| 1 | The linkage maps of Dendrobium species based on RAPD and SRAP markers显示文摘Dendrobium plants are used commonly as tonic herbs and health food in many Asian countries,especially in China.Here we report the genetic map construction of two Dendrobium species with a double pseudo-testcross strategy using random amplified polymorphic DNA(RAPD) and sequence-related amplified polymorphism(SRAP) markers.A F1 mapping population of 90 individuals was developed from a cross between D.officinale and D.hercoglossum.A total of 307 markers,including 209 RAPD and 98 SRAP,were identified and used for genetic linkage group(LG) analysis.The D.officinale linkage map consisted of 11 major linkage groups and 3 doublets,which covered 629.4 cM by a total of 62 markers with an average locus distance of 11.2 cM between two adjacent markers.The D.hercoglossum linkage map contained 112 markers mapped on 15 major and 4 minor linkage groups,spanning a total length of 1,304.6 cM with an average distance of 11.6 cM between two adjacent markers.The maps constructed in this study covered 92.7% and 82.7% of the D.hercoglossum and D.officinale genomes respectively,providing an important basis for the mapping of horticultural and medicinal traits and for the application of marker-assisted selection in Dendrobium breeding program. | Dawei Xue Shangguo Feng Hongyan Zhao Hua Jiang Bo Shen Nongnong Shi Jiangjie Lu Junjun Liu Huizhong Wang | 2010 | Journal of Genetics and Genomics2010,37,3: | 26 |
| 2 | Exploration of presence/absence variation and corresponding polymorphic markers in soybean genome显示文摘This study was designed to reveal the genome‐wide distribution of presence/absence variation(PAV) and to establish a database of polymorphic PAV markers in soybean. The 33 soybean whole‐genome sequences were compared to each other with that of Williams 82 as a reference genome. A total of 33,127 PAVs were detected and 28,912 PAV markers with their primer sequences were designed as the database NJAUSoyPAV_1.0. The PAVs scattered on whole genome while only 518(1.8%) overlapped with simple sequence repeats(SSRs) in BARCSOYSSR_1.0database. In a random sample of 800 PAVs, 713(89.13%) showed polymorphism among the 12 differential genotypes. Using 126 PAVs and 108 SSRs to test a Chinese soybean germplasm collection composed of 828 Glycine soja Sieb. et Zucc. and Glycine max(L.) Merr. accessions, the per locus allele number and its variation appeared less in PAVs than in SSRs. The distinctness among alleles/bands of PCR(polymerase chain reaction) products showed better in PAVs than in SSRs, potential in accurate marker‐assisted allele selection. | 3 | MULTI-TILLERING DWARF1, a new allele of BRITTLE CULM 12, affects plant height and tiller in rice显示文摘Plant height and tillering are crucial factors determining rice plant architecture and influencing rice grain production. In this study, multi-tillering dwarf1(mtd1), a stable multi-tiller and dwarf mutant, was screened from the ethylmethane sulfonate-treated japonica rice variety Wuyunging7. Compared with the wild type, mtd1 mutant exhibited pleiotropic phenotypes, including dwarfism, more tillers, brittle culms and delayed heading date.By employing map-based cloning strategy, the gene MTD1 was finally mapped to an approximately 66-kb region on the short arm of chromosome 9. Sequencing results showed that the gene LOC_Os09g02650(BC12) in mtd1 mutant had a single nucleotide substitution(G to A), which generated a premature translation stop. Over-expressing MTD1/BC12 coding sequence rescued all the phenotypes of mtd1 mutants including plant height and tillers, which confirms that BC12 is the mutated gene in mtd1 mutant.Quantitative reverse transcription-PCR analysis showed that MTD1/BC12 could negatively regulate the expression of MONOCULM 1, IDEAL PLANT ARCHITECTURE1 and Tillering and Dwarf 1, and control rice tillering. Remarkably, a-amylase activity analysis and gibberellic acid(GA)treatment showed that the dwarf phenotype of mtd1 mutant was dependent on GA biosynthesis pathway. | 4 | The Physalis floridana genome provides insights into the biochemical and morphological evolution of Physalis fruits显示文摘The fruits of Physalis(Solanaceae)have a unique structure,a lantern-like fruiting calyx known as inflated calyx syndrome(ICS)or the Chinese lantern,and are rich in steroid-related compounds.However,the genetic variations underlying the origin of these characteristic traits and diversity in Physalis remain largely unknown.Here,we present a high-quality chromosome-level reference genome assembly of Physalis floridana(~1.40Gb in size)with a contig N50 of~4.87Mb.Through evolutionary genomics and experimental approaches,we found that the loss of the SEP-like MADS-box gene MBP21 subclade is likely a key mutation that,together with the previously revealed mutation affecting floral MPF2 expression,might have contributed to the origination of ICS in Physaleae,suggesting that the origination of a morphological novelty may have resulted from an evolutionary scenario in which one mutation compensated for another deleterious mutation.Moreover,the significant expansion of squalene epoxidase genes is potentially associated with the natural variation of steroid-related compounds in Physalis fruits.The results reveal the importance of gene gains(duplication)and/or subsequent losses as genetic bases of the evolution of distinct fruit traits,and the data serve as a valuable resource for the evolutionary genetics and breeding of solanaceous crops. | Jiangjie Lu Meifang Luo Li Wang Kunpeng Li Yongyi Yu Weifei Yang Pichang Gong Huihui Gao Qiaoru Li Jing Zhao Lanfeng Wu Mingshu Zhang Xueyang Liu Xuemei Zhang Xian Zhang Jieyu Kang Tongyuan Yu Zhimin Li Yuannian Jiao Huizhong Wang Chaoying He | 2021 | Horticulture Research2021,8,1: | 0 |
