1The Yellow Sea green tides were dominated by one species, Ulva (Enteromorpha) prolifera, from 2007 to 2011显示文摘Green tides have occurred in the Yellow Sea successively from 2007 to 2011. Genetic analysis of the 5-year green-tide-forming algae needs to be performed to determine the source of the biomass and understand the mechanism of the green tide blooms. In this study, free-floating green algae were collected at different sites in the Yellow Sea in 2010 and 2011. Data on 182 free-floating samples and 155 attached Ulva samples from previous studies on the Yellow Sea green tides from 2007 to 2009 were also taken into consideration. Morphology observation and molecular phylogenetic analyses indicated that the Yellow Sea green tides were dominated by a single species, Ulva prolifera, from 2007 to 2011. Considering that at least five Ulva species inhabit the north coast of China, the unialgal composition of the green tides implied that (1) there may be some special physiology and propagation pathways of U. prolifera for its rapid expansion, (2) the mechanisms of the Yellow Sea green tide formation were similar for the last five years, and (3) the intra-species genetic variation and population structure of U. prolifera need to be studied to determine the exact origin of the bloom-forming biomass.ZHAO Jin JIANG Peng LIU ZhengYi WEI Wei LIN HanZhi LI FuChao WANG JinFeng QIN Song 2013Chinese Science Bulletin2013,58,19:27

4Cuprizone-induced demyelination in mice: age-related vulner-ability and exploratory behavior deficit显示文摘Schizophrenia is a mental disease that mainly affects young individuals (15 to 35 years old) but its etiology remains largely undefined. Recently, accumulating evidence indicated that demyelination and/or dysfunction of oligodendrocytes is an important feature of its pathogenesis. We hypothesized that the vulnerability of young individuals to demyelination may contribute to the onset of schizophrenia. In the present study, three different age cohorts of mice, i.e. juvenile (3 weeks), young-adult (6 weeks) and middle-aged (8 months), were subjected to a 6-week diet containing 0.2% cuprizone (CPZ) to create an animal model of acute demyelination. Then, age-related vulnerability to CPZ-induced demyelination, behavioral outcomes, and myelination-related molecular biological changes were assessed. We demonstrated: (1) CPZ treatment led to more severe demyelination in juvenile and young-adult mice than in middle-aged mice in the corpus callosum, a region closely associated with the pathophysiology of schizophrenia; (2) the higher levels of demyelination in juvenile and young-adult mice were correlated with a greater reduction of myelin basic protein, more loss of CC-1-positive mature oligodendrocytes, and higher levels of astrocyte activation; and (3) CPZ treatment resulted in a more prominent exploratory behavior deficit in juvenile and young-adult mice than in middle-aged mice. 5Drainage evolution in intermontane basins at the Qinling-Daba Mountains显示文摘River capture is of great significance to landform evolution and hominine migration.In the Qinling-Daba Mountains,there is a viewpoint that Jialing River captured Hanjiang River,but this is still controversial.In this paper,we discuss the drainage evolution processes in intermountain basins at the Qinling-Daba Mountains based on a combination of detrital zircon UPb geochronology and geomorphic indexes.We suggest that the Hanjiang River gradually captured the Jialing River from east to west,accompanied by the evolution of the ancient Yangtze River.In terms of geomorphic evidences,wide valleys did not match with discharge,and a series of wind gaps developed in the Shiquan-Ankang basin.In addition,the valley shapes and width-toheight ratios(Vf)indicate two possible rapid incisions.The hypsometric integrals(HI)reflect that the landform gradually changes from the old stage to the youth stage from west to east.Theχvalues show that the drainage divide is moving to the side of the Yuehe River,and the Yuehe River is gradually shrinking.According to the sedimentary records,the zircon U-Pb age distributions indicate the provenance change.The high-altitude terraces show three age peaks(200–250,400–505,and 700–900 Ma),with the dominant Indosinian age peak(200–250 Ma),while the modern fluvial sediments only show a single peak of Jinning(700–900 Ma).These data show that there are two major river captures:(1)The ancient Hanjiang River cut through the regional compression ridge,and then captured the Hanzhong Basin river system(a part of the ancient Jialing river system)from east to west,and(2)The southern tributary captured the trunk with the uplift of the divide in the Shiquan-Ankang Basin,forming the modern drainage pattern in the upper Hanjiang River.The activities of the regional strike-slip fault,and the associated compression uplift played a key role in the river captures,the drainage evolution,and related landforms in the Shiquan-Ankang basin.In addition,it is shown that the evolution of the upper tributary basins lagged behind the response of the trunk channel to the tectonic activities and river captures.The interconnected wide valleys caused by river capture may have provided convenient geomorphological conditions for human migration into the Qinling-Daba Mountains along those river valleys.Wanting XIE Xianyan WANG Hanzhi ZHANG Quanyu LIU Shejiang WANG Huayu LU 2021Science China Earth Sciences2021,64,11:1
6Efficacy of anti-CD20 chimeric Fab' fragment on proliferation of B lymphoma cells显示文摘The variable domain of heavy chain (VH) and light chain (VL) genes of anti-CD20 monoclonal antibody HL47 were cloned from anti-CD20 ScFv expression vector pCANBTEcd20 by PCR and ligated into vector pYZF to construct chimeric anti-CD20 Fab’ fragment expression vector pTZFcd20. Chimeric anti-CD20 Fab’ fragment was expressed in E. coli 16C9 and purified by protein G affinity chromatography. Competitive inhibition assay showed that anti-CD20 Fab’ fragment inhibited binding of HI47 to CD20 on the surface of Daudi cells. Results from MTT assay indicated that chimeric anti-CD20 Fab’fragment inhibited the proliferation of Daudi cells, IC50=69 ug/mL. 10Transcriptomic Analysis and Comparison of the Gene Expression Profiles in Fast- and Slow-Growing Pearl Oysters Pinctada fucata martensii显示文摘The pearl oyster Pinctada fucata martensii is an economically valuable shellfish that is cultured for seawater pearl pro-duction,which mainly depends on oyster growth.However,the growth mechanisms of the pearl oyster are still poorly understood.In this study,oysters were grouped with relative growth rate,including fast-growing(FG)group and slow-growing(SG)group.Oxford Nanopore Technologies(ONT)long-read sequencing was applied to investigate the molecular mechanisms involved in the growth of this species.Five alternative splicing(AS)types were analyzed in both FG and SG groups,which include alternative 3’splice site,alternative 5’splice site,exon skipping,intron retention,and mutually exclusive exon.Transcriptome analysis showed that four of five different AS events(excluding mutually exclusive exons)occurred more frequently in FG than in SG oysters,and the five main AS types exhibited different characteristics.The AS events that were detected may be involved in growth,and the difference in ex-pression of AS events between FG and SG oysters may be involved in the mechanism underlying the difference in growth.Fifty dif-ferentially expressed genes(DEGs)were identified between the FG and SG oysters.The results showed that 40 genes were signifi-cantly up-regulated in FG oysters,while 10 genes were significantly down-regulated in SG oyster.Several genes related to nutrient metabolism,shell formation,and immunity were more highly expressed in FG oysters than in SG oysters.In summary,FG oysters exhibited higher metabolic and biomineralization activities and had a more powerful immune system than SG oysters.These results provide insight into the growth of P.f.martensii that can be used to improve breeding programs.XU Hanzhi LIU Huiru ZHANG Hua HE Maoxian 2022Journal of Ocean University of China2022,21,1:0
11Disruption of ER ion homeostasis maintained by an ER anion channel CLCC1 contributes to ALS-like pathologies显示文摘Although anion channel activities have been demonstrated in sarcoplasmic reticulum/endoplasmic reticulum(SR/ER),their molecular identities and functions remain unclear.Here,we link rare variants of Chloride Channel CLIC Like 1(CLCC1)to amyotrophic lateral sclerosis(ALS)-like pathologies.We demonstrate that CLCC1 is a pore-forming component of an ER anion channel and that ALS-associated mutations impair channel conductance.CLCC1 forms homomultimers and its channel activity is inhibited by luminal Ca^(2+)but facilitated by phosphatidylinositol 4,5-bisphosphate(PIP2).We identified conserved residues D25 and D181 in CLCC1 N-terminus responsible for Ca^(2+)binding and luminal Ca^(2+)-mediated inhibition on channel open probability and K298 in CLCC1 intraluminal loop as the critical PIP2-sensing residue.CLCC1 maintains steady-state[Cl^(–)]ER and[K+]ER and ER morphology and regulates ER Ca^(2+)homeostasis,including internal Ca^(2+)release and steady-state[Ca^(2+)]ER.ALS-associated mutant forms of CLCC1 increase steady-state[Cl^(–)]ER and impair ER Ca^(2+)homeostasis,and animals with the ALS-associated mutations are sensitized to stress challenge-induced protein misfolding.Phenotypic comparisons of multiple Clcc1 loss-of-function alleles,including ALS-associated mutations,reveal a CLCC1 dosage dependence in the severity of disease phenotypes in vivo.Similar to CLCC1 rare variations dominant in ALS,10%of K298A heterozygous mice developed ALS-like symptoms,pointing to a mechanism of channelopathy dominant-negatively induced by a loss-of-function mutation.Conditional knockout of Clcc1 cell-autonomously causes motor neuron loss and ER stress,misfolded protein accumulation,and characteristic ALS pathologies in the spinal cord.Thus,our findings support that disruption of ER ion homeostasis maintained by CLCC1 contributes to ALS-like pathologies.Liang Guo Qionglei Mao Ji He Xiaoling Liu Xuejiao Piao Li Luo Xiaoxu Hao Hanzhi Yu Qiang Song Bailong Xiao Dongsheng Fan Zhaobing Gao Yichang Jia 2023Cell Research2023,33,7:0
12Modulating multidrug resistance through inhibiting of protein kinase C activity by phenothiazines显示文摘In the present study, actions of phenothiazines(PTZ) in reversing multidrug resistance(MDR) and inhibiting PKC activity were investigated. It was found that the three PTZs caused 2.49, 36.58 and 75.78 fold reversal of K562/AO2 MDR cells resistant to adriamycin, respectively, while the chemosensitizer verapamil caused 40 fold reversal in the same condition, indicating that PTZ11 is a novel reversal agent of MDR and a potential chemotherapeutic reagent for tumor therapy. PKC activity analysis in the presence of PTZs showd that PTZ6 and PTZ11 inhibited rat brain protein kinase C activity in a manner of dose_dependent. The IC 50 values were (489.77±31.4) and (113±9.64) μmol/L, respectively. PTZ7 had no inhibition on PKC activity. Further study showed that PTZ11 could reduce PMA_mediated activation of PKC in a manner of dose_dependent, suggesting that PTZ11 might compete for the high_affinity phorbol ester binding site within PKC molecule. Recently, an X_ray structure of PMA in complex with PKC Cys2 activator_binding domain was solved. We therefore decided to explore the possible binding model of PTZ11 with PKC molecule using SYBYL 6.02 program. 13Internalization and half-life of membrane-bound macrophage colony-stimulating factor显示文摘The binding of recombinant human macrophage colony-stimulating factor (M-CSF) soluble receptor (rh-M-CSF-sR) to membrane-bound macrophage colony stimulating factor (m-M-CSF) and the internalization and recycling of rh-M-CSF-sR/m-M-CSF complexes mediated by m-M-CSF were studied with a model of J6-1 cell line. The results indicated that m-M-CSF bound rh-M-CSF-sR with high affinity (Kd= 1.78×10 12 mol/L) and mediated a temperature- and energy-dependent internalization of rh-M-CSF-sR, and that internalized rh-M-CSF-sR could return to the cell surface in an m-M-CSF-bound state, suggesting that m-M-CSF may have a capability to mediate the internalization and recycling of rh-M-CSF-sR/m-M-CSF complexes. 14Preparation of Durable Emitter of Electrospray Mass Spectrometry by Covalently Coating the Fused-Silica Capillary Tip with Carbon-Nanotube Sol-Gel Composite Material显示文摘A simple approach for the preparation of emitter of electrospray ionization mass spectrometry by covalently coating the fused-silica capillary tip with the conductive carbon-nanotube sol-gel composite material(CNTSCM)is described.The CNTSCM was prepared by dispersing single-walled carbon nanotubes in the sol composed of a mixture of 3-glycidoxypropyltrimethoxysilane and 3-aminopropyltrimethoxysilane,and ethanol.The long-term sta-bility of the prepared ESI emitters was demonstrated by at least 180 h of continuous use.Signal intensity obtained by the prepared emitter was mass-flux sensitive when the flow rate was lower than 500 nL/min,while the signal in-tensity performed a concentration dependence when the flow rate was in the range of 500-800 nL/min.The use-fulness of such a prepared emitter was demonstrated by the analysis of various types of samples such as organic small molecular drugs,oligosaccharide,peptide,and protein.Hanzhi Zhang Chao Liu Xuepei Zhang Jingwu Kang 2014Chinese Journal of Chemistry2014,32,4:0
15The role of Smad6 in immunity of the pearl oyster Pinctada fucata martensii显示文摘Inhibitory Smads(I-Smads),which belong to the Smad family and inhibit bone morphogenic protein 2(BMP2)signaling by a variety of mechanisms,can suppress innate immunity responses in vertebrates.However,there are no reports for the role of Smad6 in immunity in mollusks.In this study,we showed that Smad6 of the pearl oyster Pinctada fucata martensii was located in the Smad6 cluster of the phylogenetic tree;mRNA expression of Smad6 and Smad3 was up-regulated after lipopolysaccharide and polyinosinic:polycytidylic challenge;and transcript levels of Smad6 and Smad3 showed opposite patterns during wound healing.Under salinity stress,water inflow and outflow in the gills appear to be regulated by BMP2-Smads signals,and BMP2-Smads signaling may be closely related to the immune response.Our results indicate that Smad6 is involved in immunity,that it plays a positive role in the response to immune challenge and an inhibitory role during wound healing,and that Smad6 and Smad3 may work against each other.Yu SHI Xiaolan PAN Meng XU Huiru LIU Hanzhi XU Maoxian HE 2022Journal of Oceanology and Limnology2022,40,3:0
16Switchable Kirigami Structures as Window Envelopes for Energy-Efficient Buildings显示文摘Efficient regulation of thermal radiation is an effective way to conserve energy consumption of buildings.Because windows are the least energy-efficient part of buildings,their thermal radiation regulation is highly demanded,especially in the changing environment,but is still a challenge.Here,by employing a kirigami structure,we design a variable-angle thermal reflector as a transparent envelope of windows for their thermal radiation modulation.The envelope can be easily switched between heating and cooling modes by loading different pre-stresses,which endow the envelope windows with the ability of temperature regulation,and the interior temperature of a building model can be reduced by~3.3°C under cooling mode and increased by~3.9°C under heating mode in the outdoor test.The improved thermal management of windows by the adaptive envelope provides an extra heating,ventilation,and air-conditioning energy savings percentage of 13%to 29%per year for buildings located in different climate zones around the world,making the kirigami envelope windows a promising way for energy-saving utilization.Hanzhi Yin Xishu Zhou Zhengui Zhou Rong Liu Xiwei Mo Zewen Chen Erqi Yang Zhen Huang Hao Li Hao Wu Jun Zhou Yi Long Bin Hu 2023Research2023,,4:0
17East Asian paleoclimate change in the Weihe Basin (central China) since the middle Eocene revealed by clay mineral analysis显示文摘Obtaining a continuous sedimentary record of Cenozoic East Asian paleoclimate change is key to understand the origin,evolution and driving mechanism of the East Asian monsoon and climate change.Based on the continuous Cenozoic fluvial and lacustrine sedimentary sequence in the Weihe Basin,central China,we carried out research on the content and crystal parameters of clay minerals.The paleoclimate change since the middle Eocene was reconstructed accordingly.The results show that smectite and illite are the dominant clay minerals in fluvial-lacustrine sediments of Weihe Basin.The crystallinity of illite,the chemical index of illite,the ratio of smectite to illite and chlorite decrease gradually,which indicates that chemical weathering in the Weihe River watershed stepwise weakened since the middle Eocene,under the background of a semi-arid and semi-humid climate.The formation of palygorskite may be affected by a weak diagenesis.In this case,the content of smectite and the illite crystallinity caused by the weathering in the corresponding period may be actually higher than that of the current,which indicates that the climate in the Weihe Basin region was relatively warm and humid from the middle Eocene to Pliocene.With the decrease of Cenozoic global temperature and the continuous accumulation of sediments,the intensity of chemical weathering in the Weihe Basin gradually weakened,and the East Asian monsoon climate gradually evolved until becoming dry in the Quaternary.Evidence from clay minerals also indicates that the aridification in the Weihe Basin from the late Eocene to the Oligocene may be a response to the global cooling event at the Eocene-Oligocene transition.Hengzhi LYU Huayu LU Yichao WANG Hanzhi ZHANG Yao WANG Kexin WANG Wen LAI Zhifei LIU Yanli LI Junfeng JI 2021Science China Earth Sciences2021,64,8:0
18Evaluation and prospect of scientific research capacity ranking for community healthcare centres in China显示文摘The National Health and Family Planning Commission of the People's Republic of China has proposed to improve the medical capacity of general practitioners and the establishment of general practice in recent health reform.For the first time,the ability to conduct scientific research was included in this reform,which demands community healthcare centres(CHCs)to strengthen their research capacity.The evaluation of community scientific research capacity has become an important endeavour to promote the implementation of research in CHCs.Since 2016,our research team has been working on an evaluation system and has published the scientific research capacity ranking for the top 100 CHCs in China.The latest released ranking of scientific research capacities of China CHCs has aroused great attention in the country.Qian Liu Jianwei Shi Dehua Yu Hua Jin Xuhua Ge Hanzhi Zhang Zhaoxin Wang 2020Family Medicine and Community Health2020,8,1:0
