1In vivo self-assembled small RNAs as a new generation of RNAi therapeutics显示文摘RNAi therapy has undergone two stages of development,direct injection of synthetic siRNAs and delivery with artificial vehicles or conjugated ligands;both have not solved the problem of efficient in vivo siRNA delivery.Here,we present a proof-of-principle strategy that reprogrammes host liver with genetic circuits to direct the synthesis and self-assembly of siRNAs into secretory exosomes and facilitate the in vivo delivery of siRNAs through circulating exosomes.By combination of different genetic circuit modules,in vivo assembled siRNAs are systematically distributed to multiple tissues or targeted to specific tissues(e.g.,brain),inducing potent target gene silencing in these tissues.The therapeutic value of our strategy is demonstrated by programmed silencing of critical targets associated with various diseases,including EGFR/KRAS in lung cancer,EGFR/TNC in glioblastoma and PTP1B in obesity.Overall,our strategy represents a next generation RNAi therapeutics,which makes RNAi therapy feasible.Zheng Fu Xiang Zhang Xinyan Zhou Uzair Ur-Rehman Mengchao Yu Hongwei Liang Hongyuan Guo Xu Guo Yan Kong Yuanyuan Su Yangyang Ye Xiuting Hu Wei Cheng Jinrong Wu Yanbo Wang Yayun Gu Sheng-feng Lu Dianqing Wu Ke Zen Jing Li Chao Yan Chen-Yu Zhang Xi Chen 2021Cell Research2021,31,6:10
2Effect of VC addition on the microstructure and properties of Ti(C,N)-based nano cermets显示文摘In this paper,Ti(C,N)-based nano cermets were prepared by nano particles,and the effect of VC addition on the microstructure and proper-ties of Ti(C,N)-based nano cermets was investigated.The results showed that there existed black-core grayish-rim structure as well as gray-core grayish-rim structure in VC-doped Ti(C,N)-based nano cermets.With the increase of VC addition,the number of gray cores in-creased,the lattice parameter of Ti(C,N) phase increased,the grain size decreased,the hardness and fracture toughness of Ti(C,N)-based nano cermets were enhanced,and nearly full densification could be achieved.However,excessive addition of VC to 1 wt% resulted in slight decrease in hardness and fracture toughness.Some deep dimples were found in the fracture surface of cermets with VC addition,which cor-responded to ductile fracture.YING Xiayu XIONG Ji GUO Zhixing CHEN Jianzhong XIONG Sujian YANG Mei FAN Hongyuan 2011Rare Metals2011,30,6:7

4Changes in Soil Enzyme Activity and Nutrient Content in Different Years of Continuous Cropping Tobacco Fields显示文摘In order to find out changes in the main fertility indicators of continuous cropping tobacco fields in Henan Province with years of continuous cropping,5 typical tobacco fields( Jia County,Shaoling District,Fangcheng County,Xiangcheng County,and Song County) were studied. Indicators in question included activity of main enzymes( sucrase,urease,catalase,and acid phosphatase),p H value,organic matters,total nitrogen,available phosphorus and available potassium. The results indicate that in most tobacco fields,the activity of sucrase and urease increased in two years of continuous cropping,later,it showed a declining trend with increase in years of continuous cropping; in some fields,it directly declined with increase in years of continuous cropping; the activity of catalase increased in two years of continuous cropping,later,it showed a declining trend with increase in years of continuous cropping,and it became stable after three years of continuous cropping;generally,the activity of acid phosphatase declined with increase in the years of continuous cropping within 3 years continuous cropping,later,it became stable. Except Fangcheng County,the soil p H value of other districts( counties) generally declined with increase in years of continuous cropping,and it was in the range of 5. 6-6. 8. With the increase in continuous cropping,the soil organic matters( SOM) in tobacco fields of Jia County and Song County declined,changes of SOM in tobacco fields of Fangcheng County and Xiangcheng County were not obvious,and SOM showed the trend of first decline then rise in Shaoling District. In most tobacco fields( Jia County,Xiangcheng County,and Song County),the total nitrogen showed a declining trend; in Shaoling District,it showed a trend of first decline then rise; in Fangcheng County,it showed a rising trend. The available phosphorus firstly slightly declined then rose in tobacco fields of Shaoling District and Song County; it showed a trend of first rise then decline in Jia County; there were no obvious changes in Fangcheng County and Xiangcheng County. Changes in the available potassium were different in tobacco fields. In sum,there are certain rules for changes in soil enzyme activity and nutrient content in tobacco fields in Henan Province,but changes are not completely the same. It is concluded that the site conditions and farming activities exert a certain influence on soil enzyme activity,p H value,organic matters,and nitrogen,phosphorus and potassium content. 5Superflexible and Lead-Free Piezoelectric Nanogenerator as a Highly Sensitive Self-Powered Sensor for Human Motion Monitoring显示文摘For traditional piezoelectric sensors based on poled ceramics,a low curie tem-perature(T_(c))is a fatal flaw due to the depolarization phenomenon.However,in this study,we find the low T_(c) would be a benefit for flex-ible piezoelectric sensors because small alterations of force trig-ger large changes in polarization.BaTi_(0.88)Sn_(0.12)O_(3)(BTS)with high piezoelectric coefficient and low T_(c) close to human body temperature is taken as an example for materials of this kind.Continuous piezo-electric BTS films were deposited on the flexible glass fiber fabrics(GFF),self-powered sensors based on the ultra-thin,superflexible,and polarization-free BTS-GFF/PVDF composite piezoelectric films are used for human motion sensing.In the low force region(1-9 N),the sensors have the outstanding performance with voltage sensitivity of 1.23 V N^(−1) and current sensitivity of 41.0 nA N^(−1).The BTS-GFF/PVDF sensors can be used to detect the tiny forces of falling water drops,finger joint motion,tiny surface deformation,and fatigue driving with high sensitivity.This work provides a new paradigm for the preparation of superflexible,highly sensitive and wearable self-powered piezoelectric sensors,and this kind of sensors will have a broad application prospect in the fields of medical rehabilitation,human motion monitoring,and intelligent robot.Di Yu Zhipeng Zheng Jiadong Liu Hongyuan Xiao Geng Huangfu Yiping Guo 2021Nano-Micro Letters2021,13,8:2

7The BZR1-EDS1 module regulates plant growth-defense coordination显示文摘Plants have developed sophisticated strategies to coordinate growth and immunity,but our understanding of the underlying mechanism remains limited.In this study,we identified a novel molecular module that reg-ulates plant growth and defense in both compatible and incompatible infections.This module consisted of BZR1,a key transcription factor in brassinosteroid(BR)signaling,and EDS1,an essential positive regulator of plant innate immunity.We found that EDS1 interacts with BZR1 and suppresses its transcriptional activ-ities.Consistently,upregulation of EDS1 function by a virulent Pseudomonas syringae strain or salicylic acid treatment inhibited BZR1-regulated expression of BR-responsive genes and BR-promoted growth.Furthermore,we showed that the cytoplasmic fraction of BZR1 positively regulates effector-triggered im-munity(ETI)controlled by the TIR-NB-LRR protein RPS4,which is attenuated by BZR1's nuclear transloca-tion.Mechanistically,cytoplasmic BZR1 facilitated AvrRps4-triggered dissociation of EDS1 and RPS4 by binding to EDS1,thus leading to efficient activation of RPS4-controlled ETI.Notably,transgenic expression of a mutant BZR1 that accumulates exclusively in the cytoplasm improved pathogen resistance without compromising plant growth.Collectively,these results shed new light on plant growth-defense coordina-tion and reveal a previously unknown function for the cytoplasmic fraction of BZR1.The BZR1-EDS1 mod-ule may be harnessed for the simultaneous improvement of crop productivity and pathogen resistance.Guang Qi Huan Chen Dian Wang Hongyuan Zheng Xianfeng Tang Zhengzheng Guo Jiayu Cheng Jian Chen Yiping Wang Ming-yi Bai Fengquan Liu Daowen Wang Zheng Qing Fu 2021Molecular Plant2021,14,12:1
8Model-Simulated Coastal Trapped Waves Stimulated by Typhoon in Northwestern South China Sea显示文摘In this paper, we apply an unstructured grid coastal ocean model to simulate variations in the sea level and currents forced by two typhoons in the northwestern South China Sea(SCS). The model simulations show distinct differences for the two cases in which the typhoon paths were north and south of the Qiongzhou(QZ) Strait. In both cases, coastal trapped waves(CTWs) are stimulated but their propagation behaviors differ. Model sensitivity simulations suggest the dominant role played by alongshore wind in the eastern SCS(near Shanwei) and southeast of Hainan Island. We also examine the influence of the Leizhou Peninsula by changing the coastline in simulation experiments. Based on our results, we can draw the following conclusions: 1) The CTWs stimulated by the northern typhoon are stronger than the southern CTW. 2) In the two cases, the directions of the current structures of the QZ cross-transect are reversed. The strongest flow cores are both located in the middle-upper area of the strait and the results of our empirical orthogonal function analysis show that the vertical structure is highly barotropic. 3) The simulated CTWs divide into two branches in the QZ Strait for the northern typhoon, and an island trapped wave(ITW) around Hainan Island for the southern typhoon. 4) The Leizhou Peninsula plays a significant role in the distribution of the kinetic energy flux between the two CTW branches. 10Axial compression behavior of CFRP-confined rectangular concrete-filled stainless steel tube stub column显示文摘The mechanical properties of CFRP-confined rectangular concrete-filled stainless steel tube(CFSST)stub columns under axial compression were experimentally studied.A total of 28 specimens(7 groups)were fabricated for the axial compression test to study the influences of length-to-width ratio,CFRP constraint coefficient,and the thickness of stainless steel tube on the axial compression behavior.The specimen failure modes,the stress development of stainless steel tube and CFRP wrap,and the load-strain ratio curves in the loading process were obtained.Meanwhile,the relationship between axial and transverse deformations of each specimen was analyzed through the typical relative load-strain ratio curves.A bearing capacity prediction method was proposed based on the twin-shear strength theory,combining the limit equilibrium state of the CFRP-confined CFSST stub column under axial compression.The prediction method was calibrated by the test data in this study and other literature.The results show that the prediction method is of high accuracy.Hongyuan TANG Ruizhong LIU Xin ZHAO Rui GUO Yigang JIA 2021Frontiers of Structural and Civil Engineering2021,15,5:1

17Real-time analysis of inflammatory cytokines and regulatory gene expression in tissues surrounding the hematoma after intracerebral hemorrhage显示文摘BACKGROUND:Secondary lesions can occur in tissues surrounding the hematoma following intracerebral hemorrhage,with the presence of inflammatory reactions,cytokine expression and apoptosis. These have been confirmed in animal studies. Our study sought to determine whether these could be detected in human tissues surrounding the hematoma following intracerebral hemorrhage. OBJECTIVE: To investigate expression of inflammatory cytokines,Bax and Bcl-x,and identify neural cell apoptosis in tissues surrounding the hematoma,and to analyze the correlation between them and pathological damage in intracerebral hemorrhage patients. DESIGN,TIME AND SETTING: This histopathology,controlled study was performed at the Department of Neurosurgery,Sichuan People’s Hospital,China,from January 2003 to January 2005. PARTICIPANTS: Brain tissues 1 cm from the hematoma in 30 intracerebral hemorrhage patients served as the experimental group. Brain tissues located away from the hematoma in 7 patients served as the control group. METHODS: TUNEL was used to detect neural cell apoptosis. Immunohistochemistry (labeled dextran polymer) and RT-PCR were used to measure tumor necrosis factor-α,interleukin-1β,interleukin-6,Bax and Bcl-x protein and mRNA expression. Pathological changes in brain tissues surrounding the hematoma were observed following HE staining. MAIN OUTCOME MEASURES: Neural cell apoptosis,inflammatory cytokines,Bax and Bcl-x protein and mRNA expression,pathological changes in brain tissues surrounding the hematoma. RESULTS: Brain tissues surrounding the hematoma were mildly damaged within 6 hours,severely damaged at 24-72 hours,and significantly improved 1 week following intracerebral hemorrhage. Expression of tumor necrosis factor-α protein and mRNA,interleukin-1β and interleukin-6 mRNA was not significant in tissues surrounding the hematoma,which was identical to the control group within 6 hours after intracerebral hemorrhage. This expression was significantly higher compared with the control group from 12-72 hours,and gradually decreased after 72 hours. The number of apoptotic neural cells reached a peak between 12-72 hours. Tumor necrosis factor-α protein and mRNA,interleukin-1β and interleukin-6 mRNA levels were positively correlated with apoptosis,Bax protein and mRNA levels (P<0.01). CONCLUSION: Tumor necrosis factor-α,interleukin-1β,and interleukin-6 levels are highly correlated with apoptosis. 19Simulation and Analysis of Back Siltation in a Navigation Channel Using MIKE 21显示文摘The channel back-siltation problem has been restricting the development of channels,and its monitoring is limited by funds and natural conditions.Moreover,predicting the channel back-siltation situation in a timely and accurate manner is difficult.Hence,a numerical simulation of the back-siltation problem in the sea area near the channel is of great significance to the maintenance of a channel.In this study,the back siltation of a deep-water channel in the Lanshan Port area of the Port of Rizhao after dredging is predicted.This paper relies on the MIKE 21 software to establish the wave,tidal current,and sediment numerical models and uses measured data from two observation stations in the study area for verification.On this basis,taking one month as an example,the entire project channel was divided into five sections,and three observation points were set on each section.The results show that the area with offshore siltation is located in the northerly direction of the artificial anti-wave building.Siltation occurred on the northern seabed in the sea a little farther from the shore.Siltation occurred on the seabed surface far away from the shoreline,and with the increase in the distance from the shoreline,the amount of siltation in the south,center,and north became gradually closed,and the results can be used to guide actual engineering practices.This study will play a positive role in promoting the dredging project of Rizhao Lanshan Port.ZHANG Kuncheng LI Qingjie ZHANG Jing SHI Hongyuan YU Jing GUO Xinchang DU Yonggang 2022Journal of Ocean University of China2022,21,4:0
20Transforming bulk alkenes and alkynes into fine chemicals enabled by single-atom site catalysis显示文摘The addition and substitution reactions across a variety of alkenes and alkynes have been playing an important role in organic synthesis.In particular,catalyses enabled by homogeneous complexes as well as heterogenous nanomaterials have been much received attentions over decades.Along with these blooming progresses in these fields,single-atom site catalysts(SACs)exhibit outstanding performances in terms of reactivity and selectivity,thus providing a new powerful strategy to upgrade these bulk chemicals.Herein,we summarize the reactions of alkenes and alkynes enabled by SACs,in which catalytic hydrogenation,hydrosilylation,hydroboration,hydroformylation,and cross coupling have been included.Moreover,preparations and fine structures of the corresponding SACs have been described,therefore providing a better understanding and overview to address the origin of catalytic activity in the reported examples.Ping Guo Hongyuan Liu Jie Zhao 2022Nano Research2022,15,9:0
