| 1 | Determination of quorum-sensing signal substances in water and solid phases of activated sludge systems using liquid chromatography–mass spectrometry显示文摘The detection of acyl homoserine lactones(AHLs) in activated sludge is essential for clarifying their function in wastewater treatment processes. An LC–MS/MS method was developed for the detection of AHLs in both the aqueous and solid phases of activated sludge. In addition, the effects of proteases and extracellular polymeric substances(EPS) on the detection of AHLs were evaluated by adding protease inhibitors and extracting EPS,respectively. Recoveries of each AHL were improved by adding 50 μL of protease inhibitor,and recoveries were also improved from 0 to 56.9% to 24.2%–105.8% by EPS extraction.Applying the developed method to determine the type and concentration of AHLs showed that C4-HSL, C_6-HSL, C_8-HSL and 3-oxo-C_8-HSL were widely detected in a suspended activated sludge system. The dominant AHL was C_8-HSL, with a highest concentration of304.3 ng/L. | 2 | Unintended targeting of Dmp1-Cre reveals a critical role for Bmpr1a signaling in the gastrointestinal mesenchyme of adult mice显示文摘Cre/lox P technology has been widely used to study cell type-specific functions of genes. Proper interpretation of such data critically depends on a clear understanding of the tissue specificity of Cre expression. The Dmp1-Cre mouse, expressing Cre from a 14-kb DNA fragment of the mouse Dmp1 gene, has become a common tool for studying gene function in osteocytes, but the presumed cell specificity is yet to be fully established. By using the Ai9 reporter line that expresses a red fluorescent protein upon Cre recombination, we find that in2-month-old mice, Dmp1-Cre targets not only osteocytes within the bone matrix but also osteoblasts on the bone surface and preosteoblasts at the metaphyseal chondro-osseous junction. In the bone marrow, Cre activity is evident in certain stromal cells adjacent to the blood vessels, but not in adipocytes. Outside the skeleton, Dmp1-Cre marks not only the skeletal muscle fibers, certain cells in the cerebellum and the hindbrain but also gastric and intestinal mesenchymal cells that express Pdgfra. Confirming the utility of Dmp1-Cre in the gastrointestinal mesenchyme, deletion of Bmpr1 a with Dmp1-Cre causes numerous large polyps along the gastrointestinal tract, consistent with prior work involving inhibition of BMP signaling.Thus, caution needs to be exercised when using Dmp1-Cre because it targets not only the osteoblast lineage at an earlier stage than previously appreciated, but also a number of non-skeletal cell types. | Joohyun Lim Joseph Burclaff Guangxu He Jason C Mills Fanxin Long | 2017 | Bone Research2017,5,1: | 4 |
| 3 | Inducible expression of Wnt7b promotes bone formation in aged mice and enhances fracture healing显示文摘There remain unmet clinical needs for safe and effective bone anabolic therapies to treat aging-related osteoporosis and to improve fracture healing in cases of nonunion or delayed union. Wnt signaling has emerged as a promising target pathway for developing novel bone anabolic drugs. Although neutralizing antibodies against the Wnt antagonist sclerostin have been tested,Wnt ligands themselves have not been fully explored as a potential therapy. Previous work has demonstrated Wnt7b as an endogenous ligand upregulated during osteoblast differentiation, and that Wnt7b overexpression potently stimulates bone accrual in the mouse. The earlier studies however did not address whether Wnt7b could promote bone formation when specifically applied to aged or fractured bones. Here we have developed a doxycycline-inducible strategy where Wnt7b is temporally induced in the bones of aged mice or during fracture healing. We report that forced expression of Wnt7b for 1 month starting at 15 months of age greatly stimulated trabecular and endosteal bone formation, resulting in a marked increase in bone mass. We further tested the effect of Wnt7b on bone healing in a murine closed femur fracture model. Induced expression of Wnt7b at the onset of fracture did not affect the initial cartilage formation but promoted mineralization of the subsequent bone callus. | 4 | Differential involvement of Wnt signaling in Bmp regulation of cancellous versus periosteal bone growth显示文摘Bone morphogenetic proteins(Bmp) are well-known to induce bone formation following chondrogenesis,but the direct role of Bmp signaling in the osteoblast lineage is not completely understood. We have recently shown that deletion of the receptor Bmpr1 a in the osteoblast lineage with Dmp1-Cre reduces osteoblast activity in general but stimulates proliferation of preosteoblasts specifically in the cancellous bone region,resulting in diminished periosteal bone growth juxtaposed with excessive cancellous bone formation.Because expression of sclerostin(SOST), a secreted Wnt antagonist, is notably reduced in the Bmpr1 adeficient osteocytes, we have genetically tested the hypothesis that increased Wnt signaling might mediate the increase in cancellous bone formation in response to Bmpr1 a deletion. Forced expression of human SOST from a Dmp1 promoter fragment partially rescues preosteoblast hyperproliferation and cancellous bone overgrowth in the Bmpr1 a mutant mice, demonstrating functional interaction between Bmp and Wnt signaling in the cancellous bone compartment. To test whether increased Wnt signaling can compensate for the defect in periosteal growth caused by Bmpr1 a deletion, we have generated compound mutants harboring a hyperactive mutation(A214 V) in the Wnt receptor Lrp5. However, the mutant Lrp5 does not restore periosteal bone growth in the Bmpr1 a-deficient mice. | 5 | Synthesis and electrochemical properties of Zn_3V_3O_8 as novel anode material显示文摘Zn_3V_3O_8 two-dimensional micro sheets are successfully synthesized by combination of solvothermal method and heat treatment. The X-ray diffraction(XRD) and field emission scanning electron microscopy(SEM) are used to characterize the structure and morphology of the samples, and the battery testing system is used to investigate its electrochemical performance. The results show that the Zn_3V_3O_8 is successfully obtained using mix solvent(water:ethylene glycol =1:1), and the sample takes on morphology of two-dimensional sheets. The initial discharge capacity for Zn_3V_3O_8 two-dimensional micro sheets is 752.0 mAh/g at 100 mA/g in the voltage range of 0.01-3 V. After heat treatment in a tube furnace at 550 ℃, the sample had the initial discharge capacity as 1152.0 mAh/g and maintained it at 901.4 mAh/g after 100 cycles. | 12 | Triterpene-Enriched Olive Extract as an Immunopotentiator in Black Sea Bream(Acanthopagrus schlegelii)显示文摘Recently,the use of natural immunopotentiators,such as plant extracts,is predicted to offer high disease-prevention potential in aquaculture.However,few reports on the immunomodulatory impacts of olive extract(OE)on teleost are available.Therefore,the effects of dietary intake of triterpene-enriched OE on black sea bream(Acanthopagrus schlegelii)were investigated in this study.Our data showed that total blood cell counts,in vivo content of lysozyme,activities of antiprotease and myeloperoxidase,and contents of IL-4 and IL-6 were significantly up-regulated by dietary intake of triterpene-enriched OE.Additionally,the hampered immune response induced by cadmium exposure was significantly mitigated by the administration of OE as were indicated by partially or completely rescued immune-relating parameters.Furthermore,the expressions of immune-related genes encoding NF-κB inhibitor alpha(IkBα),tumor necrosis factorα(TNFα),cyclooxygenase-2(COX-2),and proto-oncogene protein c-fos(FOS)were found to be significantly up-regulated by the dietary intake of OE.In general,the results suggested that the dietary intake of triterpene-enriched OE has immune enhancing effect in black sea bream.Such effect may be realized by 1)increasing the total counts of diverse blood cells;2)activating nonspecific immune biomolecules;and 3)affecting signaling pathways such as IKK and ERK and subsequently inducing IL-4 and IL-6 biosyntheses. | RONG Jiahuan HAN Yu ZHA Shanjie TANG Yu SHI Wei GUAN Xiaofan DU Xueying HE Maolong LIU Guangxu | 2020 | Journal of Ocean University of China2020,19,2: | 1 |

The results of tests find the optimal temperatures of Bi_(2)WO_(6) and Bi_(2)MoO_(6) are 180 and 160 °C(BW-180, BM-160). Then, BW-180 and BM-160 are further compounded with different contents of CuS. The results of photoelectrochemical(PEC) tests show that CuS can improve the PEC performance of semiconductor materials, and it has better performance when CuS mass fraction is 5%. These maybe the photoelectron potentials generated by Cu S/Bi_(2)XO_(6)(X = Mo, W) heterojunction reduce the combination of photogenerated electrons and holes. When the PEC sensor based on 5%-CuS/BW-180 detects Cr(VI), it has a linear range of 1–80 μmol/L with detection limit of 0.95 μmol/L, while the PEC sensor based on 5%-CuS/BM-160 detects Cr(VI) has a linear range of 0.5–230 μmol/L and a detection limit of 0.12 μmol/L.
