| 3 | A homologous and molecular dual-targeted biomimetic nanocarrier for EGFR-related non-small cell lung cancer therapy显示文摘The abnormal activation of epidermal growth factor receptor(EGFR)drives the development of non-small cell lung cancer(NSCLC).The EGFR-targeting tyrosine kinase inhibitor osimertinib is frequently used to clinically treat NSCLC and exhibits marked efficacy in patients with NSCLC who have an EGFR mutation.However,free osimertinib administration exhibits an inadequate response in vivo,with only~3%patients demonstrating a complete clinical response.Consequently,we designed a biomimetic nanoparticle(CMNP^(@Osi))comprising a polymeric nanoparticle core and tumor cell-derived membrane-coated shell that combines membrane-mediated homologous and molecular targeting for targeted drug delivery,thereby supporting a dual-target strategy for enhancing osimertinib efficacy.After intravenous injection,CMNP^(@Osi)accumulates at tumor sites and displays enhanced uptake into cancer cells based on homologous targeting.Osimertinib is subsequently released into the cytoplasm,where it suppresses the phosphorylation of upstream EGFR and the downstream AKT signaling pathway and inhibits the proliferation of NSCLC cells.Thus,this dual-targeting strategy using a biomimetic nanocarrier can enhance molecular-targeted drug delivery and improve clinical efficacy. | Bin Xu Fanjun Zeng Jialong Deng Lintong Yao Shengbo Liu Hengliang Hou Yucheng Huang Hongyuan Zhu Shaowei Wu Qiaxuan Li Weijie Zhan Hongrui Qiu Huili Wang Yundong Li Xianzhu Yang Ziyang Cao Yu Zhang Haiyu Zhou | 2023 | Bioactive Materials2023,,9: | 1 |
| 4 | Orthogonal modular biosynthesis of nanoscale conjugate vaccines for vaccination against infection显示文摘Conjugate vaccines represent one of the most effective means for controlling the occurrence of bacterial diseases.Although nanotechnology has been greatly applied in the field of vaccines,it is seldom used for conjugate vaccine research because it is very difficult to connect polysaccharides and nanocarriers.In this work,an orthogonal and modular biosynthesis method was used to produce nanoconjugate vaccines using the SpyTag/SpyCatcher system.When SpyTag/SpyCatcher system is combined with protein glycosylation technology,bacterial O-polysaccharide obtained from Shigela flexneri 2a can be conjugated onto the surfaces of different virus-like particles(VLPs)in a biocompatible and controlled manner.After confirming the excellent lymph node targeting and humoral immune activation abilities,these nanoconjugate vaccines further induced efficient prophylactic effects against infection in a mouse model.These results demonstrated that natural polysaccharide antigens can be easily connected to VLPs to prepare highly efficient nanoconjugate vaccines.To the best of the researchers1 knowledge,this is the first time VLP-based nanoconjugate vaccines are produced efficiently,and this strategy could be applied to develop various pathogenic nanoconjugate vaccines. | Xin Li Chao Pan Peng Sun Zhehui Peng Erling Feng Jun Wu Hengliang Wang Li Zhu | 2022 | Nano Research2022,15,2: | 1 |

| 6 | Screening and identification of Shigella flexneri 2a virulence-related genes induced after invasion of epithelial cells显示文摘An in vivo expression technology (IVET) was applied to screen s.flexneri 2a genes induced after invasion of epithelial cells, and virulence-related genes were further identified by mutational analysis. Thirteen intracellular induced genes were identified with a HeLa cell infection model. Of these, two were identified as alkylation-related genes; one was related to metabolism; one encoded a transcriptional regulator; three were identified as insertion elements; three ap- peared to be antisense to genes involved in the transmethylation,biosyntheseis, and phos- photransferase system;and three were predicted to encode polypeptides with unknown functions. Intracellular survival assavs showed that the mutants of alkA,citC and wcaJ genes had lower capability of intracellular replication or survival than the the wild-type strain.The results indicated that alkA, citC and wcaJ genes could take part in the intracellular survival or replication of S. flexneri 2a and the capability of intracellular survival or replication could be one of the major virulence elements. However, the yaiC mutant was able to survive in the murine infection assay but almost not in HeLa cell infection assay. Very possibly, yaiC gene was involved in the other mechanism of S. flexneri virulence. | 9 | A novel aberrant splicing of G_(sa) transcript in human leukemia cell lines显示文摘The α subunit of the stimulatory G protein, Gsα, is involved in the stimulation of adenylate cyclase pathway of signal transduction. The various Gsα mRNAs are generated either by alternative splicing or by using alternative promoter. A novel aberrant splicing of Gsα transcript in human leukemia cell lines is reported. The entire coding region of Gsα gene obtained, referred to as GsαLeu, was amplified by PCR with cDNA from Jurkat human leukemia cDNA library or the reverse-transcribed first strand cDNA from human leukemia cell line HL-60 as a template. The result of DNA sequencing indicated that,compared with originally reported Gsα, GsαLeu has two in-frame deletions, one in amino acid residues 23—249 and the other in amino acid residues 254—260, suggesting that G(sαLeu) may play an important role in leukemia. | 10 | Bautype: Capsule and Lipopolysaccharide Serotype Prediction for Acinetobacter baumannii Genome显示文摘Acinetobacter baumannii has three clusters of genes for polysaccharide synthesis.Capsular polysaccharides and lipopolysaccharides are major virulence determinants in many Gram-negative bacteria,and play key roles in the overall fitness and virulence in A.baumannii,meanwhile poly-beta-1-6-N-acetylglucosamine was critical for biofilm formation.A variety of studies have been conducted which focused on the surveillance of antimicrobial resistance genes and clones in A.baumannii.However,a tool to perform serotype prediction in A.baumannii is still lacking.Here we present Bautype,a tool for whole genome sequencingbased K and O locus typing in A.baumannii.Using Bautype,users can upload and analyze the genome assembled in a web browser,with the results being displayed in a tabular or graphical format.We demonstrated the high accuracy of predicting K and O locus diversity using Bautype when analyzing more than 3000 A.baumannii genomes.Using our software,we were able to analyze K and O locus diversity among 593 genomes belonging to ST208(3 K and 8 O).These diverse loci can serve as epidemiological markers for Multi-drug Resistance(MDR)strain subtyping.We also identified 45 novel O locus types.Our findings highlight the importance in monitoring the capsule type and LOS of A.baumannii strains for which Bautype is a tailored clinical software.Bautype is available at http://BauType.net. | Xiaoting Hua Qian Liang Li Fang Jintao He Meixia Wang Wenjie Hong Sebastian Leptihn Hengliang Wang Yunsong Yu Huan Chen | 2020 | Infectious Microbes & Diseases2020,2,1: | 0 |
| 11 | Preliminary study on discovery of a novel molecule that interacts with CPP32 using two-hybrid system显示文摘Of ICE protease family, CPP32 (apopain or Yama) plays a central role in different apoptotic pathways. To study the molecules regulating CPP32, yeast two_hybrid system was used to identify proteins (peptides) that interact with CPP32. First, the CPP32 gene was cloned into plasmid vector pGBT9. The resulting recombinant plasmid was designated as pGBT9/CPP. The pGBT9 /CPP plasmid was transformed into the yeast strain HF7C, then the leukemia library was introduced. The transformation mixture was plated on medium lacking Trp, Leu and His in the initial screen. Colonies growing on the selection medium were further assayed for β galactosidase activity. Within 42 Trp +Leu +His + colonies only 5 turned blue in the presence of X_Gal. Plasmid DNA from 5 positive yeast colonies was prepared respectively and used to transform HB101 by electroporation. The transformation mixture was plated on medium lacking Leu to be selected for the library plasmid. Finally, only one library plasmid, designated as pY1, was determined to be truly positive by retransformation of pGBT9/CPP and the library plasmid into HF7C. | 13 | Biosynthesis and Immunological Evaluation of a Dual-Antigen Nanoconjugate Vaccine Against Brucella melitensis显示文摘Brucellosis,caused by Brucella,is one of the most common zoonosis.However,there is still no vaccine for human use.Although some live attenuated vaccines have been approved for animals,the protection effect is not ideal.In this study,we developed a dual-antigen nanoconjugate vaccine containing both polysaccharide and protein antigens against Brucella.First,the antigenic polysaccharide was covalently coupled to the outer membrane protein Omp19 using protein glycan coupling technology,and then it was successfully loaded on a nano-carrier through the SpyTag/SpyCatcher system.After confirming the efficient immune activation and safety performance of the dual-antigen nanoconjugate vaccine,the potent serum antibody response against the two antigens and remarkable protective effect in non-lethal and lethal Brucella infection models were further demonstrated through different routes of administration.These results indicated that the dual-antigen nanoconjugate vaccine enhanced both T helper 1 cell(Th1)and Th2 immune responses and protected mice from Brucella infection.Furthermore,we found that this protective effect was maintained for at least 18 weeks.To our knowledge,this is the first Brucella vaccine bearing diverse antigens,including a protein and polysaccharide,on a single nanoparticle.Thus,we also present an attractive technology for co-delivery of different types of antigens using a strategy applicable to other vaccines against infectious diseases. | Jing Huang Yufei Wang Kangfeng Wang Shulei Li Peng Sun Yan Guo Jiankai Liu Ruifu Yang Ming Zeng Chao Pan Hengliang Wang Li Zhu | 2023 | Engineering2023,,10: | 0 |
