| 1 | CAS FGOALS-f3-L Model Datasets for CMIP6 Historical Atmospheric Model Intercomparison Project Simulation显示文摘The outputs of the Chinese Academy of Sciences(CAS) Flexible Global Ocean–Atmosphere–Land System(FGOALS-f3-L) model for the baseline experiment of the Atmospheric Model Intercomparison Project simulation in the Diagnostic,Evaluation and Characterization of Klima common experiments of phase 6 of the Coupled Model Intercomparison Project(CMIP6) are described in this paper. The CAS FGOALS-f3-L model, experiment settings, and outputs are all given. In total,there are three ensemble experiments over the period 1979–2014, which are performed with different initial states. The model outputs contain a total of 37 variables and include the required three-hourly mean, six-hourly transient, daily and monthly mean datasets. The baseline performances of the model are validated at different time scales. The preliminary evaluation suggests that the CAS FGOALS-f3-L model can capture the basic patterns of atmospheric circulation and precipitation well, including the propagation of the Madden–Julian Oscillation, activities of tropical cyclones, and the characterization of extreme precipitation. | 2 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 3 | In vitro and in vivo activity of D-serine in combination with β-lactam antibiotics against methicillin-resistant Staphylococcus aureus显示文摘As D-amino acids play important roles in the physiological metabolism of bacteria,combination of D-amino acids with antibiotics may provide synergistic antibacterial activity. The aim of the study was to evaluate in vitro and in vivo activity of D-serine alone and in combination withβ-lactams against methicillin-resistant Staphylococcus aureus(MRSA) strains, and to explore the possible sensitization mechanisms. The activity of D-serine, β-lactams alone and in combinations was evaluated both in vitro by standard MICs, time–kill curves and checkerboard assays, and in vivo by murine systemic infection model as well as neutropenic thigh infection model. An in vitro synergistic effect was demonstrated with the combination of D-serine and β-lactams against MRSA standard and clinical strains.Importantly, the combinations enhanced the therapeutic efficacy in the animal models as compared toβ-lactam alone groups. Initial mechanism study suggested possible revision of D-alanine-D-alanine residue to D-alanine-D-serine in peptidoglycan by adding of D-alanine in the medium, which may cause decreased affinity to PBPs during transpeptidation. In conclusion, D-serine had synergistic activity in combination with β-lactams against MRSA strains both in vitro and in vivo. | 4 | Investigation into the flotation response of refractory molybdenum ore to depressant mixtures:A case study显示文摘In this paper, bulk flotation followed by separation was investigated to concentrate purified molybdenite product from Jinduicheng molybdenum ores(Shanxi province, China). The bench scale tests mainly focussed on separation of molybdenite from other sulfide minerals using the new type of depressants.The effect of each single depressant, including organic depressant-modified dextrin(MD), P-Nokes reagent(PN) and sodium trithiocarbonate(ST), and their mixtures on galena, chalcopyrite and other sulfide ores, was examined in turn by changing the concentrations used in cleaner flotation tests. Closed circuit experiments were carried out under the optimal condition and satisfying recovery and grade of molybdenite concentrate could be achieved(86.294% and 53.157%, respectively). | 6 | Up-regulation of glycolipid transfer protein by bicyclol causes spontaneous restriction of hepatitis C virus replication显示文摘Bicyclol is a synthetic drug for hepatoprotection in clinic since 2004. Preliminary clinical observations suggest that bicyclol might be active against hepatitis C virus(HCV) with unknown mechanism. Here, we showed that bicyclol significantly inhibited HCV replication in vitro and in hepatitis C patients. Using bicyclol as a probe, we identified glycolipid transfer protein(GLTP) to be a novel restrictive factor for HCV replication. The GLTP preferentially bound host vesicle-associated membrane protein-associated protein-A(VAP-A) in competition with the HCV NS5 A, causing an interruption of the complex formation between VAP-A and HCV NS5 A. As the formation of VAP-A/NS5 A complex is essential for viral RNA replication, up-regulation of GLTP by bicyclol reduced the level of VAP-A/NS5 A complex and thus inhibited HCV replication. Bicyclol also exhibited an inhibition on HCV variants resistant to direct-acting antiviral agents(DAAs) with an efficacy identical to that on wild type HCV. In combination with bicyclol, DAAs inhibited HCV replication in a synergistic fashion. | 7 | Joining of Zirconia and Ti-6Al-4V Using a Ti-based Amorphous Filler显示文摘Polycrystalline ZrO2-3 mol.%Y2O3 was brazed to Ti-6Al-4V by using a Ti47Zr28Cu14Ni11(at.%) amorphous ribbon at 1123-1273 K in a high vacuum. The influences of brazing temperature on the microstructure and shear strength of the joints were investigated. The interfacial microstructures can be described as ZrO2/TiO+TiO2+Cu2Ti4O+Ni2Ti4O/α-Ti+(Ti,Zr)2(Cu,Ni) eutectic/acicular Widmanst¨aten structure/Ti-6Al-4V alloy. With the increase in the brazing temperature, the thickness of the TiO+TiO2+Cu2Ti4O+Ni2Ti4O layer reduced, the content of the α-Ti+(Ti,Zr)2(Cu,Ni) eutectic phase decreased, while that of the coarse α-Ti phase gradually increased. The shear strength of the joints did not show a close relationship with the thickness of the TiO+TiO2+Cu2Ti4O+Ni2Ti4O layer. The rapidly solidified microstructure in the alloyed layer was studied. The results showed that the alloying element distributed in the alloyed layer is inhomogeneous. The dendrite containing relatively high Ni was identified as Al3Ni phase and the areas between the dendrites are rich in Al content. Featureless with cell structure in Al-rich areas was firstly displayed by z-contrast image. | 13 | Serological Investigation of Laboratory-Confirmed and Suspected Ebola Virus Disease Patients During the Late Phase of the Ebola Outbreak in Sierra Leone显示文摘This study aimed to investigate the serological characteristics of Ebola virus(EBOV) infection during the late phase of the Ebola outbreak in Sierra Leone. In total, 877 blood samples from 694 suspected Ebola virus disease(EVD) cases assessed from March to December 2015, were analyzed via real-time reverse transcription polymerase chain reaction(RT-PCR) for viral RNA and enzyme-linked immunosorbent assay(ELISA) and Luminex to detect antibodies against EBOV. Viral load and EBOV-specific IgM/IgG titers displayed a declining trend during March to December 2015. Viral RNA load decreased rapidly at earlier stages after disease onset, while EBOV-specific IgM and IgG still persisted in 58.1%(18/31) and 93.5%(29/31) of the confirmed EVD patients and in 3.8%(25/663) and 17.8%(118/663) of the RNA-negative suspected patients in the later phase, respectively. | 14 | Protective effects of sodium butyrate on rotavirus inducing endoplasmic reticulum stress-mediated apoptosis via PERK-eIF2αsignaling pathway in IPEC-J2 cells显示文摘Background:Rotavirus(RV)is a major pathogen that causes severe gastroenteritis in infants and young animals.Endoplasmic reticulum(ER)stress and subsequent apoptosis play pivotal role in virus infection.However,the protective mechanisms of intestinal damage caused by RV are poorly defined,especially the molecular pathways related to enterocytes apoptosis.Thus,the aim of this study was to investigate the protective effect and mechanism of sodium butyrate(SB)on RV-induced apoptosis of IPEC-J2 cells.Results:The RV infection led to significant cell apoptosis,increased the expression levels of ER stress(ERS)markers,phosphorylated protein kinase-like ER kinase(PERK),eukaryotic initiation factor 2 alpha(eIF2α),caspase9,and caspase3.Blocking PERK pathway using specific inhibitor GSK subsequently reversed RV-induced cell apoptosis.The SB treatment significantly inhibited RV-induced ERS by decreasing the expression of glucose regulated protein 78(GRP78),PERK,and eIF2α.In addition,SB treatment restrained the ERS-mediated apoptotic pathway,as indicated by downregulation of C/EBP homologous protein(CHOP)mRNA level,as well as decreased cleaved caspase9 and caspase3 protein levels.Furthermore,siRNA-induced GPR109a knockdown significantly suppressed the protective effect of SB on RV-induced cell apoptosis.Conclusions:These results indicate that SB exerts protective effects against RV-induced cell apoptosis through inhibiting ERS mediated apoptosis by regulating PERK-eIF2αsignaling pathway via GPR109a,which provide new ideas for the prevention and control of RV. | Ye Zhao Ningming Hu Qin Jiang Li Zhu Ming Zhang Jun Jiang Manyi Xiong Mingxian Yang Jiandong Yang Linyuan Shen Shunhua Zhang Lili Niu Lei Chen Daiwen Chen | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 2 |
