| 1 | Inuence of oxytetracycline on the structure and activity of microbial community in wheat rhizosphere soil显示文摘The microbial community composition in wheat rhizosphere was analyzed by detecting colony forming units (CFUs) in agar plates. The total CFUs in rhizosphere were 1.04 × 109/g soil with 9.0 × 108/g bacteria, 1.37 × 108/g actinomyces and 3.6 × 106/g fungi. The 10 dominant bacteria were isolated from wheat rhizosphere and were grouped into genus Bacillus according to their full length 16S rRNA gene sequences. Although belonging to the same genus, the isolated strains exhibited dierent sensitivities to oxytetracycline. When a series of the rhizosphere soil was exposed under various concentrations of oxytetracycline, the microbial community structure was highly aected with significant decline of CFUs of bacteria and actinomyces (22.2% and 31.7% at 10 mg/kg antibiotic, respectively). This inhibition was clearly enhanced with the increase exposure dosage of antibiotic and could not be eliminated during 30 d incubation. There was no obvious inuence of this treatment on fungi population. | 2 | Hydrogen peroxide-induced changes in intracellular pH of guard cells precede stomatal closure显示文摘Epidermal bioassay demonstrated that benzylamine, a membrane-permeable weak base, can mimick hydrogen peroxide (H2O2) to induce stomatal closure, and butyric acid, a membrane-permeable weak acid, can partly abolish the H2O2-induced stomatal closure. Confocal pH mapping with the probe 5-(and-6)- carboxy seminaphthorhodafluor- 1 - acetoxymethylester (SNARF-1-AM) revealed that H2O2 leads to rapid changes in cytoplasmic and vacuolar pH in guard cells of Viola faba L, i. e. alkalinization of cytoplasmic areas occur red in parallel with a decrease of the vacuolar pH, and that butyric acid pretreatment can abolish alkalinization of cytoplasmic areas and acidification of vacuolar areas of guard cells challenged with H2O2. | 3 | Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. | 5 | Quick image-processing method of UAV without control points data in earthquake disaster area显示文摘The methods of unmanned aerial vehicles images splicing rapidly and rectifying without control points data, only using auxiliary data recorded by unmanned aerial vehicle (UAV) system, were researched. Two algorithms were proposed, including UAV images auto- splicing rapidly after image blocking and UAV images rectifying based on pseudo center points of auxiliary data. When SIFT algorithm for UAV images splicing is applied, the accuracy is very high, but the efficiency is very low. The splicing efficiency is improved greatly based on SIFT algorithm after image blocking. | 6 | Suitability evaluation for land reclamation in mining area:A case study of Gaoqiao bauxite mine显示文摘Suitability evaluation plays an important role in land reclamation because the choice of evaluation methods affects the accuracy and objectivity of the suitability evaluation results. Furthermore, it influences the decision-making related to land reclamation. An improved method, which is called limit comprehensive conditions method, was developed after different suitability evaluation methods were studied. Based on this method, the reclaimed land of the Gaoqiao bauxite mining area was evaluated. The Gaoqiao mining area was divided into seven evaluation units that were evaluated respectively by selecting evaluation factors and establishing grade standards. The results show that the proposed method is more applicable and easier to handle. Moreover, its evaluation results are more scientific compared with the traditional evaluation methods. | 10 | Construction and selection of the natural immune Fab antibody phage display library from patients with colorectal cancer显示文摘AIM: To construct the natural immune Fab antibody phage display libraries of colorectal cancer and to select antibodies related with colorectal cancer.METHODS: Extract total RNA from tissue of local cancer metastasis lymph nodes of patients with colorectal cancer.RT-PCR was used to amplify the heavy chain Fd and light chain к and the amplification products were inserted successively into the vector pComb3 to construct the human libraries of Fab antibodies. They were then panned by phage display technology. By means of Dot immunoblotting and ELISA, the libraries were identified and the Fab phage antibodies binding with antigens of colorectal cancer were selected.RESULTS: The amplified fragments of Fd and к gained by RT-PCR were about 650bp. Fd and к PCR products were subsequently inserted into the vector pComb3, resulting in a recombination rate of 40% and the volume of Fab phage display library reached 1.48 x 106. The libraries were enriched about 120-fold by 3 cycles of adsorption-elution- multiplication (panning). Dot immunoblotting showed Fab expressions on the phage libraries and ELISA showed 5clones of Fab phage antibodies which had binding activities with antigens of colorectal cancer.CONCLUSION: The natural immune Fab antibody phage display libraries of colorectal cancer were constructed. | 11 | Heat shock induction of a 65 kDa ATP-binding proteinase in rat C6 glioma cells显示文摘The 45, 55, 65 and 100 kDa ATP-binding proteinases (ATP-BPases) of the heat-shocked (44 ℃ for 30 min, recovery for 12h) rat C6 glioma cells were purified by DEAE-ionexchange and ATP-affinity chromatography. Their molecular masses, isoelectric points (pI), pH-optima and other properties were analyzed by native proteinase gels.It was shown that the 65 kDa ATP-BPase is specifically induced by heat shock and not detectable in control cells.Its N-terminal 1-9 amino acid sequence was determined by Edman degradation, but no homologies to other proteins in the protein data bases were found. 30 and 31 kDa proteinases can be cleaved from the 45, 55 and 65 kDa proteinases to which they are linked. | 12 | Marek’s disease virus-encoded microRNAs: genomics, expression and function显示文摘MicroRNAs (miRNAs) are the recently discovered small non-coding RNA molecules that have post-transcriptional regulatory functions in many important biological processes. A large number of miRNAs have been found to be encoded by viral genomes, especially in herpesviruses. Previous research regarding miRNAs encoded by herpesviruses, including Marek’s disease virus (MDV), has demonstrated their involvement in lytic replication, latent infection, T-lymphocyte transformation and tumorigenesis. MDV is an oncogenic alphaherpesvirus, with the ability to induce tumors in natural hosts; however, formation of these tumors can be prevented by immunization with attenuated or nonpathogenic forms of the virus. Marek’s disease is considered to be a good biomedical model for investigating the biology, genetics, and immunology of tumorigenesis. In this paper, we review the discovery and identification of MDV-encoded miRNAs, along with their genomics, expression profiles, and currently known functions. | 19 | Lead Isotopic Steep-Dipping Zone and Mineralization—An Example from Mineral Deposits Concentrated Area in East Qinling,China显示文摘Lead isotopic geochemical steep-dipping zone usually exists on inhomogeneous boundaries of earth blocks. Its crossing with the geophysical gradient zone often convergently occurs at giant deposits. Deep structures or concealed structural planes obviously have the coupling relationship with the convergent area of mineral deposits. The geochemical steep-dipping zone is usually distributed along the boundary of ancient continental blocks. Its crossing effect with geophysical gradient zone is usually presented as depression or swell of Moho discontinuity on the crossing direction with the ancient continental margin, which would lead to form deep fractures of earth crust at block margins or lead to adjustment of earth crust texture. The deep hydrothermal liquid would rise up along the structural planes to form the convergent areas of mineral deposits. For example, Luonan- Luanchuan area in east Qinling is a typical crossed area of the geochemical steepdipping zone and geophysical gradient zone. The mineral deposit belt extends along EW direction. It was controlled by the geochemical steepdipping zone equidistantly distributed along NE direction like a string of beads controlled by a gravity gradient zone in NE direction and a mantle depression slope. Along a plunging mantle syncline on EW plunging direction, from the east to the west, checkform was distributed which controls synergic crustmantle granoporphyry rocks.
