| 2 | Tat-functionalized Ag-Fe_3O_4 nano-composites as tissue-penetrating vehicles for tumor magnetic targeting and drug delivery显示文摘In this paper, we prepared a dual functional system based on dextrin-coated silver nanoparticles which were further attached with iron oxide nanoparticles and cell penetrating peptide(Tat), producing Tat-modified Ag-Fe_3O_4 nanocomposites(Tat-FeAgNPs). To load drugs, an –SH containing linker, 3-mercaptopropanohydrazide, was designed and synthesized. It enabled the silver carriers to load and release doxorubicin(Dox) in a pH-sensitive pattern. The delivery efficiency of this system was assessed in vitro using MCF-7 cells, and in vivo using null BalB/c mice bearing MCF-7 xenograft tumors. Our results demonstrated that both Tat and externally applied magnetic field could promote cellular uptake and consequently the cytotoxicity of doxorubicin-loaded nanoparticles,with the IC_(50) of Tat-FeAgNP-Dox to be 0.63 mmol/L. The in vivo delivery efficiency of Tat-FeAgNP carrying Cy5 to the mouse tumor was analyzed using the in vivo optical imaging tests, in which TatFeAgNP-Cy5 yielded the most efficient accumulation in the tumor(6.772.4% ID of Tat-FeAgNPs).Anti-tumor assessment also demonstrated that Tat-FeAgNP-Dox displayed the most significant tumor-inhibiting effects and reduced the specific growth rate of tumor by 29.6%(P ? 0.009), which could be attributed to its superior performance in tumor drug delivery in comparison with the control nanovehicles. | Ergang Liu Meng Zhang Hui Cui Junbo Gong Yongzhuo Huang Jianxin Wang Yanna Cui Weibing Dong Lu Sun Huining He Victor C.Yang | 2018 | Acta Pharmaceutica Sinica B2018,8,6: | 5 |
| 3 | Nose-to-brain delivery of macromolecules mediated by cell-penetrating peptides显示文摘Brain delivery of macromolecular therapeutics(e.g., proteins) remains an unsolved problem because of the formidable blood–brain barrier(BBB). Although a direct pathway of nose-to-brain transfer provides an answer to circumventing the BBB and has already been intensively investigated for brain delivery of small drugs,new challenges arise for intranasal delivery of proteins because of their larger size and hydrophilicity. In order to overcome the barriers and take advantage of available pathways(e.g., epithelial tight junctions, uptake by olfactory neurons, transport into brain tissues, and intra-brain diffusion), a low molecular weight protamine(LMWP) cell-penetrating peptide was utilized to facilitate nose-to-brain transport. Cell-penetrating peptides(CPP)have been widely used to mediate macromolecular delivery through many kinds of biobarriers. | 4 | Effects of cooling rate on the microstructure and electrical properties of Dy_2O_3-doped ZnO-based varistor ceramics显示文摘The microstructure, electrical properties, and density of Dy2O3-doped ZnO-based varistor ceramics, prepared us-ing high-energy ball milling (HEBM) and sintered at 800°C, were investigated by increasing the cooling rate in the order of H (slow cooling in furnace) → L (cooling in furnace) → K (cooling in air). With the increase in cooling rate, the grain size and density decreased, the breakdown voltage (V1 mA / mm) increased, and the nonlinear coefficient (α) and leakage current (IL) exhibited extremum. The sample with the cooling type L showed the best properties with the breakdown voltage of 2650 V/mm, α of 20.3, IL of 5.2 μA, and density of 5.42 g/cm3. The barrier height (φB), donor concentration (Nd), density of the interface states (Ns), and barrier width (ω) all exhibited extremum during the alteration in cooling rate. | 13 | Hypertension knowledge in urban elderly patients:compari-son between adherents to traditional Chinese medicine and Western medicine显示文摘Objective To compare knowledge about hypertension between elderly Chinese urban patients with preferences for either traditional Chinese medicine (TCM) or Western medicine (WM). Methods Elderly (≥ 65 years old) patients with hypertension who prefer TCM treatment (n=112) or WM (n=126) were questioned about hypertension. Their answers were compared. Results Only 32.6% of participants correctly identified hypertension as a main risk factor of coronary heart disease and stroke, 22.3% of patients answered that the main purpose of hypertension control was preventing cardiovascular disease. Other major reasons for these patients to seek medical treatment for their hypertension included: persuasion by physicians or their family members (21.6%), alleviating symptoms such as headache and dizziness (16.8%), lowering blood pressure without knowing specific reason (12.4%). The predictors for poor knowledge of hypertension were similar irrespective of preference for WM or TCM treatment, and included those with lower levels of education and older age. Television and newspaper (46.8%) were the most frequent sources of hypertension information for both groups. Among those who preferred TCM treatment, 'TCM has fewer side effects than WM' and'TCM cures disease while WM only alleviates symptoms' were common beliefs held. Conclusion This study shows that knowledge of hypertension is similar among Chinese urban patients with preferences for either WM or TCM treatment and that misunderstandings about hypertension are common among the elderly patients. In order to control hypertension effectively, public health education is necessary. | 14 | Improved method for synthesis of low molecular weight protamine–siRNA conjugate显示文摘RNAi technology has aroused wide public interest due to its high efficiency and specificity to treat multiple types of diseases. However, the effective delivery of siRNA remains a challenge due to its large molecular weight and strong anionic charge. Considering their remarkable functions in vivo and features that are often desired in drug delivery carriers, biomimetic systems for siRNA delivery become an effective and promising strategy. Based on this, covalent attachment of synthetic cell penetrating peptides(CPP) to siRNA has become of great interest. We developed a monomeric covalent conjugate of low molecular weight protamine(LMWP, a well-established CPP) and siRNA via a cytosol-cleavable disulfide linkage using PEG as a crosslinker. Results showed that the conjugates didn't generate coagulation, and exhibited much better RNAi potency and intracellular delivery compared with the conventional charge-complexed CPP/siRNA aggregates. Three different synthetic and purification methods were compared in order to optimize synthesis efficiency and product yield. The methodology using hetero-bifunctional NHS–PEG–OPSS as a crosslinker to synthesize LMWP–siRNA simplified the synthesis and purification process and produced the highest yield. | 15 | Abnormal characteristics and effectiveness evaluation of the micro-seismic signal before the Debao MS4.8 earthquake显示文摘The Debao MS4.8 earthquake occurred in western Guangxi on August 5,2021,near where the Jingxi MS5.2 earthquake occurred in 2019.To study the increasing seismicity in western Guangxi,it is necessary to determine whether there was an anomaly related to the earthquake source near the Pingxiang gravity station,which is located approximately 100 km from the epicenter of the Debao MS4.8 earthquake.In this study,the R-value scoring method was used to analyze the anomaly and evaluate the prediction efficiency of the double frequency(DF)micro-seismic signal vertical displacement(referred to as vertical displacement,VD)and the absolute value of monthly extreme rate(referred to as the monthly rate).Results show that earthquakes larger than MS4.0 in the 350 km range from the Pingxiang station tend to coincide with yearly typhoons,and the VD of micro-seismic signals correspondingly changes from low to high.The Debao MS4.8 earthquake occurred during a gradual VD increase from 0.05×10^(-6)to 0.10×10^(-6)m.When discussing the relationships among R,the rate threshold,and the effective duration of prediction,the rate threshold of the micro-seismic signal converges from 0.00039×10^(-6)to 0.00031×10^(-6)m/month,the effective duration of prediction is approximately 6-10 months,and R also converges from 0.29 to 0.31.By comparing the results of three gPhone gravity stations in Guangxi,we found that the increase of short-term VD before the Debao earthquake was related to the enhancement of the DF micro-seismic signal excited by the typhoon.When the typhoon track was perpendicular to the coastline of China,the possibility of an earthquake occurring was increased.This study provides evidence and reference for the future occurrence period of earthquakes above MS4.0 in western Guangxi. | Jin Wei Huining Huang Ying Jiang Ziwei Liu | 2023 | Geodesy and Geodynamics2023,14,6: | 0 |

| 20 | Geochemistry and Geochronology of the Jinghong Ophiolites:Implications for the Tectonic Evolution of the Eastern Paleo-Tethys显示文摘The Jinghong mafic-ultramafic complex,exposed in the eastern margin of the Lancangjiang tectonic belt,is related to the subduction of the Paleo-Tethys Ocean.Its petrogenesis plays a key role in constraining the tectonic evolution of the eastern Paleo-Tethys Ocean in southwestern China.In this study,we present petrological,geochemical and geochronological results of the Jinghong complex rocks,in order to decipher their origin and tectonic significance.The Jinghong mafic-ultramafic complex was composed of peridotite,gabbro,basalt and minor plagiogranite.Whole-rock geochemical data of the mafic rocks indicate that they have both MORB and IAB affinities and plot in the back-arc basin basalt(BABB)field in the FeO^(*)/MgO vs.TiO_(2) diagram.Combined with their trace element characteristics,it can be concluded that the Jinghong mafic-ultramafic complex represents an ophiolite suite that was formed in a back-arc ocean basin.Precise LA-ICP-MS zircon U-Pb dating yielded weighted mean ^(206)Pb/^(238)U ages of 298.4±1.7 Ma,294.3±1.6 Ma,and 292.8±2.0 Ma for gabbroic rocks from this complex,which indicates that the Jinghong ophiolites were formed during the early Permian(298-293 Ma).We propose that during subduction of the main Paleo-Tethys Ocean,a back-arc ocean basin was formed at the east of the Lancangjiang tectonic belt. | ZHU Jianjiang LIU Fulai WANG Fang XIE Shiwen CAI Jia JI Lei WANG Huining | 2021 | Acta Geologica Sinica(English Edition)2021,95,5: | 0 |
