| 1 | The Yellow Sea green tides were dominated by one species, Ulva (Enteromorpha) prolifera, from 2007 to 2011显示文摘Green tides have occurred in the Yellow Sea successively from 2007 to 2011. Genetic analysis of the 5-year green-tide-forming algae needs to be performed to determine the source of the biomass and understand the mechanism of the green tide blooms. In this study, free-floating green algae were collected at different sites in the Yellow Sea in 2010 and 2011. Data on 182 free-floating samples and 155 attached Ulva samples from previous studies on the Yellow Sea green tides from 2007 to 2009 were also taken into consideration. Morphology observation and molecular phylogenetic analyses indicated that the Yellow Sea green tides were dominated by a single species, Ulva prolifera, from 2007 to 2011. | 3 | Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. | 6 | Cuprizone-induced demyelination in mice: age-related vulner-ability and exploratory behavior deficit显示文摘Schizophrenia is a mental disease that mainly affects young individuals (15 to 35 years old) but its etiology remains largely undefined. Recently, accumulating evidence indicated that demyelination and/or dysfunction of oligodendrocytes is an important feature of its pathogenesis. We hypothesized that the vulnerability of young individuals to demyelination may contribute to the onset of schizophrenia. In the present study, three different age cohorts of mice, i.e. juvenile (3 weeks), young-adult (6 weeks) and middle-aged (8 months), were subjected to a 6-week diet containing 0.2% cuprizone (CPZ) to create an animal model of acute demyelination. Then, age-related vulnerability to CPZ-induced demyelination, behavioral outcomes, and myelination-related molecular biological changes were assessed. We demonstrated: (1) CPZ treatment led to more severe demyelination in juvenile and young-adult mice than in middle-aged mice in the corpus callosum, a region closely associated with the pathophysiology of schizophrenia; (2) the higher levels of demyelination in juvenile and young-adult mice were correlated with a greater reduction of myelin basic protein, more loss of CC-1-positive mature oligodendrocytes, and higher levels of astrocyte activation; and (3) CPZ treatment resulted in a more prominent exploratory behavior deficit in juvenile and young-adult mice than in middle-aged mice. | 7 | Combinational biosynthesis and characterization of a fluorescent 82β-phycocyanin of Spirulina platensis显示文摘To biosynthesize fluorescent Spirulina platensis (Sp)β -phycocyanin (PC) in Escherichia coli, a BLASTP search for homologs of the cpeS gene, a chromophore lyase, was performed against the Synechocystis sp. PCC 6803 (S6) proteome. A highly homologous gene, slr2049, was obtained from the S6 genome. Sites 82 and 153 in -phycocyanin of Sp were modified by site-directed muta- genesis. Two recombinant expression vectors were constructed and transformed into E. coli BL21: (i) pCDF-cpcB (C153A)- slr2049-sll0583-ho1-pcyA; and (ii) pCDF-cpcB (C82I)-slr2049-sll0583-ho1-pcyA. Lyases encoded by the genes slr2049 and sll0583 catalyzed the linking of Sp 82β -PC to phycocyanobilin (PCB), and fluorescent CpcB (C153A)-PCB was generated. We present a strategy for the co-expression of multiple genes in a single expression vector to identify the function of an unknown gene. | 8 | Root-associated(rhizosphere and endosphere) microbiomes of the Miscanthus sinensis and their response to the heavy metal contamination显示文摘The plant root-associated microbiomes, including both the rhizosphere and the root endosphere microbial community, are considered as a critical extension of the plant genome. Comparing to the well-studied rhizosphere microbiome, the understanding of the root endophytic microbiome is still in its infancy. Miscanthus sinensis is a pioneering plant that could thrive on metal contaminated lands and holds the potential for phytoremediation applications. Characterizing its root-associated microbiome, especially the root endophytic microbiome, could provide pivotal knowledge for phytoremediation of mine tailings. In the current study, M. sinensis residing in two Pb/Zn tailings and one uncontaminated site were collected. The results demonstrated that the metal contaminant fractions exposed strong impacts on the microbial community structures. Their influences on the microbial community, however, gradually decreases from the bulk soil through the rhizosphere soil and finally to the endosphere, which resulting in distinct root endophytic microbial community structures compared to both the bulk and rhizosphere soil. Diverse members affiliated with the order Rhizobiales was identified as the core microbiome residing in the root of M. sinensis. In addition, enrichment of plant-growth promoting functions within the root endosphere were predicted, suggesting the root endophytes may provide critical services to the host plant. | 11 | Alternative splicing of MaMYB16L regulates starch degradation in banana fruit during ripening显示文摘The alternative splicing of select genes is an important mechanism to regulate responses to endogenous and environmental signals in plants.However,the role of alternative splicing in regulating fruit ripening remains unclear.Here,we discovered that MaMYB16L,an R1-type MYB transcription factor,undergoes alternative splicing and generates two transcripts,the full-length isoform MaMYB16L and a truncated form MaMYB16S,in banana fruit.During banana fruit ripening,the alternative splicing process intensifies with downregulated MaMYB16L and upregulated MaMYB16S.Moreover,MaMYB16L is a transcriptional repressor that directly binds with the promoters of many genes associated with starch degradation and MaDREB2,a positive ripening regulator,and represses their expression.In contrast,MaMBY16S lacks a DNA-binding domain but competitively combines and forms non-functional heterodimers with functional MaMYB16L.MaMYB16L-MaMYB16S heterodimers decrease the binding capacity and transrepression activity of MaMYB16L.The downregulation of MaMYB16L and the upregulation of MaMYB16S,that is,a decreased ratio of active to non-active isoforms,facilitates the activation of ripening-related genes and thereby promotes fruit ripening.Furthermore,the transient overexpression of MaMYB16S promotes banana fruit ripening,whereas the overexpression of MaMYB16L delays this process.Therefore,the alternative splicing of MaMYB16L might generate a self-controlled regulatory loop to regulate banana fruit ripening. | Guoxiang Jiang Dandan Zhang Zhiwei Li Hanzhi Liang Rufang Deng Xinguo Su Yueming Jiang Xuewu Duan | 2021 | Journal of Integrative Plant Biology2021,63,7: | 2 |

| 13 | Microstructure characterization and thermal properties of the waste-styrene-butadiene-rubber (WSBR)-modified petroleum-based mesophase asphalt显示文摘In this work, waste-styrene-butadiene-rubber(WSBR)-modified petroleum-based mesophase asphalt was prepared through a co-carbonization process. The influence of contents of WSBR and carbonization temperature on the properties of mesophase asphalt was investigated. The chemical constituents,microstructure and thermal property of the samples were characterized. The results show that using WSBR as modifier can significantly promote the formation of mesophase. When the temperature is constant, the addition of WSBR results in more optically anisotropic crystal structure in the samples, and a better thermal stability. When the content of WSBR is invariable, with increasing temperature, the content of anisotropic structure in mesophase asphalt becomes higher and more uniform.
