| 2 | Extraction and back-extraction behaviors of 14 rare earth elements from sulfuric acid medium by TODGA显示文摘The unique physical and chemical properties of rare earth elements lay the foundation for their extensive application. N,N,N',N' Tetra-octyl-3-oxopentanediamide(TODGA) is excellent in its ability of extracting rare earth elements and it is favored for green initiative. In this paper, the extraction and back-extraction of14 rare earth elements by TODGA were studied. Experiments show that in conditions of 6 mol/L sulfuric acid, the extraction temperature of 25 ℃,the phase ratio of 1:1 and 0.04 mol/LTODGA(aviation kerosene as the diluent), the extraction rates of 14 rare earth elements including lanthanum, cerium, praseodymium,neodymium, europium, gadolinium, terbium, dysprosium, holmium, erbium, thulium, ytterbium, lutetium, and yttrium were 99.00%-99.73%. Mixed with hydrochloric acid and nitric acid(HCl 3.5 mol/L, HNO_30.5 mol/L), the recoveries of the 14 rare earth elements are 91.52%-99.91% when the extraction temperature is 25 ℃ and the ratio is 1:1. The following application is based on the optimum conditions above with practical samples(from the roasting production line of China North Rare Earth High-tech Company Limited) for extraction and back-extraction experiments. | 3 | Fecal metabolomics reveals the positive effect of ethanol extract of propolis on T2DM mice显示文摘A large number of studies have shown that propolis has positive effects in the treatment of type 2 diabetes mellitus(T2DM).However,there are have only been a few reports that are based on an ultra-high performance liquid chromatography-quadrupole-time-of-flight-mass spectrometry(UPLC-Q-TOF-MS)analysis of the fecal metabolomics of ethanol extract of propolis(EEP)in the treatment of T2DM.The present investigation was designed to screen potential biomarkers of T2DM by the metabonomic method and to explain the possible anti-diabetes mechanism of EEP according to the changes in the biomarkers.The results showed that EEP improved the body weight(BW)of T2DM mice,lowered blood sugar levels,and significantly restored blood biochemical indicators related to T2DM,such as fasting insulin(FINS),homeostasis model assessment of insulin resistance(HOMA-IR),aspartate transaminase(AST),and alanine aminotransferase(ALT).Liver pathology showed that EEP reversed liver damage caused by T2DM.Metabolomics data identified 27 potential biomarkers in fecal samples.EEP effectively regulated the dysfunction in the metabolic pathways of glycerophospholipids,sphingolipids,riboflavins,and sterol lipids caused by T2DM.In summary,our research results revealed positive effects of EEP in the treatment of T2DM and provided potential candidate markers for further research and in the clinical treatment of T2DM. | Chunmei Wang Huiting Zhao Kai Xu Yali Du Jinjia Liu Jinfei Wang Yusuo Jiang | 2023 | Food Science and Human Wellness2023,12,1: | 1 |

| 7 | TREM-2 Drives Development of Multiple Sclerosis by Promoting Pathogenic Th17 Polarization显示文摘Multiple sclerosis(MS)is a neuroinflammatory demyelinating disease,mediated by pathogenic T helper 17(Th17)cells.However,the therapeutic effect is accompa-nied by the fluctuation of the proportion and function of Th17 cells,which prompted us to find the key regulator of Th17 differentiation in MS.Here,we demonstrated that the triggering receptor expressed on myeloid cells 2(TREM-2),a modulator of pattern recognition receptors on innate immune cells,was highly expressed on pathogenic CD4-positive T lymphocyte(CD4^(+)T)cells in both patients with MS and experimental autoimmune encephalomyelitis(EAE)mouse models.Conditional knockout of Trem-2 in CD4^(+)T cells significantly alleviated the disease activity and reduced Th17 cell infiltration,activation,differentiation,and inflammatory cytokine production and secretion in EAE mice.Furthermore,with Trem-2 knockout in vivo experiments and in vitro inhibitor assays,the TREM-2/zeta-chain associated protein kinase 70(ZAP70)/signal transducer and activator of transcription 3(STAT3)signal axis was essential for Th17 activation and differentiation in EAE progression.In conclusion,TREM-2 is a key regulator of pathogenic Th17 in EAE mice,and this sheds new light on the potential of this therapeutic target for MS. | Siying Qu Shengfeng Hu Huiting Xu Yongjian Wu Siqi Ming Xiaoxia Zhan Cheng Wang Xi Huang | 2024 | Neuroscience Bulletin2024,40,1: | 0 |
| 8 | Effects of p38 MAPK inhibitor on the rat pain behavior and proinflammatory cytokines in a metastatic bone cancer pain model显示文摘Objective:To observe the effects of p38 mitogen activated protein kinase(MAPK) inhibitor SB203580 by intra-thecal injection on the pain behavior and the spinal proinflammatory cytokines in a rat model of bone cancer pain induced by breast cancer cells. Methods:Eleven rats were used to establish the models of bone cancer pain,six rats were treated by intrathecal SB203580 injection,and the other 5 were as the controls. The paw withdrawal latency(PWL),histology and the spinal levels of IL-1β and TNF-α were detected. Results:All the 11 rats presented evident bone destruction and thermal hyperalgesia after intra-tibial injection of breast cancer cells. No effect of SB203580 on the bone destruction was observed. However,following intrathecal injection of SB203580,the left PWLs(12.12 ± 1.26 s at 16 days and 12.99 ± 1.65 s at 19 days) were significant higher than that of controls(9.05 ± 1.08 s at 16 days and 8.55 ± 1.60 s at 19 days),P < 0.05. Meanwhile,intrathecal injection of SB203580 evidently reduced the levels of spinal IL-1β and TNF-α. | 9 | Inhibition of Ku80 by RNAi enhances the radiosensitivity of cervical carcinoma cell line SiHa显示文摘Objective: To study the radiosensitivity of SiHa cells with Ku80 inhibition by RNAi. Methods: pKu80-siRNA and pNeg-siRNA were constructed and then transfected into SiHa cells. Western blot and RT-PCR were applied to measure the expression of Ku80. After 10 Gy irradiation with 6 MV X-ray, cells were harvested at 20 h, 28 h and 48 h, and analyzed by flow cytometry for apoptosis rate. The SF2 and α, β values of cells were acquired by clone formation array. Results: Two stable transfection cell clones SiHa/Ku80-siRNA and SiHa/Neg-siRNA were screened from the pKu80-siRNA and pNeg-siRNA transfected cells. Western blot and RT-PCR indicated that the expression of Ku80 was suppressed by the Ku80-siRNA. The apoptosis rates of SiHa/Ku80-siRNA were higher than control cells at 28 h and 48 h after X-ray irradiation (P < 0.05). The SF2 value of SiHa/Ku80-siRNA (0.422) was lower than that of SiHa/Neg-siRNA cells (0.587) or untransfected cells (0.583), and the α value of SiHa/Ku80-siRNA was 0.295 ± 0.016, higher than that of SiHa/Neg-siRNA (0.176 ± 0.010) or control cells (0.188 ± 0.011). | 12 | Effectiveness and safety of red yeast rice predominated by monacolin Kβ-hydroxy acid form for hyperlipidemia treatment and management显示文摘OBJECTIVE:To assess the lipid-lowering activity and safety of a dietary supplement containing monacolin Kβ-hydroxy acid form(MKA),Heye(Folium Nelumbinis),and Cangzhu(Rhizoma Atractylodis Lanceae),compared to lifestyle modifications.METHODS:Totally 117 subjects with moderate to severe dyslipidemia(according to Chinese guidelines)and low CV risk were randomly assigned into three treatment groups:lifestyle modification(LM),LM plus a low dosage of MKA,LM plus a high dosage of MKA,and treated for 60 d.The primary endpoint was the reduction of low density lipoprotein cholesterol(LDL-C)and total cholesterol(TC).Safeties along with Traditional Chinese Medicine Syndromes were assessed through the study.RESULTS:A low dosage of MKA along with lifestyle modifications caused a significant decrease in LDL-C by 15.6%on average(95%CI,9.6%to 21%)with,a decrease in TC by 15.3%on average(95%CI,9.26%to 21.4%),and a decrease in non-HDL-C by 35.4%(95%CI,25.76%to 41.34%).Weak evidence of a reduction of triglycerides but an increment of HDL-C was observed in patients with severe hyperlipidemia.No severe adverse events occurred during the study.CONCLUSION:Our results confirm the LDL-C and TC lowering properties of MKA is clinically meaningful.It also produces a significant reduction of non-HDL-C,and slightly effects on TG and HDL-C as well. | CHEN Benjian HUANG Xiaodan PENG Huiting LI Yishi CAO Yongtao WU Huanlin XU Danping | 2022 | Journal of Traditional Chinese Medicine2022,42,2: | 0 |
