1The combination of chidamide with the CHOEP regimen in previously untreated patients with peripheral T-cell lymphoma: a prospective, multicenter, single arm, phase 1b/2 study显示文摘Objective:To assess the efficacy and safety of the novel histone deacetylase inhibitor,chidamide,in combination with cyclophosphamide,doxorubicin,vincristine,etoposide,and prednisone(Chi-CHOEP)for untreated peripheral T-cell lymphoma(PTCL).Methods:A prospective,multicenter,single arm,phase 1 b/2 study was conducted.A total of 128 patients with untreated PTCL(18–70 years of age)were enrolled between March 2016 and November 2019,and treated with up to 6 cycles with the Chi-CHOEP regimen.In the phase 1 b study,3 dose levels of chidamide were evaluated and the primary endpoint was determination of the maximumtolerated dose and recommended phase 2 dose(RP2 D).The primary endpoint of the phase 2 study was 2-year progression-free survival(PFS).Results:Fifteen patients were enrolled in the phase 1 b study and the RP2 D for chidamide was determined to be 20 mg,twice a week.A total of 113 patients were treated at the RP2 D in the phase 2 study,and the overall response rate was 60.2%,with a complete response rate of 40.7%.At a median follow-up of 36 months,the median PFS was 10.7 months,with 1-,2-,and 3-year PFS rates of 49.9%,38.0%,and 32.8%,respectively.The Chi-CHOEP regimen was well-tolerated,with grade 3/4 neutropenia occurring in approximately two-thirds of the patients.No unexpected adverse events(AEs)were reported and the observed AEs were manageable.Conclusions:This large cohort phase 1 b/2 study showed that Chi-CHOEP was well-tolerated with modest efficacy in previously untreated PTCL patients.Wei Zhang Liping Su Lihong Liu Yuhuan Gao Quanshun Wang Hang Su Yuhuan Song Huilai Zhang Jing Shen Hongmei Jing Shuye Wang Xinan Cen Hui Liu Aichun Liu Zengjun Li Jianmin Luo Jianxia He Jingwen Wang O.A.O’Connor Daobin Zhou 2021Cancer Biology & Medicine2021,18,3:4

3Genomes of 12 fig wasps provide insights into the adaptation of pollinators to fig syconia显示文摘Figs and fig pollinators are one of the few classic textbook examples of obligate pollination mutualism.The specific dependence of fig pollinators on the relatively safe living environment with sufficient food sources in the enclosed fig syconia implies that they are vulnerable to habitat changes.However,there is still no extensive genomic evidence to reveal the evolutionary footprint of this long-term mutually beneficial symbiosis in fig pollinators.In fig syconia,there are also non-pollinator species.The non-pollinator species differ in their evolutionary and life histories from pollinators.We conducted comparative analyses on 11 newly sequenced fig wasp genomes and one previously published genome.The pollinators colonized the figs approximately 66.9 million years ago,consistent with the origin of host figs.Compared with nonpollinators,many more genes in pollinators were subject to relaxed selection.Seven genes were absent in pollinators in response to environmental stress and immune activation.Pollinators had more streamlined gene repertoires in the innate immune system,chemosensory toolbox,and detoxification system.Our results provide genomic evidence for the differentiation between pollinators and nonpollinators.The data suggest that owing to the long-term adaptation to the fig,some genes related to functions no longer required are absent in pollinators.Jinhua Xiao Xianqin Wei Yi Zhou Zhaozhe Xin Yunheng Miao Hongxia Hou Jiaxing Li Dan Zhao Jing Liu Rui Chen Liming Niu Guangchang Ma Wenquan Zhen Shunmin He Jianxia Wang Xunfan Wei Weihao Dou Zhuoxiao Sui Haikuan Zhang Shilai Xing Miao Shi Dawei Huang 2021Journal of Genetics and Genomics2021,48,3:1

8Expression and function of long non-coding RNA DLX6-AS1 in endometrial cancer显示文摘Background:LncRNA DLX6-AS1 has been uncovered to exert effects on various cancers.Nevertheless,the impacts of DLX6-AS1 on endometrial cancer(EC)development remained obscure.The study explored the influence of DLX6-AS1 on EC progression via the microRNA(miR)-374a-3p/zinc-finger protein(ZFX)axis.Methods:EC cell lines were collected and DLX6-AS1,miR-374a-3p,and ZFX levels in EC cell lines were detected.The EC cells were transfected with DLX6-AS1,miR-374a-3p,and ZFX constructs to examine the biological functions of EC cells.The xenograft model was established for detecting tumor growth.Rescue experiments were conducted to verify the interaction of DLX6-AS1,miR-374a-3p,and ZFX in EC cells.Results:DLX6-AS1 and ZFX levels were elevated,while miR-374a-3p exhibited a reduced level in EC cells.Silencing DLX6-AS1 and elevated miR-374a-3p expressions repressed the biological activities of EC cells.Reduced DLX6-AS1 repressed tumor development.MiR-374a-3p silencing reversed the impacts of DLX6-AS1 silencing,while ZFX overexpression abrogated the impacts of miR-374a-3p elevation on EC cell growth.Mechanically,DLX6-AS1 was found to bind to miR-374a-3p,and miR-374a-3p targeted ZFX.Conclusion:DLX6-AS1 depletion restricts the malignant phenotype of EC cells.The study might provide novel therapeutic biomarkers for EC treatment.WEI SHI JIANXIA LIN RONG JIN XIANJING XIE YAN LIANG 2023BIOCELL2023,47,4:0
9Determination of Chlorpyrifos in Human Blood by Gas Chromatography-Mass Spectrometry显示文摘Gas chromatography-mass spectrometry method was developed for the qualitative and quantitative analyses of chlorpyrifos in human blood samples.The chlorpyrifos and parathion(internal standard)in human blood were extracted with a mixed solvent of hexane and acetonitrile.Chlorpyrifos was well separated from the internal standard.The linear range of chlorpyrifos was 0.01-2 μg/ml in blood.The limit of detection and limit of quantification were estimated at 0.002 and 0.01μg/ml,respectively.The inter-and intra-day precisions,accuracy,and recovery were assessed to verify this method.The results showed that the developed method is rapid,sensitive,and reliable.It is suitable for the determination of chlorpyrifos in forensic toxicological analysis and clinical diagnosis.Xinhua Dai Fei Fan Yi Ye Fan Chen Zhigui Wu Xiang Lu Qingtao Wei Jianxia Chen Youyi Yan Linchuan Liao 2017Journal of Forensic Science and Medicine2017,3,1:0

11An optimized finite difference method based on a polar coordinate system for regional-scale irregular topography显示文摘The finite difference method(FDM)is an important numerical approach for simulating the propagation of seismic waves,and some FDMs can be used to study the impact of the Earth’s curvature and topography over large distances.To efficiently model the effects of the Earth’s irregular topography on the propagation of seismic waves,here we optimize a previously proposed grid mesh method and develop a novel two-dimensional boundary-conforming FDM based on a curvilinear polar coordinate system.This method efficiently simulates the propagation of seismic waves in an arc-shaped model with large variations in surface topography.Our method was benchmarked against other reported methods using several global-scale models.The consistency of the results confirms the validity of our proposed optimization strategy.Furthermore,our findings indicate that the proposed optimization strategy improves computational efficiency.Jianxia Xu Wei Zhang Xiaofei Chen 2021Earthquake Science2021,34,4:0
