3Exploring the molecular basis of heterosis for plant breeding显示文摘Since approximate a century ago, many hybrid crops have been continually developed by crossing two inbred varieties. Owing to heterosis(hybrid vigor) in plants, these hybrids often have superior agricultural performances in yield or disease resistance succeeding their inbred parental lines. Several classical hypotheses have been proposed to explain the genetic causes of heterosis. During recent years, many new genetics and genomics strategies have been developed and used for the identifications of heterotic genes in plants. Heterotic effects of the heterotic loci and molecular functions of the heterotic genes are being investigated in many plants such as rice, maize, sorghum, Arabidopsis and tomato.More and more data and knowledge coming from the molecular studies of heterotic loci and genes will serve as a valuable resource for hybrid breeding by molecular design in future. This review aims to address recent advances in our understanding of the genetic and molecular mechanisms of heterosis in plants. 8Ultraselective carbon molecular sieve membrane for hydrogen purification显示文摘Hydrogen is a green clean fuel and chemical feedstock. Its separation and purification from hydrogencontaining mixtures is the key step in the production of hydrogen with high purity(>99.99%). In this work, carbon molecular sieve(CMS) membranes with ultrahigh permselectivity for hydrogen purification were fabricated by high-temperature(700–900 ℃) pyrolysis of polymeric precursor of phenolphthaleinbased cardo poly(arylene ether ketone)(PEK-C). The evolution of the microstructural texture and ultramicroporous structure and gas separation performance of the CMS membrane were characterized via TG-MS, FT-IR, XRD, TEM, CO2 sorption analysis and gas permeation measurements. CMS membranes prepared at 700 ℃ exhibited amorphous turbostratic carbon structures and high H2 permeability of 5260 Barrer with H2/CH4, H2/N2 and H2/CO selectivities of 311, 142, 75, respectively. When carbonized at900 ℃, the CMS membrane with ultrahigh H2/CH4 selectivity of 1859 was derived owing to the formation of the dense and ordered carbon structure. 9Single-cell Immune Landscape of Human Recurrent Miscarriage显示文摘t Successful pregnancy in placental mammals substantially depends on the establishment of maternal immune tolerance to the semi-allogenic fetus.Disorders in this process are tightly associated with adverse pregnancy outcomes including recurrent miscarriage(RM).However,an indepth understanding of the systematic and decidual immune environment in RM remains largely lacking.In this study,we utilized single-cell RNA-sequencing(scRNA-seq)to comparably analyze the cellular and molecular signatures of decidual and peripheral leukocytes in normal and unexplained RM pregnancies at the early stage of gestation.Integrative analysis identifies 22 distinct cell clusters in total,and a dramatic difference in leukocyte subsets and molecular properties in RM cases is revealed.Specifically,the cytotoxic properties of CD8^(+)effector T cells,nature killer(NK),and mucosal-associated invariant T(MAIT)cells in peripheral blood indicates apparently enhanced pro-inflammatory status,and the population proportions and ligand–receptor interactions of the decidual leukocyte subsets demonstrate preferential immune activation in RM patients.The molecular features,spatial distribution,and the developmental trajectories of five decidual NK(dNK)subsets have been elaborately illustrated.In RM patients,a dNK subset that supports embryonic growth is diminished in proportion,while the ratio of another dNK subset with cyto toxic and immune-active signature is significantly increased.Notably,a unique pro-inflammatory CD56^(+)CD16^(+)dNK subset substantially accumulates in RM decidua.These findings reveal a comprehensive cellular and molecular atlas of decidual and peripheral leukocytes in human early pregnancy and provide an in-depth insight into the immune pathogenesis for early pregnancy loss.Feiyang Wang Wentong Jia Mengjie Fan Xuan Shao Zhilang Li Yongjie Liu Yeling Ma Yu-Xia Li Rong Li Qiang Tu Yan-Ling Wang 2021Genomics, Proteomics & Bioinformatics2021,19,2:8

11Biothiol-specific fluorescent probes with aggregation-induced emission characteristics显示文摘Biothiols are important species in physiological processes such as regulating protein structures, redox homeostasis and cell signalling. Alternation in the biothiol levels is associated with various pathological processes, therefore non-invasive fluorescent probes with high specificity to biothiols are highly desirable research utilities. Meanwhile, fluorescent probes with aggregationinduced emission properties(AIEgens) possess unique photophysical properties that allow modulation of the sensing process through controlling the aggregation-disaggregation or the intramolecular rotational motions of the fluorophores. Herein we review the recent progress in the development of biothiol-specific AIEgens. 12The IL-1R/TLR signaling pathway is essential for efficient CD8+ T-cell responses against hepatitis B virus in the hydrodynamic injection mouse model显示文摘The outcome of hepatitis B viral(HBV)infection is determined by the complex interactions between replicating HBV and the immune system.While the role of the adaptive immune system in the resolution of HBV infection has been studied extensively,the contribution of innate immune mechanisms remains to be defined.Here we examined the role of the interleukin-1 receptor/Toll-like receptor(IL-1R/TLR)signaling pathway in adaptive immune responses and viral clearance by exploring the HBV mouse model.Hydrodynamic injection with a replication-competent HBV genome was performed in wild-type mice(WT)and a panel of mouse strains lacking specific innate immunity component expression.We found higher levels of HBV protein production and replication in Tlr2^(−/−),Tlr23479^(−/−),3d/Tlr24^(−/−),Myd88/Trif^(−/−)and Irak4^(−/−)mice,which was associated with reduced HBV-specific CD8+T-cell responses in these mice.Importantly,HBV clearance was delayed for more than 2 weeks in 3d/Tlr24^(−/−),Myd88/Trif^(−/−)and Irak4^(−/−)mice compared to WT mice.HBV-specific CD8+T-cell responses were functionally impaired for producing the cytokines IFN-γ,TNF-αand IL-2 in TLR signaling-deficient mice compared to WT mice.In conclusion,the IL-1R/TLR signaling pathway might contribute to controlling HBV infection by augmenting HBV-specific CD8+T-cell responses.Zhiyong Ma Jia Liu Weimin Wu Ejuan Zhang Xiaoyong Zhang Qian Li Gennadiy Zelinskyy Jan Buer Ulf Dittmer Carsten J Kirschning Mengji Lu 2017Cellular & Molecular Immunology2017,14,12:6
13Enhanced magnetic properties in chemically inhomogeneous Nd-Dy-Fe-B sintered magnets by multi-main-phase process显示文摘Homogeneous substitution of Dy for Nd in the hard magnetic 2:14:1 phase can effectively enhance coercivity to ensure the high temperature operation,however,inevitably deteriorate remanence at expense.In this work,we performed a comparative investigation of the two magnets prepared by multimain-phase(co-sintering Nd_(2)Fe_(14)B and(Nd,Dy)_(2)Fe_(14)B powders)and single-main-phase(sintering(Nd,Dy)_(2)Fe_(14)B powders)approaches.The comparative investigation reveals that at the same Dy substitution level(2.16 wt%),such chemically inhomogeneous multi-main-phase magnet possesses better roomtemperature magnetic properties as well as thermal stability than those of the single-main-phase one with homogenous Dy distribution in the matrix grains.Room-temperature magnetic properties H_(Cj)=1664 kA/m,B_(r)=1.33 T and(BH)_(max)=350.4 kJ/m^(3)for the multi-main-phase magnet are all better than those for the single-main-phase magnet with H_(Cj)=1536 kA/m,B_(r)=1.29 T and(BH)_(max)=318.4 kj/m^(3).In addition,over the temperature range from 295 to 423 K,both the temperature coefficients of coercivity and remanence for the multi-main-phase magnet are also lower than that for the single-main-phase magnet.Such superior magnetic performance is attributed to the short-range magnetic interactions inside individual 2:14:1 phase grains and the long-range magnetostatic interactions between adjacent grains with inhomogeneous Dy distribution.Our work provides a feasible approach of enhancing coercivity and retaining energy product simultaneously in the Nd-Dy-Fe-B permanent magnets.Xiaolian Liu Mengjie Pan Pei Zhang Tianyu Ma Lizhong Zhao Lingwei Li 2021Journal of Rare Earths2021,39,5:5
14A detached petal disc assay and virus-induced gene silencing facilitate the study of Botrytis cinerea resistance in rose flowers显示文摘Fresh-cut roses(Rosa hybrida)are one of the most important ornamental crops worldwide,with annual trade in the billions of dollars.Gray mold disease caused by the pathogen Botrytis cinerea is the most serious fungal threat to cut roses,causing extensive postharvest losses.In this study,we optimized a detached petal disc assay(DPDA)for artificial B.cinerea inoculation and quantification of disease symptoms in rose petals.Furthermore,as the identification of rose genes involved in B.cinerea resistance could provide useful genetic and genomic resources,we devised a virusinduced gene silencing(VIGS)procedure for the functional analysis of B.cinerea resistance genes in rose petals.We used RhPR10.1 as a reporter of silencing efficiency and found that the rose cultivar‘Samantha’showed the greatest decrease in RhPR10.1 expression among the cultivars tested.To determine whether jasmonic acid and ethylene are required for B.cinerea resistance in rose petals,we used VIGS to silence the expression of RhLOX5 and RhEIN3(encoding a jasmonic acid biosynthesis pathway protein and an ethylene regulatory protein,respectively)and found that petal susceptibility to B.cinerea was affected.Finally,a VIGS screen of B.cinerea-induced rose transcription factors demonstrated the potential benefits of this method for the high-throughput identification of gene function in B.cinerea resistance.Collectively,our data show that the combination of the DPDA and VIGS is a reliable and highthroughput method for studying B.cinerea resistance in rose.Xiaoqian Cao Huijun Yan Xintong Liu Dandan Li Mengjie Sui Jie Wu Hongqiang Yu Zhao Zhang 2019Horticulture Research2019,6,1:5
15A novel artificial nerve graft for repairing longdistance sciatic nerve defects:a self-assembling peptide nanofiber scaffold-containing poly (lactic-co-glycolic acid) conduit显示文摘In this study, we developed a novel artificial nerve graft termed self-assembling peptide nanofiber scaffold(SAPNS)-containing poly(lactic-co-glycolic acid)(PLGA) conduit(SPC) and used it to bridge a 10-mm-long sciatic nerve defect in the rat. Retrograde tracing, behavioral testing and histomorphometric analyses showed that compared with the empty PLGA conduit implantation group, the SPC implantation group had a larger number of growing and extending axons, a markedly increased diameter of regenerated axons and a greater thickness of the myelin sheath in the conduit. Furthermore, there was an increase in the size of the neuromuscular junction and myofiber diameter in the target muscle. 16Discrimination of False Negative Results in RT-PCR Detection of SARS-CoV-2 RNAs in Clinical Specimens by Using an Internal Reference显示文摘Reverse transcription-polymerase chain reaction(RT-PCR)is an essential method for specific diagnosis of SARS-CoV-2 infection.Unfortunately,false negative test results are often reported.In this study,we attempted to determine the principal causes leading to false negative results of RT-PCR detection of SARS-CoV-2 RNAs in respiratory tract specimens.Multiple sputum and throat swab specimens from 161 confirmed COVID-19 patients were tested with a commercialfluorescent RT-PCR kit targeting the ORF1 ab and N regions of SARS-CoV-2 genome.The RNA level of a cellular housekeeping gene ribonuclease P/MRP subunit p30(RPP30)in these specimens was also assessed by RT-PCR.Data for a total of 1052 samples were retrospectively re-analyzed and a strong association between positive results in SARS-CoV-2 RNA tests and high level of RPP30 RNA in respiratory tract specimens was revealed.By using the ROC-AUC analysis,we identified Ct cutoff values for RPP30 RT-PCR which predicted false negative results for SARS-CoV-2 RT-PCR with high sensitivity(95.03%–95.26%)and specificity(83.72%–98.55%)for respective combination of specimen type and ampli-fication reaction.Using these Ct cutoff values,false negative results could be reliably identified.Therefore,the presence of cellular materials,likely infected host cells,are essential for correct SARS-CoV-2 RNA detection by RT-PCR in patient specimens.RPP30 could serve as an indicator for cellular content,or a surrogate indicator for specimen quality.In addition,our results demonstrated that false negativity accounted for a vast majority of contradicting results in SARS-CoV-2 RNA test by RT-PCR.Yafei Zhang Changtai Wang Mingfeng Han Jun Ye Yong Gao Zhongping Liu Tengfei He Tuantuan Li Mengyuan Xu Luping Zhou Guizhou Zou Mengji Lu Zhenhua Zhang 2020Virologica Sinica2020,35,6:5

20Complementation of Wild-Type and Drug-Resistant Hepatitis B Virus Genomes to Maintain Viral Replication and Rescue Virion Production under Nucleos(t)ide Analogs显示文摘As the open reading frames of hepatitis B virus(HBV)genomes are overlapping,resistance mutations(MTs)in HBV polymerase may result in stop codon MTs in hepatitis B surface proteins,which are usually detected as a mixed population with wild-type(WT)HBV.The question was raised how the coexistence of nucleos(t)ide analogs(NAs)resistance MTs and WT sequences affects HBV replication.In the present study,HBV genomes with frequently detected reverse transcriptase(RT)/surface truncation MTs,rtA181T/sW172^*,rtV191I/sW182^*and rtM204I/sW196^*,were phenotypically characterized alone or together with their WT counterparts in different ratios by transient transfection in the absence or presence of Nas.In the absence of Nas,RT/surface truncation MTs impaired the expression and secretion of HBV surface proteins,and had a dose-dependent negative effect on WT HBV virion secretion.However,in the presence of Nas,coexistence of MTs with WT maintained viral replication,and the presence of WT was able to rescue the production of MT HBV virions.Our findings reveal that complementation of WT and MT HBV genomes is highly effective under drug treatment.Chunchen Wu Baolin Li Xiaoyong Zhang Kaitao Zhao Yingshan Chen Yifei Yuan Yan Liu Rongjuan Chen Dongping Xu Xinwen Chen Mengji Lu 2019Virologica Sinica2019,34,4:4
