| 1 | Rheological properties and viscosity reduction of South China Sea crude oil显示文摘The rheological properties of South China Sea(SCS) crude oil were studied. A group of synthetic long-chain polymers, including octadecyl acrylate-maleic anhydride bidodecyl amide copolymer(VR-D),octadecyl acrylate-maleic anhydride bioctadecyl amide copolymer(VR-O) and octadecyl acrylate-maleic anhydride phenly amide copolymer(VR-A), were employed to serve as viscosity reducers(VRs). Their performance was evaluated by both experimental and computational methodologies. The results suggest that the SCS crude oil has low wax content yet high resin and asphaltene contents, which lead to high viscosity through formation of association structures. Additionally, the SCS crude oil appears to be a pseudoplastic fluid showing linear shear stress-shear rate dependence at low temperature. Interestingly, it gradually evolves into a Newtonian fluid with exponential relationship between shear stress and shear rate at higher temperature. Synthetic VRs demonstrate desirable and effective performance on improvement of the rheological properties of SCS crude oil. Upon the introduction of 1000 ppm VR-O,which is synthesized by using octadecylamine in the aminolysis reaction, the viscosity of SCS crude oil is decreased by 44.2% at 15 °C and 40.2% at 40 °C. | 6 | Spatial-temporal variability of snow cover over the Amur River Basin inferred from MODIS daily snow products in recent decades显示文摘MODIS snow products MOD10A1\MYD10A1 provided us a unique chance to investigate snow cover as well as its spatial-temporal variability in response to global changes from regional and global perspectives.By means of MODIS snow products MOD10A1\MYD10A1 derived from an extensive area of the Amur River Basin,mainly located in the Northeast part of China,some part in far east area of the former USSR and a minor part in Republic of Mongolia,the reproduced snow datasets after removal of cloud effects covering the whole watershed of the Amur River Basin were generated by using 6 different cloud-effect-removing algorithms.The accuracy of the reproduced snow products was evaluated with the time series of snow depth data observed from 2002 to 2010 within the Chinese part of the basin,and the results suggested that the accuracies for the reproduced monthly mean snow depth datasets derived from 6 different cloud-effect-removing algorithms varied from 82%to 96%,the snow classification accuracies(the harmonic mean of Recall and Precision)was higher than 80%,close to the accuracy of the original snow product under clear sky conditions when snow cover was stably accumulated.By using the reproduced snow product dataset with the best validated cloud-effect-removing algorithm newly proposed,spatial-temporal variability of snow coverage fraction(SCF),the date when snow cover started to accumulate(SCS)as well as the date when being melted off(SCM)in the Amur River Basin from 2002 to 2016 were investigated.The results indicated that the SCF characterized the significant spatial heterogeneity tended to be higher towards East and North but lower toward West and South over the Amur River Basin.The inter-annual variations of SCF showed an insignificant increase in general with slight fluctuations in majority part of the basin.Both SCS and SCM tended to be slightly linear varied and the inter-annual differences were obvious.In addition,a clear decreasing trend in snow cover is observed in the region.Trend analysis(at 10%significance level)showed that 71%of areas between 2,000 and 2,380 m a.s.l.experienced a reduction in duration and coverage of annual snow cover.Moreover,a severe snow cover reduction during recent years with sharp fluctuations was investigated.Overall spatial-temporal variability of Both SCS and SCM tended to coincide with that of SCF over the basin in general. | XiaoLin Lu WanChang Zhang ShuHang Wang Bo Zhang QuanFu Niu JinPing Liu Hao Chen HuiRan Gao | 2020 | Research in Cold and Arid Regions2020,12,6: | 1 |

| 9 | Luminescent gold nanocluster-based sensing platform for accurate H_(2)S detection in vitro and in vivo with improved anti-interference显示文摘Gold nanoclusters(Au NCs)are promising luminescent nanomaterials due to their outstanding optical properties.However,their relatively low quantum yields and environment-dependent photoluminescence properties have limited their biological applications.To address these problems,we developed a novel strategy to prepare chitosan oligosaccharide lactate(Chi)-functionalized Au NCs(Au NCs@Chi),which exhibited emission with enhanced quantum yield and elongated emission lifetime as compared to the Au NCs,as well as exhibited environment-independent photoluminescence properties.In addition,utilizing the free amino groups of Chi onto Au NCs@Chi,we designed a FRET-based sensing platform for the detection of hydrogen sulfide(H_(2)S).The Au NCs and the specific H_(2)S-sensitive merocyanine compound were respectively employed as an energy donor and acceptor in the platform.The addition of H_(2)S induced changes in the emission profile and luminescence lifetime of the platform with high sensitivity and selectivity.Utilization of the platform was demonstrated to detect exogenous and endogenous H_(2)S in vitro and in vivo through wavelength-ratiometric and time-resolved luminescence imaging(TLI).Compared to previously reported luminescent molecules,the platform was less affected by experimental conditions and showed minimized autofluorescence interference and improved accuracy of detection. | Qi Yu Pengli Gao Kenneth Yin Zhang Xiao Tong Huiran Yang Shujuan Liu Jing Du Qiang Zhao Wei Huang | 2017 | Light(Science & Applications)2017,6,1: | 1 |

| 12 | Transcriptome profiling reveals phase-specific gene expression in the developing barley inflorescence显示文摘The shape of an inflorescence varies among cereals,ranging from a highly branched panicle in rice to a much more compact spike in barley(Hordeum vulgare L.)and wheat(Triticum aestivum L.).However,little is known about the molecular basis of cereal inflorescence architecture.We profiled transcriptomes at three developmental stages of the barley main shoot apex—spikelet initiation,floral organ differentiation,and floral organ growth—and compared them with those from vegetative seedling tissue.Transcript analyses identified 3688 genes differentially transcribed between the three meristem stages,with a further 1394 genes preferentially expressed in reproductive compared with vegetative tissue.Coexpression assembly and Gene Ontology analysis classified these 4888 genes into 28 clusters,revealing distinct patterns for genes such as transcription factors,histone modification,and cell-cycle progression specific for each stage of inflorescence development.We also compared expression patterns of VRS(SIX-ROWED SPIKE)genes and auxin-,gibberellic acid-and cytokinin-associated genes between two-rowed and six-rowed barley to describe regulators of lateral spikelet fertility.Our findings reveal barley inflorescence phase-specific gene expression,identify new candidate genes that regulate barley meristem activities and flower development,and provide a new genetic resource for further dissection of the molecular mechanisms of spike development. | Huiran Liu Gang Li Xiujuan Yang Hendrik N.J.Kuijer Wanqi Liang Dabing Zhang | 2020 | The Crop Journal2020,8,1: | 1 |

| 15 | Variant rs8400 enhances ALKBH5 expression through disrupting miR-186 binding and promotes neuroblastoma progression显示文摘Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3p caused by this genetic variation in ALKBH5 promotes the occurrence and development of neuroblastoma through the ALKBH5-SPP1 axis. | Qian Guan Huiran Lin Wenfeng Hua Lei Lin Jiabin Liu Linqing Deng Jiao Zhang Jiwen Cheng Zhonghua Yang Yong Li Jun Bian Haixia Zhou Suhong Li Li Li Lei Miao Huimin Xia Jing He Zhenjian Zhuo | 2023 | Chinese Journal of Cancer Research2023,35,2: | 0 |
| 16 | PCR Detection of Streptococcus agalactiae in Dairy Cows Suffering from Mastitis显示文摘[Objective] This study aimed to identify Streptococcus agalactiae and lay the foundation for the prevention and control of mastitis in dairy cows. [Method]Ten strains were isolated from milk samples produced by diseased dairy cows suffering from mastitis for morphologic observation,culture characteristic investigation,biochemical identification and Lancefield grouping. The isolated strains were identified at the molecular level by nested-PCR. [Result] Among the ten isolates,six strains were β-hemolytic and Gram-positive on blood agar,belonging to Lancefield group B,which were identified as Streptococcus agalactiae by biochemical identification and nested-PCR. After overnight incubation,the coincidence rate between results of nested-PCR detection and biochemical identification reached 100%. | 17 | PTBP2-Mediated Alternative Splicing of IRF9 Controls Tumor-Associated Monocyte/Macrophage Chemotaxis and Repolarization in Neuroblastoma Progression显示文摘The recurrence and metastasis of children with mediastinal neuroblastoma(NB)are also occurred after surgery,chemotherapy,or radiotherapy.Strategies targeting the tumor microenvironment have been reported to improve survival;however,thorough investigations of monocytes and tumor-associated macrophages(Mϕs)with specialized functions in NB are still lacking.Our data first demonstrated polypyrimidine tract binding protein 2(PTBP2)as a possible identifier in patients with mediastinal NB screened by proteomic profiling and that PTBP2 predicted good outcomes.Functional studies revealed that PTBP2 in NB cells induced the chemotactic activity and repolarization of tumor-associated monocytes and Mϕs,which,in turn,inhibited NB growth and dissemination.Mechanistically,PTBP2 prevents interferon regulatory factor 9 alternative splicing and upregulates signal transducers and activators of transcription 1 to stimulate C-C motif chemokine ligand 5(CCL5)and interferon-stimulated gene factor-dependent type I interferon secretion,to induce monocyte/Mϕs chemotaxis,and to sustain monocytes in a proinflammatory phenotype.Our study defined a critical event of PTBP2-induced monocytes/Mϕs in NB progression and revealed that RNA splicing occurred by PTBP2 benefits immune compartmentalization between NB cells and monocytes.This work revealed the pathological and biological role of PTBP2 in NB development and indicates that PTBP2-induced RNA splicing benefits immune compartmentalization and predicted a favorable prognosis in mediastinal NB. | Jue Tang Jing He Huiqin Guo Huiran Lin Meng Li Tianyou Yang Hai-Yun Wang Di Li Jiabin Liu Le Li Huimin Xia Zhenjian Zhuo Lei Miao | 2023 | Research2023,,3: | 0 |
