| 2 | Effect of Lamb Age and in vitro Culture System of Oocytes on JIVET Technology显示文摘[Objectives] This study was conducted to investigate the effects of lamb age and in vitro culture system of oocytes on the results of juvenile in vitro embryo transfer( JIVET). [Methods]Ten Dorper × small-tailed Han lambs aged 5 to 10 weeks were induced to superovulate via i. p. injection of pregnant mare's serum gonadotropin( PMSG). The oocytes were matured in basal maturation solution or modified maturation solution,which was prepared by adding 200 μmol/L cysteine to the basal maturation solution. Then,the oocytes were fertilized in fertilization medium I containing 2% estrus sheep serum( ESS) or fertilization medium II containing 3 mg/ml bull serum albumin( BSA). Finally,the number of oocytes,oocyte maturation rate and cleavage rate of the lambs of different ages were determined. [Results]The average number of oocytes recovered per lamb was( 111. 00 ± 16. 97),( 139. 50 ± 28. 99),( 108. 50 ± 17. 68) and( 42. 00 ± 11. 31) for5-,7-,8-and 10-week-old Dorper × small-tailed Han lambs,respectively. The number of oocytes obtained from 5-,7-and 8-week-old lambs was significantly higher than that from 10-week-old lambs( P < 0. 05),but there was no significant difference among 5-,7-and 8-week-old lambs( P > 0. 05). The maturation rate of oocytes cultured in modified maturation solution was 3. 64% higher than that in basal maturation solution. The cleavage rate of oocytes in fertilization medium I was very significantly higher than that in fertilization medium II( P < 0. 01). | 3 | Manganese(Ⅲ) acetate-mediated radical reaction of [60]fullerene with bromoacetic acid,3-chloropropionic acid or 1-naphthylacetic acid显示文摘The manganese(Ⅲ) acetate-mediated radical reaction of [60]fullerene (C60) with three carboxylic acids,that is,bromoacetic acid,3-chloropropionic acid,and 1-naphthylacetic acid,was investigated. The reaction of C60 with bromoacetic acid in the presence of 4-(dimethylamino)pyridine afforded the anticipated C60-fused lactone,while the reaction with 3-chloropropionic acid unexpectedly led to the formation of a novel C60-fused lactone with the loss of one molecule of HCl. Interestingly,the reaction with 1-naphthylacetic acid under similar conditions gave both C60-fused lactone and 1,4-adduct,yet each of them could be selectively obtained by controlling reaction conditions. | 4 | Deep white matter hyperintensity is spatially correlated to MRI-visible perivascular spaces in cerebral small vessel disease on 7 Tesla MRI显示文摘Background The association between perivascular space(PVS)and white matter hyperintensity(WMH)has been unclear.Normal-appearing white matter(NAWM)around WMH is also found correlated with the development of focal WMH.This study aims to investigate the topological connections among PVS,deep WMH(dWMH)and NAWM around WMH using 7 Tesla(7T)MRI.Methods Thirty-two patients with non-confluent WMHs and 16 subjects without WMHs were recruited from our department and clinic.We compared the PVS burden between patients with and without WMHs using a 5-point scale.Then,the dilatation and the number of PVS within a radius of 1 cm around each dWMH were compared with those of a reference site(without WMH)in the contralateral hemisphere.In this study,we define NAWM as an area within the radius of 1 cm around each dWMH.Furthermore,we assessed the spatial relationship between dWMH and PVS.Results Higher PVS scores in the centrum semiovale were found in patients with>5 dWMHs(median 3)than subjects without dWMH(median 2,p=0.014).We found there was a greater dilatation and a higher number of PVS in NAWM around dWMH than at the reference sites(p<0.001,p<0.001).In addition,79.59%of the dWMHs were spatially connected with PVS.Conclusion dWMH,NAWM surrounding WMH and MRI-visible PVS are spatially correlated in the early stage of cerebral small vessel disease.Future study of WMH and NAWM should not overlook MRI-visible PVS. | Yajing Huo Yilin Wang Cen Guo Qianyun Liu Lili Shan Mingyuan Liu Haibo Wu Guanwu Li Huihui Lv Lingdan Lu Yintin Zhou Jianfeng Feng Yan Han | 2023 | Stroke & Vascular Neurology2023，8，2： | 2 |

| 16 | Effects of Metal Ions and EDTA on Free Radical Reaction Intermediates of Laccase-catalyzed Oxidation of Wood Powder from Szemao Pine显示文摘Certain activator was expected to be developed to improve the free radical intermediates relative intensity, and thus to enhance the adhesion between wood fibers when fiberboard was made by laccase treated wood fibers. Reactive oxygen species (ROS) was detected in laccase-catalyzed oxidation of wood powder using ESR spin trapping technique. The effects of five metal ions (Cu2+, K+, Fe2+, Mg2+, Fe3+) and EDTA on ROS relative strengths were investigated under the condition of pH 5.6 by electron spin resonance spectrometry. The result shows that Cu2+ slightly activates the free radical reaction, and Fe2+ ion has a significant inhibitive effect on the ROS relative strength in the suspension liquid. There is a marked inhibition of the ROS relative intensity when 0.5 mmol/L EDTA is used. Methods: We detected the types and expression of cytokines secreted by neutrophils by cDNA array and RT-PCR. We also analyzed the changes of signal transduction in this process by detecting the expression of toll like receptor 4 (TLR4) and the inhibitor factor of κBα (I-κBα) expressed by neutrophils. The activity of NF-κB DNA-binding in neutrophils was measured by electrophoretic mobility shift assay (EMSA). Results: Cefodizime increased the neutrophils production of TNF-α, IL-1β and the mRNA expression of TLR4 in the early stage of Kle. p stimulation in mice, which seemed corresponding to the enhanced NF-κB DNA-binding activity.
