| 1 | Effect of moxa smoke produced during combustion of Aiye(Folium Artemisiae Argyi) on behavioral changes in mice inhaling the smoke显示文摘OBJECTIVE: To evaluate the effects that inhalation of the combustion products of the traditional Chinese herb Aiye(Folium Artemisiae Argyi) has on the central nervous system.METHODS: Forty Kunming mice(half male) were randomly assigned(n = 10/group) to a control group(C) or one of three moxa smoke concentration groups(% opacity): low(L1; 0.4%), medium(M1; 2%), and high(H1; 15%). Mice in the latter three groups were exposed to moxa smoke in a dynamic gas exposure cabinet for 20 min per day for7 days. Mice in control group were placed in the same cabinet without any intervention. For the sleep experiments, another 50 mice were divided into five groups of 10 mice each: a saline-injected control group, L1 + pentobarbital sodium(PS)-injected group, M1+PS group, H1(15%)+PS group,and a positive control group(10 mg/kg, chlorpromazine, p.o.). The weight, general activities, locomotor activities, rotarod performance, sleep duration,and sleeping rate induced by a subthreshold dose of pentobarbital sodium were recorded in the mice,and the composition of moxa smoke was analyzed using headspace gas chromatography(GC-HS).RESULTS: A low concentration of smoke significantly decreased the frequency of locomotor activities and the time for which the mice remaining on the rotarod; however, a high smoke concentration significantly prolonged the pentobarbital-induced sleeping time and sleeping rate.CONCLUSION: The concentration-dependent relaxing effects of moxa smoke on the Central Nervous System(CNS) were confirmed. Moreover, GC-HS analysis showed that the component present in the highest concentration in moxa smoke was eucalyp-tol, an essential oil well recognized for its soothing effects on the CNS. | 2 | Effects of moxibustion or moxa smoke on serum lipids and carotid plaque stability in atherosclerotic mice显示文摘Objective:To observe the effects of moxibustion or moxa smoke on serum lipids,aorta and liver pathology,and carotid plaque stability in atherosclerosis.Methods:Fifty-four 8-week-old ApoE^-/- mice were randomly divided into three groups(untreated,moxibustion,and moxa smoke)and received a high-fat diet.Eighteen wild-type C57 BL/6 mice of the same age were used as controls.The intervention(none,moxibustion between the nipples,or 10 e15 mg/m^3 moxa smoke)was applied to restrained mice 20 min per day,six days per week,for 12 weeks.At the end of the experimental period,we measured serum lipids and apolipoprotein,stained thoracic aortas and livers to observe pathological changes,and used immunohistochemical staining to assess the levels of a-smooth muscle actin,CD68,tumor necrosis factor-α,nuclear transcription factor-κB,and P38 mitogen-activated protein kinase.We also measured the levels of matrix metalloproteinase-2 and 9 and tissue metalloproteinase inhibitor-1.Results:After 12 weeks,lipid metabolism disorder and atherosclerotic plaques were observed in the ApoE^-/- mice.Moxibustion or moxa smoke reduced the levels of serum total cholesterol,triglycerides,low density lipoprotein,and very low density lipoprotein but did not affect the levels of high density lipoprotein,apolipoprotein A1,or oxidized low density lipoprotein.Moxibustion or moxa smoke suppressed pathological changes in thoracic aortas and livers,increased fiber cap thickness,the fiber cap thickness/intimal medial thickness ratio,and collagen area percentage,and reduced extracellular lipids.Treatment with moxibustion or moxa smoke increased a-smooth muscle actin and reduced CD68 and the vulnerability index,suppressed tumor necrosis factor-α and nuclear transcription factor-κB expression,and did not affect P38 mitogen-activated protein kinase expression.Treatment lowered the levels of matrix metalloproteinase-2 and 9 and increased those of tissue metalloproteinase inhibitor-1.Conclusion:Moxibustion or moxa smoke exert protective effects in serum lipid profiles and carotid plaque stability in atherosclerotic mice by regulating plaque stability,inflammatory factors,and matrix metalloproteinases. | Rui Zhang Lue Ha Rui He Yueping Huang Hao Wang Baixiao Zhao | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,3: | 2 |

| 4 | Lead acetate in drinking water is toxic to hippocampal tissue Measuring relative protein changes using tissue array detection显示文摘BACKGROUND: Lead can cause structural changes in the hippocampus, followed by damage to learning and memory functions, but its specific mechanisms are not yet clear. OBJECTIVE: To observe long-term toxicity of high-dose lead in drinking water on hippocampal tissue in rats, and analyze the potential association of oxidative damage, cell apoptosis, and pathology. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Center for Medical Experiment, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from May 2007 to October 2008. MATERIALS: Rabbit anti Bcl-2, Bax, and inducible nitric oxide synthase (iNOS) polyclonal antibodies were purchased from Santa Cruz Biotechnology, USA. An streptavidin-peroxidase immunohistochemistry kit and concentrated DAB kit were purchased from Beijing Zhongshan Biotechnology Company Limited, China. Crystal violet was purchased from Sigma, USA. METHODS: A total of 72 Wistar rats, aged 3 months, were randomly divided into control, low-, middle-, and high-dose lead groups, with 18 rats per group. Animal models were established through free drinking water contaminated by Pb2+ for 1, 3, and 6 months, respectively. MAIN OUTCOME MEASURES: The general toxicity of lead was dynamically observed; the levels of Pb2+ in the blood and brain tissue homogenate were detected using atomic absorption method; pathological changes were observed using hematoxylin-eosin staining and tigroid body staining; the protein expression levels of Bcl-2, Bax, and iNOS were dynamically observed using streptavidin-peroxidase immunohistochemistry of the hippocampus. RESULTS: Lead exposure reduced autonomic activities, produced a slumped appearance, slow responses, and lusterless fur, especially in the high-dose group. The amount of ingestion and hydroposia showed a decreasing trend, especially in middle- and high-dose groups. Lead levels in whole blood and brain homogenate were higher than controls (P < 0.01). Lead caused degeneration of hippocampal neurons and pyknosis, with fewer tigroid bodies, especially in high-dose lead group. Bcl-2 expression decreased with increasing lead dose (P < 0.01), whereas lead dose-dependently increased Bax levels (P < 0.01) and iNOS levels (P < 0.05). CONCLUSION: High levels of Pb2+ may disrupt hippocampal structure by passing through the blood brain barrier.
