1Combination of mild therapeutic hypothermia and adipose-derived stem cells for ischemic brain injury显示文摘Mild therapeutic hypothermia has been shown to mitigate cerebral ischemia, reduce cerebral edema, and improve the prognosis of patients with cerebral ischemia. Adipose-derived stem cell-based therapy can decrease neuronal death and infiltration of inflammatory cells, exerting a neuroprotective effect. We hypothesized that the combination of mild therapeutic hypothermia and adipose-derived stem cells would be neuroprotective for treatment of stroke. A rat model of transient middle cerebral artery occlusion was established using the nylon monofilament method. Mild therapeutic hypothermia(33°C) was induced after 2 hours of ischemia. Adipose-derived stem cells were administered through the femoral vein during reperfusion. The severity of neurological dysfunction was measured by a modified Neurological Severity Score Scaling System. The area of the infarct lesion was determined by 2,3,5-triphenyltetrazolium chloride staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The regeneration of microvessels and changes in the glial scar were detected by immunofluorescence staining. The inflammatory responses after ischemic brain injury were evaluated by in situ staining using markers of inflammatory cells. The expression of inflammatory cytokines was measured by reverse transcription-polymerase chain reaction. Compared with mild therapeutic hypothermia or adipose-derived stem cell treatment alone, their combination substantially improved neurological deficits and decreased infarct size. They synergistically reduced the number of TUNEL-positive cells and glial fibrillary acidic protein expression, increased vascular endothelial growth factor levels, effectively reduced inflammatory cell infiltration and down-regulated the m RNA expression of the proinflammatory cytokines interleukin-1β, tumor necrosis factor-α and interleukin-6. 2Pharmacology of mangostins and their derivatives:A comprehensive review显示文摘Mangosteen(Garcinia mangostana Linn.) is a well-known tropical tree indigenous to Southeast Asia. Its fruit's pericarp abounds with a class of isoprenylated xanthones which are referred as mangostins. Numerous in vitro and in vivo studies have shown that mangostins and their derivatives possess diverse pharmacological activities, such as antibacterial, antifungal, antimalarial, anticarcinogenic, antiatherogenic activities as well as neuroprotective properties in Alzheimer's disease(AD). This review article provides a comprehensive review of the pharmacological activities of mangostins and their derivatives to reveal their promising utilities in the treatment of certain important diseases, mainly focusing on the discussions of the underlying molecular targets/pathways, modes of action, and relevant structure-activity relationships(SARs). 4Hydrogen-rich water protects against inflammatory bowel disease in mice by inhibiting endoplasmic reticulum stress and promoting heme oxygenase-1 expression显示文摘AIM To investigate the therapeutic effect of hydrogen-rich water(HRW) on inflammatory bowel disease(IBD) and to explore the potential mechanisms involved.METHODS Male mice were randomly divided into the following four groups: control group, in which the mice received equivalent volumes of normal saline(NS) intraperitoneally(ip); dextran sulfate sodium(DSS) group, in which the mice received NS ip(5 m L/kg body weight, twice per day at 8 am and 5 pm) for 7 consecutive days after IBD modeling; DSS + HRW group, in which the mice received HRW(in the same volume as the NS treatment) for 7 consecutive days after IBD modeling; and DSS + HRW + Zn PP group, in which the mice received HRW(in the same volume as the NS treatment) and ZnP P [a heme oxygenase-1(HO-1) inhibitor, 25 mg/kg] for 7 consecutive days after IBD modeling. IBD was induced by feeding DSS to the mice, and blood and colon tissues were collected on the 7th d after IBD modeling to determine clinical symptoms, colonic inflammation and the potential mechanisms involved.RESULTS The DSS + HRW group exhibited significantly attenuated weight loss and a lower extent of disease activity index compared with the DSS group on the 7th d(P < 0.05). HRW exerted protective effects against colon shortening and colonic wall thickening in contrast to the DSS group(P < 0.05). The histological study demonstrated milder inflammation in the DSS + HRW group, which was similar to normal inflammatory levels, and the macroscopic and microcosmic damage scores were lower in this group than in the DSS group(P < 0.05). The oxidative stress parameters, including MDA and MPO in the colon, were significantly decreased in the DSS + HRW group compared with the DSS group(P < 0.05). Simultaneously, the protective indicators, superoxide dismutase and glutathione, were markedly increased with the use of HRW. Inflammatory factors were assessed, and the results showed that the DSS + HRW group exhibited significantly reduced levels of TNF-α, IL-6 and IL-1β compared with the DSS group(P < 0.05). In addition, the pivotal proteins involved in endoplasmic reticulum(ER) stress, including p-e IF2α, ATF4, XBP1 s and CHOP, were dramatically reduced after HRW treatment in contrast to the control group(P < 0.05). Furthermore, HRW treatment markedly up-regulated HO-1 expression, and the use of Zn PP obviously reversed the protective role of HRW. In the DSS + HRW + ZnP P group, colon shortening and colonic wall thickening were significantly aggravated, and the macroscopic damage scores were similar to those of the DSS + HRW group(P < 0.05). 5Design,synthesis and evaluation of novel cis-p-menthane type Schiff base compounds as effective herbicides显示文摘p-Menthane type monoterpene derivatives were identified as bio-based compounds with high herbicidal activities. In order to search novel p-menthane type monoterpene derivatives in good performance, a series of novel cis-p-menthane type Schiff base derivatives were designed and synthesized. All target products were easily available novel compounds and characterized by FT-IR,1H NMR,13 C NMR and ESI+-MS. Their pre-emergence herbicidal activities against annual ryegrass were evaluated. 8Melatonin ameliorates bleomycin-induced pulmonary fibrosis via activating NRF2 and inhibiting galectin-3 expression显示文摘Pulmonary fibrosis(PF)is a chronic interstitial lung disease with no effective therapies.Galectin-3(Gal-3),a marker of oxidative stress,plays a key role in the pathogenesis of PF.Fibroblast-myofibroblast differentiation(FMD)is an important source of fibrotic cells in PF.Previous studies showed that melatonin(MT)exerted anti-fibrotic effect in many diseases including PF through its antioxidant activity.In the present study we investigated the relationships among Gal-3,NRF2,ROS in FMD and their regulation by MT.We established an in vitro model of FMD in TGF-β1-treated human fetal lung fibroblast1(HFL1)cells and a PF mouse model via bleomycin(BLM)intratracheal instillation.We found that Gal-3 expression was significantly increased both in vitro and in vivo.Knockdown of Gal-3 in HFL1 cells markedly attenuated TGF-β1-induced FMD process and ROS accumulation.In TGF-β1-treated HFL1 cells,pretreatment with NRF2-specific inhibitor ML385(5μM)significantly increased the levels of Gal-3,α-SMA and ROS,suggesting that the expression of Gal-3 was regulated by NRF2.Treatment with NRF2-activator MT(250μM)blockedα-SMA and ROS accumulation accompanied by reduced Gal-3 expression.In BLM-induced PF model,administration of MT(5 mg·kg^(−1)·d^(−1),ip for 14 or 28 days)significantly attenuated the progression of lung fibrosis through up-regulating NRF2 and down-regulating Gal-3 expression in lung tissues.These results suggest that Gal-3 regulates TGF-β1-induced pro-fibrogenic responses and ROS production in FMD,and MT activates NRF2 to block FMD process by down-regulating Gal-3 expression.This study provides a useful clue for a clinical strategy to prevent PF.Yue-jiao Lan Ming-han Cheng Hui-min Ji Yu-qian Bi Yong-yue Han Chong-yang Yang Xuan Gu Jian Gao Hong-liang Dong 2023Acta Pharmacologica Sinica2023,44,5:2
9Attempted preparation of La_(0.5)Ba_(0.5)MnO_(3-δ) leading to an in-situ formation of manganate nanocomposites as a cathode for proton-conducting solid oxide fuel cells显示文摘A La_(0.5)Ba_(0.5)MnO_(3-δ) oxide was prepared using the sol-gel technique.Instead of a pure phase,La_(0.5)Ba_(0.5)MnO_(3-δ) was discovered to be a combination of La_(0.7)Ba_(0.3)MnO_(3-δ) and BaMnO_(3).The in-situ production of La_(0.7)Ba_(0.3)MnO_(3-δ)+BaMnO_(3) nanocomposites enhanced the oxygen vacancy(Vo)formation compared to single-phase La_(0.7)Ba_(0.3)MnO_(3-δ) or BaMnO_(3),providing potential benefits as a cathode for fuel cells.Subsequently,La_(0.7)Ba_(0.3)MnO_(3-δ)+BaMnO_(3) nanocomposites were utilized as the cathode for proton-conducting solid oxide fuel cells(H-SOFCs),which significantly improved cell performance.At 700 C,H-SOFC with a La_(0.7)Ba_(0.3)MnO_(3-δ)+BaMnO_(3) nanocomposite cathode achieved the highest power density(1504 mW·cm^(-2))yet recorded for H-SOFCs with manganate cathodes.This performance was much greater than that of single-phase La_(0.7)Ba_(0.3)MnO_(3-δ)or BaMnO_(3) cathode cells.In addition,the cell demonstrated excellent working stability.First-principles calculations indicated that the La_(0.7)Ba_(0.3)MnO_(3-δ)/BaMnO_(3) interface was crucial for the enhanced cathode performance.The oxygen reduction reaction(ORR)free energy barrier was significantly lower at the La_(0.7)Ba_(0.3)MnO_(3-δ)/BaMnO_(3) interface than that at the La_(0.7)Ba_(0.3)MnO_(3-δ) or BaMnO_(3) surfaces,which explained the origin of high performance and gave a guide for the construction of novel cathodes for H-SOFCs.Rui Zhou Yanru Yin Hailu Dai Xuan Yang Yueyuan Gu Lei Bi 2023Journal of Advanced Ceramics2023,12,6:1

18Modified treatment of knee osteoarthritis complicated with femoral varus deformity:A case report显示文摘BACKGROUND Surgical treatment of knee osteoarthritis(KOA)complicated by extra-articular deformity has always been controversial regardless of whether it is simultaneous or staged.Simultaneous total knee arthroplasty(TKA)combined with supracondylar osteotomy without plate for treatment of KOA complicated by femoral varus deformity has not been reported in the literature.CASE SUMMARY A 53-year-old Chinese woman complained of left knee pain for 6 years that worsened for 4 mo during her visit on April 3,2020,accompanied by instability in walking,which seriously affected quality of life.According to her medical history and preoperative imaging,the patient was diagnosed with left KOA with varus deformity.We used the angular center of rotation principle for osteotomy of the femur deformity and placed a poststabilized femur prosthesis into the knee joint.At the same time,a 13 mm×130 mm femur extension rod was used instead of a steel plate to fix the end of the femur osteotomy,reducing the possible complications caused by steel plate implantation and reducing the economic burden on patients.The operation successfully solved two major problems of KOA and varus deformity,and the clinical and imaging evaluation of postoperative followup were satisfactory.CONCLUSION TKA and supracondylar femoral osteotomy can be used for simultaneous KOA treatment and deformity correction.Sheng-Ming Xu Wei Li De-Bao Zhang Huang-Yi Bi Gui-Shan Gu 2022World Journal of Clinical Cases2022,10,6:1
