.We thus report a novel ambigrammatic mycovirus that promotes the virulence of its fungal host.The results are a significant addition to our understanding of virosphere diversity and offer insights for sustainable wheat rust disease control.Yanhui Zhang Xiaofei Liang Mengxin Zhao Tuo Qi Hualong Guo Jing Zhao Jie Zhao Gangming Zhan Zhensheng Kang Li Zheng 2023Plant Communications2023,4,3:1
5Determination of heterozygosity for avirulence/virulence loci through sexual hybridization of Puccinia striiformis f. sp. tritici显示文摘Wheat stripe rust caused by Puccinia striiformis f. sp. tritici is one of the most devastating diseases of wheat worldwide and resistant cultivars are vital for its management. Therefore, investigating the heterozygosity of the pathogen is important because of rapid virulence changes in isolates heterozygous for avirulence/virulence.An isolate of P. striiformis f. sp. tritici was selfed on Berberis shensiana to determine the heterozygosity for avirulence/virulence loci. One hundred and twenty progeny isolates obtained from this selfing were phenotyped using 25 lines of wheat containing Yrgenes and genotyped with 96 simple sequencing repeat markers, with51 pathotypes and 55 multi-locus genotypes being identified. All of these were avirulent on lines with Yr5,Yr10, Yr15, Yr24 and Yr26 and virulent on lines with Yr17,Yr25 and YrA, indicating that the parental isolate was homozygously avirulent or homozygously virulent for these loci. Segregation was found for wheat lines with Yr1,Yr2, Yr4, Yr6, Yr7, Yr8, Yr9, Yr27, Yr28, Yr32, Yr43, Yr44,YrExp2, YrSp, YrTr1, YrTye and YrV23.
