2Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s显示文摘Ulcerative colitis(UC)is a chronic inflammatory disease of the gastrointestinal tract,which is closely related to gut barrier dysfunction.Emerging evidence shows that interleukin-22(IL-22)derived from group 3 innate lymphoid cells(ILC3s)confers benefits on intestinal barrier,and IL-22 expression is controlled by aryl hydrocarbon receptor(AhR).Previous studies show that baicalein protects the colon from inflammatory damage.In this study we elucidated the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice.Mice were administered baicalein(10,20,40 mg·kg^(-1)·d^(-1),i.g.)for 10 days;the mice freely drank 3%dextran sulfate sodium(DSS)on D1-D7 to induce colitis.We showed that baicalein administration simultaneously ameliorated gut inflammation,decreased intestinal permeability,restored tight junctions of colons possibly via promoting AhR/IL-22 pathway.Co-administration of AhR antagonist CH223191(10 mg/kg,i.p.)partially blocked the therapeutic effects of baicalein in colitis mice,whereas AhR agonist FICZ(1μg,i.p.)ameliorated symptoms and gut barrier function in colitis mice.In a murine lymphocyte line MNK-3,baicalein(5-20μM)dose-dependently increased the expression of AhR downstream target protein CYP1A1,and enhanced IL-22 production through facilitating AhR nuclear translocation,these effects were greatly diminished in shAhR-MNK3 cells,suggesting that baicalein induced IL-22 production in AhR-dependent manner.To further clarify that,we constructed an in vitro system consisting of MNK-3 and Caco-2 cells,in which MNK-3 cell supernatant treated with baicalein could decrease FITC-dextran permeability and promoted the expression of tight junction proteins ZO-1 and occluding in Caco-2 cells.In conclusion,this study demonstrates that baicalein ameliorates colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s,thus providing a potential therapy for UC.Yan-yang Li Xiao-jing Wang Yu-lin Su Qing Wang Shao-wei Huang Zeng-feng Pan Yan-ping Chen Jun-jie Liang Mei-ling Zhang Xue-qian Xie Zhi-yun Wu Jin-yan Chen Lian Zhou Xia Luo 2022Acta Pharmacologica Sinica2022,43,6:14
3Chinese herbal medicine reduces mortality in patients with severe and critical coronavirus disease 2019: a retrospective cohort study显示文摘This study aimed to evaluate the efficacy of Chinese herbal medicine(CHM)in patients with severe/critical coronavirus disease 2019(COVID-19).In this retrospective study,data were collected from 662 patients with severe/critical COVID-19 who were admitted to a designated hospital to treat patients with severe COVID-19 in Wuhan before March 20,2020.All patients were divided into an exposed group(CHM users)and a control group(non-users).After propensity score matching in a 1:1 ratio,156 CHM users were matched by propensity score to 156 non-users.No significant differences in seven baseline clinical variables were found between the two groups of patients.All-cause mortality was reported in 13 CHM users who died and 36 non-users who died.After multivariate adjustment,the mortality risk of CHM users was reduced by 82.2%(odds ratio 0.178,95%CI 0.076–0.418;P<0.001)compared with the non-users.Secondly,age(odds ratio 1.053,95%CI 1.023–1.084;P<0.001)and the proportion of severe/critical patients(odds ratio 0.063,95%CI 0.028–0.143;P<0.001)were the risk factors of mortality.These results show that the use of CHM may reduce the mortality of patients with severe/critical COVID-19.Guohua Chen Wen Su Jiayao Yang Dan Luo Ping Xia Wen Jia Xiuyang Li Chuan Wang Suping Lang Qingbin Meng Ying Zhang Yuhe Ke An Fan Shuo Yang Yujiao Zheng Xuepeng Fan Jie Qiao Fengmei Lian Li Wei Xiaolin Tong 2020Frontiers of Medicine2020,14,6:10

5Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 6Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 7An AGAMOUS-like factor is associated with the origin of two domesticated varieties in Cymbidium sinense (Orchidaceae)显示文摘Cymbidium has been artificially domesticated for centuries in Asia,which produced numerous cultivated varieties.Flowers with stamenoid tepals or those with multiple tepals have been found in different species of Cymbidium;however,the molecular basis controlling the formation of these phenotypes is still largely unknown.Previous work demonstrated that AGAMOUS/AG lineage MADS genes function in floral meristem determinacy as well as in reproductive organs development in both dicots and monocots,indicating a possible relationship with the origin of two flower varieties in Cymbidium.Here,we characterized and analyzed two AG lineage paralogues,CsAG1 and CsAG2,from Cymbidium sinense,both of which were highly expressed in the gynostemium column of a standard C.sinense.Interestingly,we detected ectopic expression of CsAG1 rather than CsAG2 in all floral organs of a stamenoid-tepal variety and significant down-regulation of CsAG1 in a variety with multiple tepals.Over-expression of CsAG1 in wild type Arabidopsis resulted in petal-to-stamen homeotic conversion,suggesting a conserved C-function of CsAG1 in the development of Cymbidium flower.Altogether,our results supported a hypothesis that disruption of a single AG-like factor would be associated with the formation of two domesticated varieties in C.sinense.Shihao Su Xiaoyu Shao Changfa Zhu Jiayin Xu Yuhuan Tang Da Luo Xia Huang 2018Horticulture Research2018,5,1:4
8Microstructural evolution of Al-Cu-Li alloys with different Li contents by coupling of near-rapid solidification and two-stage homogenization treatment显示文摘Microstructural improvement of Al-Cu-Li alloys with high Li content plays a critical role for the acquisition of excellent mechanical properties and ultra-low density.In this regard,the Al-Cu-Li alloy castings with high Li content from 1.5 wt.%to 4.5 wt.%were prepared by near-rapid solidification,followed by two-stage homogenization treatment(490℃/16 h and 530℃/16 h).The microstructural evolution and solidification behavior of the as-cast and homogenized alloys with different Li contents were systematically studied by combining experiments with calculations by Pandat software.The results indicate that with the increase of Li content,the grain sizes decrease,the solution ability of Cu in the matrixα-Al phase increases,while the content of secondary dendrites increases and the precipitated phases change from low melting point phases to high melting point phases under the near-rapid solidification.Additionally,by the coupling of near-rapid solidification and two-stage homogenization,the metastable precipitated phases(Al7Cu4Li and AlCu3)can be dissolved effectively in the alloys with Li content of 1.5 wt.%-2.5 wt.%;moreover,the stable precipitated phases(Al6CuLi3 and Al2CuLi)uniformly distribute at the grain boundaries in the alloys with Li content of 3.5 wt.%-4.5 wt.%.As a result,the refined and homogenized microstructure can be obtained.Lei Luo Liang-shun Luo Zhi-ping Li Hong-ying Xia Yan-qing Su Liang Wang Jing-jie Guo Heng-zhi Fu 2020China Foundry2020,17,3:3
9Artificial antigen-presenting cells plus IL-15 and IL-21 efficiently induce melanoma-specific cytotoxic CD8^+CD28^+ T lymphocyte responses显示文摘Objective:To develop a novel artificial antigen-presenting system for efficiently inducing melanoma-specific CD8+CD28+ cytotoxic T lymphocyte(CTL) responses.Methods:Cell-sized Dynabeads? 11The first data release(DR1) of the LAMOST regular survey显示文摘The Large sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST) general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio(SNR) ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters: late A,FGK-type stars with high quality spectra(1 061 918 entries), A-type stars(100 073 entries), and M-type stars(121 522 entries). This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. 12Effect of growth rate on microstructure and microhardness of directionally solidified Ti-44Al-5Nb-1.5Cr-1.5Zr-1Mo-0.1B alloy显示文摘The effect of growth rates (V=2-50 μm·s-1) on microstructure and microhardness of directionally solidified Ti-44Al-5Nb-1.5Cr-1.5Zr-1Mo-0.1B (at.%) alloy at a constant temperature gradient (G=18 K·mm-1) was investigated. Results indicated that β phase was the primary phase of the directionally solidified Ti-44Al-5Nb-1.5Cr-1.5Zr-1Mo-0.1B alloy. As the growth rate increases, the solid/liquid interface turns from cellular growth to dendric growth. The interlamellar spacing (λs) decreases with the increase of growth rate according to the relationship of λs=3.39V -0.31. The solidification segregation occurs due to the enrichment of β-stabilizing element Nb, Cr in primary β phase during solidification;moreover, the degree of the segregation increases with the growth rate, resulting in the emergence of B2 phase in lamellar colonies at high growth rates. 16Effects of low ambient temperatures and dietary vitamin C supplementation on pulmonary vascular remodeling and hypoxic gene expression of 21-d-old broilers显示文摘The objective of this study was to evaluate the effects of low ambient temperature(LAT) and dietary vitamin C(VC)supplementation on pulmonary vascular remodeling(PVR) and the relative expression of hypoxia inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF) and its receptor 2(VEGFR-2) mRNA of lungs in 21-d-old broilers.4001-d-old male Cobb broilers were assigned randomly to 4 treatments as follows for 21 d:1) LAT and a basal diet;2) LAT and a basal diet supplemented with 1 000 mg kg^(-1) VC(LAT+VC);3) normal ambient temperature(NAT) and a basal diet;4) NAT and a basal diet supplemented with 1 000 mg kg^(-1) VC(NAT+VC).Each treatment was composed of 10 replicates of 10 birds per replicate.Samples of lung were collected after the broilers were killed at d 21.LAT increased the ratio of vessel wall area to vessel total area(WA/TA,%) and mean media thickness in pulmonary arterioles(mMTPA,%)(P<0.05).Dietary VC supplementation decreased mMTPA(P<0.05),but had no effect on the WA/TA.LAT increased(P<0.05) the relative mRNA expression of HIF-1α,VEGF and VEGFR-2,while adding VC to the diet could decrease(P<0.05) their relative mRNA expression.A significant positive correlation existed between the level of VEGF mRNA expression and the value of WA/WT(P<0.05) or mMTPA(P<0.05).These results suggested LAT resulted in pulmonary vascular remodeling,and the increase of HIF-1a,VEGF and VEGFR-2 mRNA expression,and dietary VC supplementation can alleviate pulmonary vascular remodeling in broiler by affecting these gene expression.ZENG Qiu-feng YANG Xia ZHENG Ping ZHANG Ke-ying LUO Yu-heng DING Xue-mei BAI Shi-ping WANG Jian-ping XUAN Yue SU Zhuo-wei 2016Journal of Integrative Agriculture2016,15,1:1

19Application of the Mathieu combined tunnel technique for repairing glans dehiscence after failed hypospadias repair显示文摘Repairing glans dehiscence after failed hypospadias repair is challenging for pediatric surgeons.Here,we introduced and evaluated a newly modified Mathieu technique,Mathieu combined tunnel(MCT),which involves multiple custom-designed flaps for the shortage of flap source material after repeated operations;we also constructed a tunnel to avoid the glans incision that may carry new risks of dehiscence.This retrospective study included 26 patients who were consecutively admitted to the First Affiliated Hospital of Sun Yat-Sen University(Guangzhou,China)for glans dehiscence repair after failed hypospadias repair from October 2014 to October 2020;sixteen patients underwent surgery using the MCT(MCT group)and ten patients underwent surgery using the tubularized incised plate(TIP)technique(TIP group).The operative time,blood loss,postoperative complications,normal urethral meatus rate,success rate,and Hypospadias Objective Penile Evaluation(HOPE)score were compared between the two groups.The MCT group achieved an overall satisfactory penile appearance and voiding function,with a higher rate of normal urethral meatus(15/16,93.8%)and a lower rate of glans dehiscence(1/16,6.2%),compared with the TIP group(70.0%and 30.0%,respectively).However,these differences were not statistically significant,possibly because of the limited number of patients(all P>0.05).Mean postoperative HOPE scores were similar in the MCT group(mean±standard deviation:8.83±0.89)and TIP group(8.94±0.57)(P>0.05).No significant differences were found between the two groups in terms of blood loss and success rate,nor in the rates of various complications(e.g.,fistula,urethral stricture,and glans dehiscence).In conclusion,the MCT technique appears to be feasible and reliable for repairing glans dehiscence after failed hypospadias repair.Qi-Gen Xie Kai Xia Xiang-Ping Li Peng Luo Zuo-Qing Li Cheng Su Chun-Hua Deng 2022Asian Journal of Andrology2022,24,3:1
