2Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32
3Endostatin inhibits hypertrophic scarring in a rabbit ear model显示文摘Objective:The present study was designed to use an in vivo rabbit ear scar model to investigate the efficacy of systemic administration of endostatin in inhibiting scar formation.Methods:Eight male New Zealand white rabbits were randomly assigned to two groups.Scar model was established by making six full skin defect wounds in each ear.For the intervention group,intraperitoneal injection of endostatin was performed each day after the wound healed(about 15 d post wounding).For the control group,equal volume of saline was injected.Thickness of scars in each group was measured by sliding caliper and the scar microcirculatory perfusion was assessed by laser Doppler flowmetry on Days 15,21,28,and 35 post wounding.Rabbits were euthanatized and their scars were harvested for histological and proteomic analyses on Day 35 post wounding.Results:Macroscopically,scars of the control group were thicker than those of the intervention group.Significant differences between the two groups were observed on Days 21 and 35(p<0.05).Scar thickness,measured by scar elevation index(SEI) at Day 35 post wounding,was significantly reduced in the intervention group(1.09±0.19) compared with the controls(1.36±0.28).Microvessel density(MVD) observed in the intervention group(1.73±0.94) was significantly lower than that of the control group(5.63±1.78) on Day 35.The distribution of collagen fibers in scars treated with endostatin was relatively regular,while collagen fibers in untreated controls were thicker and showed disordered alignment.Western blot analysis showed that the expressions of type I collagen and Bcl-2 were depressed by injection of endostatin.Conclusions:Our results from the rabbit ear hypertrophic scar model indicate that systemic application of endostatin could inhibit local hypertrophic scar formation,possibly through reducing scar vascularization and angiogenesis.Our results indicated that endostatin may promote the apoptosis of endothelial cells and block their release of platelet-derived growth factor(PDGF) and fibroblast growth factor(FGF),thereby controlling collagen production by fibroblasts.Blood vessel-targeted treatment may be a promising strategy for scar therapy.Hai-tao REN Hang HU Yuan LI Hong-fei JIANG Xin-lei HU Chun-mao HAN 2013Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2013,14,3:17
4The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 5Aloe-emodin exerts cholesterol-lowering effects by inhibiting proprotein convertase subtilisin/kexin type 9 in hyperlipidemic rats显示文摘Hyperlipidemia(HPL)characterized by metabolic disorder of lipids and cholesterol is one of the important risk factors for cardiovascular diseases.Proprotein convertase subtilsin/kexin type 9(PCSK9)is a potent circulating regulator of LDL through its ability to induce degradation of the low-density lipoprotein cholesterol receptor(LDLR)in the lysosome of hepatocytes.Aloe-emodin(AE)is one of potentially bioactive components of Chinese traditionalmedicine Daming capsule.In this study we evaluated the HPL-lowering efficacy of AE in both in vivo and in vitro HPL models.High-fat diet-induced rats were treated with AE(100 mg/kg per day,ig)for 6 weeks.We found that AE administration significantly decreased the levels of total cholesterol(TC)and LDL in the serum and liver tissues.Moreover,AE administration ameliorated HPL-induced hepaticlipid aggregation.But AE administration did not significantly inhibit HMG-CoA reductase activity in the liver of HPL rats.A cellular model of HPL was established in human hepatoma(HepG2)cells treated with cholesterol(20 μg/mL)and 25-hydroxycholesterol(2 μg/mL),which exhibited markedly elevated cholesterol levels.The increased cholesterol levels could be reversed by subsequent treatment with AE(30 μM).In both the in vivo and in vitro HPL models,we revealed that AE selectively suppressed the stero-regulatory element-binding protein-2(SREBP-2)and hepatocyte nuclear factor(HNF)1α-mediated PCSK9 signaling,which in turn upregulated LDL receptor(LDLR)and promoted LDL uptake.This study demonstrates that AE reduces cholesterol content in HPL rats by inhibiting the hepatic PCSK9/LDLR pathway.Zhen-li Su Peng-zhou Hang Juan Hu Yu-yang Zheng Han-qi Sun Jing Guo Ke-yu Liu Zhi-min Du 2020Acta Pharmacologica Sinica2020,41,8:14

7Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 8Platelet count combined with right liver volume and spleen volume measured by magnetic resonance imaging for identifying cirrhosis and esophageal varices显示文摘AIM: To determine whether the combination of platelet count(PLT) with spleen volume parameters and right liver volume(RV) measured by magnetic resonance imaging(MRI) could predict the Child-Pugh class of liver cirrhosis and esophageal varices(EV).METHODS: Two hundred and five cirrhotic patients with hepatitis B and 40 healthy volunteers underwent abdominal triphasic-enhancement MRI and laboratory examination of PLT in 109/L. Cirrhotic patients underwent endoscopy for detecting EV. Spleen maximal width(W), thickness(T) and length(L) in mm together with spleen volume(SV) and RV in mm3 were measured by MRI, and spleen volume index(SI) in mm3 was obtained by W × T × L. SV/PLT, SI/PLT and RV × PLT/SV(RVPS) were calculated and statistically analyzed to assess cirrhosis and EV.RESULTS: SV/PLT(r = 0.676) and SI/PLT(r = 0.707) increased, and PLT(r =-0.626) and RVPS(r =-0.802) decreased with the progress of Child-Pugh class(P < 0.001 for all). All parameters could determine the presence of cirrhosis, distinguish between each class of Child-Pugh class, and identify the presence of EV [the areas under the curve(AUCs) = 0.661-0.973]. 9On design of physical random access channel in high-speed railway LTE systems显示文摘For the uplink in the long-term evolution(LTE)systems,the physical random access channel(PRACH)adopts Zadoff–Chu(ZC) sequences as preamble sequences,and each user can choose a unique cyclic shift for user identification.In high-mobility scenario,the Doppler frequency shift raises up the false alarm probability by inducing the correlation peak offsetting at the receiver side.Traditional detection method utilizes multiple search windows for searching the potential uplink users.When the Doppler frequency shift is larger than the subcarrier spacing,the energy of correlation peak leaks outside the search window and increases the false alarm probability.In this paper,we present a novel design for PRACH preamble sequence for high-speed railway scenario.Through numerical observation,we find out that the offset of the correlation peaks only depends on the initial root sequence number and the length of ZC sequences.Therefore,the root number of ZC sequences should be carefully designed to generate a preamble sequence to fight against the Doppler frequency shift.Consequently,those offsets of the correlation peaks are concentrated within a single search window,resulting in lower complexity of detection and lower probability of false alarm for random access in LTE uplink.Gang Wu Hang Yuan Wei Guo Su Hu Shaoqian Li 2014Chinese Science Bulletin2014,59,35:10
10Albumin and magnetic resonance imaging-liver volume to identify hepatitis B-related cirrhosis and esophageal varices显示文摘AIM:To investigate whether liver lobe volume and albumin(ALB) could predict the presence and severity of liver cirrhosis,and esophageal varices.METHODS:Seventy-one cirrhotic patients with hepatitis B and 21 healthy individuals were enrolled in this study.All the participants underwent abdominal enhanced magnetic resonance imaging to measure each liver lobe volume,and biochemical workup for testing ALB and Child-Pugh class.All cirrhotic patients underwent upper gastrointestinal endoscopy to show the presence of cirrhotic esophageal varices.Right liver lobe volume(RV),left medial liver lobe volume(LMV),left lateral liver lobe volume(LLV),and caudate lobe volume(CV) were measured using enhanced magnetic resonance imaging.The ratios of RV to ALB(RV/ALB),LMV to ALB(LMV/ALB),LLV to ALB(LLV/ALB) and CV to ALB(CV/ALB) were calculated.Statistical analyses were performed to determine whether and how the combination of liver lobe volume measured using magnetic resonance imaging and albumin could predict the presence and severity of liver cirrhosis,and the presence of esophageal varices.RESULTS:RV,LMV,LLV and CV decreased(r =-0.51-0.373; all P < 0.05),while RV/ALB increased(r = 0.424; P < 0.05),with the progress of Child-Pugh classof liver cirrhosis.RV,LMV,CV,LLV/ALB and CV/ALB could identify presence of liver cirrhosis; LLV and LMV could distinguish Child-Pugh class A from B; RV,LMV,LLV,CV,RV/ALB and LLV/ALB could distinguish class A from C; RV and LLV/ALB could differentiate B from C; and RV,RV/ALB and CV/ALB could identify presence of esophageal varices(all P < 0.05).Among these parameters,CV/ALB could best identify the presence of liver cirrhosis,with an area under receiver operating characteristic curve(AUC) of 0.860,a sensitivity of 82.0% and a specificity of 83.0%.LLV could best distinguish class A from B,with an AUC of 0.761,a sensitivity of 74.4% and a specificity of 73.1%.RV could best distinguish class A from C,with an AUC of 0.900,a sensitivity of 90.3% and a specificity of 84.5%.LLV/ALB could best distinguish class B from C,with an AUC of 0.900,a sensitivity of 93.8% and a specificity of 81.5%.RV/ALB could best identify esophageal varices,with an AUC of 0.890,a sensitivity of 80.0% and a specificity of 83.5%.CONCLUSION:The combination of liver lobe volume and ALB has potential to identify presence and severity of cirrhosis,and presence of esophageal varices.Hang Li Tian-Wu Chen Zhen-Lin Li Xiao-Ming Zhang Cheng-Jun Li Xiao-Li Chen Guang-Wen Chen Jia-Ni Hu Yong-Quan Ye 2015World Journal of Gastroenterology2015,21,3:9

13Daytime HONO formation in the suburban area of the megacity Beijing, China显示文摘Nitrous acid(HONO),as a primary precursor of OH radicals,has been considered one of the most important nitrogencontaining species in the atmosphere.Up to 30%of primary OH radical production is attributed to the photolysis of HONO.However,the major HONO formation mechanisms are still under discussion.During the Campaigns of Air Quality Research in Beijing and Surrounding Region(CAREBeijing2006)campaign,comprehensive measurements were carried out in the megacity Beijing,where the chemical budget of HONO was fully constrained.The average diurnal HONO concentration varied from 0.33 to 1.2 ppbv.The net OH production rate from HONO,POH(HONO)net,was on average(from 05:00 to 19:00)7.1×106 molecule/(cm3 s),2.7 times higher than from O3 photolysis.This production rate demonstrates the important role of HONO in the atmospheric chemistry of megacity Beijing.An unknown HONO source(Punknown)with an average of 7.3×106molecule/(cm3 s)was derived from the budget analysis during daytime.Punknown provided four times more HONO than the reaction of NO with OH did.The diurnal variation of Punknown showed an apparent photo-enhanced feature with a maximum around 12:00,which was consistent with previous studies at forest and rural sites.Laboratory studies proposed new mechanisms to recruit NO2 and J(NO2)in order to explain a photo-enhancement of of Punknown.In this study,these mechanisms were validated against the observation-constraint Punknown.The reaction of exited NO2 accounted for only 6%of Punknown,and Punknown poorly correlated with[NO2](R=0.26)and J(NO2)[NO2](R=0.35).These results challenged the role of NO2 as a major precursor of the missing HONO source.YANG Qiang SU Hang LI Xin CHENG YaFang LU KeDing CHENG Peng GU JianWei GUO Song HU Min ZENG LiMin ZHU Tong ZHANG YuanHang 2014Science China Chemistry2014,57,7:8

17Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles.Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long 2020Genomics, Proteomics & Bioinformatics2020,18,6:7
18Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 20Clinical applications of exosome membrane proteins显示文摘Extracellular vesicles(EVs)are small membranous particles that can mediate cell-to-cell communication and which are divided into at least three categories according to their subcellular origin and size:exosomes,microvesicles,and apoptotic bodies.Exosomes are the smallest(30–150 nm)of these EVs,and play an important role in EV-mediated cell-to-cell interactions,by transferring proteins,nucleic acids and,lipids from their parental cells to adjacent or distant cells to alter their phenotypes.Most exosome studies in the past two decades have focused on their nucleic acid composition and their transfer ofmRNAs and microRNAs to neighboring cells.However,exosomes also carry specific membrane proteins that can identify the physiological and pathological states of their parental cells or indicate their preferential target cells or tissues.Exosome membrane protein expression can also be directly employed or modified to allow exosomes to serve as drug delivery systems and therapeutic platforms,including in targeted therapy approaches.This review will briefly summarize information on exosome membrane proteins components and their role in exosome–cell interactions,including proteins associated with specific cell-interactions and diseases,and the potential for using exosome membrane proteins in therapeutic targeting approaches.Qian Hu Hang Su Juan Li Christopher Lyon Wenfu Tang Meihua Wan Tony Ye Hu 2020Precision Clinical Medicine2020,3,1:6
