| 1 | Combined obeticholic acid and apoptosis inhibitor treatment alleviates liver fibrosis显示文摘Obeticholic acid(OCA), the first FXR-targeting drug, has been claimed effective in the therapy of liver fibrosis. However, recent clinical trials indicated that OCA might not be effective against liver fibrosis, possibly due to the lower dosage to reduce the incidence of the side-effect of pruritus. Here we propose a combinatory therapeutic strategy of OCA and apoptosis inhibitor for combating against liver fibrosis. CCl4-injured mice, D-galactosamine/LPS(GalN/LPS)-treated mice and cycloheximide/TNFα(CHX/TNFα)-treated HepG2 cells were employed to assess the effects of OCA, or together with IDN-6556, an apoptosis inhibitor. OCA treatment significantly inhibited hepatic stellate cell(HSC)activation/proliferation and prevented fibrosis. Elevated bile acid(BA) levels and hepatocyte apoptosis triggered the activation and proliferation of HSCs. OCA treatment reduced BA levels but could not inhibit hepatocellular apoptosis. An enhanced anti-fibrotic effect was observed when OCA was co-administrated with IDN-6556. Our study demonstrated that OCA inhibits HSCs activation/proliferation partially by regulating BA homeostasis and thereby inhibiting activation of HSCs. | 3 | Autophagy enhanced by curcumin ameliorates inflammation in atherogenesis via the TFEB-P300-BRD4 axis显示文摘Disturbance of macrophage-associated lipid metabolism plays a key role in atherosclerosis.Crosstalk between autophagy deficiency and inflammation response in foam cells(FCs) through epigenetic regulation is still poorly understood. Here, we demonstrate that in macrophages, oxidized lowdensity lipoprotein(ox-LDL) leads to abnormal crosstalk between autophagy and inflammation,thereby causing aberrant lipid metabolism mediated through a dysfunctional transcription factor EB (TFEB)-P300-bromodomain-containing protein 4(BRD4) axis. ox-LDL led to macrophage autophagy deficiency along with TFEB cytoplasmic accumulation and increased reactive oxygen species generation. This activated P300 promoted BRD4 binding on the promoter regions of inflammatory genes, consequently contributing to inflammation with atherogenesis. Particularly, ox-LDL activated BRD4-dependent super-enhancer associated with liquid-liquid phase separation(LLPS) on the regulatory regions of inflammatory genes. Curcumin(Cur) prominently restored FCs autophagy by promoting TFEB nuclear translocation, optimizing lipid catabolism, and reducing inflammation. The consequences of P300 and BRD4 on super-enhancer formation and inflammatory response in FCs could be prevented by Cur. Furthermore, the anti-atherogenesis effect of Cur was inhibited by macrophage-specific Brd4 overexpression or Tfeb knock-out in Apoe knock-out mice via bone marrow transplantation. | 4 | In vivo degradation and endothelialization of an iron bioresorbable scaffold显示文摘Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds(BRSs).In particular,it is urgent to establish a nondestructive approach to examine in vivo degradation of a new-generation coronary stent for interventional treatment based on mammal experiments;otherwise it is not available to semi-quantitatively monitor biodegradation in any clinical trial.Herein,we put forward a semi-quantitative approach to measure degradation of a sirolimus-eluting iron bioresorbable scaffold(IBS)based on optical coherence tomography(OCT)images;this approach was confirmed to be consistent with the present weight-loss measurements,which is,however,a destructive approach.The IBS was fabricated by a metal-polymer composite technique with a polylactide coating on an iron stent.The efficacy as a coronary stent of this new bioresorbable scaffold was compared with that of a permanent metal stent with the name of trade mark Xience,which has been widely used in clinic.The endothelial coverage on IBS was found to be greater than on Xience after implantation in a rabbit model;and our well-designed ultrathin stent exhibited less individual variation.We further examined degradation of the IBSs in both minipig coronary artery and rabbit abdominal aorta models.The present result indicated much faster iron degradation of IBS in the rabbit model than in the porcine model.The semi-quantitative approach to detect biodegradation of IBS and the finding of the species difference might be stimulating for fundamental investigation of biodegradable implants and clinical translation of the next-generation coronary stents. | Wenjiao Lin Hongjie Zhang Wanqian Zhang Haiping Qi Gui Zhang Jie Qian Xin Li Li Qin Haifeng Li Xiang Wang Hong Qiu Xiaoli Shi Wei Zheng Deyuan Zhang Runlin Gao Jiandong Ding | 2021 | Bioactive Materials2021,6,4: | 3 |

| 6 | Pollination ecology in China from 1977 to 2017显示文摘China is one of most biodiverse countries in the world, containing at least 10% of all angiosperm species.Therefore, we should anticipate a diverse, pollinator fauna. China also has a long history of applied ethnobiology, including a sustainable agriculture based on apiculture and plant-pollinator interactions.However, the science of pollination ecology is a far younger sub-discipline in China, compared to in the West. Chinese studies in pollination ecology began in the 1970 s. For this review, we compiled a complete reference database(>600 publications) of pollination studies in China. Using this database, we identified and analyzed gaps and limitations in research on the pollination systems of native and naturalized species. Specifically, we asked the following questions: 1) What do we know about the pollination systems of native, Chinese species? 2) How does Chinese pollination ecology compare with the development of pollination research abroad and which aspects of research should be pursued by Chinese anthecologists in the near future? 3) What research on pollination in China will advance our understanding and contribute to our ongoing analyses of endemism and conservation? Subsequently, we segregated and identified prospective lines of future research that are unique to China and can only be done in China.
