3PI3K/Akt/mTOR signaling orchestrates the phenotypic transition and chemo-resistance of small cell lung cancer显示文摘Small cell lung cancer(SCLC)is a phenotypically heterogeneous disease with an extremely poor prognosis,which is mainly attributed to the rapid development of resistance to chemotherapy.However,the relation between the growth phenotypes and chemo-resistance of SCLC remains largely unclear.Through comprehensive bioinformatic analyses,we found that the heterogeneity of SCLC phenotype was significantly associated with different sensitivity to chemotherapy.Adherent or semiadherent SCLC cells were enriched with activation of the PI3K/Akt/mTOR pathway and were highly chemoresistant.Mechanistically,activation of the PI3K/Akt/mTOR pathway promotes the phenotypic transition from suspension to adhesion growth pattern and confers SCLC cells with chemo-resistance.Such chemo-resistance could be largely overcome by combining chemotherapy with PI3K/Akt/mTOR pathway inhibitors.Our findings support that the PI3K/Akt/mTOR pathway plays an important role in SCLC phenotype transition and chemo-resistance,which holds important clinical implications for improving SCLC treatment.Xuefeng Li Cheng Li Chenchen Guo Qiqi Zhao Jiayu Cao Hsin-Yi Huang Meiting Yue Yun Xue Yujuan Jin Liang Hu Hongbin Ji 2021Journal of Genetics and Genomics2021,48,7:5
4Dendritic BiVO4 decorated with MnOx co-catalyst as an efficient hierarchical catalyst for photocatalytic ozonation显示文摘An appropriate co-catalyst can significantly promote the photocatalytic efficacy, but this has been seldom studied in the visible-light photocatalysis combined with ozone, namely photocatalytic ozonation. In this work, a dendritic bismuth vanadium tetraoxide (BiVO4) material composited with highly dispersed MnOv nanoparticles was synthesized, and its catalytic activity is 86.6% higher than bare BiVO4 in a visible light and ozone combined process. Catalytic ozonation experiments, ultraviolet- visible (UV-Vis) diffuse reflectance spectra and photoluminescence spectra jointly indicate that MnOA plays a triple role in this process. MnOv strengthens the light adsorption and promotes the charge separation on the composite material, and it also shows good activity in catalytic ozonation. The key reactive species in this process is OH, and various pathways for its generation in this process is proposed. 5SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX.Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He 2022Cellular & Molecular Immunology2022,19,1:4
6Influence of nano-diamond content on the microstructure,mechanical and thermal properties of the ZK60 composites显示文摘The effect of nano diamond(ND)content on the microstructure,mechanical properties,and thermal conductivity of ZK60+x(x=0,0.05,0.1,0.15,0.2 wt.%)ND composites were investigated.The microstructures of ND/ZK60 composites were observed,which indicated that the nanoscale MgZn_(2) and ND particles distributed evenly in theα-Mg matrix.The tensile yield strength(TYS)and compressive yield strength(CYS)of the composites first increased remarkably and then decreased with further increasing the ND content.Due to the surface area of the matrix-diamond interface increased and the grains sizes of composites decreased with the amount of ND increase,which cause the coefficient of thermal expansion(CTE)of the composites reduced significantly.Meanwhile,the thermal conductivity of the composite material decreases from 129 W·m^(−1)·K^(−1) to 116 W·m^(−1)·K^(−1) with the content of ND increasing from 0.05%to 2.0%.The thermal conductivity of the composites increases to the maximum and then decrease with the increase of temperature(in temperature range of 273-573 K).The ZK60+0.05 ND showed superior mechanical and thermal conductivity property,TYS of 343.97 MPa,CYS of 341.74 MPa,elongation of 15.71%,CTE of 7.3×10^(−6)K^(−1),and thermal conductivity of 129 W·m^(−1)·K^(−1) at room temperature.It is demonstrated that the ND content has an obvious influence on the microstructure,mechanical properties,and thermal conductivity of ND/ZK60 composites.Hongbin Ma Jinhui Wang Hongyu Wang Ningning Dong Junwei Zhang Peipeng Jin Yong Peng 2022Journal of Magnesium and Alloys2022,10,2:3

8Distinct synergetic effects in the ozone enhanced photocatalytic degradation of phenol and oxalic acid with Fe^(3+)/TiO_2 catalyst显示文摘In this work, phenol and oxalic acid(OA) degradation in an ozone and photocatalysis integrated process was intensively conducted with Fe^(3+)/TiO_2 catalyst. The ferrioxalate complex formed between Fe^(3+) and oxalate accelerated the removal of OA in the ozonation, photolysis and photocatalytic ozonation process, for its high reactivity with ozone and UV. Phenol was degraded in ozonation and photolysis with limited TOC removal rates, but much higher TOC removal was achieved in photocatalytic ozonation due to the generation of ·OH. The sequence of UV light and ozone in the sequential process also influences the TOC removal, and ozone is very powerful to oxidize intermediates catechol and hydroquinone to maleic acid. Fenton or photo-Fenton reactions only played a small part in Fe^(3+)/TiO_2catalyzed processes, because Fe^(3+) was greatly reduced but not regenerated in many cases.The synergetic effect was found to be highly related with the property of the target pollutants. 13Evidence fusion procedure based on hybrid DSm model显示文摘Dezert-Smarandache(DSm) theory, a new information fusion theory, is widely applied in image processing, multiple targets tracking identification, and other areas for its excellent processing ability of imperfect information. However, earlier research on DSm theory mainly focused on one sort of questions. An evidence fusion procedure is proposed based on the hybrid DSm model to compensate for a lack of research on the entire information procedure of DSm theory. This paper analyzes the evidence fusion procedure, as well as correlative node input and output information. Key steps and detailed procedures of evidence fusion are also discussed. 14Evaluation of SARS-CoV-2-Neutralizing Nanobody Using Virus Receptor Binding Domain-Administered Model Mice显示文摘Due to the rapid spread of coronavirus disease 2019(COVID-19),there is an urgent requirement for the development of additional diagnostic tools for further analysis of the disease.The isolated nanobody Nb11-59 binds to the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)reeptor-binding domain[RBD]with high afinity to neutralize the virus and block the angiotensin-converting enzyme 2-(ACE2-)RBD interaction.Here,we introduce a novel nanobody-based radiotracer named^(68)Ga-Nb1159.The radiotracer retained high affinity for the RBD and showed reliable radiochemical characteristics both in vitro and in vivo.Predinical positron emission tomography(PET)studies of Ga-Nb1159 in mice revealed is rapid dearance from circulation and robust uptake into the renal and urinary systems.Fortunately,^(68)Ga-Nb1159 could speifcally reveal the distribution of the RBD in mice.This sudy also helped to evaluate the pharmacodynamic efects of the neutralizing nanobody.Moreover,^(68)Ga-Nb1159 may be a promising l0ol lo explore the distribution of the RBD and improve the understanding of the virus.In particular,this study identified a novel molecular radioagent and established a reliable evaluation method for speifially investigating the RBD through noninvasive and visual PET technology.Song Liu Guanghui Li Lei Ding Jin Ding Qian Zhang Dan Li Xingguo Hou Xiangxing Kong Jing Zou Shiming Zhang Hongbin Han Yakun Wan Zhi Yang Hua Zhu 2022Research2022,,2:2

17Optimization of Cottonseed Meal De-gossypol Process by Extrusion via Genetic Algorithm Based on Artificial Neural Network显示文摘[Objective] The paper aimed to optimize cottonseed meal de-gossypol process by extrusion. [Method]The artificial neural network( ANN) was used to stimulate the degradation of free gossypol in cottonseed meal by extrusion process,and a three-layer back propagation neural network was established to predict the degradation of free gossypol. [Result] The result of 10-fold cross validation showed that the ANN with the training function as traingdx at hidden layer with eight neurons gave the smallest mean square error( MSE). ANN predicted results were very close to the experimental results with correlation coefficient( R2) of0. 994 1 and RMSE of 0. 497 1. A genetic algorithm( GA) based on the established neural network model was also used for optimizing de-gossypol process. The results of GA showed that the optimal conditions of de-gossypol were puffing temperature 131℃,water ratio 51%,rotational speed 158 r / min,and feeding speed 136 kg / h,while under this condition the degradation rate of free gossypol was 90. 50%,which was close to the predicted result of GA with the small average relative error of 1. 38%.
