Rare earth element geochemistry of hydrothermal deposits from Southwest Indian Ridge显示文摘The REE compositions of hydrothermal deposits and basalt samples from the Southwest Indian Ridge (SWIR) were determined with ICP-MS.The results show that there are significant differences between different types of samples although all samples show relative LREE enrichment. The contents of REE in hydrothermal sulfides and alterated rocks samples are lower (from 7.036 × 10 6 to 23.660 × 10 6),while those in the white chimney deposits are relatively higher (ranging from 84.496 × 10 6 to 103.511 × 10 6).Both of them are lower than basalts.Chondrite-normalized REE distribution patterns show that sulfides and alterated rocks samples are characterized by significant positive Eu anomalies. On the contrary, white chimney deposits have obvious negative Eu anomalies, which may be caused by abundant calcite existing in the white chimney samples. Both the content and distribution pattern of REE in sulfides suggest that REE most possibly is originally derived from hydrothermal fluids, but influenced by the submarine reducing ore-forming environment, seawater convection, mineral compositions as well as the constraint of mineral crystallizations. | CAO Zhimin CAO Hong TAO Chunhui LI Jun YU Zenghui SHU Liping | 2012 | Acta Oceanologica Sinica2012,31,2: | 6
Exploration of presence/absence variation and corresponding polymorphic markers in soybean genome显示文摘This study was designed to reveal the genome‐wide distribution of presence/absence variation(PAV) and to establish a database of polymorphic PAV markers in soybean. The 33 soybean whole‐genome sequences were compared to each other with that of Williams 82 as a reference genome. A total of 33,127 PAVs were detected and 28,912 PAV markers with their primer sequences were designed as the database NJAUSoyPAV_1.0. The PAVs scattered on whole genome while only 518(1.8%) overlapped with simple sequence repeats(SSRs) in BARCSOYSSR_1.0database. In a random sample of 800 PAVs, 713(89.13%) showed polymorphism among the 12 differential genotypes. Using 126 PAVs and 108 SSRs to test a Chinese soybean germplasm collection composed of 828 Glycine soja Sieb. et Zucc. and Glycine max(L.) Merr. accessions, the per locus allele number and its variation appeared less in PAVs than in SSRs. The distinctness among alleles/bands of PCR(polymerase chain reaction) products showed better in PAVs than in SSRs, potential in accurate marker‐assisted allele selection. The association mapping results showed SSR t PAV was more powerful than any single marker systems. The NJAUSoyPAV_1.0 database has enriched the source of PCR markers, and may fit the materials with a range of per locus allele numbers, if jointly used with SSR markers. | Yufeng Wang Jiangjie Lu Shouyi Chen Liping Shu Reid G.Palmer Guangnan Xing Yan Li Shouping Yang Deyue Yu Tuanjie Zhao Junyi Gai | 2014 | Journal of Integrative Plant Biology2014,56,10: | 5
The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability. However,it remains largely unknown about how lncRNAs are regulated. Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein, designated as dBiFP,and show that the dBiFP protein is highly destabilized, compared with the commonly-used eGFP protein. We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore, our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders. | Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu | 2018 | Genes & Diseases2018,5,1: | 4
Plasmonic anapole states of active metamolecules显示文摘Anapole states, accompanied by strong suppression of light scattering, have attracted extensive attention in recent years due to their supreme performance in enhancing both linear and nonlinear optical effects. Although both low-and high-order anapole states are observed in the dielectric particles with high refractive index, so far few studies have touched on the topic of plasmonic anapole states. Here we demonstrate theoretically and numerically that the ideal plasmonic anapole states(strong suppression of electric dipole scattering) can be achieved in metallic metamolecules via increasing the coupling strength between Cartesian electric dipole and toroidal dipole moments of the system. The increasing coupling is based on compensation of ohmic losses in a plasmon system by introducing of a gain material, the influence of which is well described by the extended coupled oscillator model. Due to suppression of dipole radiation losses, the excitation of anapole states in plasmonic systems can result in enhancement of the near fields in subwavelength spatial regions outside of nanoparticles. That is especially important for developments of nonlinear nanophotonic and plasmonic devices and active functional metamaterials, which provide facilities for strong light energy concentration at the nanoscale. Development of the considered anapole effect with increase of metamolecule components is discussed. | GUI-MING PAN FANG-ZHOU SHU LE WANG LIPING SHI ANDREY BEVLYUKHIN | 2021 | Photonics Research2021,9,5: | 3
Impaired Parahippocampal Gyrus-Orbitofrontal Cortex Circuit Associated with Visuospatial Memory Deficit as a Potential Biomarker and Interventional Approach for Alzheimer Disease显示文摘The parahippocampal gyrus-orbitofrontal cortex(PHG-OFC)circuit in humans is homologous to the postrhinal cortex(POR)-ventral lateral orbitofrontal cortex(vlOFC)circuit in rodents. Both are associated with visuospatial malfunctions in Alzheimer’s disease(AD).However, the underlying mechanisms remain to be elucidated. In this study,we explored the relationship between an impaired POR-vlOFC circuit and visuospatial memory deficits through retrograde tracing and in vivo local field potential recordings in 5XFAD mice, and investigated alterations of the PHG-OFC circuit by multi-domain magnetic resonance imaging(MRI)in patients on the AD spectrum. We demonstrated that an impaired glutamatergic POR-vlOFC circuit resulted in deficient visuospatial memory in 5XFAD mice. Moreover,MRI measurements of the PHG-OFC circuit had an accuracy of 77.33%for the classification of amnestic mild cognitive impairment converters versus non-converters.Thus,the PHG-OFC circuit explains the neuroanatomical basis of visuospatial memory deficits in AD,thereby providing a potential predictor for AD progression and a promising interventional approach for AD. | Lin Zhu Zan Wang Zhanhong Du Xinyang Qi Hao Shu Duan Liu Fan Su Qing Ye Xuemei Liu Zheng Zhou Yongqiang Tang Ru Song Xiaobin Wang Li Lin Shijiang Li Ying Han Liping Wang Zhijun Zhang | 2020 | Neuroscience Bulletin2020,36,8: | 2
Preparation and Properties of Bio-Based Flame Retardant Polyvinyl Alcohol显示文摘Polyvinyl alcohol (PVA) has been widely used in the fields of medical, food and packaging due to its excellentbiocompatibility, good fiber-forming and film-forming properties. However, the high flammability of PVA hasgreatly limited its wider applications. The flame-retardant PVA was prepared by melt blending of a bio-basedflame retardant (prepared from lignin, phosphoric acid and carbamide) with thermoplastic PVA (TPVA). Thechemical structure, morphology, thermal properties, mechanical properties, fire property and fluidity of thisflame retardant PVA were investigated by Fourier transform infrared spectrometer(FTIR), field emission scanning electron microscope(SEM), thermogravimetric analyzer(TGA), impact tester, universal testing machine,horizontal-vertical burning tester, limiting oxygen indexer(LOI) and melt flow rate meter(MFR). The resultsshowed that the prepared flame retardant had good compatibility with the PVA substrate; The impact strength, melt flow rate, fire property and char residue of this PVA material increased with the content of bio-based flameretardant. When the content of flame retardant was of 20%, the five indices including impact strength, meltflow rate, UL-94 level, LOI and char residual were 11.3 KJ/m^(2), 21.2 g/10 min, V-0 UL-94 level, 33.1%, and19.2%, respectively. This research can promote the high-value utilization of lignin and the application ofPVA in the fields of fire protection. | You Shu Qionglin Luo Hongwei Lin Yuejun Ouyang Xiaomin Zhang Liping Sheng Shengpei Su | 2021 | Journal of Renewable Materials2021,9,5: | 1
Multiplexed imaging of tumor immune microenvironmental markers in locally advanced or metastatic non-small-cell lung cancer characterizes the features of response to PD-1 blockade plus chemotherapy显示文摘Background:Although programmed cell death 1(PD-1)blockade plus chemotherapy can significantly prolong the progression-free survival(PFS)and overall survival(OS)in first-line settings in patients with driver-negative advanced non-small-cell lung cancer(NSCLC),the predictive biomarkers remain undetermined. Here,we investigated the predictive value of tumor immune microenvironmental marker expression to characterize the response features to PD-1 blockade plus chemotherapy. Methods:Tumor tissue samples at baseline were prospectively collected from 144 locally advanced or metastatic NSCLC patients without driver gene alterations who received camrelizumab plus chemotherapy or chemotherapy alone.Tumor immune microenvironmental markers, including PD-1 ligand(PDL1),CD8,CD68,CD4 and forkhead box P3,were assessed using multiplex immunofluorescence(mIF)assays.Kaplan-Meier curveswere used to determine treatment outcome differences according to their expression status.Mutational profiles were compared between tumors with distinct expression levels of these markers and their combinations. Results:Responders had significantly higher CD8/PD-L1(P=0.015)or CD68/PD-L1 co-expression levels(P=0.021)than non-responders in the camrelizumab plus chemotherapy group, while no difference was observed in the chemotherapy group.Patients with high CD8/PD-L1 or CD68/PD-L1 co-expression level was associated with significantly longer PFS(P=0.002,P=0.024;respectively)and OS(P=0.006,P=0.026;respectively)than those with low co-expression in camrelizumab plus chemotherapy group. When comparing survival in the camrelizumab plus chemotherapy with chemotherapy by CD8/PD-L1 co-expression stratification, significantly better PFS(P=0.003)and OS(P=0.032)were observed in high co-expression subgroups. The predictive value of CD8/PD-L1 and CD68/PD-L1 co-expression remained statistically significant for PFS and OS when adjusting clinicopathological features.Although the prevalence of TP53 or KRAS mutations was similar between patients with and without CD8/PD-L1 or CD68/PD-L1 co-expression,the positive groups had a significantly higher proportion of TP53/KRAS co-mutations than the negative groups(both 13.0%vs.0.0%,P=0.023).Notably,enriched PI3K(P=0.012)and cell cycle pathway(P=0.021)were found in the CD8/PD-L1 co-expression group. Conclusion:Tumor immune microenvironmental marker expression, especially CD8/PD-L1 or CD68/PD-L1 co-expression,was associated with the efficacy of PD-1 blockade plus chemotherapy as first-line treatment in patients with advanced NSCLC. | Fengying Wu Tao Jiang Gongyan Chen Yunchao Huang Jianying Zhou Lizhu Lin Jifeng Feng Zhehai Wang Yongqian Shu Jianhua Shi Yi Hu Qiming Wang Ying Cheng Jianhua Chen Xiaoyan Lin Yongsheng Wang Jianan Huang Jiuwei Cui Lejie Cao Yunpeng Liu Yiping Zhang Yueyin Pan Jun Zhao LiPing Wang Jianhua Chang Qun Chen Xiubao Ren Wei Zhang Yun Fan Zhiyong He Jian Fang Kangsheng Gu Xiaorong Dong Tao Zhang Wei Shi Jianjun Zou Xuejuan Bai Shengxiang Ren Caicun Zhou | 2022 | Cancer Communications2022,42,12: | 1

Exploration of Zinc(II)Complexes as Potent Inhibitors Against Protein Tyrosine Phosphatase IB显示文摘Although protein tyrosine phosphatases(PTPs)do not contain any metals, their activities can be inhibited by some metal complexes. Here we investigated the inhibition of two zinc complexes with Schiff base ligands against PTPs activity to explore their effect on the cellular metabolism. It has been found that they are potent inhibitors against four recombinant PIPs5 including protein tyrosine phosphatase 1B(PTP1B),T cell protein tyrosine phosphatase(TCPTP),megakaryocyte protein tyrosine phosphatase 2(PTP-MEG2)S and Src-homology phosphatase 1(SHP-1),with exception of Src-homology phosphatase 2(SHP-2).Moreover,tlaey showed moderate selective inhibition against PTP1B with the IG50 values of 0.15 and 0.36μmol/L.Meanwhile,the complexes also inhibited cellular phosphatase activities efficiently.Comparing the inhibitory potency over PTPs mediated by the zinc ion,we found that zinc complexes might be easily developed into potent and selective inhibitors against certain PTP by rationally modifying the organic ligands moieties. | LI Xinhua YUAN Caixia LU Liping ZHU Miaoli XING Shu FU Xueqi | 2019 | Chemical Research in Chinese Universities2019,35,2: | 1
Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 11 | Effect of Dihydrotestosterone on Costimulatory Molecules in a Mouse Model of Graves’ Disease显示文摘Objective Graves’disease is the most common autoimmune thyroid disease and its prevalence and clinical manifestations are disparate between females and males.Costimulatory molecules play an essential role in regulating autoimmune responses. The objective of this study was to determine if expression of inhibitory molecules was correlated with treatment by dihydrotestosterone(DHT)in an in vivo BALB/c mouse model of experimental autoimmune Graves’disease. Methods Female BALB/c mice were immunized three times with thyroid stimulating hormone receptor A-subunit encoded by adenovirus to establish a Graves’disease model.Three different doses of DHT or a matching placebo were administered by implantation of slow-release pellets a week before the first immunization.Four weeks after the third immunization, the mice were euthanatized, and then the spleen and thymus were removed.Total thyroxine and free thyroxine levels in serum of mice were detected using a radioimmunoassay kit.Real-time polymerase chain reaction was performed to estimate the expression of costimulatory molecules in lymphocytes from the spleen and thymus.Flow cytometry was used to analyze the percentage of CD4^+T cells in splenic lymphocytes.Quantitative data were compared with unpaired t-tests.Correlation between two variables was analyzed using Analysis of Variance. Results Treatment with DHT can dramatically reduce total thyroxine and free thyroxine levels.Higher expression of programmed death-1 was found in the spleen of Graves’disease mice receiving 5 mg of DHT treatment(0.635±0.296 vs.0.327±0.212;t=2.714,P=0.014),similarly,T-cell immunoglobulin domain and mucin domain 3(TIM-3)in both the spleen(1.004±0.338 vs.0.646±0.314;t=2.205,P=0.022)and the thymus(0.263±0.127 vs.0.120±0.076;t=3.221,P=0.004)also increased after 5 mg of DHT treatment compared with the parallel placebo model mice. Moreover, the percentage of CD4^+T cells declined in the splenic lymphocytes of Graves’disease mice treated with 5 mg of DHT(19.90%±3.985%vs.24.05%±2.587%;t=2.804,P=0.012).A significant negative association was observed between expression of TIM-3 in the spleen and serum levels of total thyroxine(r=-0.7106,P=0.014)as well as free thyroxine(r=-0.6542,P=0.029).Conclusion This study demonstrates that DHT can ameliorate experimental autoimmune Graves’disease, which may occur by up-regulating expression of programmed death-1 and TIM-3 and inhibiting development of CD4^+T cells. | Lianye Liu Bingyin Shi Fengyi Zhao Peng Hou Shu Liu Xiaomei Liu Liping Wu | 2020 | Chinese Medical Sciences Journal2020,35,3: | 0
Histology, physiology, and transcriptomic and metabolomic profiling reveal the developmental dynamics of annual shoots in tree peonies (Paeonia suffruticosa Andr.)显示文摘The development of tree peony annual shoots is characterized by“withering”,which is related to whether there are bud points in the leaf axillaries of annual shoots. However, the mechanism of“withering”in tree peony is still unclear. In this study,Paeonia ostii‘Fengdan’and P.suffruticosa‘Luoyanghong’were used to investigate dynamic changes of annual shoots through anatomy, physiology, transcriptome, and metabolome. The results demonstrated that the developmental dynamics of annual shoots of the two cultivars were comparable. The withering degree of P.suffruticosa‘Luoyanghong’was higher than that of P.ostii‘Fengdan’,and their upper internodes of annual flowering shoots had a lower degree of lignin deposition,cellulose,C/N ratio, showing no obvious sclerenchyma, than the bottom ones and the whole internodes of vegetative shoot, which resulted in the“withering”of upper internodes.A total of 36 phytohormone metabolites were detected,of which 33 and 31 were detected in P.ostii‘Fengdan’and P.suffruticosa‘Luoyanghong’,respectively.In addition,302 and 240 differentially expressed genes related to lignin biosynthesis, carbon and nitrogen metabolism, plant hormone signal transduction, and zeatin biosynthesis were screened from the two cultivars.Furtherly,36 structural genes and 40 transcription factors associated with the development of annual shoots were highly co-expressed,and eight hub genes involved in this developmental process were identified.Consequently, this study explained the developmental dynamic on the varied annual shoots through multi-omics,providing a theoretical foundation for germplasm innovation and the mechanized harvesting of tree peony annual shoots. | Ningning Tong Qingyan Shu Baichen Wang Liping Peng Zheng’an Liu | 2023 | Horticulture Research2023,10,9: | 0
Combined genome-wide association studies and expression quantitative trait locus analysis uncovers a genetic regulatory network of floral organ number in a tree peony (Paeonia suffruticosa Andrews) breeding population显示文摘Great progress has been made in our understanding of floral organ identity determination and its regulatory network in many species;however, the quantitative genetic basis of floral organ number variation is far less well understood for species-specific traits from the perspective of population variation. Here, using a tree peony(Paeonia suffruticosa Andrews,Paeoniaceae)cultivar population as a model, the phenotypic polymorphism and genetic variation based on genome-wide association studies(GWAS)and expression quantitative trait locus(eQTL)analysis were analyzed.Based on 24 phenotypic traits of 271 representative cultivars, the transcript profiles of 119 cultivars were obtained, which indicated abundant genetic variation in tree peony. In total,86 GWAS-related cis-eQTLs and 3188 trans-eQTL gene pairs were found to be associated with the numbers of petals, stamens, and carpels. In addition,19 floral organ number-related hub genes with 121 cis-eQTLs were obtained by weighted gene co-expression network analysis, among which five hub genes belonging to the ABCE genes of the MADS-box family and their spatial–temporal co-expression and regulatory network were constructed. These results not only help our understanding of the genetic basis of floral organ number variation during domestication, but also pave the way to studying the quantitative genetics and evolution of flower organ number and their regulatory network within populations. | Liping Peng Yang Li Wanqing Tan Shangwei Wu Qing Hao Ningning Tong Zhanying Wang Zheng’an Liu Qingyan Shu | 2023 | Horticulture Research2023,10,7: | 0

Etching mechanism of barium strontium titanate (BST) thin films in CHF_3/Ar plasma显示文摘Reactive ion etching was used to etch barium strontium titanate thin films in a CHF3/Ar plasma.BST surfaces before and after etching were analyzed by X-ray photoelectron spectroscopy to investigate the reaction ion etching mechanism, and chemical reactions had occurred between the F plasma and the Ba,Sr and Ti metal species.Fluorides of these metals were formed and remained on the surface during the etching process.Ti was almost completely removed because the TiF4 by-product is volatile.Minor quantities of Ti?F could still be detected by narrow scan X-ray photoelectron spectra, and Ti?F was thought to be present in the form of a metal-oxy-fluoride.These species were investigated from O1s spectra, and a fluoride-rich surface was formed during etching.BaF2 and SrF2 residues were difficult to remove because of their high boiling point. The etching rate was limited to 12.86 nm/min.C?F polymers were not found on the surface, indicating that the removal of BaF2 and SrF2 was important for further etching.A 1-min Ar/15 plasma physical sputtering was carried out for every 4 min of surface etching, which effectively removed remaining surface residue.Sequential chemical reaction and sputtered etching is an effective etching method for barium strontium titanate films. | DAI LiPing WANG ShuYa SHU Ping ZHONG ZhiQin WANG Gang ZHANG GuoJun | 2011 | Chinese Science Bulletin2011,56,21: | 0
Expert recommendations on collection and annotation of otoscopy images for intelligent medicine显示文摘Middle and outer ear diseases are common otological diseases worldwide.Otoscopy and otoendoscopy exami-nations are essential first steps in the evaluation of patients with otological diseases.Misdiagnosis often occurs when the doctor lacks experience in interpreting the results of otoscopy or otoendoscopy, leading to delays in treatment or complications.Using deep learning to process otoscopy images and developing otoscopic artificial-intelligence-based decision-making systems will become a significant trend in the future. However, the uneven quality of otoscopy images is among the major obstacles to development of such artificial intelligence systems, and no standardized process for data acquisition, and annotation of otoscopy images in intelligent medicine has yet been fully established. The standards for data storage and data management are unified with those of other specialties and are introduced in detail here. This expert recommendation criterion improved and standardized the collection and annotation procedures for otoscopy images and fills the current gap in otologic intelligent medicine;it would thus lay a solid foundation for the standardized collection, storage, and annotation of oto-scopy images and the application of training algorithms, and promote the development of automatic diagnosis and treatment for otological diseases. The full text introduced image collection(including patient preparation, equipment standards, and image storage),image annotation standards, and quality control. | Yuexin Cai Junbo Zeng Liping Lan Suijun Chen Yongkang Ou Linqi Zeng Qintai Yang Peng Li Yubin Chen Qi Li Hongzheng Zhang Fan Shu Guoping Chen Wenben Chen Yahan Yang Ruiyang Li Anqi Yan Haotian Lin Yiqing Zheng | 2022 | Intelligent Medicine2022,2,4: | 0
