1Abundance of antibiotic resistance genes and their association with bacterial communities in activated sludge of wastewater treatment plants: Geographical distribution and network analysis显示文摘Wastewater treatment plants(WWTPs) are deemed reservoirs of antibiotic resistance genes(ARGs). Bacterial phylogeny can shape the resistome in activated sludge. However, the co-occurrence and interaction of ARGs abundance and bacterial communities in different WWTPs located at continental scales are still not comprehensively understood. Here, we applied quantitative PCR and Miseq sequence approaches to unveil the changing profiles of ARGs(sul1, sul2, tet W, tet Q, tet X), int I1 gene, and bacterial communities in 18 geographically distributed WWTPs. The results showed that the average relative abundance of sul1 and sul2 genes were 2.08 × 10^(-1) and 1.32 × 10^(-1) copies/16 S rRNA copies, respectively. The abundance of tet W gene was positively correlated with the Shannon diversity index(H′), while both studied sul genes had significant positive relationship with the int I1 gene. The highest average relative abundances of sul1, sul2, tet X, and int I1 genes were found in south region and oxidation ditch system. Network analysis found that 16 bacterial genera co-occurred with tet W gene. Co-occurrence patterns were revealed distinct community interactions between aerobic/anoxic/aerobic and oxidation ditch systems. The redundancy analysis model plot of the bacterial community composition clearly demonstrated that the sludge samples were significant differences among those from the different geographical areas,and the shifts in bacterial community composition were correlated with ARGs. 2Biogeographic distribution patterns of algal community in different urban lakes in China: Insights into the dynamics and co-existence显示文摘Urban lake ecosystems are significant for social development,but currently we know little about the geographical distribution of algal community in urban lakes at a large-scale.In this study,we investigated the algal community structure in different areas of urban lakes in China and evaluated the influence of water quality parameters and geographical location on the algal community.The results showed that obvious differences in water quality and algal communities were observed among urban lakes in different geographical areas.Chlorophyta was the dominant phylum,followed by cyanobacteria in all areas.The network analysis indicated that algal community composition in urban lakes of the western and southern area showed more variations than the eastern and northern areas,respectively.Redundancy analysis and structural equation model revealed that nutrients and p H were dominant environmental factors that affected the algal community,and they showed higher influence than that of iron,manganese and COD Mn concentration.Importantly,algal community and density exhibited longitude and latitude relationship.In general,these results provided an ecological insight into large-scale geographical distributions of algal community in urban lakes,thereby having potential applications for management of the lakes.Haihan Zhang Rongrong Zong Huiyan He Kaiwen Liu Miaomiao Yan Yutian Miao Ben Ma Xin Huang 2021Journal of Environmental Sciences2021,33,2:9
3Cow, yak, and camel milk diets differentially modulated the systemic immunity and fecal microbiota of rats显示文摘Cow milk is most widely consumed; however,non-cattle milk has gained increasing interest because of added nutritive values. We compared the health effects of yak,cow,and camel milk in rats. By measuring several plasma immune factors,significantly more interferon-c was detected in the camel than the yak(P = 0.0020) or cow(P = 0.0062) milk group. Significantly more Ig M was detected in the yak milk than the control group(P = 0.0071). The control group had significantly less interleukin 6 than the yak(P = 0.0499)and cow(P = 0.0248) milk groups. The fecal microbiota of the 144 samples comprised mainly of the Firmicutes(76.70 ± 11.03%),Bacteroidetes(15.27 ± 7.79%),Proteobacteria(3.61 ± 4.34%),and Tenericutes(2.61 ± 2.53%) phyla. Multivariate analyses revealed a mild shift in the fecal microbiota along the milk treatment. We further identified the differential microbes across the four groups. At day 14,22 and 28 differential genera and species were identified(P = 0.0000–0.0462),while 8 and 11 differential genera and species(P = 0.0000–0.0013) were found at day 28. Some short-chain fatty acid and succinate producers increased,while certain health-concerned bacteria(Prevotella copri,Phascolarctobacterium faecium,and Bacteroides uniformis) decreased after 14 days of yak or camel milk treatment. 5Oxaliplatin,Fluorouracil and Leucovorin (FOLFOX) as First-line Chemotherapy for Metastatic or Recurrent Colorectal Cancer Patients显示文摘OBJECTIVE To investigate the efficiency and safety of the oxaliplatin, fluorouracil(5-FU)and leucovorin regimen(FOLFOX)in previously untreated patients with metastatic or recurrent colorectal cancer. METHODS Previously untreated patients with metastatic or recurrent colorectal cancer received 100 mg/m2 of oxaliplatin intravenously(IV)over 2 h on day 1,and IV 400 mg/m2 of leucovorin over 2 h followed by a bolus of 400 mg/m2 of 5-FU.Then 2,600~3,000 mg/m2 of 5-FU was administered by continuous infusion over 46 h. RESULTS An evaluated response rate was determined for 97 of 105 treated patients.The overal response rate was 35.1%,9 patients(9.3%) had a complete response and 25 patients(25.8%)a partial response.Thirty-two patients(33.0%)developed stable disease and 32.0%of the patients progressed.The median time to progression(TTP)was 7.7 months and the median overal survival 20.5 months.One and 2-year survival rates were 68%and 32%.Toxic effects based on the National Cancer Institute-Common Toxicity Criteria(NCI-CTC),reaching grade 3/4 were:neutropenia 12.3%, anemia 11.3%,vomiting 4.1%and diarrhea 7.2%.Grade 3 neuropathy was 5.1%.The overall survival rate of patients who had received a radical resection was superior to the patients who had not received a operation,or had received a pal iative resection(P=0.0658).The serum levels of CEA,ALP and LDH had no relationship with survival(P>0.05). 18SP2509, an inhibitor of LSD1, exerts potential antitumor effects by targeting the JAK/STAT3 signaling显示文摘Hyperactivation of Janus kinase(JAK)/signal transducer and activator of transcription(STAT)signaling promotes tumorigenesis and cancer progression.STAT3 participates in the essential processes of cell proliferation,survival,and differentiation in many types of tumors.In the present study,SP2509 was identified as a potent inhibitor of the JAK/STAT3 signaling pathway by high-throughput drug screening based on a STAT3-driven luciferase expression system.Our results indicated that SP2509 inhibits constitutive STAT3 activation and the expression of STAT3-driven downstream genes.Bcl-xL,c-Myc,and Cyclin D1 were downregulated after treatment with SP2509.In addition,SP2509 specifically inhibits JAK activity,which could cause cell cycle arrest,inhibit cell growth,and induce apoptosis of various cancer cells.These results confirmed that SP2509 inhibits tumor progression by suppressing the expression of JAK/STAT3 signaling and STAT3-related downstream genes.Moreover,we demonstrated that SP2509 inhibits tumor growth in vivo and induces cell death in vitro.SP2509-mediated inhibition of STAT3 phosphorylation is dependent on its original target lysine-specific demethylase 1 in cancer cells.In summary,our results indicate that SP2509 is a novel inhibitor of JAK/STAT3 signaling.Huiyan Zhen Xinxin Zhang Lei Zhang Mingming Zhou Liangliang Lu Lihong Wu Na He Juan Wang Rui Li Yan Guo 2021Acta Biochimica et Biophysica Sinica2021,53,8:0

20Liquid-liquid two-phase flow in a wire-embedded concentric microchannel: Flow pattern and mass transfer performance显示文摘In this work, flow pattern and mass transfer of liquid-liquid two-phase flow in a wire-embedded concentric microchannel are studied using toluene-water system. Droplet flow, slug flow, oval flow and annular flow are observed in the wire-embedded concentric microchannel. The effects of embedded wires and physical properties on flow patterns are investigated. The embedded wire insert is conducive to the formation of annular flow. The flow pattern distribution regions are distinguished by the Caaq(capillary number)±We_(org)(Weber number) flow pattern map. When Weorg<0.001, slug flow is the main flow pattern, and when Weorg>0.1, annular flow is the main flow pattern. Oval flow and droplet flow are between We_(org)= 0.001-0.1, and oval flow is transformed into droplet flow with the increase of Caaq. The effect of flow rate, phase ratio, initial acetic acid concentration, insert shape and flow patterns on mass transfers are studied.
