1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 2Taxus chinensis ameliorates diabetic nephropathy through down-regulating TGF-β1/Smad pathway显示文摘Diabetic nephropathy(DN) is one of the common microvascular complications of diabetes mellitus. Renal fibrosis is closely related to the deterioration of renal function. The present study aimed to investigate protective effect of Taxus chinensis on high-fat diet/streptozotocin-induced DN in rats and explore the underlying mechanism of action. The rat DN model was established via feeding high fat diet for 4 weeks and subsequently injecting streptozotocin(30 mg·kg^(-1) body weight) intraperitoneally. The rats with blood glucose levels higher than 16.8 mmol·L^(-1) were selected for experiments. The DN rats were treated with Taxus chinensis orally(0.32, 0.64, and 1.28 g·kg^(-1)) once a day for 8 weeks. Taxus chinensis significantly improved the renal damage, which was indicated by the decreases in 24-h urinary albumin excretion rate, blood serum creatinine, and blood urea nitrogen. Histopathological examination confirmed the protective effect of Taxus chinensis. The thickness of glomerular basement membrane was reduced, and proliferation of mesangial cells and podocytes cells and increase in mesangial matrix were attenuated. Further experiments showed that Taxus chinensis treatment down-regulated the expression of TGF-β1 and α-SMA, inhibited phosphorylation of Smad2 and Smad3. 3Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). Our work dem on strates the utility of the con versi on betwee n hepatocyte and liver proge nitor-like cells for studying HBV biology and an tiviral therapies. 4Microstructures and mechanical properties of the third generation automobile steels fabricated by ART-annealing显示文摘The mechanical properties dependence on the microstructure was reviewed and analyzed,and the ultrafine grained duplex microstructure of BCC matrix and large fractioned austenite was given as one of the optimum structures to develop the third generation steel with high strength and high ductility.The medium-Mn steels with different carbon contents processed by austenite reverted transformation(ART-annealing) were studied thoroughly to fabricate the ultrafine duplex steels with large fractioned austenite.The lamellar typed ultrafine structure,the granular typed ultrafine duplex structure and the corresponding mechanical properties of the medium-Mn steels processed by ART-annealing were demonstrated in this paper.It was revealed that the duplex structure with large fraction of austenite and ultrafine grain structure is capable of producing steels with excellent combination of strength and ductility,i.e.,Rm A about 30-50 GPa%,which is about two times of that of the conventional automobile steels and close to that of the TWIP steels.It was concluded that the ART-annealing of the medium-Mn steels would be at least one of the promising ways to fabricate the third generation automobile steels in the near future.CAO WenQuan WANG Chang WANG CunYu SHI Jie WANG MaoQiu DONG Han WENG YuQing 2012Science China(Technological Sciences)2012,55,7:13

613-Methyltetradecanoic acid mitigates cerebral ischemia/reperfusion injury显示文摘13-Methyltetradecanoic acid can stabilize cell membrane and have anti-inflammatory,antioxidant and anti-apoptotic effects.Previous studies mainly focused on peripheral nerve injury,but seldom on the central nervous system.We investigated whether these properties of 13-methyltetradecanoic acid have a neuroprotective effect on focal cerebral ischemia/reperfusion injury,and detected the expression of basic fibroblast growth factor and vascular endothelial growth factor.This study established rat models of middle cerebral artery occlusion/reperfusion injury by ischemia for 2 hours and reperfusion for 24 hours.At the beginning of reperfusion,13-methyltetradecanoic acid 10,40 or 80 mg/kg was injected into the tail vein.Results found that various doses of 13-methyltetradecanoic acid effectively reduced infarct volume,mitigate cerebral edema,and increased the m RNA and protein expression of basic fibroblast growth factor and vascular endothelial growth factor at 24 hours of reperfusion.The effect was most significant in the 13-methyltetradecanoic acid 40 and 80 mg/kg groups.The findings suggest that 13-methyltetradecanoic acid can relieve focal ischemia/reperfusion injury immediately after reperfusion,stimulate the upregulation of basic fibroblast growth factor and vascular endothelial growth factor to exert neuroprotective effects.Juan Yu Li-nan Yang Yan-yun Wu Bao-hua Li Sheng-mei Weng Chun-lan Hu Yong-ling Han 2016Neural Regeneration Research2016,11,9:8
7Autologous transplantation with fewer fibers repairs large peripheral nerve defects显示文摘Peripheral nerve injury is a serious disease and its repair is challenging. A cable-style autologous graft is the gold standard for repairing long peripheral nerve defects; however, ensuring that the minimum number of transplanted nerve attains maximum therapeutic effect remains poorly understood. In this study, a rat model of common peroneal nerve defect was established by resecting a 10-mm long right common peroneal nerve. Rats receiving transplantation of the common peroneal nerve in situ were designated as the in situ graft group. Ipsilateral sural nerves(10–30 mm long) were resected to establish the one sural nerve graft group, two sural nerves cable-style nerve graft group and three sural nerves cable-style nerve graft group. Each bundle of the peroneal nerve was 10 mm long. To reduce the barrier effect due to invasion by surrounding tissue and connective-tissue overgrowth between neural stumps, small gap sleeve suture was used in both proximal and distal terminals to allow repair of the injured common peroneal nerve. At three months postoperatively, recovery of nerve function and morphology was observed using osmium tetroxide staining and functional detection. The results showed that the number of regenerated nerve fibers, common peroneal nerve function index, motor nerve conduction velocity, recovery of myodynamia, and wet weight ratios of tibialis anterior muscle were not significantly different among the one sural nerve graft group, two sural nerves cable-style nerve graft group, and three sural nerves cable-style nerve graft group. These data suggest that the repair effect achieved using one sural nerve graft with a lower number of nerve fibers is the same as that achieved using the two sural nerves cable-style nerve graft and three sural nerves cable-style nerve graft. 9Edaravone attenuates paraquat-induced lung injury by inhibiting oxidative stress in human type Ⅱalveolar epithelial cells显示文摘BACKGROUND:Edaravone(3-methyl-1-penyl-2-pyrazolin-5-one) is a potent free-radical scavenger and has the antioxidant ability to inhibit lipid peroxidation.The study aimed to examine the effect of edaravone on protecting the acute injury of human type II alveolar epithelial cells(A549cells) induced by paraquat(PQ) and the change of production of reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD).METHODS:A549 cells were cultured and divided into PQ group(group P),edaravone-treated group(group E) and normal control group(group C).The cells in group P were exposed to paraquat(600 umol/L),and the cells in group E were treated with edaravone(100 umol/L) additionally,and no drug intervention was given to the cells in group C.Real-time monitoring by LSCM was used to detect the cell response and the intracellular dynamic change of ROS level in A549 cells after administration of PQ and edaravone.And the levels of SOD and MDA were detected respectively by biochemistry colorimetry.Data were expressed as mean ± standard error of the mean.Statistical analysis was carried out with the soft SPSS 16.0.RESULTS:The concentration of intracellular ROS significantly increased when PQ was given to A549 cells.But after administration of edaravone,the concentration of intracellular ROS was decreased.Compared to the PQ group,the levels of SOD in the edaravone group were significantly increased while the levels of MDA were markedly decreased.CONCLUSIONS:Paraquat can increase the oxidative stress,and induce the lipid peroxidation of A549 cells.Edaravone has the effect to scavenge reactive oxygen species,and to protect against the PQ-induced lung toxicity.Zhi-qiang Cheng Ji-yuan Han Peng Sun Yu-ying Weng Jiao Chen Guo-yan Wu Hong-xia Ma 2012World Journal of Emergency Medicine2012,3,1:8
10Appraisal of the Quality and Contents of Clinical Practice Guidelines for Hypertension Management in Chinese Medicine: A Systematic Review显示文摘Objective: To evaluate the quality and consistency of recommendations in the clinical practice guidelines(CPGs) for hypertension in Chinese medicine(CM). Methods: CM CPGs were identified from 5 electronic databases and hand searches through related handbooks published from January 1990 to December 2013. Three reviewers independently appraised the CPGs based on the Appraisal of Guidelines for Research and Evaluation(AGREE Ⅱ) instrument, and compared the CPGs' recommendations on CM syndrome pattern classification and treatment. Results: Five CM CPGs for hypertension were included. The quality score of the evidence-based(EB) guideline was higher than those of the consensus-based with no explicit consideration of evidence-based(CB-EB) and the consensus-based(CB) guidelines. Three out of five patterns in the CPGs were recommended by the EB guideline. Tianma Gouteng Formula(天麻钩藤复方) in the EB guideline was recommended mostly for hypertension patients with pattern of ascendant hyperactivity of Gan(Liver)-yang and pattern of yin deficiency with yang hyperactivity in the CPGs. Acupuncture and massage were recommended for Grade Ⅰ and Grade Ⅱ hypertension with severe symptoms weakening the quality of life in the EB guideline. For Grade Ⅰ and Grade Ⅱ hypertension, CM could be used alone, while for Grade Ⅲ hypertension, they should be used in combination with Western medicines. 11Effects of total pressures and equivalence ratios on kerosene/air rotating detonation engines using a paralleling CE/SE method显示文摘In this paper,the kerosene/air rotating detonation engines(RDE)are numerically investigated,and the emphasis is laid on the effects of total pressures and equivalence ratios on the operation characteristics of RDE including the initiation,instabilities,and propulsive performance.A hybrid MPI t OpenMP parallel computing model is applied and it is proved to be able to obtain a more effective parallel performance on high performance computing(HPC)systems.A series of cases with the total pressure of 1 MPa,1.5 MPa,2 MPa,and the equivalence ratio of 0.9,1,1.4 are simulated.On one hand,the total pressure shows a significant impact on the instabilities of rotating detonation waves.The instability phenomenon is observed in cases with low total pressure(1 MPa)and weakened with the increase of the total pressure.The total pressure has a small impact on the detonation wave velocity and the specific impulse.On the other hand,the equivalence ratio shows a negligible influence on the instabilities,while it affects the ignition process and accounts for the detonation velocity deficit.It is more difficult to initiate rotating detonation waves directly in the lean fuel operation condition.Little difference was observed in the thrust with different equivalence ratios of 0.9,1,and 1.4.The highest specific impulse was obtained in the lean fuel cases,which is around 2700 s.The findings could provide insights into the understanding of the operation characteristics of kerosene/air RDE.Fang Wang Chun-sheng Weng Yu-wen Wu Qiao-dong Bai Quan Zheng Han Xu 2021Defence Technology(防务技术)2021,17,6:7
12Third generation high strength low alloy steels with improved toughness显示文摘On-line thermo mechanical controlled processing(TMCP) was conducted to develop the third generation high strength low alloy(HSLA) steel with high toughness economically.The ultra-low carbon content ensured a high level of upper shelf energy while ultrafine lath martensitic structure transformed from super-thin pancaked austenite during controlled rolling and cooling.The reduction of martensite block size decreased ductile-to-brittle transition temperature(DBTT) and compensated the strength loss due to carbon reduction.Consequently,the excellent balance of strength and toughness values was obtained as 950-1060 MPa for yield strength,180 J for Charpy V-notch impact absorbed energy at 30℃,which is much superior to that of traditional martensitic steel.Two mechanisms for the refinement of lath martensite block were proposed:One is the austenite grain refinement in the direction of thickness,and the other is the reduction in the fraction of sub-block boundaries with small misorientation and the increase in the fraction of block boundaries with large misorientation,possibly due to austenite hardening.SUN XinJun LI ZhaoDong YONG QiLong YANG ZhiGang DONG Han WENG YuQing 2012Science China(Technological Sciences)2012,55,7:7
13Chondrogenesis mediates progression of ankylosing spondylitis through heterotopic ossification显示文摘Ankylosing spondylitis(AS)is chronic inflammatory arthritis with a progressive fusion of axial joints.Anti-inflammatory treatments such as anti-TNF-αantibody therapy suppress inflammation but do not effectively halt the progression of spine fusion in AS patients.Here we report that the autoimmune inflammation of AS generates a microenvironment that promotes chondrogenesis in spine ligaments as the process of spine fusion.Chondrocyte differentiation was observed in the ligaments of patients with earlystage AS,and cartilage formation was followed by calcification.Moreover,a large number of giant osteoclasts were found in the inflammatory environment of ligaments and on bony surfaces of calcified cartilage.Resorption activity by these giant osteoclasts generated marrow with high levels of active TGF-β,which induced new bone formation in the ligaments.Notably,no Osterix+osteoprogenitors were found in osteoclast resorption areas,indicating uncoupled bone resorption and formation.Even at the late and maturation stages,the uncoupled osteoclast resorption in bony interspinous ligament activates TGF-βto induce the progression of ossification in AS patients.Osteoclast resorption of calcified cartilage-initiated ossification in the progression of AS is a similar pathologic process of acquired heterotopic ossification(HO).Our finding of cartilage formation in the ligaments of AS patients revealed that the pathogenesis of spinal fusion is a process of HO and explained why anti-inflammatory treatments do not slow ankylosing once there is new bone formation in spinal soft tissues.Thus,inhibition of HO formation,such as osteoclast activity,cartilage formation,or TGF-βactivity could be a potential therapy for AS.Tao Yu Jianguo Zhang Wei Zhu Xiao Wang Yun Bai Bin Feng Qianyu Zhuang Chang Han Shengru Wang Qimiao Hu Senbo An Mei Wan Shiwu Dong Jianzhong Xu Xisheng Weng Xu Cao 2021Bone Research2021,9,2:7

15Deep origin of Cenozoic volcanoes in Northeast China revealed by 3-D electrical structure显示文摘The three-dimensional(3-D)electrical structure of the upper-mantle was used to examine the deep origins of and relationship among the Cenozoic volcanoes located in Northeast China(NEC).High-quality,long-period magnetotelluric(LMT)full-impedance tensor data were collected in NEC and subjected to 3-D Gauss-Newton inversion in order to construct a resistivity model.The resulting model reveals the presence of multiple localized low-resistivity anomalies(LRAs)within the high resistivity lithosphere beneath NEC.These LRAs partially coincide with Cenozoic volcanoes on the surface.Three LRAs that form a larger,annular LRA were observed in the deep upper mantle beneath the Songliao Basin,whereas vein-like LRAs were found in the asthenosphere that connect the lithosphere and deep upper mantle.Petrophysical analyses suggest that the LRAs may have been caused by fluid-induced melting.Based on our electrical model,we propose that,following dehydration of the subducted Western Pacific slab into the mantle transition zone(MTZ)beneath NEC,the released water migrated upward and caused partial melting at the top of the MTZ beneath the Songliao Basin.Under the effect of buoyancy,the melted mantle formed a thermal upwelling that caused melting of asthenosphere before diapiring at the base of the dry lithosphere.The magma then penetrated structural boundaries(such as thinner,weaker,or activated suture zones)and finally reached the Earth's surface.This melting and upwelling of hot mantle materials may have resulted in large-scale volcanism in the region throughout the Cenozoic,including the eruption of Changbai Mountain and Halaha Volcanoes.Our results suggest that the Cenozoic NEC volcanoes may all share a similar mode of genesis,and probably originated from the annular LRA in the deep upper mantle.Shiwen LI Aihua WENG Jianping LI Xuanlong SHAN Jiangtao HAN Yu TANG Yanhui ZHANG Xueqiu WANG 2020Science China Earth Sciences2020,63,4:4
16Constraint-induced movement therapy promotes brain functional reorganization in stroke patients with hemiplegia显示文摘Stroke patients with hemiplegia exhibit flexor spasms in the upper limb and extensor spasms in the lower limb, and their movement patterns vary greatly. Constraint-induced movement therapy is an upper limb rehabilitation technique used in stroke patients with hemiplegia; however, studies of lower extremity rehabilitation are scarce. In this study, stroke patients with lower limb hemiplegia underwent conventional Bobath therapy for 4 weeks as baseline treatment, followed by constraint-induced movement therapy for an additional 4 weeks. The 10-m maximum walking speed and Berg balance scale scores significantly improved following treatment, and lower extremity motor function also improved. The results of functional MRI showed that constraint-induced movement therapy alleviates the reduction in cerebral functional activation in patients, which indicates activation of functional brain regions and a significant increase in cerebral blood perfusion. 18Curcumin-incorporated 3D bioprinting gelatin methacryloyl hydrogel reduces reactive oxygen species-induced adipose-derived stem cell apoptosis and improves implanting survival in diabetic wounds显示文摘Background:Gelatin methacryloyl(GelMA)hydrogels loaded with stem cells have proved to be an effective clinical treatment for wound healing.Advanced glycation end product(AGE),interacting with its particular receptor(AGER),gives rise to reactive oxygen species(ROS)and apoptosis.Curcumin(Cur)has excellent antioxidant activity and regulates intracellular ROS production and apoptosis.In this study,we developed a Cur-incorporated 3D-printed GelMA to insert into adiposederived stem cells(ADSCs)and applied it to diabetic wounds.Methods:GelMA hydrogels with Cur were fabricated and their in vitro effects on ADSCs were investigated.We used structural characterization,western blot,ROS and apoptosis assay to evaluate the antioxidant and anti-apoptotic activity,and assessed the wound healing effects to investigate the mechanism underlying regulation of apoptosis by Cur via the AGE/AGER/nuclear factor-κB(NF-κB)p65 pathway.Results:A 10%GelMA scaffold exhibited appropriate mechanical properties and biocompatibility for ADSCs.The circular mesh structure demonstrated printability of 10%GelMA and Cur-GelMA bioinks.The incorporation of Cur into the 10%GelMA hydrogel showed an inhibitory effect on AGEs/AGER/NF-κB p65-induced ROS generation and ADSC apoptosis.Furthermore,Cur-GelMA scaffold promoted cell survival and expedited in vivo diabetic wound healing.Conclusions:The incorporation of Cur improved the antioxidant activity of 3D-printed GelMA hydrogel and mitigated AGE/AGER/p65 axis-induced ROS and apoptosis in ADSCs.The effects of scaffolds on wound healing suggested that Cur/GelMA-ADSC hydrogel could be an effective biological material for accelerating wound healing.Sizhan Xia Tingting Weng Ronghua Jin Min Yang Meirong Yu Wei Zhang Xingang Wang Chunmao Han 2022Burns & Trauma2022,10,1:4

20Microglia modulation with 1070-nm light attenuates Aβ burden and cognitive impairment in Alzheimer’s disease mouse model显示文摘Photobiomodulation,by utilizing low-power light in the visible and near-infrared spectra to trigger biological responses in cells and tissues,has been considered as a possible therapeutic strategy for Alzheimer’s disease(AD),while its specific mechanisms have remained elusive.Here,we demonstrate that cognitive and memory impairment in an AD mouse model can be ameliorated by 1070-nm light via reducing cerebralβ-amyloid(Aβ)burden,the hallmark of AD.The glial cells,including microglia and astrocytes,play important roles in Aβclearance.Our results show that 1070-nm light pulsed at 10 Hz triggers microglia rather than astrocyte responses in AD mice.The 1070-nm lightinduced microglia responses with alteration in morphology and increased colocalization with Aβare sufficient to reduce Aβload in AD mice.Moreover,1070-nm light pulsed at 10 Hz can reduce perivascular microglia and promote angiogenesis to further enhance Aβclearance.Our study confirms the important roles of microglia and cerebral vessels in the use of 1070-nm light for the treatment of AD mice and provides a framework for developing a novel therapeutic approach for AD.Lechan Tao Qi Liu Fuli Zhang Yuting Fu Xi Zhu Xiaofu Weng Hongbin Han Yong Huang Yuanzhen Suo Liang Chen Xiaoling Gao Xunbin Wei 2021Light(Science & Applications)2021,10,10:4
