| 1 | Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. | 2 | WHEAT STRIPE RUST AND INTEGRATION OF SUSTAINABLE CONTROL STRATEGIES IN CHINA显示文摘Stripe (yellow) rust caused by Puccinia striiformis f. sp. tritici occurs in almost allwheat-producing regions of the world. Severe countrywide epidemics in Chinahave caused substantial yield losses. Growing resistant cultivars is the beststrategy to control this disease but the pathogen can overcome resistance inwheat cultivars. The high variation in the virulence of the pathogen combinedwith the large areas of susceptible wheat cultivars enables the pathogenpopulation to increase rapidly and disperse over long distances under favorableenvironmental conditions, resulting in severe pandemics within croppingseasons. Current stripe rust control measures are based on many years ofresearch including the underlying epidemiology regarding year-to-year survivalof the pathogen, pathways of pathogen dispersal within seasons and years, therole of P. striiformis sexual hybridization, the use of resistance sources inbreeding programs, and year-round surveillance of national wheat crops that arepresent in different parts of the country throughout the year. All these strategiesdepend on accurate prediction of epidemics, more precise use of fungicides tomeet national requirements and better deployment of resistance genes. | 3 | Protocol for analyzing protein liquid-liquid phase separation显示文摘Numerous cellular functions occur in spatially and temporally confined regions. Recent studies have shown that membrane-less organelles and compartments in the cell are assembled via liquid-liquid phase separation (LLPS). In vitro LLPS assays using recombinant expressed and purified proteins are necessary for us to further understand how the assembly of phase-separated compartments is regulated in cells. However;uniform standards and protocols are lacking for these in vitro studies. Here, we describe a step-by-step protocol commonly used to investigate in vitro LLPS using purified proteins. This protocol includes expression and purification of the studied proteins, induction of LLPS of the purified proteins, and studies of the biophysical properties of the liquid droplets formed by LLPS. | 4 | Influence of mild hypothermia on vascular endothelial growth factor and infarct volume in brain tissues after cerebral ischemia in rats显示文摘BACKGROUND: It has been demonstrated that mild hypothermia has obvious protective effect on both whole and local cerebral ischemia. However, the definite mechanism is still unclear for the brain protection of mild hypothermia on cerebral edema, inhibiting inflammatory reaction, stabilizing blood brain barrier, etc. OBJECTIVE: To investigate the effect of mild hypothermia on the expression of vascular endothelial growth factor and the infarct volume after cerebral ischemia in rats, and analyze the brain protective mechanism of mild hypothermia. DESIGN: A randomized grouping and controlled animal trial. SETTING: Department of Neurology, People's Hospital of Yunyang Medical College. MATERIALS: Twenty adult male SD rats of clean degree, weighing (250±30) g, were provided by the animal experimental center, School of Medicine, Wuhan University. The kits for SP immunohistochemistry were purchased from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd. METHODS: The experiments were carried out in the laboratory of Department of Neurology, Renmen Hospital of Wuhan University from May to July 2005. ① The 20 rats were divided randomly into normal temperature group (n =10) and mild hypothermia group (n =10). Models of permanent middle cerebral artery occlusion were established with modified nylon suture embolization. The rats were assessed with the Longa standards: 0 point for without nerve dysfunction; 1 for mild neurological deficit (fore claws could no extend completely); 2 for moderate neurological deficit (circling towards the affected side); 3 for severe neurological deficit (tilting towards the affected side); 4 for coma and unconscious; 1-3 points represented that models were successfully established. The rats of the normal temperature group were fed at room temperature, and those in the mild hypothermia group were induced by hypothermia from 2 hours postoperatively, and the rectal temperature was kept at 34-35 ℃ for 72 hours. ② Measurement of infarct volume: All the rats were anesthetized by intraperitoneal injection overdose sodium pentobarbital 7 days postoperatively, and then the heads were cut down to harvest brain. The brain tissues were placed into -20 ℃ refrigerator for 20 minutes, coronal sections of 2 mm were prepared. The infarct sites were not stained, whereas normal brain tissues were stained as red. The infarct volumes were calculated by using MPLAS-500 multimedia color pathological image&word analytical system. ③ Counting positive cells of vascular endothelial growth factor protein: The brains were harvested by cutting heads, then coronal sections of 2 mm were prepared. Routine dehydration, hyalinization, wax immersion and embedding were performed, then the detected with SP immunohistochemistry, the kits were purchased from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd. The cells whose cytoplasm was yellow-brown were positive ones, a single sample as a unit, peri-ischemic site and ischemic core were selected, and the corresponding sites in controlateral hemisphere were taken as controls. Five visual fields were selected from each site to be observed under microscope, the cells were counted, and the average number of positive cells was calculated in each group. The numbers of positive cells were determined with the image analytical apparatus. MAIN OUTCOME MEASURES: Number of the positive cells of vascular endothelial growth factor protein; Infarct volume of rat brain tissue. RESULTS: All the 20 rats were involved in the analysis of results. ① Number of positive cells of vascular endothelial growth factor protein in brain tissue: It was obviously lower in the mild hypothermia group than in the normal temperature group [(24.02±5.05), (36.07±2.69) cells/high power visual field, P < 0.01]. ② Comparison of infarct volume of brain tissue: After MCAO, it was obviously smaller in the mild hypothermia group than in the normal temperature group [(153.25±23.14), (253.45±36.21) mm3, P < 0.01]. CONCLUSION: Mild hypothermia can inhibit the expression of vascular endothelial growth factor and decrease the volume of cerebral infarction. | 5 | Synthesis and biological evaluation of ^(18)F-FB-NGA as a hepatic asialoglycoprotein receptor PET imaging agent显示文摘Asialoglycoprotein receptor(ASGP-R)is a hepatic membrane receptor that uniquely exists on the surface of mammalian hepatocytes,and has been used as target of liver functional imaging agents for many years.We labeled the Galactosyl-neoglycoalbumin(NGA)with 18F to get a PET molecular probe 18F-FB-NGA and evaluated its ability as a liver functional PET imaging agent.The 18F-FB-NGA was prepared with NGA by conjugation with Nsuccinimidyl-4-18F-fluorobenzoate(18F-SFB)and purified with PD-10 desalting column.The radiolabeling yield and radiochemical purity of 18F-FB-NGA were determined by radio-HPLC.Starting with 18F-F–,the total time for 18F-FB-NGA was about 120±10 min.The decay-corrected radiochemical yield is about 25–30%.The radiochemical purity of purified 18F-FB-NGA was more than 98%.Labeled with 185–1850 MBq 18F-SFB,the specific activity of 18F-FBNGA was estimated to be 7.83–78.3 TBq/mmol.Biodistribution of 18F-FB-NGA in normal mice was investigated after injection through the tail vein.The results showed that the liver accumulated 39.47±3.42 and 12.12±6.11%ID/g at 10 and 30 min after injection,respectively.Dynamic MicroPET images in mice were acquired with and without block after injection of the radiotracer,respectively.High liver activity accumulation was observed at 5 min after injection in normal group.On the contrary,the liver accumulation was significantly lower after block,indicating the specific binding to ASGP-R.18F-FB-NGA is probably a potential PET liver imaging agent. | GU Xiaobo CAI Gangming JIANG Mengjun ZHOU Yaoyuan ZHANG Rongjun | 2013 | Nuclear Science and Techniques2013,24,6: | 2 |

| 20 | Determination of heterozygosity for avirulence/virulence loci through sexual hybridization of Puccinia striiformis f. sp. tritici显示文摘Wheat stripe rust caused by Puccinia striiformis f. sp. tritici is one of the most devastating diseases of wheat worldwide and resistant cultivars are vital for its management. Therefore, investigating the heterozygosity of the pathogen is important because of rapid virulence changes in isolates heterozygous for avirulence/virulence.An isolate of P. striiformis f. sp. tritici was selfed on Berberis shensiana to determine the heterozygosity for avirulence/virulence loci. One hundred and twenty progeny isolates obtained from this selfing were phenotyped using 25 lines of wheat containing Yrgenes and genotyped with 96 simple sequencing repeat markers, with51 pathotypes and 55 multi-locus genotypes being identified. All of these were avirulent on lines with Yr5,Yr10, Yr15, Yr24 and Yr26 and virulent on lines with Yr17,Yr25 and YrA, indicating that the parental isolate was homozygously avirulent or homozygously virulent for these loci. Segregation was found for wheat lines with Yr1,Yr2, Yr4, Yr6, Yr7, Yr8, Yr9, Yr27, Yr28, Yr32, Yr43, Yr44,YrExp2, YrSp, YrTr1, YrTye and YrV23.
