7Expression and purification of recombinant human hemangiopoietin in Escherichia coli显示文摘Objective:To express the soluble recombinant hemangiopoietin protein in E.coli BL21(DE3).Methods:Using human fetal live cDNA as a template,a partial cDNA fragment of HAPO coding N-terminal region was subcloned into plasmids pTrc99,pQE60 and pET32c to construct different recombinant prokaryotic expression systems.After selecting,the soluble rhHAPO fusion protein was expressed stably in E.coli BL21(DE3) by vector pET32c-HAPO and further isolated by nickelnitrilotriacetic acid(NTA) affinity chromatography.After cleavage with enterokinase,the rhHAPO protein was applied to Fast Flow SP sepharose column.Results:The rhHAPO protein had a purity of more than 95% and a good bioactivity based on the cell adhesion assay in ECV304 cells.Conclusion:We have established a protein engineering system to produce rhHAPO which may provide the possibility for clinical application.Ren Qian Ma Fengxia Chen Zhong Lu Shihong Han Zhibo Liu Yongjun Xu Bin Zhang Xiangyu Han Zhongchao 2008Journal of Medical Colleges of PLA(China)2008,23,3:0

9白血病骨髓微环境中GATA-1、GATA-2和GATA-3基因表达模式(英文)显示文摘Objective: The aim of the study was to investigate the expression pattern of hematopoietic transcription factor GATA-1, -2 and -3 genes in leukemic bone marrow (BM) microenvironment [including bone marrow stromal cells (BMSCs) and BM hematopoietic cells]. Methods: Mononuclear cells were isolated from BM of patients with acute myeloid leukemia (AML), chronic myelogenous leukemia (CML), or acute lymphoblastic leukemia (ALL). Adherent cells (BMSCs) and nonadherent cells (BM hematopoietic cells) were collected after long-term culture in vitro. The semi-quantitative expression levels of GATA genes in the BMSCs or BM hematopoietic cells from patients with leukemia were analyzed by using RT-PCR-ELISA and compared with normal controls. Results: The expression level of GATA-1 gene in the BMSCs from CML group was significantly lower than that of the normal controls. The expression level of GATA-3 gene in the BMSCs from ALL was higher than that of the normal controls, but that from CML was lower than the normal controls. Dominant expression of GATA-3 gene was found in the normal BM hematopoietic cells. The dominant expression of GATA-2 gene was found in the normal BMSCs and the BMSCs from CML, whereas the dominant expression of GATA-3 gene was detected in the BMSCs from AML. 10Embryonic stem cell-derived extracellular vesicles rejuvenate senescent cells and antagonize aging in mice显示文摘Aging is a degenerative process that leads to tissue dysfunction and death.Embryonic stem cells(ESCs)have great therapeutic potential for age-related diseases due to their capacity for self-renewal and plasticity.However,the use of ESCs in clinical treatment is limited by immune rejection,tumourigenicity and ethical issues.ESC-derived extracellular vesicles(EVs)may provide therapeutic effects that are comparable to those of ESCs while avoiding unwanted effects.Here,we fully evaluate the role of ESC-EVs in rejuvenation in vitro and in vivo.Using RNA sequencing(RNA-Seq)and microRNA sequencing(miRNA-Seq)screening,we found that miR-15b-5p and miR-290a-5p were highly enriched in ESC-EVs,and induced rejuvenation by silencing the Ccn2-mediated AKT/mTOR pathway.These results demonstrate that miR-15b-5p and miR-290a-5p function as potent activators of rejuvenation mediated by ESC-EVs.The rejuvenating effect of ESC-EVs was further investigated in vivo by injection into aged mice.The results showed that ESC-EVs successfully ameliorated the pathological age-related phenotypes and rescued the transcriptome profile of aged mice.Our findings demonstrate that ESC-EVs treatment can rejuvenate senescence both in vitro and in vivo and suggest the therapeutic potential of ESC-EVs as a novel cell-free alternative to ESCs for age-related diseases.Lu Yu Hang Wen Chang Liu Chen Wang Huaxin Yu Kaiyue Zhang Qingsheng Han Yue Liu Zhongchao Han Zongjin Li Na Liu 2023Bioactive Materials2023,,11:0
