1Interleukin-1β is a potential therapeutic target for periodontitis: a narrative review显示文摘Interleukin(IL)-1β, a pro-inflammatory cytokine, was elevated and participates in periodontitis. Not only the link between IL-1β and periodontitis was proved by clinical evidence, but also the increased IL-1β triggers a series of inflammatory reactions and promotes bone resorption. Currently, IL-1β blockage has been therapeutic strategies for autoimmune and autoinflammatory diseases such as rheumatoid arthritis, cryopyrin-associated periodic syndromes, gout and type II diabetes mellitus. It is speculated that IL-1β be a potential therapeutic target for periodontitis. 2Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE.Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu 2019Cellular & Molecular Immunology2019,16,3:15
3Evaluation of soil washing process with carboxymethyl-β-cyclodextrin and carboxymethyl chitosan for recovery of PAHs/heavy metals/fluorine from metallurgic plant site显示文摘Polycyclic aromatic hydrocarbons(PAHs)/heavy metals/fluorine(F) mixed-contaminated sites caused by abandoned metallurgic plants are receiving wide attention. To address the associated environmental problems,this study was initiated to investigate the feasibility of using carboxymethyl-β-cyclodextrin(CMCD) and carboxymethyl chitosan(CMC) solution to enhance ex situ soil washing for extracting mixed contaminants. Further,Tenax extraction method was combined with a first-three-compartment model to evaluate the environmental risk of residual PAHs in washed soil. In addition,the redistribution of heavy metals/F after decontamination was also estimated using a sequential extraction procedure. Three successive washing cycles using50 g/L CMCD and 5 g/L CMC solution were effective to remove 94.3% of total PAHs,93.2% of Pb,85.8% of Cd,93.4% of Cr,83.2% of Ni and 97.3% of F simultaneously. After the 3rd washing,the residual PAHs mainly existed as very slowly desorbing fractions,which were in the form of well-aged,well-sequestered compounds; while the remaining Pb,Cd,Cr,Ni and F mainly existed as Fe–Mn oxide and residual fractions,which were always present in stable mineral forms or bound to non-labile soil fractions. 5Sox9 augments BMP2-induced chondrogenic differentiation by downregulating Smad7 in mesenchymal stem cells(MSCs)显示文摘Cartilage injuries caused by arthritis or trauma pose formidable challenges for effective clinical management due to the limited intrinsic proliferative capability of chondrocytes.Autologous stem cell-based therapies and transgene-enhanced cartilage tissue engineering may open new avenues for the treatment of cartilage injuries.Bone morphogenetic protein 2(BMP2)induces effective chondrogenesis of mesenchymal stem cells(MSCs)and can thus be explored as a potential therapeutic agent for cartilage defect repair.However,BMP2 also induces robust endochondral ossification.Although the precise mechanisms through which BMP2 governs the divergence of chondrogenesis and osteogenesis remain to be fully understood,blocking endochondral ossification during BMP2-induced cartilage formation may have practical significance for cartilage tissue engineering.Here,we investigate the role of Sox9-donwregulated Smad7 in BMP2-induced chondrogenic differentiation of MSCs.We find that overexpression of Sox9 leads to a decrease in BMP2-induced Smad7 expression in MSCs.Sox9 inhibits BMP2-induced expression of osteopontin while enhancing the expression of chondrogenic marker Col2a1 in MSCs.Forced expression of Sox9 in MSCs promotes BMP2-induced chondrogenesis and suppresses BMP2-induced endochondral ossification.Constitutive Smad7 expression inhibits BMP2-induced chondrogenesis in stem cell implantation assay.Mouse limb explant assay reveals that Sox9 expands BMP2-stimulated chondrocyte proliferating zone while Smad7 promotes BMP2-intitated hypertrophic zone of the growth plate.Cell cycle analysis indicates that Smad7 induces significant early apoptosis in BMP2-stimulated MSCs.Taken together,our results strongly suggest that Sox9 may facilitate BMP2-induced chondrogenesis by downregulating Smad7,which can be exploited for effective cartilage tissue engineering.Chen Zhao Wei Jiang Nian Zhou Junyi Liao Mingming Yang Ning Hu Xi Liang Wei Xu Hong Chen Wei Liu Lewis L.Shi Leonardo Oliveira Jennifer Moriatis Wolf Sherwin Ho Aravind Athiviraham H.M.Tsai Tong-Chuan He Wei Huang 2017Genes & Diseases2017,4,4:7
6Fate of PM2.5-bound PAHs in Xiangyang, central China during 2018 Chinese spring festival: Influence of fireworks burning and air-mass transport显示文摘Variations of levels,possible source and air mass transmission were investigated for 16 USEPA priority-controlled PAHs in PM2.5 during 2018 Chinese Spring Festival(CSF)in Xiangyang City,central China which is the North-South pollutant airmass transport channel of China.Totally 37 samples were collected.Mass concentrations of 16 PAHs for the Pre–CSF day(Pre–CSFD),during the CSF day(CSFD)and after the CSF day(Af–CSFD)are 33.78±17.68 ng/m3,22.98±6.49 ng/m3,and 8.99±4.44 ng/m3,respectively.High resolution samples showed that 16 PAHs are higher in the morning(06:00–11:00)or afternoon(11:30–16:30),than those in the evening(17:00–22:00)and at night(22:30–05:30),whereas the result is reversed during the CSFD.Fireworks burning can obviously increase the mass concentration of PAHs.Air mass trajectory indicated that Xiangyang is a sink area of pollutants for northwest and southeast,and the sources of the northeast and southwest.The air mass only can be transmitted out through northeast and southwest.It is effective for improvement of air quality in Wuhan and Hunan to control fireworks emission in Henan and local areas.Fireworks burning was an important source for PAHs during CSFD,biomass,coal combustion,and traffic emission were the main sources of PAHs for Pre–CSFD and Af–CSFD periods.The health risk on the CSFD was higher than the acceptable levels,especially during the intensive fireworks burning,the risk value far exceed 1.0×10^-4,controlling burning fireworks is required.Tianpeng Hu Yao Mao Weijie Liu Cheng Cheng Mingming Shi Zhanle Chen Qian Tian Jiaquan Zhang Shihua Qi Xinli Xing 2020Journal of Environmental Sciences2020,32,11:6

Although a number of certificateless aggregate signature schemes have been proposed, all of them are based on the classic number theory problem,which are no longer secure in the quantum era. In this paper, a certificateless sequential aggregate signature over number theory research unit lattice is proposed, which is proven to be secure in random oracle model. 8Feasibility of Tea Saponin-Enhanced Soil Washing in a Soybean Oil-Water Solvent System to Extract PAHs/Cd/Ni Efficiently from a Coking Plant Site显示文摘Mixed contaminated brownfield sites have brought serious risks to human health and environmental safety. With the purpose of removing polycyclic aromatic hydrocarbons(PAHs) and heavy metals from a coking plant site, an innovative technology for ex-situ washing was developed in the present work. The combination of 15.0 mL L^(-1) soybean oil and 7.5 g L^(-1) tea saponin proved an effective method to extract co-pollutants from soil. After two consecutive washing cycles, the efficiency rates of removal for 3-, 4-,5(+6)-ring, and total PAHs, Cd, and Ni were approximately 98.2%, 96.4%, 92.3%, 96.3%, 94.1%, and 89.4%, respectively. Meanwhile,as evaluated by Tenax extraction method and metal stability indices, the residual PAHs and heavy metals after consecutive washing mainly existed in the form with extremely low bioaccessibility in the soil. Thus, in the soil after two washing cycles, there appeared limited environmental transfer risk of co-pollutants. Moreover, a subsequent precipitation method with alkaline solution and PAHdegrading strain Sphingobium sp. PHE9 inoculation effectively removed 84.6%–100% of Cd, 82.5%–91.7% of Ni, and 92.6%–98.4% of PAHs from the first and second washing solvents. The recovered solvents also exhibited a high recycling effectiveness. 9Lactobacillus reuteri-derived extracellular vesicles maintain intestinal immune homeostasis against lipopolysaccharideinduced inflammatory responses in broilers显示文摘Background:Lactobacillus reuteri strains are widely used as probiotics to prevent and treat inflammatory bowel disease by modulating the host’s immune system.However,the underlying mechanisms by which they communicate with the host have not been clearly understood.Bacterial extracellular vesicles(EVs)have been considered as important mediators of host-pathogen interactions,but their potential role in commensals-host crosstalk has not been widely studied.Here,we investigated the regulatory actions of EVs produced by L.reuteri BBC3,a gut-associated commensal bacterium of Black-Bone chicken,in the development of lipopolysaccharide(LPS)-induced intestinal inflammation in a chicken model using both in vivo and in vitro experiments.Results:L.reuteri BBC3 produced nano-scale membrane vesicles with the size range of 60-250 nm.Biochemical and proteomic analyses showed that L.reuteri BBC3-derived EVs(LrEVs)carried DNA,RNA and several bioactive proteins previously described as mediators of other probiotics’beneficial effects such as glucosyltransferase,serine protease and elongation factor Tu.In vivo broiler experiments showed that administration of LrEVs exerted similar effects as L.reuteri BBC3 in attenuating LPS-induced inflammation by improving growth performance,reducing mortality and decreasing intestinal injury.LrEVs suppressed the LPS-induced expression of pro-inflammatory genes(TNF-α,IL-1β,IL-6,IL-17 and IL-8),and improved the expression of anti-inflammatory genes(IL-10 and TGF-β)in the jejunum.LrEVs could be internalized by chicken macrophages.In vitro pretreatment with LrEVs reduced the gene expression of TNF-α,IL-1βand IL-6 by suppressing the NF-κB activity,and enhanced the gene expression of IL-10 and TGF-βin LPS-activated chicken macrophages.Additionally,LrEVs could inhibit Th1-and Th17-mediated inflammatory responses and enhance the immunoregulatory cells-mediated immunosuppression in splenic lymphocytes of LPS-challenged chickens through the activation of macrophages.Finally,we revealed that the reduced content of both vesicular proteins and nucleic acids attenuated the suppression of LrEVs on LPS-induced inflammatory responses in ex vivo experiments,suggesting that they are essential for the LrEVs-mediated immunoregulation.Conclusions:We revealed that LrEVs participated in maintaining intestinal immune homeostasis against LPSinduced inflammatory responses in a chicken model.Our findings provide mechanistic insight into how commensal and probiotic Lactobacillus species modulate the host’s immune system in pathogens-induced inflammation.Rujiu Hu Hua Lin Mimi Wang Yuezhen Zhao Haojing Liu Yuna Min Xiaojun Yang Yupeng Gao Mingming Yang 2021Journal of Animal Science and Biotechnology2021,12,3:5
10Pravastatin alleviates lipopolysaccharide-induced placental TLR4 over-activation and promotes uterine arteriole remodeling without impairing rat fetal development显示文摘Preeclampsia is associated with over-activation of the innate immune system in the placenta,in which toll-like receptor 4(TLR4) plays an essential part.With their potent anti-inflammatory effects,statins have been suggested as potential prevention or treatment of preeclampsia,although evidence remains inadequate.Herewith,we investigated whether pravastatin could ameliorate preeclampsia-like phenotypes in a previously established lipopolysaccharide(LPS)-induced rat preeclampsia model,through targeting the TLR4/NF-κB pathway.The results showed that pravastatin reduced the blood pressure [maximum decline on gestational day(GD) 12,(101.33±2.49) mmHg vs.(118.3±1.37) mmHg,P<0.05] and urine protein level [maximum decline on GD9,(3,726.23± 1,572.86) μg vs.(1,991.03 ±609.37)μg,P< 0.05],which were elevated following LPS administration.Pravastatin also significantly reduced the rate of fetal growth restriction in LPS-treated rats(34.10% vs.8.99%,P<0.05).Further pathological analyses suggested a restoration of normal spiral artery remodeling in preeclampsia rats by pravastatin treatment.These effects of pravastatin were associated with decreased TLR4/NF-κB protein levels in the placenta and IL-6/MCP-1 levels in serum.Additionally,no obvious abnormalities in fetal liver,brain,and kidney were found after administration of pravastatin.These results provide supportive evidence for use of pravastatin in preventing preeclampsia.Muyi Yang Zhenyu Diao Zhiyin Wang Guijun Yan Guangfeng Zhao Mingming Zheng Anyi Dai Yimin Dai Yali Hu 2018The Journal of Biomedical Research2018,32,4:5
11Genetic improvement of heat tolerance in wheat:Recent progress in understanding the underlying molecular mechanisms显示文摘As a cool season crop, wheat(Triticum aestivum L.) has an optimal daytime growing temperature of 15 ℃ during the reproductive stage. With global climate change, heat stress is becoming an increasingly severe constraint on wheat production. In this review, we summarize recent progress in understanding the molecular mechanisms of heat tolerance in wheat. We firstly describe the impact of heat tolerance on morphology and physiology and its potential effect on agronomic traits. We then review recent discoveries in determining the genetic and molecular factors affecting heat tolerance, including the effects of phytohormone signaling and epigenetic regulation. 12Ablation of Akt2 and AMPKα2 rescues high fat diet-induced obesity and hepatic steatosis through Parkin-mediated mitophagy显示文摘Given the opposing effects of Akt and AMP-activated protein kinase(AMPK)on metabolic homeostasis,this study examined the effects of deletion of Akt2 and AMPKα2 on fat diet-induced hepatic steatosis.Akt2–Ampkα2 double knockout(DKO)mice were placed on high fat diet for 5 months.Glucose metabolism,energy homeostasis,cardiac function,lipid accumulation,and hepatic steatosis were examined.DKO mice were lean without anthropometric defects.High fat intake led to adiposity and decreased respiratory exchange ratio(RER)in wild-type(WT)mice,which were ablated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.High fat intake increased blood and hepatic triglycerides and cholesterol,promoted hepatic steatosis and injury in WT mice.These effects were eliminated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet promoted fat accumulation,and enlarged adipocyte size,the effect was negated in DKO mice.Fat intake elevated fatty acid synthase(FAS),carbohydrate-responsive element-binding protein(CHREBP),sterol regulatory element-binding protein 1(SREBP1),peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),peroxisome proliferator-activated receptor-α(PPARα),PPARγ,stearoyl-CoA desaturase 1(SCD-1),phosphoenolpyruvate carboxykinase(PEPCK),glucose 6-phosphatase(G6Pase),and diglyceride O-acyltransferase 1(DGAT1),the effect was absent in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet dampened mitophagy,promoted inflammation and phosphorylation of forkhead box protein O1(FoxO1)and AMPKα1(Ser^(485)),the effects were eradicated by DKO.Deletion of Parkin effectively nullified DKO-induced metabolic benefits against high fat intake.Liver samples from obese humans displayed lowered microtubule-associated proteins 1A/1B light chain 3B(LC3B),Pink1,Parkin,as well as enhanced phosphorylation of Akt,AMPK(Ser^(485)),and FoxO1,which were consolidated by RNA sequencing(RNAseq)and mass spectrometry analyses from rodent and human livers.These data suggest that concurrent deletion of Akt2 and AMPKα2 offers resilience to fat diet-induced obesity and hepatic steatosis,possibly through preservation of Parkin-mediated mitophagy and lipid metabolism.Shuyi Wang Jun Tao Huaguo Chen Machender R.Kandadi Mingming Sun Haixia Xu Gary D.Lopaschuk Yan Lu Junmeng Zheng Hu Peng Jun Ren 2021Acta Pharmaceutica Sinica B2021,11,11:4

14Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. 15An Attribute-Based Signature Scheme from Lattice Assumption显示文摘Inspired by the framework of Boyen, in this paper, an attribute-based signature(ABS) scheme from lattice assumption is proposed. In this attribute-based signature scheme, an entity's attributes set corresponds to the concatenation of a lattice matrix with the sum of some random matrices, and the signature vector is generated by using the Preimage Sampling algorithm. 16Foraging habitat selection of overwintering Black-necked Cranes in the farming area surrounding the Caohai Wetland, Guizhou Province, China显示文摘Background: Understanding how overwintering birds choose foraging habitats is very important for conservation management. The overwintering Black-necked Crane(Grus nigricollis) feeds on crop remains in farmlands;thus, reasonable conservation management of this type of farmland that surrounds wetlands is critical for the overwintering populations of the Black-necked Crane;however, it is not clear how the Black-necked Crane chooses the foraging land in the farmland.Methods: A thorough field positioning survey of all foraging sites in farmland areas around the Caohai Wetland and a-sampling analysis of habitat selection by the Black-necked Crane were conducted during the winters from 20162017 and 2017-2018.Results: Multiple factors contributed to the selection of foraging habitat in farmlands, i.e., food factors(crop remains and tillage methods) > human disturbance factors(distance to road and settlement) > topography factors(slope aspect), listed according to the strength of influence. Additionally, Black-necked Cranes tend to choose farmland sites where there was no machine tillage, the crop remains were > 500 g/m^2, the distance to residences ranged from 100 to 500 m, the distance to roads ranged from 50 to 100 m, and the slopes exhibited western or eastern aspects. As the winters progressed, the volume of the edible crop remains declined, and the influences of the other main factors also changed, i.e., the factors of human disturbance(distance to road and settlement) became less important, while the effect of the food factor(crop remains) was strengthened. Thus, the foraging sites near the road became more important.Conclusion: The farming area surrounding the Caohai Wetland is very important for the overwintering Black-necked Crane. 17Lattice-based sequential aggregate signatures with lazy verification显示文摘This paper proposes the first lattice-based sequential aggregate signature(SAS) scheme with lazy verification that is provably secure in the random oracle model. As opposed to large integer factoring and discrete logarithm based systems, the security of the construction relies on worst-case lattice problem, namely, under the small integer solution(SIS) assumption. Generally speaking, SAS schemes enable any group of signers ordered in a chain to sequentially combine their signatures such that the size of the aggregate signature is much smaller than the total size of all individual signatures. Unlike prior such proposals, the new scheme does not require a signer to retrieve the keys of other signers and verify the aggregate-so-far before adding its own signature, and the signer can add its own signature to an unverified aggregate and forward it along immediately, postponing verification until load permits or the necessary public keys are obtained. 18Present Situation and Control of Outbreaks of Algal Blooms in the Three Gorges Reservoir显示文摘After water storage in the Three Gorges Reservoir Region,there are no outbreaks of algal blooms in the main stream of the reservoir region,but the density of algae increases obviously. Outbreaks of algal blooms mainly appeared in the tributaries of the reservoir region such as the Xiangxi River,Daning River,Shennong River and Xiaojiang River,but they did not occur every year. The reasons for outbreaks of algal blooms in the tributaries are shown as follows: the existence of sources of algae(blue-green algae) in the Three Gorges Reservoir is the root cause,and the sources include sources existing and being produced in the reservoir and sources from upstream main stream and its tributaries and other related lakes and reservoirs,of which the sources are mainly from the Dianchi Lake; slight or moderate eutrophication of water is the basic condition;hydrologic and hydrodynamic conditions and suitable temperature are conducive to proliferation and aggregation of algae(blue-green algae) after the operation of the Three Gorges Reservoir until outbreaks of algal blooms appear. Outbreaks of algal blooms in the tributaries of the Three Gorges Reservoir Region mainly appear in backwater reaches; they mainly occur in the tributaries in the north of the reservoir region and near to the dam;they mainly appear from March to July; the dominant species of algae( blue-green algae) in the Three Gorges Reservoir are Pyrrophyta,Bacillariophyta and Chlorophyta,but they tend to change into blue-green algae and other algae. 20Uterine artery embolization in association with methotrexate infusion for the treatment of tubal ectopic pregnancy显示文摘Objective To investigate the safety, feasibility, and effectiveness of uterine artery embolization in association with methotrexate(MTX) infusion for the treatment of tubal ectopic pregnancy. Methods Fifty-one patients with tubal ectopic pregnancy were referred for interventional management. All patients received super-selective arteriography of the uterine artery, were infused with 50–100 mg methotrexate(MTX) through a catheter, and underwent embolization of the uterine artery with a gel-foam pledge. Clinical presentation, findings of physical examination, β-HCG values, and the size of the ectopic mass were documented for comparison. The concentration of MTX in blood was evaluated at 0.5, 6, 12, 24, 36, and 48 hours after the procedure. Results Forty-seven out of the 51 patients had clinical resolution of their tubal pregnancy(92.2%). The average time for the β-HCG value to decrease and come back to normal was 9.16 ± 2.54 days(mean +/-SD). MTX levels in peripheral blood could not be detected for patients who received 50 or 75 mg MTX at 36 hours after the procedure, while the MTX level was 0.01 μmol/L at 48 hours after the procedure for patients who received 100 mg. Out of the 4 cases whose ectopic mass size was ≥5 cm, 3 failed to respond to the treatment; however, those whose ectopic mass size was ≤5 cm responded positively to the treatment, regardless of the β-HCG concentration and abdominal bleeding, except for 1 patient who had to undergo laparoscopy for severe abdominal pain and who showed a reduction in her β-HCG level. Conclusion Uterine artery embolization in association with methotrexate infusion is safe and effective in the treatment of tubal ectopic pregnancy, especially for those women with mild to moderate bleeding, or for those at risk of a major hemorrhage.
