| 4 | Analysis of components of conserved 'backbone sequences' among genomes of Shigella spp. strains显示文摘Difference in the genomic compositions of prokaryotes is the basis of the diversity in their biological characters. However, besides their flora-or strain-specific genes,those floras with closer relationship in the evolution also have conserved “backbone sequences”, which reveal the marks of their origin and evolution, and these “backbone sequences”are just the basis of their elementary living abilities and common biological properties. Shigella is very closely related to E. coli in the origin and evolution, and may turn out to belong to the same genus. In this study, a microarray containing E. coli K-12 whole genome and Sf301 specific ORFs is used to investigate the genomic components of four Shigella strains. The results indicate that 16%-22% K-12 ORFs sequences are not detected in the genome of Shigella strains while the genome of Shigella contains at least 2800 conserved ORFs, which compose the common “backbone sequences”.Advanced analysis indicated that the “backbone sequences”are the essential components in maintaining the normal physiological activities of intestinal bacteria. This new bird possesses the following unique combination of features: a long rostrum, with some teeth in the front; short nasal; slender jugal; Y- shaped furcula, with expanded distal end of the hypocleidum; cake-like sternum, with a low and caudally distributed keel; strut-like and caudally concave coracoid. | 8 | A fully automated centrifugal microfluidic system for sample-to-answer viral nucleic acid testing显示文摘The outbreak of virus-induced infectious diseases poses a global public-health challenge.Nucleic acid amplification testing(NAAT)enables early detection of pandemic viruses and plays a vital role in preventing onward transmission.However,the requirement of skilled operators,expensive instrumentation,and biosafety laboratories has hindered the use of NAAT for screening and diagnosis of suspected patients.Here we report development of a fully automated centrifugal microfluidic system with sample-in-answer-out capability for sensitive,specific,and rapid viral nucleic acid testing.The release of nucleic acids and the subsequent reverse transcription loop-mediated isothermal amplification(RT-LAMP)were integrated into the reaction units of a microfluidic disc.The whole processing steps such as injection of reagents,fluid actuation by rotation,heating and temperature control,and detection of fluorescence signals were carried out automatically by a customized instrument.We validate the centrifugal microfluidic system using oropharyngeal swab samples spiked with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)armored RNA particles.The estimated limit of detection for armored RNA particles is 2 copies per reaction,the throughput is 21 reactions per disc,and the assay sample-to-answer time is approximately 70 min.This enclosed and automated microfluidic system efficiently avoids viral contamination of aerosol,and can be readily adapted for virus detection outside the diagnostic laboratory. | Fei Tian Chao Liu Jinqi Deng Ziwei Han Lu Zhang Qinghua Chen Jiashu Sun | 2020 | Science China Chemistry2020,63,10: | 4 |
| 9 | The impact of biofuel plantation on biodiversity: a review显示文摘Because plants convert solar energy into chemical energy stored in organisms, biomass production as an energy source can help to reduce the world's reliance on fossil energy and mitigate global warming. Biofuel production is a fast-growing industry that represents a new type of large-scale human disturbance on ecosystems. Thus, the benefits of biofuel production bring environmental risks that include its potential impact on biodiversity, which is still an open question. In this review, we start first with a brief overview of the evolution of biofuel concept; second, we review the state of biofuel production across the continents,with a major emphasis on the main species used and their major feedstock. For which, we found significant differences for land use and environmental cultural management of biofuel plantation between tropical and temperate regions.Third, we summarize the impacts of biofuel plantation on biodiversity at multiple scales, based on the case studied with respect to the corresponding issues. At the genetic level,introgression and contamination by aggressive genotypes are a primary risk. At the species level, habitat pollution, degradation, and disturbance caused by intensive managementof biofuel plantation significantly raise the risk of habitat fragmentation, native extinction, and bio-invasion. At the ecosystem level, the large-scale homogeneous landscape of biofuel plantation results in simplified community and food web that severely damage ecosystem services, including ecosystem diversity. Finally, we compare the current and potential benefits and risks of biofuel plantations for the practical application of a biofuel industry of China. | 11 | Comparative genome analysis of deleted genes in Shigella flexneri 2a strain 301显示文摘Comparative genome analysis is performed between Shigella flexneri 2a strain 301 and its close relatives, the nonpathogenic E. Coli K-12 strain MG1655. Result shows that there are 136 DNA segments whose size is larger than 1000 bp absent from Shigella flexneri 2a strain 301, which is up to 717253 bp in total length. These deleted segments altogether contain 670 open reading frames (ORFs). Prediction of these ORFs indicates that there are 40% genes of unknown function. The other genes of definite functions encode metabolic enzymes, structure proteins, transcription regulatory factors and some elements correlated with horizontal transfer. Here we compare the complete genomic sequences of the two closely related species, which differ in pathogenic phenotype. | 13 | Evaluation of menopausal status among breast cancer patients with chemotherapy-induced amenorrhea显示文摘Objective: In patients with chemotherapy-induced amenorrhea(CIA), the menopausal status is ambiguous and difficult to evaluate. This study aimed to establish a discriminative model to predict and classify the menopausal status of breast cancer patients with CIA.Methods: This is a single center hospital-based study from 2013 to 2016. The menopausal age distribution and accumulated incidence rate of CIA are described. Multivariate models were adjusted for established and potential confounding factors including age, serum concentration of estradiol(E2) and follicle-stimulating hormone(FSH),feeding, pregnancy, parity, abortions, and body mass index(BMI). The odds ratio(OR) and 95% confidence interval(95% CI) of different risk factors were estimated.Results: A total of 1,796 breast cancer patients were included in this study, among whom, 1,175(65.42%) were premenopausal patients and 621(34.58%) were post-menopause patients. Five hundred and fifty patients were included in CIA analysis, and a cumulative CIA rate of 81.64% was found in them. Age(OR: 1.856, 95% CI:1.732-1.990), serum concentration of E2(OR: 0.976, 95% CI: 0.972-0.980) and FSH(OR: 1.060, 95% CI:1.053-1.066), and menarche age(OR: 1.074, 95% CI: 1.009-1.144) were found to be associated with the patients' menopausal status. According to multivariate analysis, the discriminative model to predict the menopausal status is Logit(P)=-28.396+0.536 Age-0.014 E2+0.031 FSH. The sensitivities for this model were higher than 85%, and its specificities were higher than 89%.Conclusions: The discriminative model obtained from this study for predicting menstrual state is important for premenopausal patients with CIA.
