| 1 | Morphological and functional changes of mitochondria in apoptotic esophageal carcinoma cells induced by arsenic trioxide显示文摘AIM:To demonstrate that mitochondrial morphological andfunctional changes are an important intermediate link in thecourse of apoptosis in esophageal carcinoma calls inducedby As_2O_3.METHODS:The esophageal carcinoma call line SHEEC1,established in our laboratory,was cultured in 199 growthmedium,supplemented with 100mL·L^(-1)calf serum and3 mol·L^(-1)As_2O_3(the same below).After 2,4,6,12,24 hof drug adding,the SHEECl calls were collected for light-and electron-microscopic examination.The mitochondriawere labeled by Rhodamine fluorescence probe and thefluorescence intensity of the mitochondria was measured byflow cytorneter and cytofluorimetric analysis.Further,themitochondrial transmembrane potential(MTP,ΔΨm)change was also calculated.RESULTS:The mitochondrial morphological change afteradding As_2O_3 could be divided into three stages.In theearly-stage(2-6h)after adding As203,an adaptiveproliferation of mitochondria appeared;in the mid-stage(6-12 h)a degenerative change was observed;and in the late-stage(12-24 h)the mitochondria swelled with outermembrane broken down and then calls death with apoptoticchanges of nucleus.The functional change of themitochondria indicated by fluorescent intensity,whichreflected the MTP status of mitochondria,was in accordancewith morphological change of the mitochondria.Thefluorescent intensity increased at early-stage,decUned inmid-stage and decreased to the lowest in the late-stage.24h after As_2O_3 adding,the cell nucleus showed typicalapoptotic changes.CONCLUSION:Under the inducement of As_2O_3,the earlyapoptotic changes of SHEEC1 cells ware the apparentmorphological and functional changes of mitochondria,afterwards the nucleus changes followed.It is consideredthat changes of mitochondria are an important intermediatelink in the course of apoptosis of esophageal carcinoma cells induced by As_2O_3. | Zhong-Ying Shen Jian Shen Qiao-Shan Li Department of Pathology,Medical College of Shantou University,Shantou 515031,Guandong Province,China Cai-Yun Chen Central Lab.Medical College of Shantou University Jiong-Yu Chen Central Lab.of Tumor Hospital,Medical College of Shantou University Yi Zeng Institute of Virology,Chinese Academy of Preventive Medicine,Beijing 100052,China | 2002 | World Journal of Gastroenterology2002,8,1: | 35 |
| 2 | Expression of p27,cyclin E and cyclin A in hepatocellular carcinoma and its clinical significance显示文摘AIM: TO investigate the expression of p27, cyclin E andcyclin A in hepatocellular carcinoma (HCC) and its potential clinical significance.METHODS: Expression of p27, cyclin E and cyclin A in 45 HCC specimens and 30 adjacent noncancerous lesions obtained from 45 patients during surgery was examined by immunohistochemical SABC assay. The diameter of tumor ranged from 1 cm to 19 cm (d≤5 cm, 9 samples; 5 cm10 cm, 17 samples). The tumors were graded according to the criteria described by EdmondsonSteiner: well-differentiated HCC group (Grade Ⅰ+Ⅱ), 26 samples; poorly-differentiated HCC group (Grade Ⅲ+Ⅳ),19 samples. According to the clinical-pathologic features,19 samples were poorly encapsulated, 15 samples had portal invasion of cancer, 11 samples had extrahepatic metastasis,and 12 samples had intrahepatic metastasis. All of the samples were classified as the invasive and metastatic group,while the remaining was classified as the non-invasive and non- metastatic group.RESULTS: The average labeling index (LI) of p27 in HCC lesions was significantly higher than that in adjacent noncancerous lesions (45.87±14.21 vs33.77±12.92, t=3.745,P<0.001). The LI of p27 was associated with differentiation,invasiveness and metastasis of the tumors (34.46±12.29 vs 52.80±11.36, t=5.17; 41.42±12.86 vs51.44±14.10, t=2.48;P<0.05). Cyclin E was overexpressed in 16 cases (35.6 %)while cyclin A was overexpressed in 21 cases (46.7 %) in HCC lesions. No overexpression of cyclin E or cyclin A could be observed in adjacent non-carcinoma lesions and normal liver tissues. The overexpressions of cyclin E and cyclin A were correlated with differentiation, tumor thrombus,invasiveness and metastasis (P<0.05). Expression of cyclin E was significantly correlated with expression of cyclin A (r=0.329, P<0.05). The LI of p27 was significantly decreased in cyclin E, cyclin A positive groups (40.33±11.91 vs 49.50±13.76, t=3.05; 38.86±11.19 vs 52.57±12.62,t=3.89; P<0.05).CONCLUSION: p27, cyclin E, cydin A play cooperative roles in HCC tumorigenesis, differentiation, invasiveness and metastasis. | 3 | In vivo gene expression profiling of the entomopathogenic fungus Beauveria bassiana elucidates its infection stratagems in Anopheles mosquito显示文摘The use of entomopathogenic fungi to control mosquitoes is a promising tool for reducing vector-borne disease transmission.To better understand infection stratagems of insect pathogenic fungi,we analyzed the global gene expression profiling of Beauveria bassiana at 36,60,84 and 108 h after topical infection of Anopheles stephensi adult mosquitoes using RNA sequencing(RNA-Seq).A total of 5,354 differentially expressed genes(DEGs) are identified over the course of fungal infection.When the fungus grows on the mosquito cuticle,up-regulated DEGs include adhesion-related genes involved in cuticle attachment,Pthll-like GPCRs hypothesized to be involved in host recognition,and extracellular enzymes involved in the degradation and penetration of the mosquito cuticle.Once in the mosquito hemocoel,the fungus evades mosquito immune system probably through up-regulating expression of |3-l,3-glucan degrading enzymes and chitin synthesis enzymes for remodeling of cell walls.Moreover,six previous unknown SSCP(small secreted cysteine-rich proteins) are significantly up-regulated,which may serve as 'effectors' to suppress host defense responses.B.bassiana also induces large amounts of antioxidant genes to mitigate host-generated exogenous oxidative stress.At late stage of infection,B.bassiana activates a broad spectrum of genes including nutrient degrading enzymes,some transporters and metabolism pathway components,to exploit mosquito tissues and hemolymph as a nutrient source for hyphal growth.These findings establish an important framework of knowledge for further comprehensive elucidation of fungal pathogenesis and molecular mechanism of Beauveria-mosqaito interactions. | Yiling Lai Huan Chen Ge Wei Guandong Wang Fang Li Sibao Wang | 2017 | Science China(Life Sciences)2017,60,8: | 5 |
| 4 | Variation of Yellow River Runoff and Its Influence on Salinity in Laizhou Bay显示文摘Salinity in estuaries has an important influence on sediment deposition,delta formation,and fishery economy.The change of runoff in Yellow River(YR),which is the largest river along the Bohai Sea,has an important effect on the salinity of the sea,particularly in Laizhou Bay.In this study,measured runoff data in Lijin hydrological station from 1950 to 2018 are adopted to examine the change of YR runoff into the sea.The study concludes that the runoff of YR is distributed unevenly throughout the year and the amount of runoff mainly concentrates during the flood season.Results show a decreasing trend at a significant level ofα=0.95,with a range of−6.17×10^9 m^3 yr−1.Three groups of runoff are set by using the Finite-Volume Coastal Ocean Model numerical model to analyze the change of salinity in Laizhou Bay.These modeled effects were compared with water level observations made by Bayuquan,Tanggu,and Penglai gauge stations and salinity surveys conducted by Longkou gauge station.Model results and observations of salinity distributions and coastal water level showed good agreement.We can conclude that the distribution of salinity in Laizhou Bay is high in the east and low in the west.Furthermore,it responded quickly to the change of the runoff from YR.Under conditions of high river discharge,the low-salinity zone increased in horizontal direction and reached the bottom in vertical direction. | SHI Hongyuan LI Qingjie SUN Jiacheng GAO Guandong SUI Yi QIAO Shouwen YOU Zaijin | 2020 | Journal of Ocean University of China2020,19,6: | 3 |

Under simple supporting condition, the mechanical and electrical characteristics have been analyzed by using Piezoelectric-elastic theory, Kirchhoff 's thin shell vibration theory, Rayleigh-Ritz's theory and equivalent circuit method. The approximate solution and series resonance frequency equation have been given. Under no load, equivalent circuit, correlation parameters of cymbal transducer and the relations between the ratio of cavity depth to radius of Cymbal transducer with resonance frequency, electromechanical coupling coefficient of cymbal transducer have been researched. The best electromechanical coupling coefficient of cymbal transducer has been gained from the results of numerical analysis. | 20 | Synthesis of high-entropy MXenes with high-efficiency electromagnetic wave absorption显示文摘High-entropy MXenes,as a new emerging class of materials,possess diverse compositions,unexpected physicochemical characteristics,and great potentials for electromagnetic(EM)wave absorption.Herein,two single-to-few-layer high-entropy MXenes,(Mo_(0.25)Cr_(0.25)Ti_(0.25)V_(0.25))3 C2 T_(x)and(Mo_(0.2)Cr_(0.2)Nb_(0.2)Ti_(0.2)V_(0.2))_(4)C_(3)T_(x),were synthesized for the first time.During the exfoliation and delamination processes,the structural,morphological,and compositional evolutions were analyzed,verifying the successful formation of single-to-few-layer two-dimensional MXene nanosheets.Investigations indicate that with the filling content of only 35 wt%,MXene powder filled composites exhibit high-efficiency EM wave absorption performances.The f-(Mo_(0.25)Cr_(0.25)Ti_(0.25)V_(0.25))_(3)C_(2)T_(x)possesses the minimum reflection loss(RL min)of−45.0 dB with the matching thickness of 1.52 mm and the maximum effective absorption bandwidth(EAB)of 5.6 GHz at 1.65 mm thickness.Also,f-(Mo_(0.2)Cr_(0.2)Nb_(0.2)Ti_(0.2)V_(0.2))_(4)C_(3)T_(x)can attain an RL min of−52.8 dB with the thickness of 1.58 mm and an optimum EAB value of 3.6 GHz at 1.50 mm.The satisfactory EM wave absorption efficiency and bandwidth,thin matching thickness,and low filling content prove the lightweight advantage and great application potential of high-entropy MXenes in EM wave absorption.In this work,the high-entropy strategy is applied to tune the EM wave absorption performances for MXenes.Furthermore,high-entropy engineering is expected to provide control and tunability of many other properties,such as electrochemical,catalytic,and mechanical behaviors. | Linjing Qiao Jianqiang Bi Guandong Liang Yao Yang Hongyi Wang Shaoyin Wang | 2023 | Journal of Advanced Ceramics2023,12,10: | 0 |
