2Bioactivity of five components of Chinese herbal formula Jiangzhi granules against hepatocellular steatosis显示文摘OBJECTIVE: This study aims to evaluate the bioactivity of five components of the traditional Chinese medicine complex prescription Jiangzhi granules against hepatocellular steatosis. METHODS: The five major components, including protopanaxadiol, tanshinone IIA, emodin, chlorogenic acid, and nuciferine, were extracted from Jiangzhi granules. Their cytotoxicity was assessed to determine the safe dose of each component for HepG2 cells. HepG2 cellular steatosis was induced using 1 mmol/L of free fatty acids (FFAs) for 24 h, and then treated with each component at high, intermediate, and low doses (500, 50, and 5 μmol/L), respectively for another 24 h. The effects on HepG2 steatosis were observed directly under optical phase microscopy, or through oil red O staining and Nile red assays. In addition, the levels of reactive oxygen species (ROS) in the steatotic HepG2 cells with and without high-dose protopanaxadiol treatment were measured using fluorescent dye 2′,7′-dichlorodihydrofluorescein diacetate staining. RESULTS: No obvious cytotoxicity was observed in the HepG2 cells incubated with each of the five components at up to 500 μmol/L. At 24 h after incubation with FFAs, the HepG2 cells swelled and many lipid droplets accumulated. The lipid content was attenuated after 24 h of incubation with protopanaxadiol, tanshinone IIA, and emodin at 500 or 50 μmol/L (P<0.05), especially with 500 μmol/L protopanaxadiol (P<0.01). In addition, the ROS level was elevated in steatotic cells, but decreased after intervention with 500 μmol/L protopanaxadiol (P<0.05). CONCLUSION: Protopanaxadiol, tanshinone IIA, and emodin alleviate hepatocellular steatosis in a dose-dependent manner, and oxidative stress regulation may partially contribute to the effects of protopanaxadiol.Hai-yan Song Li Zhang Jie-lu Pan Li-li Yang Guang Ji 2013Journal of Integrative Medicine2013,11,4:17
3Three-dimensional Culture of Human Airway Epithelium in Matrigel for Evaluation of Human Rhinovirus C and Bocavirus Infections显示文摘Objective Newly identified human rhinovirus C(HRV-C) and human bocavirus(HBoV) cannot propagate in vitro in traditional cell culture models; thus obtaining knowledge about these viruses and developing related vaccines are difficult. Therefore, it is necessary to develop a novel platform for the propagation of these types of viruses. Methods A platform for culturing human airway epithelia in a three-dimensional(3 D) pattern using Matrigel as scaffold was developed. The features of 3 D culture were identified by immunochemical staining and transmission electron microscopy. Nucleic acid levels of HRV-C and HBoV in 3 D cells at designated time points were quantitated by real-time polymerase chain reaction(PCR). Levels of cytokines, whose secretion was induced by the viruses, were measured by ELISA. Results Properties of bronchial-like tissues, such as the expression of biomarkers CK5, ZO-1, and PCK, and the development of cilium-like protuberances indicative of the human respiration tract, were observed in 3 D-cultured human airway epithelial(HAE) cultures, but not in monolayer-cultured cells. Nucleic acid levels of HRV-C and HBoV and levels of virus-induced cytokines were also measured using the 3 D culture system. 4Mechanochemical synthesis and high-capacity performances of transition-metal borides as aqueous anode materials显示文摘Transition-metal borides MB0.5(M = Co,Mo,V) were synthesized by high-speed mechanical ball-milling of the corresponding elemental metals and boron,and investigated as aqueous anode materials.The as-synthesized borides can achieve an excellent discharge capacity,about twice that of their parent transition metals.The metal boride electrodes also exhibit polarizations about 100-300 mV lower than those of their parent metals.The galvanostatic discharge curve of CoB0.5 shows a single discharge voltage plateau as a result of simultaneous electro-oxidation of elemental cobalt and/or amorphous cobalt boride.Both MoB0.5 and VB0.5 show two well-defined voltage plateaus,corresponding to the electro-oxidation of the corresponding metal and boride.These results show that the coexisting transition metal and boride in the metal borides co-activate each other in the ball-milling process,thereby significantly enhancing their electrochemical performances.WANG YaDong GUANG XianYong PAN Mu 2012Chinese Science Bulletin2012,57,32:5

7Inverse relations between Helicobacter pylori infection and risk of esophageal precancerous lesions in drinkers and peanut consumption显示文摘BACKGROUND Helicobacter pylori(H. pylori) is a Gram-negative bacterium found in the upper digestive tract. Although H. pylori infection is an identified risk factor for gastric cancer, its role in esophageal squamous cell carcinoma(ESCC) remains a topic of much debate.AIM To evaluate the association between H. pylori infection and the risk of precancerous lesions of ESCC, and further explore the association between dietary factors and the risk of H. pylori infection.METHODS Two hundred patients with esophageal precancerous lesions(EPL) aged 63.01 ± 6.08 years and 200 healthy controls aged 62.85 ± 6.03 years were included in this case-control study. Epidemiological data and qualitative food frequency data were investigated. Enzyme-linked immunosorbent assay measuring serum immunoglobulin G antibodies was used to determine H. pylori seropositivity. An unconditional logistic regression model was used to assess the association between H. pylori infection and EPL risk dichotomized by gender, age, and the use of tobacco and alcohol, as well as the association between dietary factors and the risk of H. pylori infection.RESULTS A total of 47(23.5%) EPL cases and 58(29.0%) healthy controls had positive H. pylori infection. An inverse relation between H. pylori infection and the risk of EPL was found in the group of drinkers after adjustment for covariates [odds ratio(OR) = 0.32, 95% confidence interval(95%CI): 0.11-0.95]. Additionally, peanut intake was significantly associated with a decreased risk of H. pylori infection(OR = 0.39, 95%CI: 0.20-0.74).CONCLUSION Our study suggested that H. pylori infection may decrease the risk of EPL for drinkers in a rural adult Chinese population, and the consumption of peanut may reduce the risk of H. pylori infection. 8Determination of levels of adenosine phosphates in blood by ion chromatography显示文摘A new method was developed for simultaneous determination of levels of AMP,ADP and ATP in blood by anion-exchange chromatography.The three adenosine phosphates were separated by Dionex IonPac AG18(50 mm×4 mm) guard column and IonPac AS 18(250 mm×4 mm) analytical column using a gradient method and detected with a suppressed conductivity detector. 9Improved resistive switching stability of Pt/ZnO/CoO_x /ZnO/Pt structure for nonvolatile memory devices显示文摘For Pt(Ag)/ZnO single-layer/Pt structure,random 10 formation and rupture of conductive filaments composed by oxygen vacancies or metallic ions often cause dispersion problems of resistive switching(RS)parameters,which is disadvantageous to devices application.In this study,ZnO/CoOx/ZnO(ZCZ)tri-layers were utilized as the switching layers to investigate their RS properties as compared with ZnO-based single-layer devices.It is interestingly noted that Pt/ZCZ/Pt devices show quite stable bipolar RS behaviors with little resistance value fluctuations compared to Ag/ZCZ/Pt devices and Pt(Ag)/ZnO/Pt devices,which minimize the dispersion of the resistances of RS.This highly stable RS effect of Pt/ZCZ/Pt structure would be promising for high density memory devices.Guang Chen Cheng Song Feng Pan 2013Rare Metals2013,32,6:2

12Costunolide covalently targets NACHT domain of NLRP3 to inhibit inflammasome activation and alleviate NLRP3-driven inflammatory diseases显示文摘The NLRP3 inflammasome’s core and most specific protein,NLRP3,has a variety of functions in inflammation-driven diseases.Costunolide(COS)is the major active ingredient of the traditional Chinese medicinal herb Saussurea lappa and has anti-inflammatory activity,but the principal mechanism and molecular target of COS remain unclear.Here,we show that COS covalently binds to cysteine 598 in NACHT domain of NLRP3,altering the ATPase activity and assembly of NLRP3 inflammasome.We declare COS’s great anti-inflammasome efficacy in macrophages and disease models of gouty arthritis and ulcerative colitis via inhibiting NLRP3 inflammasome activation.We also reveal that theα-methylene-γ-butyrolactone motif in sesquiterpene lactone is the certain active group in inhibiting NLRP3 activation.Taken together,NLRP3 is identified as a direct target of COS for its anti-inflammasome activity.COS,especially theα-methylene-γ-butyrolactone motif in COS structure,might be used to design and produce novel NLRP3 inhibitors as a lead compound.Haowen Xu Jiahao Chen Pan Chen Weifeng Li Jingjing Shao Shanshan Hong Yi Wang Lingfeng Chen Wu Luo Guang Liang 2023Acta Pharmaceutica Sinica B2023,13,2:2
13Buckling of composite cylindrical shells with ovality and thickness variation subjected to hydrostatic pressure显示文摘The initial geometric imperfection is one of the primary factors affecting the buckling behaviors of composite cylindrical shells under hydrostatic pressure.In this study,ovality and thickness variations as two representative types of the geometric imperfections are considered.After measuring the geometric imperfections,a typical carbon fiber reinforced polymers(CFRP)cylindrical shell is tested to obtain the buckling pressure.The buckling behaviors of the shell sample are analyzed in combination with the strain responses.By using the nonlinear numerical analysis,the buckling shapes of the CFRP cylinder shells with different combinations of ovality and thickness variation are firstly discussed.The rules of influence of such imperfections on the buckling pressure are then obtained by nonlinear regression method.Finally,an empirical formula is proposed to predict the buckling pressure of the composite cylinder shells,and the calculated results from the formula are in good agreement with the numerical results.Zhun Li Ke-chun Shen Xin-hu Zhang Guang Pan 2022Defence Technology(防务技术)2022,18,5:2
