2GOLM1 restricts colitis and colon tumorigenesis by ensuring Notch signaling equilibrium in intestinal homeostasis显示文摘Intestinal epithelium serves as the first barrier against the infections and injuries that mediate colonic inflammation.Colorectal cancer is often accompanied with chronic inflammation.Differed from its well-known oncogenic role in many malignancies,we present here that Golgi membrane protein 1(G0LM1,also referred to as GP73)suppresses colorectal tumorigenesis via maintenance of intestinal epithelial barrier.G0LM1 deficiency in mice conferred susceptibility to mucosal inflammation and colitis-induced epithelial damage,which consequently promoted colon cancer.Mechanistically,depletion of G0LM1 in intestinal epithelial cells(lECs)led to aberrant Notch activation that interfered with IEC differentiation,maturation,and lineage commitment in mice.Pharmacological inhibition of Notch pathway alleviated epithelial lesions and restrained pro-tumorigenic inflammation in G0LM1-deficient mice.Therefore,G0LM1 maintains IEC homeostasis and protects against colitis and colon tumorigenesis by modulating the equilibrium of Notch signaling pathway.Yang Pu Ya Song Mengdi Zhang Caifeng Long Jie Li Yanan Wang Yinzhe Xu Fei Pan Na Zhao Xinyu Zhang Yanan Xu Jianxin Cui Hongying Wang Yan Li Yong Zhao Di Jin Hongbing Zhang 2021Signal Transduction and Targeted Therapy2021,6,5:2
3SUMOylation-modified Pelota-Hbs1 RNA surveillance complex restricts the infection of potyvirids in plants显示文摘RNA quality control nonsense-mediated decay is involved in viral restriction in both plants and animals.However,it is not known whether two other RNA quality control pathways,nonstop decay and no-go decay,are capable of restricting viruses in plants.Here,we show that the evolutionarily conserved Pelota–Hbs1 complex negatively regulates infection of plant viruses in the family Potyviridae(termed potyvirids),the largest group of plant RNA viruses that accounts for more than half of the viral crop damage worldwide.Pelota enables the recognition of the functional G1-2A6-7 motif in the P3 cistron,which is conserved in almost all potyvirids.This allows Pelota to target the virus and act as a viral restriction factor.Furthermore,Pelota interacts with the SUMO E2-conjugating enzyme SCE1 and is SUMOylated in planta.Blocking Pelota SUMOylation disrupts the ability to recruit Hbs1 and inhibits viral RNA degradation.These findings reveal the functional importance of Pelota SUMOylation during the infection of potyvirids in plants.Linhao Ge Buwei Cao Rui Qiao Hongguang Cui Shaofang Li Hongying Shan Pan Gong Mingzhen Zhang Hao Li Aiming Wang Xueping Zhou Fangfang Li 2023Molecular Plant2023,16,3:2

10Rapid divergent coevolution of Sinopotamon freshwater crab genitalia facilitates a burst of species diversification显示文摘One of the most striking radiations in brachyuran evolution is the considerable morphological diversification of the external reproductive structures of primary freshwater crabs:the male first gonopod(G1)and the female vulva(FV).However,the lack of quantitative studies,especially the lack of data on female genitalia,has seri­ously limited our understanding of genital evolution in these lineages.Here we examined 69 species of the large Chinese potamid freshwater crab genus Sinopotamon Bott,1967(more than 80%of the described species).We used a landmark-based geometric morphometric approach to analyze variation in the shape of the G1 and FV,and to compare the relative degree of variability of the genitalia with non-reproductive structures(the third maxillipeds).We found rapid divergent evolution of the genitalia among species of Sinopotamon when com­pared to non-reproductive traits.In addition,the reconstruction of ancestral groundplans,together with plot­ting analyses,indicated that the FV show the most rapid divergence,and that changes in FV traits correlate with changes in G1 traits.Here we provide new evidence for coevolution between the male and female external gen­italia of Sinopotamon that has likely contributed to rapid divergent evolution and an associated burst of specia­tion in this lineage.Fengxin YAO Boyang SHI Xiaoqi WANG Da PAN Ming BAI Jie YAN Neil CUMBERLIDGE Hongying SUN 2020Integrative Zoology2020,15,3:0

12Construction and validation of a risk-prediction model for anastomotic leakage after radical gastrectomy: A cohort study in China显示文摘Objectives:Anastomotic leakage(AL)stands out as a prevalent and severe complication following gastric cancer surgery.It frequently precipitates additional serious complications,significantly influencing the overall survival time of patients.This study aims to enhance the risk-assessment strategy for AL following gastrectomy for gastric cancer.Methods:This study included a derivation cohort and validation cohort.The derivation cohort included patients who underwent radical gastrectomy at Sir Run Run Shaw Hospital,Zhejiang University School of Medicine,from January 1,2015 to December 31,2020.An evidence-based predictor questionnaire was crafted through extensive literature review and panel discussions.Based on the questionnaire,inpatient data were collected to form a model-derivation cohort.This cohort underwent both univariate and multivariate analyses to identify factors associated with AL events,and a logistic regression model with stepwise regression was developed.A 5-fold cross-validation ensured model reliability.The validation cohort included patients from August 1,2021 to December 31,2021 at the same hospital.Using the same imputation method,we organized the validation-queue data.We then employed the risk-prediction model constructed in the earlier phase of the study to predict the risk of AL in the subjects included in the validation queue.We compared the predictions with the actual occurrence,and evaluated the external validation performance of the model using model-evaluation indicators such as the area under the receiver operating characteristic curve(AUROC),Brier score,and calibration curve.Results:The derivation cohort included 1377 patients,and the validation cohort included 131 patients.The independent predictors of AL after radical gastrectomy included age65 y,preoperative albumin<35 g/L,resection extent,operative time240 min,and intraoperative blood loss90 mL.The predictive model exhibited a solid AUROC of 0.750(95%CI:0.694e0.806;p<0.001)with a Brier score of 0.049.The 5-fold cross-validation confirmed these findings with a calibrated C-index of 0.749 and an average Brier score of 0.052.External validation showed an AUROC of 0.723(95%CI:0.564e0.882;p?0.006)and a Brier score of 0.055,confirming reliability in different clinical settings.Conclusions:We successfully developed a risk-prediction model for AL following radical gastrectomy.This tool will aid healthcare professionals in anticipating AL,potentially reducing unnecessary interventions.Jinrui Wang Xiaolin Liu Hongying Pan Yihong Xu Mizhi Wu Xiuping Li Yang Gao Meijuan Wang Mengya Yan 2024Laparoscopic, Endoscopic and Robotic Surgery2024,7,1:0

14Numerical study of laser-induced collision process in Eu-Sr in strong field显示文摘Theoretically based on the four-level model, one LICET process in Eu-Sr system in both weak and strong fields was calculated by immediate numerical integrations. Numerical results in weak field are in fair to good agreement with analytical ones.
