| 1 | Regulation of innate immune responses by rabies virus显示文摘Rabies virus(RABV)is an infectious and neurotropic pathogen that causes rabies and infects humans and almost all warm-blooded animals,posing a great threat to people and public safety.It is well known that innate immunity is the critical first line of host defense against viral infection.It monitors the invading pathogens by recognizing the pathogen-associated molecular patterns and danger-associated molecular patterns through pattern-recognition receptors,leading to the production of type I interferons(IFNα/β),inflammatory cytokines,and chemokines,or the activation of autophagy or apoptosis to inhibit virus replication.In the case of RABV,the innate immune response is usually triggered when the skin or muscle is bitten or scratched.However,RABV has evolved many ways to escape or even hijack innate immune response to complete its own replication and eventually invades the central nervous system(CNS).Once RABV reaches the CNS,it cannot be wiped out by the immune system or any drugs.Therefore,a better understanding of the interplay between RABV and innate immu-nity is necessary to develop effective strategies to combat its infection.Here,we review the innate immune responses induced by RABV and illustrate the antagonism mechanisms of RABV to provide new insights for the control of rabies. | Haili Zhang Jingbo Huang Yumeng Song Xingqi Liu Meichen Qian Pei Huang Yuanyuan Li Ling Zhao Hualei Wang | 2022 | Animal Models and Experimental Medicine2022,5,5: | 2 |
| 2 | Clay nanosheet-mediated delivery of recombinant plasmids expressing artificial miRNAs via leaf spray to prevent infection by plant DNA viruses显示文摘Whitefly-transmitted begomoviruses are economically important plant pathogens that cause severe problems in many crop plants,such as tomato,papaya,cotton,and tobacco.Tomato yellow leaf curl virus(TYLCV)is a typical monopartite begomovirus that has been extensively studied,but methods that can efficiently control begomoviruses are still scarce.In this study,we combined artificial microRNA(amiRNA)-mediated silencing technology and clay nanosheetmediated delivery by spraying and developed a method for efficiently preventing TYLCV infection in tomato plants.We designed three amiRNAs that target different regions of TYLCV to silence virus-produced transcripts.Three plant expression vectors expressing pre-amiRNAs were constructed,and recombinant plasmid DNAs(pDNAs)were loaded onto nontoxic and degradable layered double hydroxide(LDH)clay nanosheets.LDH nanosheets containing multiple pDNAs were sprayed onto plant leaves.We found that the designed amiRNAs were significantly accumulated in leaves 7 days after spraying,while the pDNAs were sustainably detected for 35 days after the spray,suggesting that the LDH nanosheets released pDNAs in a sustained manner,protected pDNAs from degradation and efficiently delivered pDNAs into plant cells.Importantly,when the LDH nanosheets coated with pDNAs were sprayed onto plants infected by TYLCV,both the disease severity and TYLCV viral concentration in sprayed plants were significantly decreased during the 35 days,while the levels of H_(2)O_(2) were significantly increased in those plants.Taken together,these results indicate that LDH nanosheets loaded with pDNAs expressing amiRNAs can be a sustainable and promising tool for begomovirus control. | Qili Liu Yanpeng Li Kedong Xu Dongxiao Li Haiyan Hu Feng Zhou Puwen Song Yongang Yu Qichao Wei Qian Liu Weipeng Wang Ruifang Bu Haili Sun Xiaohui Wang Jianjun Hao Honglian Li Chengwei Li | 2020 | Horticulture Research2020,7,1: | 1 |

| 10 | An inactivated recombinant rabies virus chimerically expressed RBD induces humoral and cellular immunity against SARS-CoV-2 and RABV显示文摘Many studies suggest that severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)can infect various animals and transmit among animals,and even to humans,posing a threat to humans and animals.There is an urgent need to develop inexpensive and efficient animal vaccines to prevent and control coronavirus disease 2019(COVID-19)in animals.Rabies virus(RABV)is another important zoonotic pathogen that infects almost all warmblooded animals and poses a great public health threat.The present study constructed two recombinant chimeric viruses expressing the S1 and RBD proteins of the SARS-CoV-2 Wuhan01 strain based on a reverse genetic system of the RABV SRV9 strain and evaluated their immunogenicity in mice,cats and dogs.The results showed that both inactivated recombinant viruses induced durable neutralizing antibodies against SARS-CoV-2 and RABV and a strong cellular immune response in mice.Notably,inactivated SRV-nCoV-RBD induced earlier antibody production than SRV-nCoV-S1,which was maintained at high levels for longer periods.Inactivated SRV-nCoV-RBD induced neutralizing antibodies against both SARS-CoV-2 and RABV in cats and dogs,with a relatively broadspectrum cross-neutralization capability against the SARS-CoV-2 pseudoviruses including Alpha,Beta,Gamma,Delta,and Omicron,showing potential to be used as a safe bivalent vaccine candidate against COVID-19 and rabies in animals. | Haili Zhang Hongli Jin Feihu Yan Yumeng Song Jiaxin Dai Cuicui Jiao Yujie Bai Jingxuan Sun Di Liu Shen Wang Mengyao Zhang Jilong Lu Jingbo Huang Pei Huang Yuanyuan Li Xianzhu Xia Hualei Wang | 2023 | Virologica Sinica2023,38,2: | 0 |
| 11 | Hepatic Ischemia-reperfusion Injury in Mice was Alleviated by Rac1 Inhibition - More Than Just ROS-inhibition显示文摘Background and Aims:Reducing reactive oxygen species(ROS)production has proven an effective way for allevi-ating oxidative stress during ischemia-reperfusion injury(IRI).Moreover,inhibition of Rac1 could reduce ROS pro-duction and prevent oxidative stress injury.Previous stud-ies have suggested a positive interactivation feedback loop between Rac1 and hypoxia-inducible factor(HIF)-1α,the latter being up-regulated early during ischemia.The posi-tive inter-activation between Rac1 and HIF-1αwould ag-gravate ROS production,thereby promoting IRI.This study was designed to verify the effects of Rac1 inhibition on he-patic IRI both at animal and cellular levels and to explore the interaction between Rac1 and HIF-1αduring hepatic IRI.Methods:C57B/6 mice and AML-12 cells were used for the construction of hepatic IRI animal and cell models.Rac1 inhibition was achieved by NSC23766(a specific Rac1 inhibitor).Lentiviral vectors were used for Rac1 knock-down.At designated time points,serum and liver tissues were collected from the mice and treated cells were col-lected for further analysis.Results:NSC23766 treatment significantly alleviated the hepatic IRI in mice,manifesting as lower vacuolation score and less apoptosis cells,lower ROS and serum/liver alanine aminotransferase/aspartate aminotransferase levels,and fewer activated inflammatory cells.IRI of AML-12 was also alleviated by 50μM NSC23766 or Rac1-knockdown,manifesting as reduced cell apoptosis,less extensive interruption of mitochondrial membrane po-tential,down-regulation of apoptosis,and effects on DNA damage-related proteins.Interestingly,Rac1 knockdown also down-regulated the expression level of HIF-1α.Con-clusions:Our study supports a protective effect of Rac1 inhibition on hepatic IRI.Aside from the classic topics of reducing ROS production and oxidative stress,our study showed an interaction between Rac1 and HIF-1αsignaling during hepatic IRI. | Zhilin Sha Yajie Yang Ruling Liu Haili Bao Shaohua Song Junfeng Dong Meng Guo Yuanyu Zhao Hu Liu Guoshan Ding | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 0 |
| 12 | National incidence of joint dislocation in China:a retrospective survey of 512,187 individuals显示文摘Background:Joint dislocations significantly impact public health.However,a comprehensive study on the incidence,distribution,and risk factors for joint dislocations in China is lacking.We conducted the China National Joint Dislocation Study,which is a part of the China National Fracture Study conducted to obtain the national incidence and risk factors for traumatic fractures,and to investigate the incidence and risk factors for joint dislocations.Methods:For this national retrospective epidemiological study,512,187 participants were recruited using stratified random sampling and probability-proportional-to-size method from January 19 to May 16,2015.Participants who sustained joint dislocations of the trunk,arms,or legs(skull,sternum,and ribs being excluded)in 2014 were personally interviewed to obtain data on age,educational background,ethnic origin,occupation,geographic region,and urbanization degree.The joint-dislocation incidence was calculated based on age,sex,body site,and demographic factors.The risk factors for different groups were examined using multiple logistic regression.Results:One hundred and nineteen participants sustained 121 joint dislocations in 2014.The population-weighted incidence rate of joint dislocations of the trunk,arms,or legs was 0.22(95%confidence interval[CI]:0.16,0.27)per 1000 population in 2014(men,0.27[0.20,0.34];women,0.16[0.10,0.23]).For all ages,previous dislocation history(male:OR 42.33,95%confidence interval[CI]:12.03-148.90;female:OR 54.43,95%CI:17.37-170.50)and alcohol consumption(male:OR 3.50,95%CI:1.49-8.22;female:OR 2.65,95%CI:1.08-6.50)were risk factors for joint dislocation.Sleeping less than 7 h/day was a risk factor for men.Compared with children,women aged≥15 years(female 15-64 years:OR 0.16,95%CI:0.04-0.61;female≥65 years:OR 0.06,95%CI:0.01-0.41)were less likely to sustain joint dislocations.Women with more than three children were at higher dislocation risk than women without children(OR 6.92,95%CI:1.18-40.78).Conclusions:The up-to-date data on joint dislocation incidence,distribution,and risk factors can be used as a reference for national healthcare,prevention,and management in China.Specific strategies for decreasing alcohol consumption and encouraging adequate sleeping hours should be developed to prevent or reduce dislocation incidents.Trial Registration:Chinese Clinical Trial Registry,ChiCTR-EPR-15005878. | Hongzhi Lv Wei Chen Zhiyong Hou Siming Jia Yanbin Zhu Bo Liu Xiao Chen Guang Yang Lei Liu Tao Zhang Haili Wang Bing Yin Song Liu Jialiang Guo Xiaolin Zhang Yichong Li Yingze Zhang | 2022 | Chinese Medical Journal2022,,14: | 0 |
| 13 | The therapy and mechanisms of replication-deficient recombinant adenovirus Ad-p14ARF in hepatocellular carcinoma显示文摘Objective: To study the therapeutic effect and mechanisms of recombinant adenovirus Ad-p14ARF in hepatocel- lular carcinoma cell lines. Methods: Morphology and trypan blue assay were adopted to evaluate the proliferation of different liver cancer cells after Ad-p14ARF infection. Cell apoptosis was confirmed by detecting phosphatidylserine (PS) externaliza- tion with Annexin V/PI double staining. Western blotting assay analyzed the expression of related proteins. Subcutaneous tumor model of BEL7402 was established to evaluate the therapeutic ability of Ad-p14ARF. Results: Ad-p14ARF suppressed cell growth, proliferation and promoted cell apoptosis of cancer cell lines with different genetic background. Ad-p14ARF in- hibited growth of liver cancer cells (HepG2, BEL7402) in a dose-dependent manner. Ad-p14ARF leaded to overexpression of Bax and p21, which were the downstream regulating genes of p53. Ad-p14ARF suppressed tumor growth significantly in the experimental therapy in nude mice bearing subcutaneous tumor of BEL7402. Conclusion: P14ARF gene is a powerful tumor suppressor gene to be used in cancer gene therapy.
