| 1 | Protective effects of sodium butyrate on rotavirus inducing endoplasmic reticulum stress-mediated apoptosis via PERK-eIF2αsignaling pathway in IPEC-J2 cells显示文摘Background:Rotavirus(RV)is a major pathogen that causes severe gastroenteritis in infants and young animals.Endoplasmic reticulum(ER)stress and subsequent apoptosis play pivotal role in virus infection.However,the protective mechanisms of intestinal damage caused by RV are poorly defined,especially the molecular pathways related to enterocytes apoptosis.Thus,the aim of this study was to investigate the protective effect and mechanism of sodium butyrate(SB)on RV-induced apoptosis of IPEC-J2 cells.Results:The RV infection led to significant cell apoptosis,increased the expression levels of ER stress(ERS)markers,phosphorylated protein kinase-like ER kinase(PERK),eukaryotic initiation factor 2 alpha(eIF2α),caspase9,and caspase3.Blocking PERK pathway using specific inhibitor GSK subsequently reversed RV-induced cell apoptosis.The SB treatment significantly inhibited RV-induced ERS by decreasing the expression of glucose regulated protein 78(GRP78),PERK,and eIF2α.In addition,SB treatment restrained the ERS-mediated apoptotic pathway,as indicated by downregulation of C/EBP homologous protein(CHOP)mRNA level,as well as decreased cleaved caspase9 and caspase3 protein levels.Furthermore,siRNA-induced GPR109a knockdown significantly suppressed the protective effect of SB on RV-induced cell apoptosis.Conclusions:These results indicate that SB exerts protective effects against RV-induced cell apoptosis through inhibiting ERS mediated apoptosis by regulating PERK-eIF2αsignaling pathway via GPR109a,which provide new ideas for the prevention and control of RV. | Ye Zhao Ningming Hu Qin Jiang Li Zhu Ming Zhang Jun Jiang Manyi Xiong Mingxian Yang Jiandong Yang Linyuan Shen Shunhua Zhang Lili Niu Lei Chen Daiwen Chen | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 2 |
| 2 | SHANK2 is a frequently amplified oncogene with evolutionarily conserved roles in regulating Hippo signaling显示文摘Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth.However,for a significant portion of human cancer,how Hippo signaling is perturbed remains unknown.To answer this question,we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome.In our screen,Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth.Interestingly,a mammalian homolog of Prosap,SHANK2,is the most frequently amplified gene on 11 q13,a major tumor amplicon in human cancer.Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification.More importantly,across the human cancer genome,SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon.Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator,and as a potential oncogene,SHANK2 promotes cellular transformation and tumor formation in vivo.In cancer cell lines with deregulated Hippo pathway,depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation.Taken together,these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator,commonly amplified in human cancer and potently promotes cancer.Our study for the first time illustrated oncogenic function of SHANK2,one of the most frequently amplified gene in human cancer.Furthermore,given that in normal adult tissues,SHANK2 s expression is largely restricted to the nervous system,SHANK2 may represent an interesting target for anticancer therapy. | Liang Xu Peixue Li Xue Hao Yi Lu Mingxian Liu Wenqian Song Lin Shan Jiao Yu Hongyu Ding Shishuang Chen Ailing Yang Yi Arial Zeng Lei Zhang Hai Jiang | 2021 | Protein & Cell2021,12,3: | 1 |
| 3 | Trapping precursor-level functionalities in hierarchically porous carbons prepared by a pre-stabilization route for superior supercapacitors显示文摘Versatile module design of precursor networks enables flexible functionalization of nano-carbon electrode materials to meet the adaptable energy-storage demand. Functionalized heterogeneous networks are more likely to decompose by swift temperature programming together with predesign module removal, so high functionality/network transfer from precursor to carbon is still a work in progress. A pre-stabilization route is proposed here to enhance the network strength at early pyrolysis and pin up precursor-level functionalities on the final carbon. Such strategy successfully fixes more electroactive N(4.28-8.86 wt%) into the resultant carbon microspheres compared with non-pretreated carbon(2.89wt%), as well as achieves broad ion-accessible platforms of 1575-2269 m^(2)/g with preset structural superiorities. As a result, a typical acidic device reveals an outstanding specific capacitance of 383 F/g at 10 mV/s. Taking advantage of a novel LiNO_(3)-PAM polymer electrolyte, the upgraded symmetric device displays the maximum specific capacitance of 229 F/g, along with a boosted energy density of 41.1 Wh/kg at 643.4 W/kg. | 4 | Distinct Assembly Mechanisms for Prokaryotic and Microeukaryotic Communities in the Water of Qinghai Lake显示文摘Assembly processes of prokaryotic and microeukaryotic community is an important issue in microbial ecology.However,unclear remains about the relative contribution of deterministic and stochastic processes to the shaping of prokaryotic and microeukaryotic communities in saline lake water.Here,we systematically investigated the assembly processes governing the prokaryotic and microeukaryotic communities in Qinghai Lake with the use of Illumina sequencing and a null model.The results showed that both deterministic and stochastic processes play vital roles in shaping the assemblies of prokaryotic and microeukaryotic communities,in which stochastic processes appeared to dominate(> 70%).Prokaryotic communities were mainly governed by non-dominant processes(60.4%),followed by homogeneous selection(15.8%),variable selection(13.6%) and dispersal limitation(10.2%),whereas microeukaryotes were strongly driven by non-dominant processes(68.9%),followed by variable selection(23.6%) and homogenizing dispersal(6.3%).In terms of variable selection,nutrients(e.g.,ammonium,dissolved inorganic carbon,dissolved organic carbon and total nitrogen) were the major factors influencing prokaryotic and microeukaryotic community structures.In summary,prokaryotes and microeukaryotes can be predominantly structured by different assembly mechanisms,in which stochasticity is stronger than deterministic processes.This finding helps to better comprehend the assembly of prokaryotic and eukaryotic communities in saline lakes. | Mingxian Han Jianrong Huang Jian Yang Beichen Wang Xiaoxi Sun Hongchen Jiang | 2023 | Journal of Earth Science2023,34,4: | 0 |
